摘要:
Inflammatory bowel disease (IBD) correlates with oxidative stress, inflammation, and alteration in several signal pathways, including nuclear transcription factor-kappaB (NF-κB). Pyrrolidine dithiocarbamate (PDTC), an inhibitor of NF-κB, has been widely demonstrated to exhibit an antioxidant and anti-inflammatory function. This study aimed to test the hypothesis that NF-κB inhibitor PDTC confers a beneficial role in a colitis model induced by dextran sodium sulfate (DSS) in mouse. The results showed that DSS decreased daily weight gain, induced colonic inflammation, suppressed the expression of antioxidant enzymes and tight junctions, and activated NF-κB and nuclear factor erythroid 2-related factor 2/Kelch-like ECH-associated protein 1 (Nrf2/Keap1) signaling pathways. PDTC significantly upregulated (P < 0.05) Gpx1, Gpx4, occludin, and ZO-1 expressions in the DSS-induced colitis model. Meanwhile, PDTC reversed (P < 0.05) the activation of NF-κB signal pathway caused by DSS treatment. In conclusion, PDTC could serve as an adjuvant therapy for the patient with IBD. ©2015, Springer Science+Business Media New York.

摘要:
Cyclodextrins (CDs) represent the most extensively investigated cyclic molecules due to their wide availability, facile functionalization, unique amphiphilicity and inclusion capacity. The marriage of CD chemistry with polymer science has generated novel biomaterials by integrating supermolecular host-guest chemistry and state-of-the-art polymer chemistry techniques. The current mini-review focuses on the recent progress in CD-functionalized polymers as drug carriers for cancer therapy. CD-functionalized polymers with different structures are summarized. Their application as drug carriers for cancer therapy is then highlighted. In the end, the future directions of this rapidly developing research field are discussed. © The Royal Society of Chemistry.

Both 19F MRI and optical imaging are powerful noninvasive molecular imaging modalities in biomedical applications. To integrate these two complementary imaging modalities, the design and synthesis of a novel 19F MRI/fluorescence dual-modal imaging agent is reported herein. Through Sonogashira coupling reaction between the fluorinated phenylacetylene and 1,2,4,5-tetraiodobenzene, a fluorophore with 48 symmetrical fluorines at its periphery was constructed with high efficacy. High aqueous solubility was achieved by PEGylation of the fluorophore with monodisperse PEGs. However, an unexpected self-assembly of the PEGylated amphiphilic fluorophore in water “turned off” the 19F NMR signal. However, hydrogenation of the triple bonds or introduction of branched monodisperse PEGs was able to efficiently tune the self-assembly, resulting in the “turning on” of the 19F NMR signal. One of these amphiphiles combines the advantages of label-free fluorescence, high 19F MRI sensitivity, biocompatibility, and excellent aqueous solubility. The results demonstrate the great potential of such amphiphiles for real-time 19F MRI and fluorescence dual-modality imaging.

摘要:
A new chemosensor 7-nitro-2,1,3-benzoxadiazole-4-allyl-N-(thiophen-2-ylmethyl)carbamate (NBDTC) was synthesized and utilized for palladium detection based on the Tsuji-Trost reaction. NBDTC displayed specific and ratiometric fluorescent responses toward palladium species. The chemosensor showed more than 50-fold enhancement in fluorescence intensity with the presence of PEG400 and palladium because NBDTC can be transformed to NBDT under palladium-catalyzing Tsuji-Trost reaction. NBDTC displayed high selectivity and sensitivity for palladium species with the detection limit of 1.13 x 10(-9) M. (C) 2015 Elsevier B.V. All rights reserved.

以白杨素为起始原料,通过卤代和水解反应制得中间产物7-O-羧烷基化的白杨素衍生物(6 ~ 9); 然后以1-乙基-3-(3-二甲氨基丙基)碳二亚胺(EDCI)、1-羟基苯并三氮唑(HOBt)和4-二甲氨基吡啶(DMAP)为催化体系,4个中间产物分别与甘氨酸甲酯盐酸盐进行酰胺缩合反应,制得白杨素甘氨酸甲酯类化合物12 ~ 15; 化合物12 ~ 15在pH = 10 ~ 11和室温下水解得到相应的白杨素甘氨酸类化合物(16 ~ 19). 所有目标化合物的结构均经~1H NMR,~(13)C NMR,IR 以及MS 确认. 以顺铂为阳性对照药物,采用噻唑蓝比色(MTT)法检测了目标化合物对人肝癌细胞HepG2和人胃癌细胞MGC-803的体外增殖抑制作用. 结果表明,目标化合物14 ~ 16,18和19的体外抗肿瘤活性明显强于白杨素,且化合物18(IC_(50) = 4. 36 μmol /L)对MGC-803细胞的增殖抑制作用强于阳性药物顺铂(IC_(50) = 4. 40 μmol /L).

分析研究了某铀尾矿区植物资源,并对这些植物的铀积累作用进行了研究。结果显示,该尾矿区污染土壤上植物群落较简单,共有高等植物31种,隶属12科,其中:禾本科最多,12种;其次是菊科植物,5种;凤尾蕨科和莎草科植物各3种;其他科各1种。1年生或多年生草本植物有28种,占总数的90.3%,其他3种为灌木或小乔木。对这31种植物进行了铀含量测定,结果发现,铀富集量在200 mg/kg以上的植物有14种,占总数的45%,其中富集量在600 mg/kg以上的有3种,分别是水莎草、牧草、小飞蓬。植物体内铀迁移系数大于1的有9种,其中燕麦、牧草、鼠曲草、青蒿中铀的迁移系数较大。这31种植物中,可考虑将水莎草、牧草作为超富集植物应用于铀污染土壤的修复,小飞蓬、盐肤木、枸骨、燕麦、鼠曲草、碎米莎草、龙葵等对铀污染土壤的修复具有潜在应用价值,可进一步研究。

摘要:
低压水银管是常见的灭菌工具,其一般采用的是波长为254 nm的UVC作为灭菌光源.而UVA(365 nm波长)配合发光二极管(light-emitting diodes,LED)也有明显的杀菌效果.本研究从灭菌能力,照射后细菌的光修复能力和再生长能力以及能耗等方面入手,比较短波长的UVC和长波长的UVA/LED对副溶血性弧菌的灭菌效果.结果显示,UVA/LED比UVC有更强的杀菌能力,且在受到UVA/LED照射后,副溶血性弧菌的再生长明显慢于单纯UVC处理后的细菌;对副溶血性弧菌的光修复酶基因表达水平进行检测发现,UVA/LED照射后副溶血性孤菌三种光修复酶基因的表达水平都无激活现象,而UVC处理后的细菌三种基因表达水平明显升高.我们认为UVA/LED可能抑制副溶血性弧菌的光修复现象.但在具备以上优势的同时,UVA/LED的能耗大大高于UVC,因此该技术还有待进一步改进.

Soybean is one of most important dicotyledonous food crops and is widely planted in Hunan Province, China. However, mining activity causes contamination of the soil in which soybean grows. To assess the impact of mining-induced soil contamination on soybean plants, a geoaccumulation index (I<inf>geo</inf>) was used to evaluate 20 soil samples from the mining and smelting areas of southern Hunan Province. The results indicated that Zn ranged from uncontaminated to a moderately contaminated level (I<inf>geo</inf><1), Pb was at a strongly contaminated level (I<inf>geo</inf>>3), and Cd was at an extremely contaminated level (I<inf>geo</inf>>5) across the whole study area. All of the studied soybean plants were affected by heavy metal Pb and Cd contamination, and the mean concentrations in seeds were 13.9mg/kg and 2.95mg/kg, respectively. The estimated bioconcentration factor and translocation factor showed that the soybean roots had a strong Cd bioconcentration capability and the stems had a strong translocation capability in terms of Pb, Cd, and Zn, with preferential transference of metals to the soybean leaves. The bioavailable fraction in the soil was characterized by the exchangeable fraction of heavy metals. In the present study, the bioavailable fractions of Pb, Cd, and Zn were significantly positively correlated with the concentration of these metals in soybean tissues (roots, stems, leaves, husks, and seeds). ©2013 SETAC.

摘要:
Exposure of cells to densely ionizing radiation can initiate a series of molecular and cellular events, which result in the gain and loss of functions in critical regulatory genes and cellular damage. In the present study, alpha-particle radiation-induced insulin resistance and toxic injury to 3T3- L1 preadipocyte were observed, and the mechanism underlying the action was investigated. 3T3-L1 preadipocytes were irradiated by different doses of alpha-particle at 0Gy , 0.3Gy , 0.6Gy , 1.0Gy , 1.5Gy , 2.0Gy , and 4.0Gy . Cloning efficiency was investigated to evaluate the cellular toxic effect of alpha-particle irradiation on cellular viability. Cytokinesis-block micronucleus technique ( CB-MNT ) and single-cell comet assay COMET assay were used to observe alpha-particle irradiation-induced DNA damage. Expression of caveolin-1, IRS-1 (insulin receptor substrate-1 ) and GLUT-4 ( glucose transporter-4 ) were analyzed by western blot and Reverse-transcription PCR (RT-PCR). Alpha-particle irradiation inhibited cloning efficiency, and increased micronucleus cells and the DNA damage in 3T3-L cells in a dose-dependent manner. Alpha-particle irradiation decreased the mRNA expression of GLUT-4 and IRS-1, and inhibited the protein expression of caveolin-1. The present results indicate that alphaparticle irradiation induced toxic injury and insulin resistance in 3T3-L1 preadipocyte, which might be associated with inhibition of GLUT-4 and IRS-1 expression by down-regulating caveolin-1. ©2013 Trade Science Inc. - INDIA.

摘要:
A generic and efficient homolog-targeted approach was used to expand screening and detection of target class of sulfonamides and structural analogs, based on a fast single-tube extraction/partitioning-multifunction adsorption cleanup (SEP/MAC) for class-specific fragmentation-dependent acquisition with a liquid chromatography-hybrid triple-quadrupole linear ion trap mass spectrometer (LC-QqLIT). By combining the two-stage process conducted in a single tube as one-pot protocol, the straightforward SEP/MAC procedure was optimized to offer clean extracts with reasonable recovery (71-109% with RSDs < 20%) and decreased matrix interferences (-9 to 19%) of multiresidual sulfonamide extraction from different tissue samples. The novel use of neutral loss scan of 66 Da (NLS) or precursor ion scanning of m/z 108 (PreS) in positive ion mode was found to achieve more comprehensive coverage of protonated molecular ions of a wide array of sulfonamides including N-4-acetyl and hydroxylamine metabolites plus their possible dimers. Moreover, the PreS-triggered automatically enhanced product ion spectral acquisition enabled simultaneous screening, profiling and confirmation of an unlimited number of analytes belonging to the sulfonamide class within a single analysis. The validation and application results of the generic SEP/MAC-based LC-QqLIT strategy consistently demonstrated favorable performances with acceptable accuracy (67-116%), precision (RSDs < 25%), and sensitivity (LOQs <= 7.5 ng g(-1)) to meet the acceptance criteria for all the sulfonamide-tissue combinations. Thus, the integration of the matrix-independent SEP/MAC procedure and the multiparameter matching algorithm with the unit-resolution LC-QqLIT instrument can serve as a valuable semi-targeted discovery strategy for rapid screening and reliable quantitative/confirmatory analysis of real samples. The objective of this study was to apply the "on/off" switch consisting of 3′ phosphorothioate-modified allele specific primers and exo + polymerase in single base discrimination of A1555G and C1494T mutations in the highly conserved sites of the mitochondrial 12S rRNA. The two point mutations are the hotspot mutations associated with either aminoglycoside antibiotics induced deafness or inherited nonsyndromic hearing loss. The PCR products of mitochondrial DNA (mtDNA) 12S rRNA gene were inserted into the pMD19-T vector for transformation into Escherichia coli JM109 competent cells for preparing wild-type pMD19-T/mt vector. Inverse PCR was carried out for mtDNA 12S rRNA gene C1494T and A1555G mutagenesis and DpnI endonuclease degradating methylated pMD19-T/mt vector existing in the inverse PCR products was carried out to construct the mutation-type pMD19-T/mtM vector. These constructed vectors were confirmed by DNA sequencing. Allelic specific primers targeting wild-type and mutation-type templates were designed with 3′ terminal phosphorothioate modification. Two-directional primer extension was performed using Pfu polymerases. Amplified by exo + polymerase, allelic specific primers perfectly matching wild-type allele were extended while no products were produced from primers targeting point-mutated deafness-related allele. Similarly, allelic specific primers perfectly matching point-mutated deafness-related mutation-type allele were extended and no products were yielded from primers targeting wild-type allele. No specific product was observed in the primer extension reaction mediated by on/off switch in screening the mtDNA 12S rRNA gene harboring either C1494T or A1555G mutation in 40 healthy volunteers tested. These data suggest that the "off switch" mediated by exo + polymerase is highly reliable in the diagnosis of monogenic diseases and the novel "on/off" switch has enormous applications in systematic and extended screening of the12S rRNA gene A1555G and C1494T mutations. The established assay can be widely used not only for hearing loss patients but also for normal subjects before the use of aminoglycoside antibiotics. © 2011 Springer Science+Business Media, LLC.

Atherosclerosis is an inflammatory disease characterized by the accumulation of macrophages in the arterial intima. The activated macrophages secreted more pro-inflammatory cytokines, such as tumor necrosis factor (TNF)-α, which promote the development of the disease. Apolipoprotein A-I (apoA-I), the major component of high density lipoprotein, is involved in reverse cholesterol transport of lipid metabolism. Recently, it has been found that apoA-I suppresses inflammation via repression of inflammatory cytokine expression;the mechanisms of the apoA-I-suppressive action, however, are not yet well characterized. In this study, we have for the first time found that apoA-I suppresses the expression of some inflammatory cytokines induced by lipopolysaccharide via a specific post-transcriptional regulation process, namely mRNA destabilization, in macrophages. Our further studies have also shown that AU-rich elements in the 3′-untranslated region of TNF-αmRNA are responsive to the apoA-I-mediated mRNA destabilization. The apoA-I-induced inflammatory cytokine mRNA destabilization was associated with increased expression of mRNA-destabilizing protein tristetraprolin through aJAK2/STAT3 signaling pathway-dependent manner. When blocking interaction of apoA-Iwith ATP-binding membrane cassette transporter A1 (ABCA1), a major receptor for apoA-Iin macrophages, it would almost totally abolish the effect of apoA-I on tristetraprolin expression. These results present not only a novel mechanism for the apoA-I-mediated inflammation suppression in macrophages but also provide new insights for developing strategies for modulating vascular inflammation and atherosclerosis. ©2011 by The American Society for Biochemistry and Molecular Biology, Inc.

A simple and accurate method has been developed to simultaneously separate and determine 10 bioactive compounds in citrus fruits by high-performance liquid chromatography coupled with diode array detection and electrospray ionisation mass spectrometry (HPLC-DAD-ESI/MS). This HPLC assay was performed on a reversed-phase C18 column with acetonitrile and 0.1% (v/v) aqueous formic acid as mobile phase. DAD has been performed at 273, 283 and 324 nm for quantification of the alkaloids, flavanones and hydroxycinnamic acids. MS was also employed to identify the each analyte. Ten analytes (naringin, hesperidin, neohesperidin, ferulic acid, p-coumaric acid, chlorogenic acid, caffeic acid, octopamine, synephrine and tyramine) demonstrated good linearity (r ≥ 0.9990) in a relatively wide concentration range. The method revealed high average recovery (range, 92.1-97.9%) and good precision with interday and intraday variations with less than 4.71%. The limits of detection (LODs) ranged from 0.02 to 0.11 μg/ml, while the limits of quantification (LOQs) were defined in the range of 0.08-0.39 μg/ml. The proposed method has been successfully applied to analyse three types of bioactive constituents in eight citrus hybrids pulps and eight citrus hybrids peels, which has been successfully cultivated in China. © 2011 Elsevier Ltd. A novel sensitive method has been developed for the detection of adenosine (AD) in human urine by using enhanced resonance light scattering (RLS). This method is based on the specific recognition and signal amplification of adenosine aptamer (Apt) coupled with gold nanoparticles (GNPs) via G-quartet-induced nanoparticle assembly, which was fabricated by triggering a structure switching of the 3′ terminus G-rich sequence and aptamer duplex. RLS signal linearly correlated with the concentration of adenosine over the range of 6-115 nM. The limit of detection (LOD) for adenosine is 1.8 nM with relative standard deviations (RSD) of 2.90-4.80% (n = 6). The present method has been successfully applied to determination of adenosine in real human urine, and the obtained results were in good agreement with those obtained by the HPLC method. Our investigation shows that the combination of the excellent selectivity of aptamer with the high sensitivity of the RLS technique could provide a promising potential for aptamer-based small molecule detection, and be beneficial in extending the application of RLS. © 2009 Elsevier Inc. 摘要:
Indels in evolutionary studies are rapidly decayed obeying a power law. The present study analyzed the length distribution of small insertions and deletions associated with human diseases and confirmed that the decay pattern of these small mutations is similar to that of indels when the mutation datasets are large enough. The describable decay pattern of somatic mutations may have application in the evaluation of varied penetrance of different mutations and in association study of gene mutation with carcinogenesis. © 2009 Humana Press.

The effect of the presence of ions on the aggregation behavior of natural polymer alginate in aqueous media was investigated. Through self-assembly of alginate in aqueous solutions containing Ca<sup>2+</sup> and CO<inf>3</inf> <sup>2-</sup> ions under very mild conditions, nanosized aggregates with four different morphologies, i.e., nanosphere, vesicle, nanoparticle, and nanorod, could be obtained under different ion concentrations. This study provides a simple approach to prepare organic/inorganic (O/I) hybrid natural polymer based aggregates with different morphologies. ©2009 American Chemical Society.

摘要:
We propose a short definition of GENOME: The full complement of genetic materials possessed by an intracellular parasite, a cell, or an organism. Accordingly, the human genome is the entire complement of inherited genetic materials possessed by an individual person, or possessed by a cell in an individual person. For higher species, the genomic makeup includes DNA in the nucleus and in the organelles regardless of the number of chromosomes and the homoplasmic or heteroplasmic status of the mitochondrial or chloroplastic DNA. Practically, GENOME can be referred to at the molecular, cellular, individual, and species levels, which has various implications in biotechnological research and molecular diagnostics.

To investigate the effect of shRNA-SR-PSOX (Scavenger receptor binds phosphatidylserine and oxidized lipoprotein) on smooth muscle cells(SMC) migration, we designed and synthesised the shRNA oligonucleotide targeted human SR-PSOX gene, and constructed the shRNA lentiviral expression system in vitro to infect HepG2 cell line, and the silencing efficiency was assayed by real-time reverse transcription-polymerase chain reaction, immunofluorescence staining, Western blotting and Transwell chambers. The reverse transcription-polymerase chain reaction (RT-PCR) results showed that the potent inhibition of SR-PSOX expression could reach 80%, and it was specific to the SR-PSOX -derived shRNA, not the Caveolin-1-derived shRNA (negative shRNA control). Indirect immunofluorescence and Western blot results showed the SR-PSOX expression decreased obviously in SR-PSOX-shRNA transfected group compared with control group. In addition, transwell chambers results indicated that these SR-PSOX over-expressing cells showed ˜3 fold increased migration of SMCs compared with normal SMC cells or siRNA cells. Red oil staining found that SR-PSOX over expressed SMCs had more lipid droplets, while lipid droplets in siRNA-SR-PSOX vector groups decreased significantly. All these data indicated that blockage and downregulation SR-PSOX expression is a potential therapeutic target to prevent inflammatory AS damage. ©2009 IEEE.

Aldo-keto reductase family 1 member B10 (AKR1B10) is primarily expressed in the normal human colon and small intestine but overexpressed in liver and lung cancer. Our previous studies have shown that AKR1B10 mediates the ubiquitin-dependent degradation of acetyl-CoA carboxylase-α. In this study, we demonstrate that AKR1B10 is critical to cell survival. In human colon carcinoma cells (HCT-8) and lung carcinoma cells (NCIH460), small-interfering RNA-induced AKR1B10 silencing resulted in caspase-3-mediated apoptosis. In these cells, the total and subspecies of cellular lipids, particularly of phospholipids, were decreased by more than 50%, concomitant with 2 - 3-fold increase in reactive oxygen species, mitochondrial cytochrome c efflux, and caspase-3 cleavage. AKR1B10 silencing also increased the levels of α,β-unsaturated carbonyls, leading to the 2 - 3-fold increase of cellular lipid peroxides. Supplementing the HCT-8 cells with palmitic acid (80 μM), the end product of fatty acid synthesis, partially rescued the apoptosis induced by AKR1B10 silencing, whereas exposing the HCT-8 cells to epalrestat, an AKR1B10 inhibitor, led to more than 2-fold elevation of the intracellular lipid peroxides, resulting in apoptosis. These data suggest that AKR1B10 affects cell survival through modulating lipid synthesis, mitochondrial function, and oxidative status, as well as carbonyl levels, being an important cell survival protein. ©2009 by The American Society for Biochemistry and Molecular Biology, Inc.
