Introduction: We have previously demonstrated the antagonistic role of hydrogen sulfide (H2S) in the cognitive dysfunction of streptozotocin (STZ)-induced diabetic rats. It has been confirmed that the impaired hippocampal autophagic flux has a key role in the pathogenesis of cognitive impairment and that ornithine decarboxylase (ODC)/spermidine (Spd) pathway plays an important role in the formation of memory by promoting autophagic flux. Objectives: To investigate the roles of hippocampal ODC/Spd pathway and autophagic flux in H2S-attenuated cognitive impairment in STZ-induced diabetic rats. Methods: Cognitive function is judged by the novel objective recognition task (NOR), the Y-maze, and the Morris water maze (MWM) tests. The ODC/Spd pathway in hippocampus was evaluated using the expression of ODC detected by western blot and the level of Spd assayed by GC-MS. Autophagic flux was assessed using the expressions of Beclin-1, LC3II/I, and P62 detected by western blot, and the number of autophagosomes observed by transmission electron microscope. Results: Sodium hydrosulfide (NaHS, a donor of H2S) markedly improved the autophagic flux in the hippocampus of STZ-exposed rats, as evidenced by a decrease in the number of autophagosomes as wells as downregulations in the expressions of LC3-II, Beclin-1, and P62 in the hippocampus of cotreatment with NaHS and STZ rats. NaHS also up-regulated the expression of ODC and the level of Spd in the hippocampus of STZ-induced diabetic rats. Furthermore, inhibited hippocampal ODC/Spd pathway by difluoromethylornithine (DFMO) markedly reversed the protections of NaHS against the hippocampal autophagic flux impairment as well as the cognitive dysfunction in STZ-exposed rats. Conclusion: These findings indicated that improving hippocampal autophagic flux plays a key role in H2S-attenuated cognitive impairment in STZ-induced diabetic rats, as results of up-regulating hippocampal ODC/Spd pathway. © 2020

Aldo-keto reductase family 1 member B10 (AKR1B10) is a secretory protein overexpressed in hepatocellular carcinoma (HCC). We aimed to evaluate AKR1B10 as a serum marker for detection of HCC. Herein, we conducted a cohort study that consecutively enrolled 1,244 participants from three independent hospitals, including HCC, healthy controls (HCs), benign liver tumors (BLTs), chronic hepatitis B (CHB), and liver cirrhosis (LC). Serum AKR1B10 was tested by time-resolved fluorescent assays. Data were plotted for receiver operating characteristic (ROC) curve analyses. Alpha-fetoprotein (AFP) was analyzed for comparison. An exploratory discovery cohort demonstrated that serum AKR1B10 increased in patients with HCC (1,567.3 +/- 292.6 pg/mL; n = 69) compared with HCs (85.7 +/- 10.9 pg/mL; n = 66; P < 0.0001). A training cohort of 519 participants yielded an optimal diagnostic cutoff of serum AKR1B10 at 267.9 pg/mL. When ROC curve was plotted for HCC versus all controls (HC + BLT + CHB + LC), serum AKR1B10 had diagnostic parameters of the area under the curve (AUC) 0.896, sensitivity 72.7%, and specificity 95.7%, which were better than AFP with AUC 0.816, sensitivity 65.1%, and specificity 88.9%. Impressively, AKR1B10 showed promising diagnostic potential in early-stage HCC and AFP-negative HCC. Combination of AKR1B10 with AFP increased diagnostic accuracy for HCC compared with AKR1B10 or AFP alone. A validation cohort of 522 participants confirmed these findings. An independent cohort of 68 patients with HCC who were followed up showed that serum AKR1B10 dramatically decreased 1 day after operation and was nearly back to normal 3 days after operation. Conclusion: AKR1B10 is a potent serum marker for detection of HCC and early-stage HCC, with better diagnostic performance than AFP.

目的研究正常成人上颌第一磨牙种植相关数据,为临床牙种植术等口腔外科手术及口腔解剖的教学提供参考依据。方法根据纳入标准筛选32例新鲜成人上颌骨,测量上颌第一磨牙各根根尖到上颌窦底的距离,牙槽嵴顶到上颌窦底的距离,根分叉到上颌窦底的距离,近中颊根颊面、远中颊根颊面、腭根腭面根颈、根中和根尖牙槽骨的厚度。结果上颌第一磨牙牙槽嵴顶到上颌窦底的距离为(12.57±3.19)mm,根分叉到上颌窦底的距离为(9.18±0.79)mm;上颌第一磨牙远中颊根根尖到上颌窦底的距离最远,为(3.11±0.71)mm,腭根次之,近中颊根最近,为(2.27±0.43)mm。各根牙槽骨的厚度从根颈到根中,再到根尖逐渐增厚,其中以腭根表面牙槽骨厚度最厚,近中颊根次之,远中颊根最薄。结论上颌第一磨牙缺失后行即刻种植是可行的,种植钉可向腭侧牙槽骨方向倾斜一定的角度。

<jats:title>Abstract</jats:title><jats:sec><jats:title>Objective</jats:title><jats:p>β‐catenin is one of the most critical oncogenes associated with many kinds of human cancers, especially in the human CRC. Innate immunity recognizes tumour derived damage‐associated molecular patterns (DAMPs) and primes the anti‐tumour adaptive responses. While the function of β‐catenin in CRC tumourigenesis is well established, its impact on innate immune evasion is largely unknown. The aim of this study is to characterize the role of β‐catenin in inhibiting RIG‐I‐like receptor (RLR)‐mediated IFN‐β signalling in colorectal cancer.</jats:p></jats:sec><jats:sec><jats:title>Materials and Methods</jats:title><jats:p>Immunohistochemical staining and western blotting were conducted to study the expression of β‐catenin, IRF3 and phospho‐IRF3 (p‐IRF3) in CRC samples and cell lines. Plaque assay determining virus replication was performed to assess the regulation of β‐catenin on IFN‐β signalling. The inhibition of β‐catenin on RLR‐mediated IFN‐β signalling was further studied by real‐time analyses and reporter assays in the context of lentiviral‐mediated β‐catenin stably knocking down. Lastly, co‐immunoprecipitation and nuclear fractionation assay were conducted to monitor the interaction between β‐catenin and IRF3.</jats:p></jats:sec><jats:sec><jats:title>Results</jats:title><jats:p>We found that high expression of β‐catenin positively correlated with the expression of IRF3 in CRC cells. Overexpression of β‐catenin increased the viral replication. Conversely knocking down of β‐catenin inhibited viral replication. Furthermore, our data demonstrated that β‐catenin could inhibit the expression of IFN‐β and interferon‐stimulated gene 56 (ISG56). Mechanistically, we found that β‐catenin interacted with IRF3 and blocked its nuclear translocation.</jats:p></jats:sec><jats:sec><jats:title>Conclusion</jats:title><jats:p>Our study reveals an unprecedented role of β‐catenin in enabling innate immune evasion in CRC.</jats:p></jats:sec>

摘要:
目的:探讨丰富环境(EE)对弱视小鼠初级视皮质和外侧膝状体Sortilin的表达影响。方法:选择36只21 d昆明小鼠,雌雄不限,随机分为正常组(Control) 、单眼剥夺+标准环境组(MD + SE)和单眼剥夺+丰富环境组(MD + EE),通过行为学检测各组小鼠视功能改变、免疫组织化学方法检测各组小鼠视皮质和外侧膝状体Sortilin的表达。结果:行为学结果显示:与Control组相比,MD + SE组探爪触地成功率明显降低(P < 0.05),MD + EE组探爪触地成功率显著高于MD + SE组(P < 0.05);免疫组织化学结果显示:与Control组相比,MD + SE组外侧膝状体背内侧Sortilin阳性神经元数目显著升高(P < 0.001); MD + EE组显著低于MD + SE组,但高于Control组(P < 0.001);与Control组相比,MD + SE组小鼠视皮质Sortilin阳性神经元数目显著降低(P < 0.001); MD + EE组显著高于MD + SE组(P < 0.001),但显著低于Control组(P < 0.001) 。结论:丰富环境改善弱视小鼠视功能可能通过调节视皮质和外侧膝状体Sortilin的表达实现。

Treponema pallidum subsp. pallidum membrane proteins are considered as potent inducers in the initiation and development of inflammation. In the present study, the mechanism that leads to the production of interleukin 6 (IL-6), one of the key proinflammatory cytokines, by human monocytic THP-1 cells when these cells are treated with T. pallidum flagellin FlaA2 was investigated. Stimulation with flagellin FlaA2 can induce IL-6 expression in human monocytes and augment the phosphorylation of ERK, p38, and NF-κB, but has no effect on the phosphorylation of JNK. Likewise, FlaA2-induced IL-6 production was found to be attenuated by inhibitors for ERK, p38, and NF-κB, but not by JNK inhibitor. Immunofluorescence analysis showed that flagellin FlaA2 could stimulate the translocation of IκBα from the cytosol to the nucleus, and this phenomenon could be inhibited by the specific inhibitor BAY11-7082. FlaA2–induced IL-6 expression was also proved to be abrogated by transfection with dominant negative (DN) plasmid of MyD88. We further demonstrated that transfection with DN-TLR2 was sufficient to attenuate IL-6 expression and the phosphorylation of ERK, p38, and IκBα. These results suggest that flagellin FlaA2 induces IL-6 production via signaling pathways involving TLR2, MyD88, ERK, p38, and NF-κB in monocytes, which could contribute to the pathogenesis of T. pallidum. © 2016 Elsevier Ltd

神经干细胞移植对颈上交感神经节损毁兔MAP-2、GFAP表达的影响

摘要:
目的:研究神经干细胞(NSCs)移植对颈上交感神经节(SCSG)损毁兔胶质纤维酸性蛋白(GFAP)以及微管相关蛋白2(MAP2)表达的影响。方法32只新西兰兔中8只为正常组不予处理,24只通过持针钳持续压榨兔SCSG 10 s的方法构建SCSG损毁模型,均分成模型组、培养液组和干细胞组,造模后第7天分别给予等量的生理盐水、培养液及2×106个神经干细胞,移植后第1、7、10、14、21天分别记录各组兔损伤侧瞳孔直径变化,第21天取损伤侧SCSG进行HE染色,并进行免疫组化检测MAP ̄2和GFAP的表达。结果瞳孔直径变化情况,第10天、14天、21天干细胞组瞳孔直径均大于模型组和培养液组,差异有显著性( P<0.05);HE染色,干细胞组较模型组和培养液组细胞排列整齐,大小形态较一致,细胞间质增生少;免疫组化结果显示,干细胞可增加MAP ̄2的表达,降低GFAP的表达。结论神经干细胞通过促进兔损毁侧SCSG内MAP ̄2的表达,同时抑制GFAP的表达来发挥神经功能的修复作用。

摘要:
目的 探讨视神经横断后Mü ller细胞在改善视网膜节细胞轴突再生微环境及其对光信号传递突触的保护作用.方法 实验建立视神经横断模型,随机分成正常昼夜循环组及黑暗饲养组,用免疫组织化学及Western blot法检测视神经横断后,SD大鼠在正常昼夜循环条件及黑暗条件下饲养时视网膜谷氨酰胺合成酶随饲养时间延长的表达变化;Nissl染色法观察相应视网膜节细胞的形态结构改变.结果 正常昼夜循环饲养的大鼠视网膜谷氨酰胺合成酶的表达于视神经横断后持续减少,5d时达最低值;暗饲养的大鼠视网膜谷氨酰胺合成酶的表达于视神经横断后3d时最低,5~7 d时持续增加.Nissl染色显示暗饲养组大鼠视网膜节细胞14d时存活较多.结论 正常昼夜循环条件视神经横断后Mü ller细胞谷氨酰胺合成酶的表达呈应激性反应,暗饲养能在突触溃变关键期维持视网膜内谷氨酰胺合成酶的表达强度.
