挥发曲线法测定鹅不食草挥发油的研究

摘要:
建立了鹅不食草挥发油含量测定新方法,并对水蒸汽蒸馏法一些提取条件(如粉末粗细,萃取溶剂,浸泡等)进行优化,以确定最佳提取方案。具体方法是:利用水蒸汽蒸馏法提取鹅不食草中挥发油,采用挥发曲线法测定挥发油含量。结果表明:水蒸汽蒸馏法的提取率最佳工艺是粉末先浸泡1~2 h,再进行水蒸气蒸馏法,用乙醚萃取为佳。挥发曲线法RSD为0.959%。挥发曲线法测定挥发油含量是一种成本低廉,操作简便,准确的测定方法。

摘要:
目的 比较原代宁乡花猪与其第一代仔猪血液生理生化指标的变化.方法 常规方法测定宁乡花猪F0代16头,F1代16头血液18项生理指标和13项生化指标.结果 F0代与F1代部分血液生理指标如白细胞(WBC)、红细胞(RBC)、血红蛋白(HGB)、红细胞积压(HCT)等13项有显著性差异(P<0.05);部分血液生化指标如总胆红素(TBIL),尿素氮(BUN),总蛋白(TP),白/球蛋白(ALB/GLB),葡萄糖(GLU),有显著性差异(P<0.05);其他指标均无统计学差异.结论 2代宁乡花猪之间部分血液生理生化指标有显著性差异.得到的数据可以为实验动物化研究提供相应的基础生理参考数据.

摘要:
实验鱼类作为模式生物已经被广泛地应用于药学研究,其中犹以斑马鱼为经典。斑马鱼因其独特的优点,被建立人类重大疾病模型和药物筛选平台,取得了许多有价值的研究成果。另外,青鳉、虹鳟、剑尾鱼、稀有鮈鲫、红鲫、金鱼、鲤鱼等实验鱼类也具有在药物开发与应用研究的优势。本文综述了近年来国内外实验鱼类在药物毒理学与安全性评价、环境化合物安全性评价、药物筛选与新药发现、药物药理学研究、药物代谢研究、药物制剂学研究、中药研究、再生药物研究等药学研究的一些事例,为药学研究中实验动物的选择积累有价值的资料,并为实验鱼类应用研究提供新的启示。

摘要:
ATP-binding cassette transporter A1 (ABCA1) is critical in exporting cholesterol from macrophages and plays a protective role in the development of atherosclerosis. The purpose of this study was to investigate the effects of betulinic acid (BA), a pentacyclic triterpenoid, on ABCA1 expression and cholesterol efflux, and to further determine the underlying mechanism. BA promoted ABCA1 expression and cholesterol efflux, decreased cellular cholesterol and cholesterol ester content in LPS-treated macrophages. Furthermore, we found that BA promoted ABCA1 expression via down-regulation of miR-33s. The inhibition of LPS-induced NF-kappa B activation further decreased miR-33s expression and enhanced ABCA1 expression and cholesterol efflux when compared with BA only treatment. In addition, BA suppressed I kappa B phosphorylation, p65 phosphorylation and nuclear translocation, and the transcription of NF-kappa B-dependent related gene. Moreover, BA reduced atherosclerotic lesion size, miR-33s levels and NF-kappa B activation, and promoted ABCA1 expression in apoE(-/-) mice. Taken together, these results reveal a novel mechanism for the BA-mediated ABCA1 expression, which may provide new insights for developing strategies for modulating vascular inflammation and atherosclerosis.

目的:探讨降钙素基因相关肽受体组分蛋白(RCP)在静压力诱导血管平滑肌细胞(VSMC)增殖及跨膜信号转导中的作用.方法:以鼠源性A10血管平滑肌细胞(A10VSMC)作为研究对象,用不同静态压力(0,80,100,120,140,160,180 mmHg)在不同时间(0,3,6,12,24 h)处理血管平滑肌细胞.噻唑蓝比色法(MTT)检测细胞的活性;Western Blot 检测细胞增殖核原(PCNA)和RCP的蛋白表达;RT-PCR检测RCP mRNA表达水平.分别用小RNA干扰技术(siRNA)或CGRP沉默或诱导RCP基因的表达,免疫共沉淀(Co-IP)和Western Blot检测Caveolin-1/RCP蛋白是否存在相互作用.

Objectives: The protective response against Treponema pallidum (Tp) infection of a DNA vaccine enhanced by an adjuvant CpG ODN was investigated. Results: The mucosal adjuvant CpG ODN enhanced the production of higher levels of anti-TpGpd antibodies induced by pcD/Gpd-IL-2 in rabbits. It also resulted in higher levels of secretion of IL-2 and IFN-α, and facilitated T cell proliferation and differentiation (p < 0.05). No significant difference about testing index above-mentioned was found in the intranasal immunization group of pcD/Gpd-IL-2 vaccine adjuvanted by CpG ODN when compared with the immunization by pcD/ Gpd-IL-2 vaccine intramuscular injection alone (p > 0.05). Furthermore, CpG ODN stimulated the production of mucosaspecific anti-sIgA antibodies and resulted in the lowest Tp-positive rate (6.7%) for Tp-infection of skin lesions and the lowest rates (8.3%) of ulceration lesions, thus achieving better protective effects. Methods: New Zealand rabbits were immunized with the eukaryotic vector encoding recombinant pcD/Gpd-IL-2 using intramuscular multi-injection or together with mucosal enhancement via a nasal route. The effect of the mucosal adjuvant CpG ODN was examined. Conclusions: The CpG ODN adjuvant significantly enhances the humoral and cellular immune effects of the immunization by pcD/Gpd-IL-2 with mucosal enhancement via nasal route. It also stimulates strong mucosal immune effects, thus initiating more efficient immune-protective effects. © 2013 Landes Bioscience.

Soybean is one of most important dicotyledonous food crops and is widely planted in Hunan Province, China. However, mining activity causes contamination of the soil in which soybean grows. To assess the impact of mining-induced soil contamination on soybean plants, a geoaccumulation index (I<inf>geo</inf>) was used to evaluate 20 soil samples from the mining and smelting areas of southern Hunan Province. The results indicated that Zn ranged from uncontaminated to a moderately contaminated level (I<inf>geo</inf><1), Pb was at a strongly contaminated level (I<inf>geo</inf>>3), and Cd was at an extremely contaminated level (I<inf>geo</inf>>5) across the whole study area. All of the studied soybean plants were affected by heavy metal Pb and Cd contamination, and the mean concentrations in seeds were 13.9mg/kg and 2.95mg/kg, respectively. The estimated bioconcentration factor and translocation factor showed that the soybean roots had a strong Cd bioconcentration capability and the stems had a strong translocation capability in terms of Pb, Cd, and Zn, with preferential transference of metals to the soybean leaves. The bioavailable fraction in the soil was characterized by the exchangeable fraction of heavy metals. In the present study, the bioavailable fractions of Pb, Cd, and Zn were significantly positively correlated with the concentration of these metals in soybean tissues (roots, stems, leaves, husks, and seeds). ©2013 SETAC.

摘要:
Recent studies have demonstrated that natural water that has 65% of the deuterium concentration depleted, can exhibit anti-tumor properties. However, the anti-tumor effects of DDW on various nasopharyngeal carcinoma (NPC) cells have not previously been reported. In the present study, NPC cell lines and normal preosteoblast MC3T3-E1 cells were grown in RPMI1640 media containing different deuterium concentrations (50-150. ppm). The effects of DDW on the proliferation and migration of NPC and MC3T3-E1 cells were investigated using the MTT, plate colony formation, and Transwell assays, as well as Boyden chamber arrays, flow cytometry (FCM), western blot and immunofluorescence. We found that DDW was an effective inhibitor of NPC cell proliferation, plated colony formation, migration and invasion. In contrast, the growth of normal preosteoblast MC3T3-E1 cells was promoted when they were cultured in the presence of DDW. Cell cycle analysis revealed that DDW caused cell cycle arrest in the G1/S transition, reduced the number of cells in the S phase and significantly increased the population of cells in the G1 phase in NPC cells. Western blot analysis revealed that treatment with DDW significantly increased the expression of NADPH:quinone oxidoreductase-1 (NQO1), while immunofluorescence assay analysis revealed that treatment with DDW decreased the expression of PCNA and matrix metalloproteinase 9 (MMP9) in NPC cells. These results demonstrated that DDW is a novel, non-toxic adjuvant therapeutic agent that suppresses NPC cell proliferation, migration, and invasion by inducing the expression of NQO1 and causing cell cycle arrest, as well as decreasing PCNA and MMP9 expression. © 2013 Elsevier Masson SAS.

This study was done to explore the role of microRNA-98 (miR-98) in cisplatin sensitization in human lung adenocarcinoma cell line. Differential expressions of miRNAs were analysed between cisplatin-resistant human lung adenocarcinoma cell line A549/DDP and its parental cell A549 by miRNAs microarray, of which 14 miRNAs were showed to be significantly (> 2-fold) up-regulated and 8 miRNAs had marked down-regulation (< 0.5-fold) in A549/DDP cells compared with in A549 cells. MiR-98, a member in the let-7 family, acts as a negative regulator in the expression of HMGA2 (high mobility group A2) oncogene, and it has been shown to have a nearly 3-fold decrease in A549/DDP cells. We found that elevated expression of miR-98 led to a higher sensitivity of A549/DDP cells to cisplatin, and the protein level of HMGA2, was clearly up-regulated in both A549/DDP and A549 cells by miR-98. Moreover, both Bcl-XL and Bcl-2, were down-regulated in the Pre-miR-98(TM) transfectants cells. We for the first time demonstrated that the expression of miR-98 increases cells spontaneous apoptosis and sensitizes cells to cisplatin at least in part via HMGA2 up-regulation. Our findings provided insight into some specific miRNAs in lung cancer as potential therapeutic targets.

摘要:
Despite the established efficacy of statin therapy, the risk of cardiovascular events remains high in many patients. We examined high-density lipoprotein (HDL) subclass distribution profiles among statin-treated coronary heart disease (CHD) patients undergoing percutaneous coronary intervention (PCI). Plasma HDL subclasses were measured in 85 patients with established CHD and quantified by two-dimensional gel electrophoresis and immunoblotting. In CHD patients with statin treatment, the mean value of total cholesterol (TC) reached the desirable level and the triacylglycerol level (TAG) was borderline high. Moreover, low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), apolipoproteinA-I, and apolipoproteinB-100 levels in these patients resembled those in normolipidemic healthy subjects. The HDL subclass did not show a normal distribution and was characterized by the lower large-sized HDL2b contents and higher contents of small-sized preβ1-HDL in CHD patients, compared to those in normolipidemic control subjects. Multiple stepwise regression analysis revealed that the severity of coronary stenosis, determined by the Gensini Score, was significantly and independently predicted by HDL2b and HDL3b. Statin therapy was effective in modifying plasma lipids levels, but not adequate as a monotherapy to normalize the HDL subclass distribution phenotype of patients with CHD undergoing PCI. The HDL subclass distribution may aid in risk stratification, especially in patients with CHD and therapeutic LDL-C and HDL-C levels.

TNF-α and IFN-γ are the major pro-inflammatory cytokines in the β-cell destruction. However, the underlying mechanism remains unclear. The present study used a murine insulinoma cell line MIN6 for further investigation of the effect of Caspase-3 on the cytokines-induced pancreatic β-cell apoptosis and analyzed the mechanisms involved in the activation of Caspase-3. It was showed that the combination of IFN-γ and TNF-α significantly reduced the viability of MIN6 cells and the observed cells growth inhibition was due to cell apoptosis as judged by the morphological changes under a confocal laser scanning microscopy and FACS assay of Annexin-V/7-AAD double staining. Accompanying with NF-κB activation and Bcl-2 downregulation, both the cleaved Caspase-3 and PARP, a known substrate of Caspase-3 in vivo, were observed at 24 and 12 h, respectively, after cells exposure to IFN-γ and TNF-α treatment. Pretreatment of Caspase-3 inhibitors remarkably attenuated IFN-γ- and TNF-α-induced cells apoptosis. Inhibition of NF-κB activation led to the increase in Bcl-2 expression, a significant attenuation in Caspase-3 activity, and an obvious amelioration in cells viability in IFN-γ- and TNF-α-treated MIN6 cells. Taken together, our results indicate that Caspase-3 is critical for the induction of MIN6 cells apoptosis and it's activation is further confirmed to be related to the NF-κB-mediated Bcl-2 downregulation, which may be the underlying mechanism of IFN-γ- and TNF-α-mediated MIN6 cells apoptosis. © 2013 Springer Science+Business Media New York.

Syphilis is a chronic infection caused by Treponema pallidum subsp. pallidum, and diagnosis with sensitive and specific methods is a challenging process that is important for its prevention and treatment. In the present study, we established a recombinant protein TpF1-based indirect immunoglobulin G (IgG) enzyme-linked immunosorbent assay (ELISA) and a Western blot assay for human and rabbit sera. The 20-kDa recombinant protein TpF1 was detected by Western blotting performed with sera from rabbits immunized with recombinant TpF1 and infected with the T. pallidum Nichols strain and T. pallidum clinical isolates but was not detected by Western blotting with sera from uninfected rabbits. The sensitivity of the recombinant protein was determined by screening sera from individuals with primary, secondary, latent, and congenital syphilis (n = 82). The specificity of the recombinant protein was determined by screening sera from uninfected controls (n = 30) and individuals with potentially cross-reactive infections, including Lyme disease (n = 30) and leptospirosis (n = 5). The sensitivities of TpF1-based ELISAs were 93.3%, 100%, 100%, and 100% for primary, secondary, latent, and congenital syphilis, respectively, and the specificities were all 100% for sera from uninfected controls and individuals with potentially cross-reactive infections. In Western blot assays, the sensitivities and specificities of TpF1 for human sera were all 100%. The reactivities of TpF1 with syphilitic sera were proportional to the titers of the T. pallidum particle agglutination (TPPA) assay. These data indicate that the recombinant protein TpF1 is a highly immunogenic protein in human and rabbit infections and a promising marker for the screening of syphilis. 摘要:
Depression is a common and debilitating mental illness and is often comorbid with anxiety disorders. Altered synaptic plasticity is considered to be an important mechanism underlying antidepressant drug action. It has been reported that hydrogen sulfide (H2S), the third gaseous transmitter, facilitates the induction of hippocampal long-term potentiation and augments synaptic neurotransmission, involved in the regulation of synaptic plasticity. The aim of this study was to clarify the antidepressant-like and anxiolytic-like effects of H2S. H2S (NaHS, 1.68 or 5.6 mg/kg, intraperitoneally, for 7 days) exerts a specific antidepressant-like effect in the forced swimming test of mice and rats and the tail suspension test of mice, and reduces the anxiety-like behaviors of both mice and rats in the elevated plus-maze test. These results reveal a unique antagonistic action of H2S in depressive-like and anxiety-like behaviors and suggest that elevating H2S signaling in the brain may represent a novel approach for the treatment of depressive and anxiety disorders. 摘要:
具有良好的生物相容性且生物可降解天然多糖,一直作为药物传递体系,具有如下优点:可延长药物的生物半衰期,减轻药物的毒副作用.本实验旨在制备一种果胶基载5-氟脲嘧啶(5-Fluorouracil,5-FU)纳米粒子(P-5-FU)载药系统,探讨果胶基纳米载药体系本身带有大量的半乳糖残基这种天然靶头对人肝癌细胞HepG2的靶向效果.M,ITr法测定载药果胶基纳米粒子对HepG2和A549细胞的增殖抑制作用,MTY结果显示P-5-Fu对HepG2细胞的增殖具有明显抑制作用,呈剂量依赖性,且作用较5-Fu强;而P-5-FU对A549细胞增殖亦有明显抑制作用,呈剂量依赖性,但与5-FU相比无明显差别;高效液相色谱法(HPLC)对两种细胞对载药纳米粒子的摄取情况和靶向性进行了测定.细胞摄取结果显示HepG2细胞对P-5-FU的摄取量较5-FU明显增多,而A549细胞对P-5-FU和5-FU的摄取量没有明显的差别.半乳糖饱和ASGPR结合位点后两种细胞对P-5-Fu和5-Fu的摄取量都没有明显的差别.结果表明果胶基纳米载药粒子可特异性靶向高表达的细胞.

死亡结构域相关蛋白(death domain-associated protein, DAXX)是一个多功能的核蛋白, 广泛分布于核仁、核质、染色质、早幼粒细胞白血病蛋白核体(PML-NBs)和胞浆等亚细胞区域, 对细胞的转录调控、凋亡及细胞周期等生物学活动发挥重要作用。SUMO化修饰(small ubiquitin-like modification)是一种重要的、高度保守的翻译后生物学修饰, 大量文献显示SUMO化修饰对DAXX的亚细胞定位及功能的调节具有重要意义。本文对此方面的研究进展做一综述。

目的 研究二苯乙烯苷(TSG)对过氧化氢(H2O2)诱导的人脐静脉内皮细胞凋亡的作用以及TSG是否通过调节X连锁凋亡抑制蛋白(XIAP)的表达抗内皮细胞凋亡.方法 预先用不同剂量TSG(1~ 100 μmol/L)处理人脐静脉内皮细胞24h,再用300 μmol/L H2O2作用细胞24h.采用MTT法检测细胞活力,Hoeschest染色及流式细胞仪检测细胞凋亡.RT-PCR及Western blot检测TSG、XIAP抑制剂Embelin作用细胞后XIAP的表达.结果 300 μmol/L H2O2作用HUVECs细胞24 h可有效降低细胞活力,诱导细胞凋亡;1 ~ 100 μmol/L TSG预处理24 h均可提高细胞存活率,抑制细胞凋亡;30 μmol/L Embelin预处理细胞6h后再用TSG预处理24h,细胞凋亡率较TSG预处理组明显上升,细胞活力显著降低.RT-PCR、Western blot结果显示H2O2处理后,细胞XIAP表达水平显著降低;与H2O2组相比,1~ 100 μmol/L TSG预处理24h后,XIAP表达升高;与TSG预处理组相比,Embelin预处理6h后再用TSG预处理24h,XIAP的表达降低.结论 TSG能抑制H2O2诱导的人脐静脉内皮细胞凋亡,其作用机制与上调XIAP的表达有关.

Atherosclerosis is a chronic disease characterized by the deposition of excessive cholesterol in the arterial intima. Macrophage foam cells play a critical role in the occurrence and development of atherosclerosis. The generation of these cells is associated with imbalance of cholesterol influx, esterification and efflux. CD36 and scavenger receptor class A (SR-A) are mainly responsible for uptake of lipoprotein-derived cholesterol by macrophages. Acyl coenzyme A:cholesterol acyltransferase-1 (ACAT1) and neutral cholesteryl ester hydrolase (nCEH) regulate cholesterol esterification. ATP-binding cassette transporters A1(ABCA1), ABCG1 and scavenger receptor BI (SR-BI) play crucial roles in macrophage cholesterol export. When inflow and esterification of cholesterol increase and/or its outflow decrease, the macrophages are ultimately transformed into lipid-laden foam cells, the prototypical cells in the atherosclerotic plaque. The aim of this review is to describe what is known about the mechanisms of cholesterol uptake, esterification and release in macrophages. An increased understanding of the process of macrophage foam cell formation will help to develop novel therapeutic interventions for atherosclerosis. © 2013 The Authors.
