This study aims to identify differentially expressed microRNAs (miRNAs) in gastric cancer by comparing gastric cancerous tissues with normal tissues, explore the potential roles.The miRNA expression microarray was employed on gastric cancer tissues, and apparently normal para-cancerous tissues from 3 patients undergoing radical surgery were matched. Quantitative RT-PCR was performed on the other 7 patients to validate the findings of the microarray. Furthermore, Gene Ontology (GO) analysis and enrichment analysis of KEGG Pathway were performed for 5 dysregulated candidate miRNAs, including 3 upregulated (miR-31-3p, miR-6736-3p, and miR-147b) and 2 downregulated (miR-3065-5p and miR-3921) miRNAs, in order to determine the role of miRNAs in tumorigenesis and development.Among these miRNAs, 17 miRNAs were found to be upregulated, and 19 miRNAs were found to be downregulated. The dysregulated expression of 5 candidate miRNAs, including miR-31-3p, miR-147b, miR-6736-3p, miR-3065-5p, and miR-3921, were verified by quantitative RT-PCR in the validation set. Among these miRNAs, miR-31-3p, miR-6736-3p, miR-3065-5p, and miR-3921 had 551 target gene intersections. The GO and KEGG Pathway analyses Revealed that miR-31-3p, miR-6736-3p, miR-3065-5p, and miR-3921 may participate in multiple pathophysiological processes, such as foreign substance metabolism and chemical carcinogenesis.The profile of differentially expressed miRNAs was successfully screened, and 4 miRNAs (i.e., miR-31-3p, miR-6736-3p, miR-3065-5p, and miR-3921) appeared to be involved in gastric carcinogenesis. Glutathione S-transferase Pi (GSTP1) is an isozyme encoded by the GST pi gene that plays an important regulatory role in detoxification, anti-oxidative damage, and the occurrence of various diseases. The aim of the present study was to review the association between the expression of GSTP1 and the development and treatment of various cancers, and discuss GSTP1 methylation in several malignant tumors, such as prostate, breast and lung cancer, as well as hepatocellular carcinoma; to review the association between polymorphism of the GSTP1 gene and various diseases; and to review the effects of GSTP1 on electrophilic oxidative stress, cell signal transduction, and the regulation of carcinogenic factors. Collectively, GSTP1 plays a major role in the development of various diseases. © 2020 Spandidos Publications. All rights reserved.

Diallyl disulfide (DADS), the main active component of the cancer fighting allyl sulfides found in garlic, has shown potential as a therapeutic agent in various cancers. Previous studies showed DADS induction of HL-60 cell differentiation involves down-regulation of calreticulin (CRT). Here, we investigated the mechanism of DADS-induced differentiation of human leukaemia cells and the potential involvement of CRT and CCAAT enhancer binding protein-α (C/EBPα). We explored the expression of CRT and C/EBPα in clinical samples (20 healthy people and 19 acute myeloid leukaemia patients) and found that CRT and C/EBPα expressions were inversely correlated. DADS induction of differentiation of HL-60 cells resulted in down-regulated CRT expression and elevated C/EBPα expression. In severe combined immunodeficiency mice injected with HL-60 cells, DADS inhibited the growth of tumour tissue and decreased CRT levels and increased C/EBPα invivo. We also found that DADS-mediated down-regulation of CRT and up-regulation of C/EBPα involved enhancement of reactive oxidative species. RNA immunoprecipitation revealed that CRT bound C/EBPα mRNA, indicating its regulation of C/EBPα mRNA degradation by binding the UG-rich element in the 3′ untranslated region of C/EBPα. In conclusion, the present study demonstrates the C/EBPα expression was correlated with CRT expression invitro and invivo and the molecular mechanism of DADS-induced leukaemic cell differentiation. © 2018 The Authors. 三萜类化合物含30个左右碳原子,基本骨架为多个异戊二烯,来源广泛,药用价值高,已作为一种重要化学成分运用于药物之中。通过对三萜类化合物中齐墩果酸、白桦脂酸、人参皂苷Rh2、Rg3的抗肿瘤机制展开论述,探讨三萜类化合物的抗肿瘤现状,总结三萜类化合物的抗肿瘤机制及不同三萜类化合物抗肿瘤作用的选择性和特异性,对抗肿瘤新药的研制和开发具有重要意义。

摘要:
目的探讨糖尿病足溃疡(diabetic foot ulcer,DFU)患者伤口和血清中半乳糖凝集素3(galectin-3)的表达,以期阐明galectin-3和DFU的关系。方法选取2015年1月-2016年3月Wanger 1、2级的糖尿病足患者50例,另选取同期血糖正常的外伤患者20例为对照,分别取其足部伤口边缘组织及血清。对所有入选对象均行ELISA检测血清galectin-3浓度,Western-blot法检测伤口边缘组织galectin-3的表达水平。结果 ELISA结果显示DFU组血清galectin-3水平高于外伤对照组[(5.66±2.76)vs(4.25±1.94)ng/ml](P=0.04),Western-blot结果显示galectin-3在DFU组伤口边缘组织表达增强。结论 galectin-3在DFU患者中高表达,galectin-3的高表达可能参与了糖尿病足的进展。

Background: Human Interleukin 37 (IL37), a unique anti-inflammatory cytokine of IL1 family member, plays critical roles in innate and adaptive immunity and inflammation. Objective: Preparation of high purity and tag-removed recombinant IL37 protein (rIL37) is critical for its clinical application. Method: In this study, we constructed an N-terminal cleavable GST-fused IL37 expression vector for recombinant expression. Results: Subsequent to transformation and optimization of the induction temperature, the soluble expression level of rIL37 was 306.5 mg/L of culture medium at 18 °C induction in Escherichia coli. Meanwhile, rIL37 was digested on beads by GST-HRV3C protease during GST affinity chromatography. After further purification, the purity of rIL37 was higher than 99 %. Finally, the anti-inflammatory activity of tag-removed protein was verified by the results showing that rIL37 suppressed IL1β production in PBMCs. Conclusion: This work presents a protocol to produce high purity and tag-removed rIL37 with anti-inflammatory activity, which provides the firm basis for advancing clinical application in human IL37-related inflammatory diseases. © 2018 Bentham Science Publishers.

Ginnalin A (also known as acertannin) is one of the most important phenolic compounds of several beverage Acer plants. In this study, it is reported for the first time that ginnalin A is an activator of the Nrf2 signaling pathway in human colon cancer cells. Ginnalin A, isolated from the leaves of Acer tataricum subsp. ginnala, exhibited promising preventive activity against colon cancer cells (HCT116, SW480 and SW620) with IC50 values of 24.8 μM, 22.0 μM and 39.7 μM, respectively. In addition, it significantly reduced the colony formation of these cells. Flow cytometry analysis indicated that ginnalin A suppressed cancer proliferation via the induction of cell cycle arrest at the S-phase. Real time PCR analysis demonstrated that ginnalin A can upregulate the mRNA expression levels of Nrf2-related antioxidant genes Nrf2, HO-1 and NQO1. Western blotting analysis revealed that ginnalin A promoted the Nrf2 nuclear translocation and upregulated the proteins Nrf2, HO-1 and NQO1. Moreover, the upregulation of p62 and the inhibition of Keap1 were also found by Western blotting analysis. Therefore, the activation of the Nrf2 signaling pathway was probably induced through the upregulation of p62 and the inhibition of Keap1. © 2018 The Royal Society of Chemistry.

目的:对冠心病血瘀证蛋白质组学发现的差异蛋白质进行生物信息学分析.方法:利用PANTHER、DAVID和STRING在线数据库对CNKI和PUBMED发表的冠心病血瘀证差异蛋白质进行GO功能富集分析和蛋白质相互作用网络构建.结果:冠心病血瘀证差异蛋白质细胞组分主要定位于细胞外区域、细胞、细胞器等,主要参与细胞过程、定位、多细胞生物过程等,主要涉及到催化活性、结合功能、结构分子激活等分子功能,主要参与血小板活化、补体及凝血级联反应、致心律失常性右室心肌病等信号通路,蛋白质相互作用涉及44个节点和131条边.结论:多个蛋白质、多个信号通路、多个蛋白质相互作用方式为理解冠心病血瘀证多阶段病理过程提供了帮助.

Colon cancer is one of the most common types of gastrointestinal cancers and the fourth cause of cancer death worldwide. To discover novel diagnostic biomarkers for colon cancer and investigate potential mechanisms of oncogenesis, quantitative proteomic approach using iTRAQ-tagging and 2D-LC-MS/MS was performed to characterize proteins alterations in colon cancer and non-neoplastic colonic mucosa (NNCM) using laser capture microdissection-harvested from the two types of tissues, respectively. As a result, 188 DEPs were identified, and the differential expression of two DEPs (DCN and HSPD1) was further verified by Western blotting and immunohistochemistry. KEGG pathway analysis disclosed that the DEPs were related to signaling pathways associated with cancer; furthermore, DCN and HSPD1 are in the relative central hub position among protein-protein interaction subnetwork of the DEPs. The results not only shed light on the mechanism by the DEPs contributed to colonic carcinogenesis, but also showed that DCN and HSPD1 are novel potential biomarkers for the diagnosis of colon cancer. © Ivyspring International Publisher.

为分析结肠上皮癌变进程中的差异表达蛋白质,筛选结肠癌诊断候选标志物,先采用激光捕获显微切割技术(LCM)纯化人正常结肠组织和结肠癌组织,再采用同位素标记对蛋白质进行相对和绝对定量(iTRAQ),然后结合强阳离子柱分离和反相液质联用(2D-LC-MS/MS)鉴定差异表达蛋白质。试验共鉴定了1 042个非冗余蛋白,筛选出与人结肠癌变相关的差异蛋白162个,其中在结肠癌中表达上调的蛋白质68个,表达下调的蛋白质94个,功能分析表明,这些差异蛋白质涉及内质网蛋白质加工、糖酵解/糖原异生、黏着斑细胞连接、调节微丝组装和细胞外基质受体相互作用等分子功能。进一步采用Western blotting及免疫组织化学技术(IHC)验证了其中差异蛋白S100A9的表达,证实了定量蛋白质组学结果的可靠性。研究表明:差异表达蛋白质与结肠癌变相关,并有可能成为结肠癌诊断的候选分子标志物,为进一步深入研究这些蛋白质在结肠癌发生和发展进程中的分子机制,为结肠癌的发病机制研究提供新线索。

为探讨基于团队的学习模式TBL教学法在医学生物学实验课程中的具体教学模式、评价体系及存在的问题与对策,选择南华大学船山学院2014级四年制预防医学专业学生为研究对象,采用分组方法,将班级分为若干个团队,然后以团队为基础共同学习和完成病案分析的讨论式教学方法,为高等学校医学生物学实验课程教学研究和其他医学课程教学研究提供重要参考依据。

Diallyl disulfide (DADS) has shown potential as a therapeutic agent in various cancers. Previously, calreticulin (CRT) was found to be downregulated in differentiated HL‑60 cells treated with DADS. The present study investigated the role of CRT proteins in DADS‑induced proliferation, invasion and differentiation in HL‑60 cells. The present study demonstrated that DADS treatment significantly changed the morphology of HL‑60 cells and caused the significant time‑dependent downregulation of CRT. Small interfering RNA (siRNA)‑mediated knockdown of CRT expression significantly inhibited proliferation, decreased invasion ability, increased the expression of cluster of differentiation (CD)11b and reduced the expression of CD33 in DADS‑treated HL‑60 cells. DADS also significantly affected cell proliferation, invasion and differentiation in CRT‑overexpressed HL‑60 cells. Nitroblue tetrazolium (NBT) reduction assays showed decreased NBT reduction activity in the CRT overexpression group and increased NBT reduction in the CRT siRNA group. Following treatment with DADS, the NBT reduction abilities in all groups were increased. In conclusion, the present study clearly demonstrates the downregulation of CRT during DADS‑induced differentiation in HL‑60 cells and indicates that CRT is involved in cell proliferation, invasion and differentiation in DADS‑treated HL‑60 cells. © 2016, Spandidos Publications. AIM: To discover novel biomarkers for early diagnosis, prognosis or treatment of human colorectal cancer. METHODS: iTRAQ 2D LC-MS/MS analysis was used to identify differentially expressed proteins (DEPs) in the human colonic epithelial carcinogenic process using laser capture microdissection-purified colonic epithelial cells from normal colon, adenoma, carcinoma in situ and invasive carcinoma tissues. RESULTS: A total of 326 DEPs were identified, and four DEPs (DMBT1, S100A9, Galectin-10, and S100A8) with progressive alteration in the carcinogenic process were further validated by immunohistochemistry. The DEPs were involved in multiple biological processes including cell cycle, cell adhesion, translation, mRNA processing, and protein synthesis. Some of the DEPs involved in cellular process such as "translation" and "mRNA splicing" were progressively up-regulated, while some DEPs involved in other processes such as "metabolism" and "cell response to stress" was progressively downregulated. Other proteins with up- or down-regulation at certain stages of carcinogenesis may play various roles at different stages of the colorectal carcinogenic process. CONCLUSION: These findings give insights into our understanding of the mechanisms of colorectal carcinogenesis and provide clues for further investigation of carcinogenesis and identification of biomarkers. © 2016 Baishideng Publishing Group Inc.

LIGHT recruits and activates naive T cells in the islets at the onset of diabetes. IFN-γ secreted by activated T lymphocytes is involved in beta cell apoptosis. However, whether LIGHT sensitizes IFNγ-induced beta cells destruction remains unclear. In this study, we used the murine beta cell line MIN6 and primary islet cells as models for investigating the underlying cellular mechanisms involved in LIGHT/IFNγ – induced pancreatic beta cell destruction. LIGHT and IFN-γ synergistically reduced MIN6 and primary islet cells viability; decreased cell viability was due to apoptosis, as demonstrated by a significant increase in Annexin V+ cell percentage, detected by flow cytometry. In addition to marked increases in cytochrome c release and NF-κB activation, the combination of LIGHT and IFN-γ caused an obvious decrease in expression of the anti-apoptotic proteins Bcl-2 and Bcl-xL, but an increase in expression of the pro-apoptotic proteins Bak and Bax in MIN6 cells. Accordingly, LIGHT deficiency led to a decrease in NF-κB activation and Bak expression, and peri-insulitis in non-obese diabetes mice. Inhibition of NF-κB activation with the specific NF-κB inhibitor, PDTC (pyrrolidine dithiocarbamate), reversed Bcl-xL down-regulation and Bax up-regulation, and led to a significant increase in LIGHT- and IFN-γ-treated cell viability. Moreover, cleaved caspase-9, -3, and PARP (poly (ADP-ribose) polymerase) were observed after LIGHT and IFN-γ treatment. Pretreatment with caspase inhibitors remarkably attenuated LIGHT- and IFNγ-induced cell apoptosis. Taken together, our results indicate that LIGHT signalling pathway combined with IFN-γ induces beta cells apoptosis via an NF-κB/Bcl2-dependent mitochondrial pathway. © 2016 The Authors. 为提高医学留学生教学质量,结合留学生教学现状,将TBL教学法应用于医学留学生细胞生物学全英文教学。从建立学习小组,教学实施过程、教学后评价等方面来实践TBL教学模式。并从建立教师队伍,做好课前准备、课堂氛围等方面总结TBL教学经验。同时对TBL教学法在留学生细胞生物学全英文教学中的优势以及存在的问题进行了展望。总之,TBL教学方法在留学生全英文教学中值得推广。

摘要:
目的 探讨TBL教学法在独立学院“细胞生物学”参与式教学中的应用及其教学效果.方法 将南华大学船山学院2014级药学专业41名学生按照随机数字表法分为对照组(20名)和研究组(21名).研究组采用TBL教学法,对照组采用传统教学法.观察并比较两组学生在自主学习、合作团结、沟通能力以及考核成绩等方面的差别.结果 理论考试成绩,研究组的选择题、简答题、综合分析题和实验设计题得分分别为(8.22±0.73)、(7.94±1.35)、(15.68±2.03)、(15.21±1.97)分,对照组分别为(7.17±0.87)、(6.07±1.22)、(12.24±2.48)、(11.60±1.87)分,两组比较,除选择题外,其余各题型差异均有统计学意义(P<0.05).研究组的理论成绩、实验成绩和总成绩分别为(53.78±2.19)、(34.24±2.97)、(89.85±1.29)分,对照组分别为(43.26±2.35)、(26.67±3.31)、(69.23±2.14)分,两组比较,差异均有统计学意义(P<0.05).问卷调查结果显示,研究组的学生普遍认为TBL教学法有利于多学科知识的综合应用,有利于学生学习积极性和主动性的调动,有利于学生人际交往和沟通能力的提高,有利于学习成绩的提高,有利于学生个人责任心的增强.结论 TBL教学法在参与式教学中能够提高学生的自主学习能力和学习成绩,可以在独立学院教学中推广.

科研能力的培养是现代本科生教育的重要环节。该文以美国佛罗里达大学为例,概括了美国本科生物学专业课程设置情况,着重分析了课程设置中注重学生科研气质的培养特点;并通过比较国内综合性大学生物学专业课程设置,分析了由UF本科生物学专业注重科研能力培养课程设置给予国内综合性大学生物学课程设置的启示,从而为综合性大学本科生物学专业的教学改革提供一些合理化的建议,并有助于综合性大学本科生物学专业的发展。

摘要:
目的:建立小鼠唾液腺干细胞体外分离培养方法。方法:从78周龄的C57BL/6J小鼠唾液腺分离唾液腺干细胞,采用悬浮唾液球法培养唾液腺干细胞,应用倒置显微镜动态观察细胞形态;细胞计数观察细胞生长状况;免疫荧光检测LN(层连蛋白)的表达。结果:倒置显微镜下,接种2 d后细胞(salispheres)小聚集体明显增加;接种4 d后唾液腺球直径逐渐增大,形成直径约100μm的神经球,球周围无明显刺突,存在明显细胞聚集。接种7 d后唾液腺球周围存在较多微刺,10 d后球继续增长,形状明显不同于4 d,表明处于活跃的增殖状态,唾液腺干细胞已经开始分化。生长曲线结果显示随着培养天数的增加,唾液腺球的数目呈明显递增趋势,同时,唾液腺球的直径也明显增大。免疫荧光结果显示,培养2 d后,唾液干细胞球开始表达LN,培养第4 d后,随着唾液干细胞球的增加,层连蛋白表达更明显。结论:应用该方法获得了唾液腺干细胞,具有成体干细胞的特征,具有自我更新和多向分化的潜能。

摘要:
目的 对无转移肺腺癌和有转移肺腺癌表达差异蛋白质进行生物信息学分析.方法 利用Cytoscape软件对前期发现无转移肺腺癌和有转移肺腺癌表达差异的20个蛋白质进行基因本体论(GO)功能富集分析和蛋白质相互作用网络构建.结果 20种肺腺癌转移差异表达蛋白质的主要生物学过程为葡萄糖分解和酒精分解等过程,主要分子功能为磷脂酶A2抑制剂活性和钙依赖的磷脂结合,蛋白质相互作用网络为包含245种不同蛋白质和230种不同蛋白质相互作用关系.结论 葡萄糖分解过程、酒精分解过程、磷脂酶A2抑制剂活性和钙依赖的磷脂结合及ACTB、ANXA1、ANXA2、ANXA3、VCP、NPM1、KRT1、SUMO4等关键蛋白质在人肺腺癌转移过程中发挥重要作用,为进一步研究提供线索.
