摘要:
Metalloproteins and metalloenzymes play important roles in biological systems by using the limited metal ions, complexes, and clusters that are associated with the protein matrix. The design of artificial metalloproteins and metalloenzymes not only reveals the structure and function relationship of natural proteins, but also enables the synthesis of artificial proteins and enzymes with improved properties and functions. Acknowledging the progress in rational design from single to multiple active sites, this review focuses on recent achievements in the design of artificial metalloproteins and metalloenzymes with metal clusters, including zinc clusters, cadmium clusters, iron–sulfur clusters, and copper–sulfur clusters, as well as noble metal clusters and others. These metal clusters were designed in both native and de novo protein scaffolds for structural roles, electron transfer, or catalysis. Some synthetic metal clusters as functional models of native enzymes are also discussed. These achievements provide valuable insights for deep understanding of the natural proteins and enzymes, and practical clues for the further design of artificial enzymes with functions comparable or even beyond those of natural counterparts. © 2019 by the author.

摘要:
Hyperglycemia, a key characteristic and risk factor for diabetes mellitus (DM), causes neuronal senescence. Hydrogen sulfide (H2S) is a novel neuroprotectant. The present work was to investigate the potential effect of H2S on hyperglycemia-induced neuronal senescence and the underlying mechanisms. We found that NaHS, a donor of H2S, inhibited high glucose (HG)-induced cellular senescence in HT22 cells (an immortalized mouse hippocampal cell line), as evidenced by a decrease in the number of senescence associated-β-galactosidase (SA-β-gal) positive cells, increase in the growth of cells, and down-regulations of senescence mark proteins, p16INK4a and p21CIP1. NaHS improved the autophagic flux, which is judged by a decrease in the amount of intracellular autophagosome as well as up-regulations of LC3II/I and P62 in HG-exposed HT22 cells. Furthermore, blocked autophagic flux by chloroquine (CQ) significantly abolished NaHS-exerted improvement in the autophagic flux and suppression in the cellular senescence of GH-exposed HT22 cells, which indicated that H2S antagonizes HG-induced neuronal senescence by promoting autophagic flux. We also found that NaHS up-regulated the expression of silent mating type information regulation 2 homolog 1 (SIRT1), an important anti-aging protein, in HG-exposed HT22 cells. Furthermore, inhibition of SIRT1 by sirtinol reversed the protection of H2S against HG-induced autophagic flux blockade and cellular senescence in HT22 cells. These data indicated that H2S protects HT22 cells against HG-induced neuronal senescence by improving autophagic flux via up-regulation of SIRT1, suggesting H2S as a potential treatment strategy for hyperglycemia-induced neuronal senescence and neurotoxicity. © Copyright © 2019 Wu, Chen, Wang, Tang, Huang, Kang, Li, Xie and Tang.

摘要:
Type 2 diabetes mellitus (T2DM) is a chronic degenerative endocrine and metabolic disease with high mortality and morbidity, yet lacks effective therapeutics. We recently generated a novel fusion peptide INSR-IgG4Fc, Yiminsu (YMS), to facilitate the high-affinity binding and transportation of insulin. Thus, the aim of the present study was to determine whether the novel recombinant peptide, YMS, could contribute to restoring insulin sensitivity and glycaemic control in insulin resistance models and revealing its underlying mechanism. Palmitic acid (PA)-treated LO2 cells and high fat diet (HFD)-fed mice were treated with YMS. Therapeutic effects of YMS were measured using Western blotting, ELISA, qPCR, Histology and transmission electron microscopy. We observed that YMS treatment effectively improved insulin signaling in PA-treated LO2 cells and HFD-fed mice. Notably, YMS could significantly reduce serum levels of glucose, triglycerides, fatty acids and cholesterol without affecting the serum insulin levels. Moreover, our data demonstrated that YMS could restore glucose and lipid homeostasis via facilitating insulin transportation and reactivating PI3K/Akt signaling in both PA-treated cells and liver, gastrocnemius and brown fat of HFD-fed mice. Additionally, we noticed that the therapeutic effects of YMS was similar as rosiglitazone, a well-recognized insulin sensitizer. Our findings suggested that YMS is a potentially candidate for pharmacotherapy for metabolic disorders associated with insulin resistance, particularly in T2DM.

Two-dimensional MXene Ti3C2Tx nanosheets with peroxide decoration (p-Ti3C2Tx) are synthesized by a sonication-assisted MILD etching method. The obtained MXenes can generate hydroxyl radical species and act as an initiator for free-radical polymerization of a series of acrylic monomers without the use of light illumination or co-initiators. The monomers analyzed include acrylamide, N-isopropylacrylamide (NIPAM), N,N-dimethylacrylamide, methyl methacrylate, and hydroxyethyl methacrylate. By simply mixing N-isopropylacrylamide monomers and p-Ti3C2Tx nanosheets under deoxygenated conditions, PNIPAM-based nanocomposite hydrogels are synthesized using a high concentration of the monomer. The nanocomposite hydrogels have a photothermal conversion efficiency of 34.7% and photothermal stability superior to that of pristine Ti3C2Tx. Taking advantage of the thermal responsive behavior of PNIPAM, the nanocomposite hydrogels are successfully exploited as remotely near-infrared light controlled "smart" windows, fluidic valves and photodetectors. 摘要:
A molecularly imprinted electrochemical sensor for the rapid detection of the anti-parasitic drug Secnidazole (SCZ) is reported. In this work, the build electrochemical sensor was based on a carbon paste electrode (CPE) modified with multi-wall carbon nanotubes (MWCNTs) and boron-embedded duplex molecularly imprinted composite membranes (B-DMICMs), that significantly increased the efficiency of the sensor for the detection of template molecule SCZ. Density functional theory (DFT) was employed to study the interactions between the template and monomers to select appropriate functional monomers for rational design of the B-DMICMs.The optimal experimental conditions were optimised for the factors affecting the performance of the sensor. Under the optimal conditions, the reduction peak currents of SCZ by differential pulse voltammetry increased linearly with SCZ concentration in the range from 3.0 x 10(-4) to 1.0 x 1.0(-6) mol L-1 and 1.0 x 1.0(-6) to 1.91 x 10(-8) mol L-1 with a detection limit of 1.72 x 10(-8) mol L-1 for secnidazole, which is significantly lower than those in the currently used methods and in previous reports. This method offers low cost, sensitive and effective determination of SCZ and can potentially be used for detection of SCZ in pharmaceutical and biological samples with good precision and accuracy.

摘要:
Two three-dimensional (3-D) chiral coordination polymers, [Cd(NDC)(DMF)] n (1 L and 1 D : H 2 NDC = 1,4-naphthalenedicarboxylic acid and DMF = N,N′-dimethylformamide), have been synthesized by introducing the rigid achiral dicarboxylic acid ligand under solvo-thermal conditions, without chiral template or additives. Both chiral Cd-CPs were characterized by single crystal X-ray crystallography, and structural analysis indicates that 1 L and 1 D are enantiomers based on the zigzag [Cd(COO) 2 (DMF)] n rod secondary building units, featuring the left- or right-hand 2 1 Cd-O-C-O-Cd helix chains and left- or right-hand 4 1 screw [Cd(NDC)] n columns, respectively. The FT-IR, TGA, powder X-ray diffraction and solid-state luminescence of 1 have been investigated. It is intriguing that the evidence from the circular dichroism spectroscopies confirms enantiomeric excess in the whole spontaneous resolution crystallization. © 2019, © 2019 Informa UK Limited, trading as Taylor & Francis Group.

Metal-doped MnO 2 has been successfully synthesized via chemical precipitation method. Interestingly, doping can increase not only ion conductivity but also intrinsic electron conductivity. By doping, the particle size of MnO 2 decreases, relatively appropriate pore size distribution is obtained, and physically and chemically bound water content increases, all of which contribute to ion conductivity. Meanwhile, doping has induced some defects in MnO 2 crystal, increasing the hole concentration and then intrinsic electron conductivity. As the result, metal-doped MnO 2 displays better rate performance than the un-doped. Especially, nickel-doped MnO 2 exhibits the best rate behavior. Even at 8 A g -1 , nickel-doped MnO 2 still has a specific capacitance as high as 234.0 F g -1 , larger than that of the un-doped obtained at 1 A g -1 (only 173.6 F g -1 ). In conclusion, doping is a useful way for improving the rate performance of MnO 2 . © 2019 The Authors.

摘要:
Poly(cyclotriphosphazene-co-4,4′-diaminodiphenyl ether) crosslinked microspheres with active amino groups on the surface were prepared by one-step precipitation polymerization method. The as-prepared material were characterized by means of FTIR, XPS, XRD, EDS and SEM. The effects of pH, time of contact, dosage, initial concentration and temperature on the adsorption of uranium(VI) by the crosslinked microspheres were studied, and the adsorption mechanism was elucidated. Under the optimized adsorption conditions, the adsorption removal efficiency reached 97.03%. The experimental results fit the Langmuir isotherm model and adsorption processes comply with the pseudo-second-order kinetic model. Thermodynamic data revealed that the adsorption behavior of uranium(VI) on the microspheres was spontaneous and exothermic.

With rapid development of nuclear industry, the overexploitation of uranium has brought a series of radioactive pollution problems to water, soil, atmosphere and ecosystem. Uranium and its compounds are highly carcinogenic and biologically toxic, posing a deadly threat to human health. Therefore, how to design a highly selective and sensitive sensor to detect uranyl ions in the environment and organisms has become an urgent task. This review summarizes relevant literature on the detection of uranyl ions by different sensors over the years. The detection methods can be roughly divided into three categories: optical sensors, electrochemical sensors and other sensors; further classification has also been carried out according to their detection mechanism. The detection limits and performances of different sensors are discussed to intuitively understand different capability for these methods in detecting uranyl ions. We also discuss the trend and future direction of uranium to provide a valuable reference for further developing better uranyl sensors. © 2019 Elsevier B.V.

摘要:
The thermal conductivity of isotropic graphite was investigated using Raman spectroscopy technique. In this method, the Raman laser acted simultaneously as an excitation source and a heating source. The dependences of the Raman peak position on temperature and laser power for the isotropic graphite were analyzed to establish Raman thermometry. Meanwhile, the local temperature rise induced by the laser irradiation was also theoretically described using a three-dimensional heating model, which can be related to the thermal conductivity. The laser absorptivity, the spot size, and the probing volume of Raman spectra were discussed, respectively. Besides, simulations of the localized temperature distribution were performed. On the basis of the Raman thermometry and the three-dimensional heating model, the thermal conductivity of 55 W/m K for the isotropic graphite was estimated, which was comparable with the value (60 W/m K) obtained by the laser flash technique.

Poly(cyclotriphosphazene-co-4,4′-diaminodiphenyl ether) microspheres (PZA) was based on hexachlorocyclotriphosphazene (HCCP) and 4,4′-diaminodiphenyl ether (ODA). It was synthesized via one-step precipitation method. The PZA microspheres were characterized by FT-IR, XPS, zeta potential and SEM/EDS. Through experimental research, PZA microspheres had the optimal adsorption performance with pH value 3.5, contact time of 60 min, thorium(IV) ions concentration of 30 mg L−1, and PZA microspheres of 10 mg. The maximum adsorption capacity of PZA microspheres for thorium(IV) was 29.04 mg g−1. The adsorption process could be described by the quasi-second order kinetic model and simulated by the Langmuir isotherm equation. © 2019, Akadémiai Kiadó, Budapest, Hungary.

摘要:
A series of chrysin amino acid derivatives were synthesized to evaluate for their antiproliferative activities against several cancer cell lines. Among the compounds tested, N-(2-((5-hydroxy-4-oxo-2-phenyl-4H-chromen-7-yl)oxy)octanoyl)-L-leucine methyl ester(5d) presented a good anti-proliferative activity in MDA-MB-231 and MCF-7 cells. Flow cytometry analysis showed that 5d induced apoptosis and prolonged cell cycle progression in MDA-MB-231 and MCF-7 cells. Western blot analysis showed that 5d significantly inhibited Akt phosphorylation (Ser473) in MDA-MB-231 and MCF-7 cells. In addition, 5d treatment markedly downregulated Bcl-2 and upregulated Bax in a dose-dependent manner. In vitro caspase activation assay showed that 5d induced apoptosis of MDA-MB-231 cells by enhancing caspase 3/7 activity. The regulatory effect of 5d on apoptosis of MDA-MB-231 and MCF-7 cells may be induced by mitochondrial apoptosis pathway. This study is of great significance for designing and developing more effective chrysin amino acid derivatives. [GRAPHICS] .

摘要:
Approaches to degradation of industrial dyes are desirable, of which bioremediation is more favorable. In addition to the use of native enzymes, rational design of artificial enzymes provides an alternative approach. Meanwhile, few designs can achieve a catalytic activity comparable to that of native enzymes. We have previously designed two generations of artificial dye-decolorizing peroxidases (DyPs) in myoglobin (Mb) by introduction of Tyr43 and Trp138 in the heme pocket; however, the activity is moderate. To improve the activity of the artificial DyP, we herein designed a third generation by introduction of an additional Trp (P88W) to the protein surface, named F43Y/F138W/P88W Mb. The third generation of artificial DyP was shown to exhibit a catalytic efficiency exceeding that of various native DyPs and comparable to that of the most efficient native DyPs. Titration of reactive blue 19 (RB19) and molecular docking studies revealed crucial roles of Trp88 in substrate binding and oxidation, which acts as a catalytic site. This study not only provides clues for heme protein design but also suggests that the artificial DyP has potential applications for bioremediation in the future. © 2019 American Chemical Society.

摘要:
A practical protocol for the preparation of quinoxaline-2,3(1H,4H)-diones through direct C(sp2)−H hydroxylation of quinoxalin-2(1H)-ones in recyclable DL-α-Tocopherol methoxypolyethylene glycol succinate solution (2 wt% in water) (TPGS-750-M/H2O) was developed. 摘要:
Using ethylenediamine-β-cyclodextrin and α-naphthylacetic acid as raw material, a novel inclusion complex (en-β[email protected]) was synthesized. The inclusion complex structures and properties were analyzed by reliable spectroscopic and physicochemical techniques. The stoichiometry of the inclusion complex was 1:1 and the stability constant (K S ) value was found to be 2511 M −1 at 293 K. Molecular thermodynamic parameters indicated that the spontaneous and exothermic nature of the process. The inclusion form was foreseen by Chem 3D program combined with 1 H-NMR experimental data. The solubilization curves (K 1 = 2.05) and in-vitro dissolution data demonstrated that the en-β[email protected] inclusion complex has satisfactory water solubility. © 2019, Springer Nature B.V.

Abstract: A novel Zn2+ fluorescent probe Nʹ-((4′-(diphenylamino)-4-hydroxy-[1,1′-biphenyl]-3-yl)methylene)-2-hydroxybenzohydrazide (TPA-HPH) with a larger Stokes shift (210nm) was designed and synthesized, which was seen to be highly selective and sensitive to Zn2+ ion over other metal ions in the system of MeCN:H2O (V/V = 6:4, pH = 7.0). Meanwhile, it was observed that the TPA-HPH solution changed from colorless to yellow by “naked eye” when Zn2+ was added. Under optimized conditions, the fluorescence intensity of the probe showed a good linear relationship with Zn2+ ion concentration in the range of 0–10μM with a low detection limit of 7.3 × 10−10M. The fluorescent mechanism was proposed. Most importantly, TPA-HPH could realize on-site monitor and live cell imaging for Zn2+ ion. 摘要:
A facile method is introduced to tune the aggregate morphologies of ABC triblock copolymer in selective media by combining the self-assembly and hydrogen bonding. Poly(styrene)-block-poly(1,4-butadiene)-block-poly(2-vinyl pyridine), abbreviated for PS-b-PBd-b-P2VP and used as an ABC triblock copolymer, self-assembles in toluene and methanol mixture to form discoid micelles with PBd as the disk containing part of PS domains in the core, other PS as bumps and P2VP as corona, respectively. When oleic acid (OA) is added in the assembly system, supramolecular polymer PS-b-PBd-b-P2VP(OA) is prepared by the hydrogen bonding between OA and P2VP of triblock copolymer. As a result, biscuit-like and mushroom-like micelles are formed with assistance of hydrogen bonding. Interestingly, the biscuit-like and mushroom-like micelles can transform reciprocally by fission and fusion mechanism through varying the volume ratio of toluene and methanol. Thus, it provides a simple and convenient approach to control the aggregate morphologies of block copolymers by tuning the hydrogen bonding and selective solvent content. The multicompartment micelles from ABC triblock copolymer may present potential applications in drug delivery, targeting, catalysis and others. © 2019 Elsevier B.V.

摘要:
An eco-friendly and economical route for the oxidation of 5-hydroxymethylfurfural (HMF) to 2,5-furandicarboxylic acid (FDCA) with atmospheric dioxygen as the sole oxidant under acid-, base-, metal-, and external initiator-free conditions in minimal solvent was reported. In the present reaction, the 1,2-diethoxyethylane has a dual role: reaction medium and free-radical initiator. The FDCA easily crystallizes during the reaction and was simple purified via recrystallization to provide the pure FDCA. [email protected]3O4 magnetic nanoparticles were prepared by ultrasonic-assisted method and characterized by TEM, FTIR, XRD, SEM, EDS, BET and VSM. The factors affecting the adsorption properties of uranyl ions by [email protected]3O4 were studied. Results show that the [email protected]3O4 nanoparticles have core–shell structure and superparamagnetism. Under the optimized conditions, the maximum adsorption capacity can reach 229.03 mg/g. The optimum adsorption conditions were as follows: pH = 6, temperature 80 °C, C0 = 35 mg/L, contact time 2.5 h, adsorbent dosage 10 mg. Adsorption kinetics and thermodynamic studies show that the adsorption process accords with the Freundlich isotherm adsorption model and the pseudo-second-order kinetic model. © 2019, Akadémiai Kiadó, Budapest, Hungary.

The encapsulated enzyme system by metal-organic frameworks (MOFs) exhibits great potential in biofuel cells, pharmaceuticals, and biocatalysis. However, the catalytic efficiency and the enzymatic activity are severely hampered due to enzyme leaching and deficiency of storage stability. In this study, we immobilized cytochrome c (Cyt c) into dimethylimidazole-copper (Cu(Im)2) by biomimetic mineralization, and constructed a bioinorganic hybrid material, termed Cyt [email protected](Im)2. Encapsulated Cyt c in Cu(Im)2 with a nanosheet structure exhibited significantly improved catalytic efficiency, enzymatic activity and kinetic performance. The catalytic efficiency (kcat /Km) for Cyt [email protected](Im)2 was ~20-fold higher compared to that of free Cyt c. Moreover, the increased activity was not affected by long-term storage. Based on this system, we further constructed a multi-enzyme composite with glucose-oxidase (GOx), termed GOx-Cyt [email protected](Im)2, which exhibited greatly improved enzymatic activity, stability, and excellent selectivity for the detection of low concentrations of glucose. This strategy may provide new insights into the design of enzymes with high activity and stability, as well as the construction of multi-enzyme systems. © 2019 by the authors.
