Bone morphogenetic protein 2 (BMP-2) plays a critical role in osteogenesis of MSCs. However, the use of SDF-1 and BMP-2 in bone tissue engineering is limited by their short half-lives and rapid degradation in vitro and in vivo. Methods: The chitosan oligosaccharide/ heparin nanoparticles (CSO/H NPs) were first prepared via self-assembly. Chitosan-agarose-gelatin (CAG) Scaffolds were then synthesized via gelation technology using cross-linked chitosan, agarose, and gelatin, and were modified by CSO/H NPs. The encapsulation efficiency and release kinetics of SDF-1 and BMP-2 were quantified using an enzyme-linked immunosorbent assay. A CCK-8 assays were used to evaluate biocompatibility of NP-modified scaffolds. The biological activity of the loaded SDF-1 and BMP-2 was evaluated using the transwell migration assay and osteogenic induction assay. An animal MSC recruitment model was used to study the ability of SDF-1 released from NP-modified scaffolds to induce migration of MSCs. Results: In this study, we developed a novel nanoparticle-modified CAG scaffold for the delivery of SDF-1 and BMP-2. CCK-8 assays demonstrated excellent biocompatibility of NP-modified scaffolds. In addition, we investigated the release of SDF-1 and BMP-2 from NP-modified scaffolds, and evaluated the effect of released SDF-1 on MSC migration. The effect of released BMP-2 on MSC osteogenesis was also examined. In vitro cell migration assays showed that SDF-1 released from NP-modified scaffolds retained its migration activity; osteogenesis studies demonstrated that released BMP-2 exhibited a strong ability to induce differentiation towards osteoblasts. Our in vivo recruitment assays showed continuous chemotactic response of MSCs to SDF-1 released from the NP-modified scaffold.Conclusion: The simplicity of synthesizing CSO/H NP-modified CAG scaffolds, combined with its high cytokine loading capacity and sustained release effect, renders NP-modified CAG scaffold an attractive candidate for sustained release of SDF-1 and BMP-2 to promote bone repair and regeneration. 摘要:
目的探究核因子κB(NF-κB)是否通过分拣蛋白Sortilin影响荷脂THP-1巨噬细胞脂质代谢和泡沫化。方法 NF-κB激活剂PMA或其特异性抑制剂PDTC孵育荷脂THP-1巨噬细胞,qRT-PCR检测Sortilin mRNA水平,Western blot检测Sortilin蛋白表达;沉默荷脂THP-1巨噬细胞Sortilin同时加入PMA孵育,胞内胆固醇流出行液体闪烁计数器检测,胞内脂质含量行高效液相色谱法检测,胞内脂滴情况用油红O染色显示。结果 PMA孵育处理荷脂THP-1巨噬细胞后Sortilin mRNA水平及蛋白表达上调,PDTC孵育处理后Sortilin mRNA水平及蛋白表达下调;PMA孵育处理荷脂THP-1巨噬细胞内脂质流出减少,脂质蓄积程度增加,泡沫细胞形成加剧,而PMA与Sortilin shRNA共处理后,其胞内脂质流出增多,胞内脂质蓄积程度减轻,泡沫细胞形成受抑。结论 NF-κB通过促进Sortilin表达,导致巨噬细胞内脂质流出受抑,胞内脂质蓄积程度加重,从而加剧泡沫细胞形成。

摘要:
目的研究丰富环境对辐射诱导小鼠认知功能障碍的保护作用及其可能机制。方法将45只2月龄雌性昆明小鼠采用随机数表法分为对照组、照射组和照射丰富环境组,每组15只。照射组和照射丰富环境组予以单次4 Gy全身~(137)Cs γ射线照射,照射丰富环境组辐射后连续35 d给予丰富环境刺激。新旧事物识别实验检测小鼠认知功能;免疫组织化学方法检测小鼠海马区小胶质细胞标记物IBA-1的表达;Western blot方法检测小胶质细胞激活标记物CD68及突触囊泡素(SYP)的表达。结果与对照组相比,照射组小鼠在新旧事物识别实验中新事物分辨率降低,海马区IBA-1阳性细胞数目增加,CD68蛋白表达升高,SYP蛋白表达降低(t=3.66、 6.83、 5.79、 6.84,P<0.05)。与照射组相比,照射丰富环境组小鼠新事物分辨率升高,海马区IBA-1阳性细胞数目减少,CD68蛋白表达降低,SYP蛋白表达增加(t=3.56、 7.69、 4.59、 4.06,P<0.05)。结论4 Gy单次全身~(137)Cs γ射线照射可构建放射性认知功能障碍模型,丰富环境可改善模型小鼠认知功能,其机制可能与抑制海马区小胶质细胞激活以及减少神经元突触丢失有关。

中期因子(midkine)是一种分泌型肝素结合生长因子,在炎症、肿瘤、代谢类疾病及胚胎的早期发育方面发挥着重要的作用。动脉粥样硬化(atherosclerosis)是一种常见的心血管疾病,迄今为止,其具体发病机制尚未完全阐明。研究表明,动脉粥样硬化是一种血管壁受损的慢性炎症性疾病,并与遗传、环境、脂质代谢相关。中期因子在脂质代谢、氧化应激及炎症等方面起重要作用,并与动脉粥样硬化密切相关。本文对中期因子在动脉粥样硬化发生发展中的作用的研究进展进行综述。

摘要:
Overexpression and activation of Axl receptor tyrosine kinase have been widely accepted to promote cell proliferation, chemotherapy resistance, invasion, and metastasis in several human cancers, such as lung, breast, and pancreatic cancers. Axl, a member of the TAM (Tyro3, Axl, Mer) family, and its inhibitors can specifically break the kinase signaling nodes, allowing advanced patients to regain drug sensitivity with improved therapeutic efficacy. Therefore, the research on Axl is promising and it is worthy of further investigations. In this review, we present an update on the Axl inhibitors and provide new insights into their latent application.

目的:探讨丰富环境(EE)对弱视小鼠初级视皮质和外侧膝状体Sortilin的表达影响。方法:选择36只21 d昆明小鼠,雌雄不限,随机分为正常组(Control) 、单眼剥夺+标准环境组(MD + SE)和单眼剥夺+丰富环境组(MD + EE),通过行为学检测各组小鼠视功能改变、免疫组织化学方法检测各组小鼠视皮质和外侧膝状体Sortilin的表达。结果:行为学结果显示:与Control组相比,MD + SE组探爪触地成功率明显降低(P < 0.05),MD + EE组探爪触地成功率显著高于MD + SE组(P < 0.05);免疫组织化学结果显示:与Control组相比,MD + SE组外侧膝状体背内侧Sortilin阳性神经元数目显著升高(P < 0.001); MD + EE组显著低于MD + SE组,但高于Control组(P < 0.001);与Control组相比,MD + SE组小鼠视皮质Sortilin阳性神经元数目显著降低(P < 0.001); MD + EE组显著高于MD + SE组(P < 0.001),但显著低于Control组(P < 0.001) 。结论:丰富环境改善弱视小鼠视功能可能通过调节视皮质和外侧膝状体Sortilin的表达实现。

摘要:
目的探讨三七总皂苷(PNS)对脂多糖(LPS)诱导的小鼠抑郁样行为及脑内小胶质细胞活化的影响。方法24只雄性昆明小鼠随机分为溶媒对照(NS)组、LPS组、三七总皂苷预处理(PNS + LPS)组,采用新奇环境抑制摄食实验、糖水偏好实验、悬尾实验、强迫游泳实验观察小鼠的抑郁样行为;采用免疫组织化学法检测海马、杏仁核小胶质细胞标记物离子钙接头蛋白分子1(IBA-1)的表达变化。结果LPS导致小鼠出现抑郁样行为,包括新奇环境摄食潜伏期延长(P < 0.05),糖水偏好百分比下降(P < 0.05),悬尾不动时间延长(P < 0.05),强迫游泳不动时间增加(P < 0.01),PNS干预可逆转上述抑郁行为表现(P < 0.05); LPS导致小鼠海马和杏仁核IBA-1阳性细胞数目显著增多(P < 0.0001),PNS预处理能明显减少IBA-1的阳性细胞数(P < 0.05) 。结论三七总皂苷能有效缓解LPS诱导的小鼠抑郁行为,其机制可能与抑制脑内小胶质细胞活化有关。

摘要:
Accumulating evidence indicates that microRNAs are implicated in tumor initiation and progression through negatively regulating oncogenes or tumor suppressor genes. In the present study, we report that the expression of miR-200a was significantly lower in renal cell carcinoma (RCC) specimens and RCC cell lines. Restoration of miR-200a suppressed cell growth, arrested cell cycle progression, and promoted cell apoptosis in RCC cell lines. We next used qRT-PCR array technology to identify Sirtuin 1 (SIRT1) as one of the downregulated proteins during miR-200a overexpression in 786-O cells. Following a further assay by luciferase reporter system, SIRT1 was validated as a direct target of miR-200a. Moreover, siRNA-mediated knockdown of SIRT1 could partially phenocopy the effects of miR-200a overexpression. In contrast, overexpression of truncated SIRT1 (without an endogenous 3′-UTR) could rescue the effect of miR-200a overexpression on 786-O cells, which suggested that SIRT1 3′-UTR is targeted by miR-200a specifically. These observations provide further evidence for a critical tumor-suppressive role of the miR-200a in RCC in addition to identifying a novel regulatory mechanism, which may contribute to SIRT1 upregulation in RCC. © 2017, Springer Science+Business Media, LLC.

目的 探讨分拣受体Sortilin对THP-1巨噬细胞内脂质代谢的影响.方法 用氧化型低密度脂蛋白(ox-LDL)孵育THP-1巨噬细胞.Western blot检测荷脂后THP-1巨噬细胞中Sortilin表达变化;THP-1巨噬细胞Sortilin高表达和沉默后,液体闪烁计数仪检测胞内胆固醇流出,高效液相色谱法检测细胞内脂质含量,油红O染色观察胞内脂滴情况.结果 THP-1巨噬细胞Sortilin表达呈ox-LDL剂量和时间依赖性增加;以50 mg/L ox-LDL处理24 h后THP-1巨噬细胞Sortilin表达水平达到高峰.与对照组相比,Sortilin过表达后巨噬细胞胆固醇流出减少,胞内脂质含量增加且脂滴明显增多,而Sortilin沉默后则刚好出现相反的结果.结论 Sortilin抑制巨噬细胞内胆固醇流出,促进胞内脂质蓄积.

Sortilin is closely associated with hyperlipidemia and the risk of atherosclerosis (AS). The role of sortilin and the underlying mechanism in peripheral macrophage are not fully understood. In this study, we investigated the effect of macrophage sortilin on ATP-binding cassette transporter A1 (ABCA1) expression, ABCA1-mediated cholesterol efflux, and aortic AS. Macrophage sortilin expression was upregulated by oxidized low-density lipoproteins (ox-LDLs) in both concentrationand time-dependent manners. Its expression reached the peak level when cells were incubated with 50 ?g/ml ox-LDL for 24 h. Overexpression of sortilin in macrophage reduced cholesterol efflux, leading to an increase in intracellular total cholesterol, free cholesterol, and cholesterol ester. Sortilinwas found to bind with ABCA1 protein and suppress macrophage ABCA1 expression, resulting in a decrease in cholesterol efflux from macrophages. The inhibitory effect of sortilin in cholesterol efflux was partially reversed by treatment with chloroquine, a lysosomal inhibitor. On the contrary, the ABCA1 protein level and ABCA1-mediated cholesterol efflux is increased by sortilin short hairpin RNA transfection. The fecal and biliary cholesterol 3H-sterol from cholesterolladen mouse peritoneal macrophage was reduced by sortilin overexpression through lentivirus vector (LV)-sortilin in low-density lipoprotein receptor knockout mice, which was prevented by cotreatment with chloroquine. Treatment with LV-sortilin reduced plasma high-density lipoprotein and increased plasma ox-LDL levels. Accordingly, aortic lipid deposition and plaque area were exacerbated, and ABCA1 expression was reduced in mice in response to infection with LVsortilin alone. These effects of LV-sortilin were partially reversed by chloroquine. Sortilin enhances lysosomal degradation of ABCA1 protein and suppresses ABCA1-mediated cholesterol efflux from macrophages, leading to foam cell formation and AS development. © 2019 The Author(s) .

摘要:
探讨丰富环境(Environmental Enrichment, EE)对电离辐射所致小鼠认知功能障碍的保护作用及其可能机制。将36只雌性昆明小鼠随机分为对照组(Control)、辐射组(IR)和辐射丰富环境组(IR+EE)。IR组和IR+EE组小鼠采用137Cs γ射线进行全身辐照至吸收剂量4 Gy;IR+EE组小鼠辐照后给予连续35 d EE刺激。采用新物体识别实验检测小鼠认知功能;免疫组织化学方法检测海马区神经发生标记物双皮质素(Doublecortin, DCX)及磷酸化环磷酸腺苷应答元件结合蛋白(Phosphorylated cyclic adenosine monophosphate (cAMP) response element-binding protein, p-CREB)的表达;Western blot方法检测海马区CREB及p-CREB蛋白的表达。结果表明:与对照组相比,IR组小鼠新物体分辨率明显降低(45.55 ± 5.80 vs. 2.99 ± 6.18,p<0.05),海马区DCX阳性细胞数明显减少(123.8 ± 9.3 vs. 70.2 ± 5.9,p<0.05),p-CREB/CREB表达下调(1.007 ± 0.058 vs. 0.772 ± 0.039, p<0.05);而予以EE刺激后小鼠新物体分辨率明显回升(2.99 ± 6.18 vs. 28.31 ± 7.30,p<0.05),海马区DCX阳性细胞数明显增加(70.2 ± 5.9 vs. 95.7 ±6.5,p<0.05),p-CREB/CREB表达上调(0.772 ± 0.039 vs. 1.014 ± 0.093, p<0.05)。结果提示EE可能是通过上调海马区p-CREB的表达,保护海马神经发生,进而改善辐射所致的小鼠认知功能障碍。

文章就护理专业基础与临床全方位相结合的系统解剖学教学模式进行论述,围绕护理专业系统解剖学教学为何要将基础与临床全方位相结合以及如何做到这一点作了详细探讨。通过精选教材内容、PBL教学模式、体表标志的触摸、实验教学学生自主观察并课后绘图以及课后反馈等方式可以使系统解剖学与临床护理全方位地结合在一起。相比传统护理专业系统解剖学教学模式,新的教学模式教学重点突出、学生参与积极性高涨、学生掌握知识结构更完整,对于护理专业学生学好系统解剖学并提高其自学能力、实践能力和创新能力具有重要作用。

摘要:
The present review provides a summary of recent evidence of sortilin expression, function, and regulation and its implications in lipid metabolism and development of lipid disorder diseases. As a member of the vacuolar protein sorting 10 protein (Vps10p) receptor family, sortilin mediates intracellular trafficking of diverse endogenous or exogenous protein substrates between the trans-Golgi network (TGN) and plasma membrane compartments. Recent studies reveal that sortilin regulates the expression of lipid genes, plasma lipid level, and the development of lipid disorder diseases. Sortilin promotes atherogenesis by regulating hepatic very low density lipoprotein (VLDL) secretion and plasma lipid level and subsequently macrophage lipid accumulation. Sortilin deficiency is caused by accelerated proteasome degradation under insulin resistance conditions and is thereby implicated in the hyperlipidemia of type 2 diabetes mellitus (T2DM). Sortilin facilitates hepatic cholesterol accumulation by inhibiting hepatic cholesterol catabolism, which promotes the development of nonalcoholic fatty liver disease (NAFLD). Sortilin plays an important role in lipid metabolism and represents a promising therapeutic target for lipid disorder diseases.

Background: Percutaneous cervical pedicle screw fixation has been proven to be an effective method of cervical screw instrumentation, which has the advantages of less invasiveness and low blood loss. Emerging evidence has indicated that the cervical spinous process plays an important role in percutaneous spine surgery. However, there is a limited amount of information on the fundamental research of pedicle and its associated imaging parameter measurement. The purpose of this study was to measure the anatomic data of the pedicle screw channel (PSC) using reverse engineering and three-dimensional reconstruction, and also to discuss the three-dimensional relationship between the cervical spinous process and the pedicle screw channel. Methods: Twenty adult subjects (10 males, 10 females, age range 19–46 years) were studied using the method of three-dimensional CT reconstruction and reverse engineering. The centrum was divided into 10 equal parts from front to back. The bisectors were defined as borderline depths of the centrum, from front to back, 100%, 90%, 80%, 70%, 60%, 50%, 40%, 30%, 20%, 10%, 0% of borderline depths were presented. Then, a 3D coordinate system was constructed to measure all the data, including the radius of the inscribed circle, the length of the PSC, the insertion angle, the distances from entry point to cervical spinous process and skin depth. All the indexes were measured from 70% to 90% borderline depth. Results: The radius of the inscribed circles from C3 to C7 at 90% borderline depth were 2.94 ± 0.55 mm, 3.04 ± 0.40 mm, 3.15 ± 0.36 mm, 3.28 ± 0.47 mm, 3.89 ± 0.54 mm, respectively. The lengths of the PSC were between 25 and 32 mm. The insertion angles for 70% to 90% borderline depth were 28.33°, 34.28°, 37.92°, respectively. The relationship between the PSC and spinous process was measured as the distance from the entry point to the end of the spinous process, which were, respectively, 26.91 mm, 28.18 mm, 30.03 mm, 35.67 mm, 41.99 mm from C3 to C7.The distance from the skin to the entry point of C3-7 increased gradually. Conclusions: The measurements of this study could provide detailed information for percutaneous cervical screw fixation. The data of the relationship between the cervical spinous process and the pedicle screw channel present valuable technical information for the design, optimization and clinical application of the aiming device for percutaneous cervical pedicle screw fixation. Signal transducer and activator of transcription 3 (STAT3) is a member of the family of latent cytoplasmic transcriptional factors that could regulate cell proliferation, survival, and development. It has been reported that Twist is a target gene of STAT3, and STAT3/Twist signaling plays an important role in regulating cancer progress. Here, to explore whether 8-bromo-7-methoxychrysin (BrMC) inhibits liver cancer stem-like cell (LCSLC) properties via disrupting STAT3/Twist signaling, we cultured SMMC-7721 cells in vitro, and evaluated the effects of BrMC on the stemness of spheroids by determining the sphere-forming capability and migration. The sphere formation assay results showed a concentration-dependent decrease of sphere-forming capacity in LCSLCs (P < 0.05) treated with different concentrations of BrMC. Wound-healing assays results demonstrated a concentration-dependent decline in cell migration of LCSLCs treated with different concentrations of BrMC. In addition, CD133, CD44, and ALDH1 levels were decreased in LCSLCs treated with BrMC. Treatment with different concentrations of BrMC also reduced the expressions of p-STAT3 and Twist1 proteins. The effect of BrMC was substantially enhanced by co-treatment with JSI-124, a specific inhibitor of STAT3. Ectopic expression of Twist1 attenuated the inhibitory effects of BrMC on sphere formation, migration, and expression of the markers in LCSLCs. However, it had no affect on p-STAT3 expression in LCSLCs. These results demonstrated that BrMC inhibits the stemness of LCSLCs originated from SMMC-7721 cell line by inhibiting STAT3/ Twist signal axis.
