山银花黄酮类提取物对人肝L-02细胞甘油三酯蓄积及SREBP-1c表达的影响

摘要:
目的观察山银花黄酮类提取物对人肝L-02细胞甘油三酯蓄积的影响,并初步探讨SREBP-1c蛋白及基因表达在其中的作用。方法油红O染色初步观察山银花黄酮类提取物对脂变人肝L-02细胞中脂滴形成的影响。然后以脂变人肝L-02细胞为模型,不同浓度(0、5、10、20 mg/L)山银花黄酮类提取物作用于细胞24 h以及10 mg/L山银花黄酮类提取物作用于细胞不同时间(0、12、24、48、72 h),生化试剂盒测定细胞内甘油三酯(TG)的含量,Western印迹法检测细胞内SREBP-1c蛋白表达变化,实时荧光定量PCR检测细胞内SREBP-1c mRNA表达变化。结果山银花黄酮类提取物显著降低人肝L-02细胞中脂滴含量,细胞内TG水平下降,SREBP-1c蛋白及基因表达下调,且呈现一定的剂量和时间依赖性。结论山银花中黄酮类提取物能引起人肝L-02细胞TG含量下降,且这种作用与SREBP-1c蛋白及基因表达下调有一定关系。

摘要:
This study aims to observe the effect of ω-3 polyunsaturated fatty acids on initiation and elimination of the schistosomiasis inflammatory response and liver fibrosis. The mice infected with the cercariae of Schistosoma japonicum (20 ± cercarie per mice) were separated randomly into several groups. After 60 days, liver tissue samples of all mice were sectioned. Hematoxylin-eosin (HE) staining, Masson staining, the enzyme-linked immunosorbent assay (ELISA), and flow cytometry (FCM) were performed. Through HE and Masson staining, the size of egg (ovum) granuloma and the collagen deposited in mice’s livers in ω-3 PUFAs and praziquantel mixed groups were less than that of model group and praziquantel treated group. The serum level of IL-13 and TNF-α were lower than that of model group and praziquantel treated group. The indicators of liver fibrosis, such as HA and LN in the group treated with ω-3 PUFAs and praziquantel before the release of soluble eggs antigen (SEA) into blood, were lower than that of model group and praziquantel treated group, respectively. The combined treatment of ω-3 polyunsaturated fatty acids and praziquantel conducted before the release of soluble eggs antigens into the blood decreases liver ovum granulomatous inflammation and fibrosis degree in the schistosomiasis. The mechanism of the ω-3 polyunsaturated fatty acid may be related to the adjustment of the anti-inflammatory and immune responses. © 2015, E-Century Publishing Corporation. All rights reserved.

Background Atherosclerosis is a major cause of coronary artery disease, which is characterized by cellular lipid accumulation. Lipoprotein lipase (LPL) is a key enzyme in lipid metabolism. Studies have shown that macrophage-derived LPL exhibits proatherogenic properties, and plays a major role in lipid accumulation in macrophages. Evidence suggests that oxidative stress can effectively enhance macrophage LPL production. Betulinic acid (BA) is a pentacyclic lupane triterpene with a potent antioxidant activity. In this study, we investigated whether BA affects the expression of macrophage LPL and how it regulates cellular lipid accumulation. Methods and results We revealed that BA downregulated H2O2-simulated macrophage LPL protein, mRNA levels and its activity in both concentration- and time-dependent manners. Furthermore, BA decreased LPL-involved total cholesterol and triglyceride levels in macrophages. In addition, cellular lipid staining by Oil Red O showed that BA decreased cellular lipid droplet deposition. Next, we confirmed that pretreatment with BA decreased H2O2-induced production of intracellular reactive oxygen species in a concentration-dependent manner. Further studies demonstrated that BA inhibited H2O2-induced membrane translocation of PKC, phosphorylation of ERK1/2 and c-Fos. Finally, the induction of LPL production and activity by H2O2 was abolished by BA, inhibition of PKC or ERK or depletion c-Fos, respectively. Conclusions BA, through its role of antioxidant activity, attenuated macrophage-derived LPL expression and activity induced by oxidative stress, and effectively reduced cellular lipid accumulation, likely through inhibition of the pathways involving PKC, ERK and c-Fos. These effects of BA may contribute to its mitigation of atherosclerosis and help develop BA as a therapeutic compound in treatment of atherosclerosis. © 2015 Published by Elsevier B.V.

Aim: Previous studies have demonstrated that the dysregulated-secretion of adipokines by adipocytes may contribute to obesity-associated atherosclerosis (As) and high density lipoprotein (HDL) may protect against atherogenesis through multiple pathways. This study was to explore the effect of HDL on the oxLDL uptake in inflammatory adipocytes stimulated by endotoxin lipopolysaccharide (LPS) and the possible mechanism. Methods and Results: 3T3-L1 adipocytes were cultured and induced to differentiation and maturation. Acute inflammation in adipocytes was induced by LPS (100 ng/ml) for 6 hours. The adipocytes were pretreated with HDL in various concentrations (10, 50, 100 μg/ml) for 16 hours or with specific PPARγ antagonist (GW9662, 10 μM) or agonist (Rosiglitazone, 10 μM) for 30 min before administration of LPS. The results showed that LPS significantly increased the release of inflammation-related adipokines, such as monocyte chemoattractant protein-1 (MCP-1), plasminogen activator inhibitor 1 (PAI-1), tumor necrosis factor-alpha (TNF-α), interleukin (IL)-8 and IL-6, while decreasing the release of leptin and adiponectin. Meanwhile, LPS reduced the uptake and degradation of 125I-oxLDL, and down-regulated the expression of PPARγ and CD36. Pretreatment with HDL dose-dependently affected the release of IL-8 and IL-6 and the reduced uptake and degradation of oxLDL of adipocytes stimulated by LPS, accompanied with marked upregulation of PPARγ and CD36 expression. Pretreatment with GW9662 markedly inhibited the upregulation of CD36 expression mediated by HDL (100 μg/ml), while the effects of Rosiglitazone were opposite to GW9662. Conclusions: HDL may increase oxLDL uptake of inflammatory adipocytes stimulated by LPS via upregulation of PPARγ/CD36 pathway, which may be a new mechanism of anti-atherosclerosis mediated by HDL. © 2015 Ivyspring International Publisher.

Here, we investigated the role of LXR alpha in capsaicin mediated anti-inflammatory effects. Results revealed that capsaicin inhibits LPS-induced IL-1 beta, IL-6 and TNF-alpha production in a time- and dose-dependent manner. Moreover, capsaicin increases LXR alpha expression through PPAR gamma pathway. Inhibition of LXR alpha activation by siRNA diminished the inhibitory action of capsaicin on LPS-induced IL-1 beta, IL-6 and TNF-alpha production. Additionally, LXR alpha siRNA abrogated the inhibitory action of capsaicin on p65 NF-kappa B protein expression. Thus, we propose that the anti-inflammatory effects of capsaicin are LXR alpha dependent, and LXR alpha may potentially link the capsaicin mediated PPAR gamma activation and NF-kappa B inhibition in LPS-induced inflammatory response. 内皮-间质转化(endothelial-to-mesenchymal transition,End MT)属于上皮-间质转化(epithelial-to-mesenchymal transition,EMT)的特殊类型,是内皮细胞在多种刺激因素作用下向间充质细胞转化的过程,在此过程中内皮细胞逐渐失去其形态和功能,获得增殖、迁移和合成胶原等间充质细胞表型特点.近来研究发现,内皮-间质转化在内皮功能调节,心肌、血管及瓣膜的发育和结构重塑等方面发挥着关键的作用,提示其在心血管疾病领域具有重要的研究意义.本文对内皮-间质转化的特点、功能、调节机制以及在心血管系统发育、心肌纤维化、肺动脉高压和动脉粥样硬化性血管重构等心血管疾病中的作用做一综述,以期为心血管疾病的防治提供新靶点.

Oxidized low-density lipoprotein (ox-LDL) is an independent risk factor of atherosclerosis. However, the mechanism underlying its pro-atherosclerosis roles has not yet been well explored. DNA demethylation modification, via DNA methyltransferases or ten-eleven-translocation (TET) family, is a crisis epigenetic regulation for various biological and pathological processes. This study aimed to investigate the effects of ox-LDL on macrophage autophagy and its potential epigenetic mechanism. Results showed that after treatment with 0, 10, 20, 40 or 80 mg/L ox-LDL for 24 h, the autophagy markers Beclin 1 and LC3 expression were obviously decreased at protein levels (P < 0.05). The mRNA and protein expression of TET2 was evidently decreased (P < 0.05). After pre-treatment with TET2 siRNA, the mRNA and protein levels of Beclin 1 and LC3 decreased compared with the 80 mg/L treatment group (P < 0.01). The mRNA and protein levels of Beclin 1 and LC3-II were up-regulated (P < 0.05) in the 5-aza-2′-deoxycytidine (a DNA methyltransferase inhibitor) of pretreatment group. Consistent with the Western blot results, cell immunofluorescence showed that the protein concentration of LC3-II decreased in the TET2 siRNA group and increased in the 5-aza-2′-deoxycytidine group. Taken together, these results showed that DNA demethylation modifications regulate ox-LDL-treated THP-1 macrophages autophagy and TET2 might be a novel regulator.

Atherosclerosis is a lipid disorder disease characterized by chronic blood vessel wall inflammation driven by the subendothelial accumulation of macrophages. Studies have shown that lipoprotein lipase (LPL) participates in lipid metabolism, but it is not yet known whether post-transcriptional regulation of LPL gene expression by microRNAs (miRNAs) occurs in vivo. Here, we tested that miR-467b provides protection against atherosclerosis by regulating the target gene LPL which leads to reductions in LPL expression, lipid accumulation, progression of atherosclerosis and production of inflammatory cytokines in apolipoprotein E knockout (apoE-/-) mice. Treatment of apoE-/- mice with intra-peritoneal injection of miR-467b agomir led to decreased blood plasma levels of total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), IL-1β and monocyte chemotactic protein-1 (MCP-1). Using Western blots and real time PCR, we determined that LPL expression in aorta and abdominal cavity macrophages were significantly down-regulated in the miR-467b agomir group. Furthermore, systemic treatment with miR-467b antagomir accelerated the progression of atherosclerosis in the aorta of apoE -/- mice. The present study showed that miR-467b protects apoE -/- mice from atherosclerosis by reducing lipid accumulation and inflammatory cytokine secretion via downregulation of LPL expression. Therefore, targeting miR-467b may offer a promising strategy to treat atherosclerotic vascular disease. © 2013 Elsevier Inc. All rights reserved.

Rhodococcus opacus strain PD630 (R. opacus PD630), is an oleaginous bacterium, and also is one of few prokaryotic organisms that contain lipid droplets (LDs). LD is an important organelle for lipid storage but also intercellular communication regarding energy metabolism, and yet is a poorly understood cellular organelle. To understand the dynamics of LD using a simple model organism, we conducted a series of comprehensive omics studies of R. opacus PD630 including complete genome, transcriptome and proteome analysis. The genome of R. opacus PD630 encodes 8947 genes that are significantly enriched in the lipid transport, synthesis and metabolic, indicating a super ability of carbon source biosynthesis and catabolism. The comparative transcriptome analysis from three culture conditions revealed the landscape of gene-altered expressions responsible for lipid accumulation. The LD proteomes further identified the proteins that mediate lipid synthesis, storage and other biological functions. Integrating these three omics uncovered 177 proteins that may be involved in lipid metabolism and LD dynamics. A LD structure-like protein LPD06283 was further verified to affect the LD morphology. Our omics studies provide not only a first integrated omics study of prokaryotic LD organelle, but also a systematic platform for facilitating further prokaryotic LD research and biofuel development.

摘要:
Objective Foam cell formation in the arterial wall plays a key role in the development of atherosclerosis. Recent studies showed that Urotensin II (U II) is involved in the pathogenesis of atherosclerosis. Here we examined the effects of human U II on ATP-binding cassette transporter A1 (ABCA1) expression and the underlying mechanism in THP-1 macrophages. Methods and results Cultured THP-1 macrophages were treated with U II, followed by measuring the intracellular lipid contents, cholesterol efflux and ABCA1 levels. The results showed that U II dramatically decreased ABCA1 levels and impaired cholesterol efflux. However, the effects of U II on ABCA1 protein expression and cellular cholesterol efflux were partially reversed by inhibition of extracellular signal regulated kinase 1/2 (ERK1/2) and nuclear factor kappa B (NF-κB) activity, suggesting the potential roles of ERK1/2 and NF-κB in ABCA1 expression, respectively. Conclusion Our current data indicate that U II may have promoting effects on the progression of atherosclerosis, likely through suppressing ABCA1 expression via activation of the ERK/NF-κB pathway and reducing cholesterol efflux to promote macrophage foam cell formation. © 2014 Elsevier Inc. 摘要:
Background: Accumulating evidence suggests that microRNA-590 (miR-590) has protective effects on cardiovascular diseases, but the mechanism is unknown. Interestingly, previous studies from our laboratory and others have shown that macrophage-derived lipoprotein lipase (LPL) might accelerate atherosclerosis by promoting lipid accumulation and inflammatory response. However, the regulation of LPL at the post-transcriptional level by microRNAs has not been fully understood. In this study, we explored whether miR-590 affects the expression of LPL and its potential subsequent effects on lipid accumulation and pro-inflammatory cytokine secretion in human THP-1 macrophages. Methods and results: Using bioinformatics analyses and dual-luciferase reporter assays, we found that miR-590 directly inhibited LPL protein and mRNA expression by targeting LPL 3'UTR. LPL Activity Assays showed that miR-590 reduced LPL activity in the culture media. Oil Red 0 staining and high-performance liquid chromatography assays showed that miR-590 had inhibitory effects on the lipid accumulation in human THP-1 macrophages. We also illustrated that miR-590 alleviated pro-inflammatory cytokine secretion in human THP-1 macrophages as measured by ELISA. With the method of small interfering RNA, we found that LPL siRNA can inhibit the miR-590 inhibitor-induced increase in lipid accumulation and secretion of pro-inflammatory cytokines in oxLDL-treated human THP-1 macrophages. Conclusions: MiR-590 attenuates lipid accumulation and pro-inflammatory cytokine secretion by targeting LPL gene in human THP-1 macrophages. Therefore, targeting miR-590 may offer a promising strategy to treat atherosclerotic cardiovascular diseases. 摘要:
目的:分析ECS模拟人在卓越医师临床技能教学中的教学方式和效果。方法:使用ECS模拟人对我校2010级临床医学专业卓越医师班26名学生进行教学训练,另取同班级26名进行传统课堂教学模式的学生为对照组,观察教学效果并进行问卷调查。结果:教学前两组学生考核成绩无明显差别,经ECS教学后实验组学生对临床技能的掌握程度显著提高(P<0.05),其整体成绩也显著高于对照组(P<0.05)。结论:ECS模拟人教学有助于提高学生的临床操作能力。

摘要:
Lipoprotein lipase (LPL) is a key enzyme in lipid metabolism and responsible for catalyzing lipolysis of triglycerides in lipoproteins. LPL is produced mainly in adipose tissue, skeletal and heart muscle, as well as in macrophage and other tissues. After synthesized, it is secreted and translocated to the vascular lumen. LPL expression and activity are regulated by a variety of factors, such as transcription factors, interactive proteins and nutritional state through complicated mechanisms. LPL with different distributions may exert distinct functions and have diverse roles in human health and disease with close association with atherosclerosis. It may pose a pro-atherogenic or an anti-atherogenic effect depending on its locations. In this review, we will discuss its gene, protein, synthesis, transportation and biological functions, and then focus on its regulation and relationship with atherosclerosis and potential underlying mechanisms. The goal of this review is to provide basic information and novel insight for further studies and therapeutic targets. (C) 2014 Elsevier Ireland Ltd. All rights reserved.

番茄红素对大鼠脑缺血再灌注损伤的保护作用及脑组织caspase-3表达的影响

摘要:
目的:观察番茄红素对大鼠脑缺血再灌注损伤的保护作用及脑组织天冬氨酸特异性半胱氨酸蛋白酶( caspase )-3表达的影响。方法选择雄性SD大鼠36只,利用线栓法制备SD大鼠大脑中动脉缺血模型,模型制备成功后随机分为3组各12只,对照组以羧甲基纤维素钠溶液灌胃,番茄红素组以40 mg/kg番茄红素(羧甲基纤维素钠悬溶)灌胃,银杏叶片组以40 mg/kg银杏叶片(羧甲基纤维素钠悬溶)灌胃,共14 d。按照Bederson6级5分评分标准评估各组大鼠神经行为学,显微镜下观察大鼠脑组织形态结构,Western blotting法检测脑组织caspase-3。结果番茄红素组、银杏叶片组、对照组干预14 d神经功能缺损程度评分分别为(2.25±0.17)、(2.04±0.28)、(3.76±0.42)分,番茄红素组、银杏叶片组与对照组比较,P均<0.05。对照组神经组织大片坏死,周围残存组织结构不完整,神经细胞水肿明显,大量空泡形成;番茄红素组与银杏叶片组神经组织结构明显改善、坏死区域缩小,梗死灶周围残存组织较多。番茄红素组、银杏叶片组、对照组caspase-3相对表达值分别为0.2、0.15、1,番茄红素组、银杏叶片组与对照组比较,P均<0.05。结论番茄红素对大鼠脑缺血再灌注损伤有一定的保护作用,且能引起脑组织caspase-3表达下调。

目的:观察菝葜乙酸乙酯提取物对人单核/巨噬细胞系(THP-1)源性荷脂细胞胆固醇蓄积的影响,并初步探讨类固醇调节因子结合蛋白-1(SREBP-1)蛋白表达在其中的作用。方法:以THP-1源性荷脂细胞为模型,洛伐他汀(80 mg·L-1)做阳性对照,菝葜乙酸乙酯提取物不同浓度(0,40,80,160 mg·L-1)作用于细胞24 h以及80 mg·L-1作用于细胞不同时间(0,12,24,48 h),高效液相色谱法测定细胞内总胆固醇(TC),游离胆固醇(FC),胆固醇酯(CE)的含量,油红O染色观察菝葜乙酸乙酯提取物对THP-1源性荷脂细胞中脂滴形成的影响,Western blot印迹法检测细胞内SREBP-1蛋白表达变化。结果:菝葜乙酸乙酯提取物明显降低THP-1源性荷脂细胞中脂滴含量,细胞内TC,FC,CE水平呈现一定的剂量和时间依赖性下降趋势,同时SREBP-1蛋白表达上调。结论:菝葜乙酸乙酯提取物能引起THP-1源性荷脂细胞TC,FC,CE含量下降,且这种作用与SREBP-1蛋白表达上调有一定关系。

目的:观察薯蓣皂苷对大鼠缺血再灌注脑损伤的保护作用,同时检测在此过程中脑组织caspase-3蛋白表达的变化。方法:利用线栓法制作SD大鼠大脑中动脉缺血(MCAO)模型,根据完全随机原则分为5组,分别为假手术组、模型组及薯蓣皂苷不同剂量(40,80,160 mg·kg~(-1)·d~(-1),ig)治疗组。14 d后,进行神经行为学评分,观察脑组织形态结构,检测血清TNF-α浓度,测定脑组织caspase-3蛋白的表达。结果:在薯蓣皂苷作用下,缺血再灌注模型大鼠神经行为缺损得到一定程度改善,脑组织坏死区域缩小,血清TNF-α浓度下降,脑组织caspase-3蛋白表达降低,并呈现一定的剂量依赖性。结论:薯蓣皂苷对大鼠缺血再灌注脑损伤有保护作用,且能引起缺血再灌注脑损伤大鼠脑组织caspase-3蛋白表达下调。
