Ferroptosis is a cell death process caused by metabolic dysfunction with the feature of aberrant iron accumulation. Emerging studies have identified that ferroptosis is an important biological function involving in the tumorigenesis, and targeting ferroptosis could provide promising therapeutic targets for lung cancer. However, such therapeutic strategies show limited therapeutic effect owing to drug resistance and other unknown underlying mechanisms. In this study, lysine-specific demethylase 1 (LSD1/KDM1A) was found to be significantly upregulated in lung cancer cells and tissues. The patients with KDM1A downregulation displayed the good prognosis. Using gene set enrichment analysis (GSEA), we demonstrated that KDM1A-associated genes might participate in the regulation of cell ferroptosis and Myc signaling in lung cancer. Knockdown of KDM1A inhibited the level of c-Myc and increased the concentration of malondialdehyde (MDA) and irons in human lung cancer cells H1299 and A549. Downregulation of c-Myc could facilitate KDM1A knockdown-mediated ferroptosis. Our study has elucidated the effect of KDM1A/c-Myc regulatory axis in the ferroptosis resistance of lung cancer cells.

Objectives: The contributing factors for spondylitis after percutaneous vertebroplasty (PVP) or percutaneous kyphoplasty (PKP) remain unclear.Here, we sought to investigate the factors affecting spondylitis occurrence after PVP/PKP. We also compared the clinical characteristics between patients with tuberculous spondylitis (TS) and nontuberculous spondylitis (NTS) following vertebral augmentation. Methods: Literature searches (from January 1, 1982 to October 16, 2020) using MEDLINE, EMBASE, Google Scholar and Web of science databases were conducted to identify eligible studies according to predefined criteria. The local database was also retrospectively reviewed to include additional TS and NTS patients at our center. Results: Thirty studies from the literature and 11 patients from our local institute were identified, yielding a total of 23 TS patients and 50 NTS patients for analysis. Compared with NTS group, patients in the TS group were more likely to have a history of trauma before PVP/PKP treatment. Univariate analyses of risk factors revealed pulmonary tuberculosis and diabetes were significant factors for TS after PVP/PKP. Analyzing NTS, we found obesity, a history of preoperative trauma, urinary tract infection, diabetes and multiple surgical segments (≥2) were significantly associated with its occurrence following PVP/PKP treatment. Multivariate logistic analyses showed a history of pulmonary tuberculosis and diabetes were independent risk factors for TS after PVP...

摘要:
The evaluation of rockburst damage potential plays a significant role in managing rockburst risk and guaranteeing the safety of personnel. However, it is still a challenging problem because of its complex mechanisms and numerous influencing factors. In this study, a bagged ensemble of Gaussian process classifiers (GPCs) is proposed to assess rockburst damage potential with an imbalanced dataset. First, a rockburst dataset including seven indicators and four levels is collected. To address classification problems with an imbalanced dataset, a novel model that integrates the under-sampling technique, Gaussian process classifier (GPC) and bagging method is constructed. Afterwards, the comprehensive performance of the proposed model is evaluated using the values of accuracy, precision, recall, and F-1. Finally, the methodology is applied to assess rockburst damage potential in the Perseverance nickel mine. Results show that the performance of the proposed bagged ensemble of GPCs is acceptable, and the integration of data preprocessing, under-sampling technique, GPC, and bagging method can improve the model performance. The proposed methodology can provide an effective reference for the risk management of rockburst.

Arecoline, a major alkaloid of the areca nut, has potential toxicity to the nervous system. Our previous study reveals that the neurotoxicity of arecoline involves in inhibited endogenous hydrogen sulfide (H2S) generation. Therefore, the present study investigated whether exogenous H2S protects against arecoline-induced neurotoxicity and further explore the underlying mechanisms focusing on leptin/leptin receptor signaling pathway. The cell viability was measured by CCK-8 kit. The apoptosis were detected by Hoechst 33258 and Annexin V/PI (propidium iodide) staining. The protein expressions were determined by Western blot analysis. Our results demonstrated that NaHS, an exogenous H2S donor, significantly increases the cell viability, decreases apoptosis ratio, and reduces caspase-3 activity as well as Bax/Bcl-2 ratio in PC12 cells exposed to arecoline, indicating the protection of H2S against arecoline-induced cytotoxicity and apoptosis. Also, NaHS attenuated arecoline-induced endoplasmic reticulum (ER) stress, as evidenced by the decreases in the expressions of glucose-regulated protein 78 (GRP78), C/EBP homologous protein (CHOP), and Cleaved caspase-12. Meanwhile, NaHS promoted leptin/leptin receptor signaling pathway in arecoline-exposed PC12 cells, as illustrated by upregulations of leptin and leptin receptor expressions. Furthermore, leptin tA, an antagonist of leptin receptor, obviously abolished the inhibitory effects of NaHS on arecoline-induced cytotoxicity, apoptosis, and ER stress in arecoline-exposed PC12 cells. Taken together, these results suggested that H2S prevents arecoline-induced neurotoxicity via enhancing the leptin/leptin receptor signaling pathway.

Corynebacterium accounts for around 20% of the armpit microbiome and plays an essential role in axillary osmidrosis (AO). In this study, the effects of Lactobacillus bulgaricus treatment on the microecological environment of armpits and its efficacy in the treatment of AO were investigated. A total of 10 AO patients were enrolled in this study. The patients were treated with L. bulgaricus mixed with saline on the left armpit (experimental group) and pure saline on the right armpit (control group) for 28 days. After treatment, AO severity showed a significant decrease (p = 0.013) in the experimental group compared with the control group, and the Corynebacterium abundance also showed a corresponding significant decrease (p < 0.01). Moreover, no significant variation in Staphylococcus abundance was found between these two groups. The microbe diversity is not disturbed in the treatment. Accordingly, our study demonstrates that L. bulgaricus can serve as an effective probiotic microbe for AO treatment by reducing the abundance of Corynebacterium and rebalancing the microecological environment.

In this paper, to reveal the flow characteristics of broken mineral-rock of block caving, we present the Attribute Stochastic Medium Theory that combines an attribute block model with stochastic medium theory. An attribute block model was established to fit discrete target regions. A void transfer model and data structure were established based on stochastic medium theory. Our key contribution is that we propose Attribute Stochastic Medium Theory and present the concept of draw-out index and block fragmentation index. It can be used to analyze the flow characteristics of rock block under different drawing heights and different fragmentation conditions. We implement the flow algorithm and simulation with C++ programming. The results show that mineral elemental grade information of the ore-drawing body was obtained, and when the block fragmentation index remains unchanged, with the increase of the draw-out index, the ore-drawing ellipsoid develops gradually, the eccentricity increases gradually, and the average depth of the depression increases gradually. In the isolated extraction zone (IEZ), the length of the long half-axis of the IEZ increases linearly with the increase of the ore-drawing height. But the length of the short half-axis and the eccentricity of the IEZ increase nonlinearly with the increase of the ore-drawing height, showing a general power exponential relationship. Under the condition of constant draw-out index, with an increase of the block index, the eccentricity of the ellipsoid decreases. This method could obtain the properties and flow characteristics of caving ore and rock. Combined with in situ test of ore drawing. This method could provide guidance for the design of ore drawing bottom structure and ore drawing plan.

Dendrobium nobile Lindl polysaccharides (DNLP) exhibited various biological functions. This study aimed to investigate the protective effects of DNLP on testicular spermatogenic function in streptozotocin (STZ)-induced diabetic rats in comparison with metformin. The blood glucose level was significantly increased and the homeostatic model assessment for insulin resistance (HOMA-IR) aggravated markedly in diabetic rats. The weight of testis and epididymis, and the sperm number and motility were decreased in the diabetic rats. The pathologic changes occurred in the spermatogenic tubules along with the decreased number of spermatogenic cells, downregulated proliferating cell nuclear antigen (PCNA) and Sirtuin 1 (SIRT1) expression and increased cell apoptosis in the testes. Compared with the model group, DNLP and metformin treatment significantly decreased the level of blood glucose, improved the HOMA-IR, and increased the weight of testis and epididymis, as well as the sperm number and sperm motility. Furthermore, the pathologic changes in the spermatogenic tubules improved significantly with increased number of spermatogenic cells, the upregulation of PCNA and SIRT1 and suppression of cell apoptosis in the testes. Collectively, our study for the first time examined the effects of DNLP on the male reproductive system of STZ-induced diabetic rats, and indicated that DNLP was protective against diabetes mellitus-induced testis injury via increasing the proliferation, inhibiting cell apoptosis and upregulating SIRT1 expression in testicular spermatogenic cells.

Traditional composite cathode was usually mixed with cathode materials and BaZr0.1Ce0.7Y0.2O3-delta (BZCY) to obtain thermal and chemical compatibility with proton-conducting electrolyte. Considering the poor chemical stability of BZCY in wet atmosphere, three different composite cathodes of La0.6Sr0.4Co0.2Fe0.8O3+delta (LSCF)-BZCY, LSCF-Sm0.2Ce0.8O2-delta (SCO), LSCF-La2Ce2O7+delta (LCO) were fabricated and systematically researched. Due to the high chemical stability and triple conductivity, the single cell with cathode of LSCF-LCO presented a better cell performance of 1020 mW.cm(-2) at 700 degrees C and long-term operation durability. In addition, the analysis of distribution of relaxation time method (DRT) and impedance spectrum revealed that the migration rates of proton and oxygen ion were enhanced through adjusting electrolyte material, showing great potential for future application of new cathode materials for proton-conducting solid oxide fuel cells (H-SOFCs).

The development of eco-friendly and efficient alternative binder to conventional Portland cement is a challenging issue that deserves to be paid more attention. The mineral additive-modified magnesium potassium phosphate cement mixture was introduced in sediment solidification recycled as alternative roadbed material. The feasibility of mineral additive-MKPC blend in solidifying sediment and its mechanical performance were probed into deeply through unconfined compressive strength, durability, microstructural and mineralogical tests. The results demonstrated that it existed an optimal mineral additive content where the peak strength was achieved, and an excessive amount caused the strength degradation. The unconfined compressive strength of solidified sediment increased with curing age and exhibited a growing potential. Struvite-k crystals were identified as the major cementing phase in mineral additive-MKPC solidified sediment. C-S-H gels and Ca2P2O7 c 2H2O were recognized as secondary phases for fly ash-MKPC blend and MgSiO3 for silica fume-MKPC blend. As the molar ratio of M/P and curing age grew, the total porosity was reduced due to the pore filling of mineral grains and densification of hydrated phases. The strength of solidified sediment after durability tests showed a tendency of first decline and then increase with durability period. Overall, the superior benefits including excellent strength performance, strong resistance to external environmental damage and highly densified microstructure could be expected for mineral additive-MKPC blend in sediment solidification as a promising roadbed filling.

Unnatural α-amino acids are important synthetic targets in the field of peptide science. Herein we report an efficient, versatile, and straightforward strategy for the synthesis of homophenylalanine derivatives via the nickel-catalyzed Csp3–Csp3 cross-coupling of (fluoro)benzyl bromides/chlorides with natural α-amino-acid-derived alkylzinc reagents. The current protocol features the advantages of a low-cost nickel catalyst system, synthetic convenience, and the tolerance of rich functionality and stereochemistry.
