摘要:
Bacteria can induce significant alteration in the cell transcriptome and develop many strategies to modify immune signaling for its survival. In recent years, a new class of regulatory RNAs, long noncoding RNAs (lncRNAs), has been demonstrated to play an essential role in host gene expression. Growing literature indicate that lncRNAs function as positive or negative effectors on antibacterial immunity. On the one hand, the host regulates immune-related genes at epigenetic, transcriptional, and post-transcriptional levels by lncRNAs, thereby protecting itself from pathogen invasion. On the other hand, bacteria can manipulate the host signaling pathways by regulating the host lncRNAs to escape immune clearance. In addition, some bacteria even produce lncRNAs, which are involved in the pathogenic process of pathogens. Some dysregulated lncRNAs during bacterial infections can be used as a potential diagnostic marker for infection. Understanding of gene expression regulation through lncRNAs helps illustrate bacterial pathogenesis. Here, we summarize the functions of lncRNAs and current advances of lncRNAs in different bacterial infections and look forward to the future research orientation.

摘要:
The past decades have witnessed significant progress in discovery and characterize cytosolic DNA sensing and signaling, especially the understanding of the stimulator of interferon genes (STING). This pathway to foreign nucleic acids enables the initiation of robust anti-pathogenic responses to protect the host, and provides a new understanding for therapeutic intervention in a growing infectious disease, including chlamydial infection. Chlamydiae are obligate intracellular pathogenic bacterium causing widespread human diseases such as sexually transmitted infections and respiratory tract infections. Previous studies have shown that IFN production and autophagy are well recognized as being two critical processes induced by STING, and these two processes were also activated during chlamydial infection. In this review, we summarize the important characteristics of the STING activation pathway and recent snapshots about the role of STING in chlamydial infection. Studying the role of STING in chlamydial infection could provide valuable information to further understand the pathogenesis and treatment of chlamydial infection.

摘要:
Cervical cancer (CC) is regarded as the second serious threat to women's health worldwide; it's associated with certain viruses that are transmitted through sexual intercourse. Therefore, the pathogenesis of CC remains to be studied. The identified long non-coding RNAs (lncRNAs) as a key genomic product were found to be commonly dysregulated in CC and to exert significant effects in the initiation, migration, invasion and therapeutic response of CC. Therefore lncRNAs may be used as tumor suppressor genes or oncogenes to interact with DNA, RNA or proteins for the regulation of gene expression and cell signaling pathways. The relationship between single lncRNA and CC has been discovered. However, full-scale reviews on the lncRNAs function in CC are deficiency. In this review, we describe the recent reports on the dysregulated patterns regulation of lncRNAs in CC. We also conclude the recent advances on biologic functions and molecular regulation mechanism and potential clinical application of lncRNAs in CC.

摘要:
As obligate intracellular bacterial pathogens, members of the Chlamydia genera are the pivotal triggers for a wide range of infections, which can lead to blinding trachoma, pelvic inflammation, and respiratory diseases. Because of their restricted parasitism inside eukaryotic cells, the pathogens have to develop multiple strategies for adaptation with the hostile intracellular environment-intrinsically present in all host cells-to survive. The strategies that are brought into play at different stages of chlamydial development mainly involve interfering with diverse innate immune responses, such as innate immune recognition, inflammation, apoptosis, autophagy, as well as the manipulation of innate immune cells to serve as potential niches for chlamydial replication. This review will focus on the innate immune responses against chlamydial infection, highlighting the underlying molecular mechanisms used by the Chlamydia spp. to counteract host innate immune defenses. Insights into these subtle pathogenic mechanisms not only provide a rationale for the augmentation of immune responses against chlamydial infection but also open avenues for further investigation of the molecular mechanisms driving the survival of these clinically important pathogens in host innate immunity. © 2019 Chen, Wen and Li.

摘要:
目的 探讨pORF5质粒蛋白抗凋亡分子机制,为进一步阐明沙眼衣原体致病机制提供实验依据。方法 将HeLa细胞分为两组,一组用凋亡诱导剂碳酰氰基-对-氯苯腙(CCCP)刺激30 min,另一组经pORF5质粒蛋白预处理18 h后,再用CCCP刺激30 min。然后,Western印迹检测细胞凋亡相关蛋白Bcl-2、Bax和胱天蛋白酶3(caspase-3)的表达水平;JC-1荧光探针检测HeLa细胞线粒体膜电位变化;间接免疫荧光观察细胞色素c释放情况。为了分析高迁移率族蛋白1(HMGB1)是否参与pORF5质粒蛋白的抗凋亡作用,分别用HMGB1 shRNA和对照RNA稳定转染HeLa细胞,再用pORF5质粒蛋白与CCCP共刺激两种细胞后,测定Bcl-2、Bax和活化的caspase-3蛋白的表达以及细胞色素c的释放情况。两组间比较采用配对样本t检验。结果 pORF5质粒蛋白能拮抗CCCP诱导的线粒体膜电位的下降,CCCP单独处理组Hela细胞红/绿荧光强度比率为0.4 ± 0.1,显著低于pORF5与CCCP共处理组(1.7 ± 0.3,t = 6.95,P < 0.01)。与对照组相比,pORF5与CCCP共处理组HeLa细胞Bcl-2蛋白表达水平增加(5.3 ± 0.6)倍(t = 8.62,P < 0.01),而Bax和活化的caspase-3平均表达水平分别降低79% ± 10%(t = 9.23,P < 0.01)和75% ± 8%(t = 4.26,P < 0.05)。与对照RNA转染组相比,HMGB1 shRNA转染组HeLa细胞线粒体膜电位下降(t = 11.23,P < 0.01),细胞色素c释放增加,Bcl-2的表达水平降低(t = 7.19,P < 0.05),而Bax的表达水平增加(t = 13.06,P < 0.01)。结论 沙眼衣原体pORF5质粒蛋白通过HMGB1阻断线粒体凋亡途径发挥抗凋亡作用。

OBJECTIVE: This study is to investigate the functions of newly discovered genes in Chlamydia muridarum (C. muridarum) strains with single gene differences. METHODS: Using whole genome sequencing and plaque formation assays, C. muridarum parental and passaging strains were established, and the isogenic clones expressing certain genotypes were isolated. Strains with single gene differences were obtained. Based on prediction, the valuable strains with single gene differences of tc0412, tc0668 or tc0237 were subjected to the in vitro and in vivo experiments for biological characterization and virulence analysis. RESULTS: Insertional -472840T mutation of the tc0412 gene (T28T/B3 type) matching with the nonmutant tc0668 gene and tc0237 gene with point mutations G797659T (Q117E) might slow the growth of Chlamydia due to the lack of a plasmid. The nonmutant tc0668 in the strain might induce a high incidence of hydrosalpinx in mice, while tc0668 with a G797659T point mutation was significantly attenuated. Compared with the nonmutant tc0237, the strains containing mutant tc0237 were characterized by reduced centrifugation dependence during infection. CONCLUSION: The identification and characterization of these genes might contribute to the comprehensive understanding of the pathogenic mechanism of Chlamydia. © FEMS 2019.

Previously we reported that recombinant Chlamydia muridarum macrophage infectivity potentiator (MIP) provided partial protection against C. muridarum genital tract infection in mice. On the other hand, Chlamydia trachomatis plasmid encoded Pgp3could induce the protection against C. muridarum air way infection. This study aimed to evaluate the immunogenicity of MIP and Pgp3 from C. trachomatis serovar D and further investigate whether MIP and Pgp3 provide cross-serovar protection against C. muridarum genital tract infection in mice. Our results showed that vaccination by any regimen, including MIP alone, Pgp3 alone or MIP plus Pgp3, induced specific serum antibody production and Th1-dominant cellular responses in mice. Live chlamydial shedding from the vaginal and inflammatory pathologies in the oviduct markedly reduced. However, MIP + Pgp3 vaccination did not provide better protection than the single immunization. In conclusion, this study demonstrated that both MIP and Pgp3 can induce cross-serovar protective against chlamydial genital tract infection, and provided the guide for the development of optimal multisubunit vaccines against C. trachomatis infection. © 2018 Elsevier GmbH

衣原体(Chlamydia)是一类具有独特的两相发育周期、专性细胞内寄生的革兰阴性菌,能引起人类多种疾病。pORF5是衣原体隐蔽性质粒编码的分泌性效应蛋白。近年来研究证实,pORF5质粒编码蛋白是衣原体重要的毒力蛋白,与衣原体致病密切相关。现就衣原体pORF5质粒编码蛋白的生物学特性、致炎性作用、抗凋亡作用及促自噬作用作一概述。

哺乳动物肽聚糖识别蛋白(peptidoglycan recognition proteins, PGRPs)是一类可识别肽聚糖的模式识别受体,在先天免疫应答中发挥重要的识别和调节功能。PGRPs通过与肽聚糖结合,诱导激活细菌双组分系统(two-component systems, TCSs)如CssR-CssS、CpxA-CpxR等,诱导氧化应激、硫醇应激和金属应激等反应发挥抗菌活性。现对PGRPs的抗菌活性及其与抗生素的杀菌机制进行比较,旨在为疾病的防治提供理论依据。

Chlamydia trachomatis is an important human pathogen responsible for a variety of important diseases. pORF5, a plasmid protein in C. trachomatis, has been reported to contribute to chlamydial pathogenesis. Apoptosis and autophagy are two important cellular processes, which also participate in the pathological and the physiological processes in C. trachomatis infection. The present study aimed to explore the effects of pORF5 protein on autophagy and apoptosis in HeLa cells. Stable pORF5-HeLa and vector-HeLa cell lines that were transduced with pORF5 recombination lentivirus and control lentivirus, respectively, were constructed. Immunofluorescence was used to observe the cells. Western blotting was carried out to detect expression of target proteins. Real-time polymerase chain reaction was used to determine the mRNA expression. Flow cytometry, TUNEL assay and Hoechst staining were employed to detect apoptosis. pORF5 sensitized HeLa cells to autophagy that was induced by nutrient deprivation. In addition, pORF5 increased the ratio of LC3-II/LC3-I and Beclin-1 expression, down-regulated the expression of Bax and Caspase-3 and up-regulated Bcl-2 expression to suppress apoptosis induced by TNF-α. Autophagy inhibitor 3-MA elevated apoptotic rate significantly in pORF5-HeLa cells. The study preliminarily confirms that plasmid protein pORF5 mediates the crosstalk between autophagy and apoptosis, which may contribute to persistent C. trachomatis infection in host cells. © 2019, © 2019 The Author(s). 摘要:
Chlamydia muridarum, an obligate intracellular pathogen, was used to establish a murine model of female upper genital tract infection by Chlamydia trachomatis. TC0668 in C. muridarum is a hypothetical chromosomal virulence protein that is involved in upper genital tract pathogenesis. The infection of mice with the C. muridarum TC0668-mutant (G216*) strain results in less pathological damage in the upper genital tract. In this study, an isobaric tags for relative and absolute quantitation (iTRAQ)-based quantitative proteomics analysis was performed to identify differentially expressed proteins between TC0668 wild-type (TC0668wt) and TC0668 mutant (TC0668mut) strains at 6, 12, 18, and 24 h post-infection (p.i.). Of the 550 proteins differentially expressed at 18 h p.i., 222 and 328 were up-regulated and down-regulated, respectively, inTC0668mut-infected cells. The expression of seven up-regulated proteins (encoded by SRPRB, JAK1, PMM1, HLA-DQB1, THBS1, ITPR1, and BCAP31) and three down-regulated proteins (encoded by MAPKAPK2, TRAFD1, and IFI16) from the iTRAQ analysis were validated using quantitative real-time (qRT)-PCR. The qRT-PCR results were consistent with those of iTRAQ. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses revealed that the differentially expressed proteins primarily participated in inflammatory responses, fibrosis, metabolic processes, and complement coagulation cascades, and were mainly enriched in the phosphatidylinositol 3′-kinase (PI3K)/Akt, nuclear factor kappa-B (NF-κB), and other signaling pathways. Using western-blotting and immunofluorescence detection, significant differences in activation of the PI3K/Akt and NF-κB signaling pathways were observed between the TC0668wt- and TC0668mut-infected cells. Differentially expressed proteins linked with inflammation and fibrosis were used in a protein-protein interaction network analysis. The results suggest that TC0668 may play a pivotal role in C. muridarum-induced genital pathology by inducing inflammatory responses and fibrosis, which may involve the activation of the PI3K/Akt and NF-κB signaling pathways. © Copyright © 2019 Wang, Arthur, Liu, Li, Xiang, Maxwell, Li and Zhou.

摘要:
Wnt/β-catenin信号通路是一条高度保守的细胞内信号转导通路,具有介导细胞黏附、调控增殖、凋亡、参与炎症反应等多种生物学功能,在多种细菌的感染和致病过程中发挥重要作用。研究Wnt/β-catenin信号通路激活的分子机制及其在疾病发生、发展中的作用,不但可以了解细菌的致病机制,还为其治疗提供了新的靶点和策略。现将该信号通路在衣原体、结核分枝杆菌、铜绿假单胞菌和幽门螺杆菌致病中的作用作一概述。

摘要:
目的 探讨沙眼衣原体pORF5质粒蛋白对细胞凋亡的影响,以及与MDM2-p53通路的关系,为阐明沙眼衣原体致病机制提供实验依据. 方法 用不同浓度的pORF5质粒蛋白以不同时间的刺激TNF-α预处理的HeLa细胞,采用Hoechst 33342染色及流式细胞术检测细胞凋亡情况;采用Western blot检测p53的表达水平以及鼠双微体2(MDM2)的磷酸化水平;采用间接免疫荧光法分析MDM2在细胞内的定位;分别经MDM2抑制剂Nutlin3a预处理Hela细胞1h,pORF5质粒蛋白刺激HeLa细胞24 h,采用流式细胞仪测定细胞凋亡率,分析凋亡相关蛋白Bax和Bel-2的表达水平及MDM2磷酸化水平. 结果 pORF5质粒蛋白能使TNF-α诱导的细胞凋亡率降低29%;p53蛋白的表达与pORF5浓度呈现出一定的时间和浓度依赖性,当pORF5浓度达10 μg/mL时,p53的表达水平随pORF5浓度的升高而显著降低(P<0.05);pORF5刺激Hela细胞8h后p53表达开始下调,并且随时间的延长,p53的蛋白表达显著减少(P<0.05);pORF5质粒蛋白刺激HeLa细胞15 min后,MDM2开始发生磷酸化反应,30 min达到峰值;间接免疫荧光试验检测显示pORF5质粒蛋白可促进MDM2发生核转位;MDM2抑制剂Nutlin3a能上调Bax蛋白的表达,下调Bcl-2的表达,并能促进细胞凋亡,Nutlin3a处理组细胞凋亡率较对照组增加约11%(P<0.05). 结论 pORF5质粒蛋白通过激活MDM2-p53通路促进Bcl-2蛋白的表达和降低Bax蛋白的表达抑制细胞凋亡.

衣原体感染常表现为持续性感染,能够引起人和动物的多种慢性疾病。衣原体成功逃逸宿主免疫反应是其维持持续感染的前提,目前已知衣原体可通过逃避宿主细胞溶酶体融合、调控宿主细胞免疫反应、干扰宿主细胞凋亡与自噬活动以及诱导宿主细胞蛋白质组改变等方式逃逸宿主免疫反应。现就衣原体逃逸宿主免疫反应的相关研究作一综述。

摘要:
Chlamydia trachomatis, the most common human pathogen that causes trachoma and sexually transmitted disease, has developed various strategies for inhibiting host cell apoptosis. Activation of the PI3K (phosphoinositide 3-kinase)/AKT-mediated MDM2 (murine double minute 2)-p53 pathway plays a prominent role in the apoptosis resistance arising from C. trachomatis infection. However, the precise upstream mechanisms by which C. trachomatis activates this pathway have not been adequately investigated. Here, we reveal that the secreted C. trachomatis plasmid-encoded protein Pgp3 inhibits apoptosis in HeLa cells. This process requires the activation of the PI3K/AKT signaling pathway, thereby leading to phosphorylation and nuclear entry of MDM2, and p53 degradation. PI3 K inhibitor LY294002 and MDM2 inhibitor Nutlin-3a block Pgp3-induced inhibition of HeLa cell apoptosis, suggesting a critical role for the PI3K/AKT pathway and its effect on the MDM2-p53 axis in Pgp3 anti-apoptotic activity.

目的:探讨沙眼衣原体(Chlamydia trachomatis,Ct) pORF5蛋白对细胞凋亡的影响,并进一步分析其分子机制,为阐明Ct致病机制提供实验依据。方法: pGEX-6p /pORF5重组质粒转化XL1-blue大肠杆菌,IPTG诱导表达GST-pORF5融合蛋白,融合蛋白经谷胱甘肽琼脂糖凝胶4B纯化及蛋白酶切除GST标签后得到pORF5蛋白。用不同浓度的pORF5蛋白刺激HeLa细胞,Western blot检测不同时间Bax和Bcl-2的表达水平以及PI3K/Akt磷酸化水平,Hoechst 33342及流式细胞技术分析细胞凋亡情况;HeLa细胞经PI3K/Akt特异性抑制剂LY294002预处理1 h后,再用pORF5蛋白刺激24 h,测定细胞凋亡率,并进一步分析凋亡相关蛋白Bax和Bcl-2的表达水平及PI3K/Akt磷酸化水平。结果:凋亡相关蛋白Bax和Bcl-2的表达变化与pORF5蛋白浓度呈现一定的浓度和时间依赖性,当pORF5蛋白浓度达10 μg /ml时,Bax表达下调,Bcl-2的表达上调,当升高至15 μg /ml时,Bax和Bcl-2的表达量变化最明显;15 μg /ml的pORF5蛋白刺激HeLa细胞24 h,Bax和Bcl-2表达变化最明显;流式细胞检测结果显示:pORF5蛋白刺激组较TNF-α处理组和未处理组细胞凋亡率分别降低了27.3%(P<0.01)和8.4%(P<0.05);Akt在pORF5蛋白刺激15 min后发生磷酸化,30 min后磷酸化水平达到峰值,用PI3K抑制剂LY294002预处理Hela细胞后,发现Akt的磷酸化显著减少,Bax蛋白表达明显上调,Bcl-2表达明显下调;LY294002处理组细胞凋亡率相比于对照组增加了13.0 %(P<0.01)。结论: pORF5蛋白通过激活PI3K/Akt信号通路调节Bcl-2和Bax蛋白的表达抑制细胞凋亡。

目的研究沙眼衣原体(Chlamydia trachomatis,Ct)pORF5质粒蛋白诱导THP-1细胞产生IL-1β的分子机制,为阐明Ct致病机制提供实验依据。方法将原核表达重组体pGEX-6p/pORF5转化大肠埃希菌,表达并纯化GST-pORF5融合蛋白,融合蛋白经蛋白酶酶切后制备不含GST标签的pORF5蛋白;用不同浓度的pORF5蛋白体外刺激THP-1细胞,ELISA测定不同时间IL-1β水平;分别用NALP3 siRNA、ASC siRNA、Caspase-1抑制剂和p38抑制剂预处理THP-1细胞,再用pORF5蛋白刺激THP-1细胞24h,ELISA测定IL-1β含量,Real-time PCR测定IL-1β和NALP3炎性体mRNA的表达,Western blot分析Caspase-1的表达及p38磷酸化水平。结果pORF5蛋白以剂量和时间依赖的方式刺激THP-1细胞产生IL-1β,24μg/ml pORF5蛋白刺激24h时IL-1β的表达水平达到峰值(495.1±55.5pg/ml);pORF5蛋白能促进THP-1细胞中NALP3炎性体mRNA的表达;NALP3-siRNA、ASC-siRNA及Caspase-1抑制剂预处理THP-1细胞后,IL-1β分泌量分别降低37.7%、71.3%和40.1%;p38抑制剂能降低NALP3炎性体mRNA及IL-1β分泌量(P<0.01),但抑制NALP3炎性体后p38磷酸化水平未受影响(P>0.05)。结论pORF5质粒蛋白通过激活p38MAPK和NALP3信号通路共同调控IL-1β的产生和分泌。

“互联网+”的便捷性、开放性和时效性的特性,以及虚拟现实技术交互性、沉浸性、构想性等优势,促进了两者与医学教育的深度融合。卓越医生教育是国家为培养高水平医学拔尖创新人才所实施的重大医学教育改革。文章分析了高校卓越医生培养教学存在的问题后,提出了加强虚拟仿真实验中心建设,完善“互联网+”虚拟技术平台,加强师资队伍建设和优化互联网教育资源是提高卓越医生教育教学质量和效果的重要举措,为其他医学高等院校卓越医生培养计划的实施提供参考。

问题中心式学习法(problem-based learning,PBL)是现代医学教育中不同于传统教学模式的一种全新的教学模式,该教学模式以其独特的优越性在国外医学教育改革中取得了重要成效。文章介绍了PBL教学模式在人体寄生虫学理论教学中的运用,研究并探讨在人体寄生虫学教学中推行PBL教学方法的可能性。
