Determination of uranium in water based on enzyme inhibition using a wireless magnetoelastic sensor

摘要:
The aim of this paper is to develop a wireless magnetoelastic sensing method for the determination of uranium in water based on the inhibitory effect of uranyl cation to α-amylase. In this method, a wireless sensor used for detecting uranium was fabricated by immobilizing a layer of starch gel on the surface of a magnetoelastic foil. When the sensor was in a solution containing α-amylase, the α-amylase catalyzed the hydrolyzation of starch, causing a resonance frequency shift of the sensor. Meanwhile, the catalytic hydrolyzation of starch was inhibited by uranium presented in the above solution, resulting in a decrease in the resonance frequency shift of the sensor. Consequently, the amount of uranium could be determined by measuring the resonance frequency shift. The influence of manifold variables on the determination was investigated in details. A linear range was found to be 9.2 to 103.5 μg L<sup>-1</sup> under optimal conditions with a detection limit of 3.6 μg L<sup>-1</sup>. The method was applied to the determination of uranium in environmental water samples with satisfactory results. ©2013 Copyright Taylor and Francis Group, LLC.

摘要:
奶粉用0.1%三氯乙酸溶液和2%乙酸铅溶液消除样品中的干扰物质(主要是蛋白质),用2,6-二氯靛酚钠为显色剂对牛奶中三聚氰胺进行分光光度法检测。对不同的入射波长、显色剂用量、pH值、显色时间、温度进行了条件实验。结果表明,采用2,6-二氯靛酚钠为显色剂,在25 ℃显色10 min,在634 nm有最大吸收。测定范围为40~800 μg/mL,线性相关系数r=0.998 9,加标回收率为92.66%~97.84%,RSD为0.45%~1.43%。

合成了一种Salophen与荧光素的连接物以用作铀酰的荧光探针。用水杨醛和3,4-二氨基苯甲酸合成含羧基的四齿西佛碱配体Salophen,Salophen与5-氨基荧光素缩合,生成连接物Salophen-荧光素,Salophen-荧光素与铀酰反应,生成配合物铀酰-Salophen-荧光素。用红外光谱、紫外可见吸收光谱和荧光光谱研究了Salophen-荧光素及其与铀酰的配合物的光谱性质。结果表明,Salophen-荧光素的荧光强于5-氨基荧光素,但弱于其与铀酰的配合物。一定条件下,Salophen-荧光素的荧光强度随着铀酰浓度的增加而线性地增强,表明Salophen-荧光素可作为铀酰的荧光探针。

摘要:
A new wireless sensing method for the detection of uranium in water samples has been reported in this paper. The method is based on a sandwich-type detection strategy. Salophen, a tetradentate ligand of uranyl ion, was immobilized on the surface of the polyurethane-protected magnetoelastic sensor as receptor for the capture of uranyl ion. The phosphorylated polyvinyl alcohol coated magnetic Fe3O4 nanoparticles were used as signal-amplifying tags of uranyl ion. In a procedure of determining uranium, firstly uranyl ion in sample solution was captured on the sensor surface. Then the captured uranyl bound the nanoparticle through its coordination with the phosphate group. The amount of uranium was detected through the measure of the resonance frequency shift caused by the enhanced mass loading on the sensor surface. A linear range was found to be 0.2-20.0 μg/L under optimal conditions with a detection limit of 0.11 μg/L. The method has been applied to determine uranium in environmental water samples with the relative standard deviations of 2.1-3.6 % and the recoveries of 98.0-101.5 %. The present technique is one of the most suitable techniques for assay of uranium at trace level in environmental water samples collected from different sources. © 2013 Akadémiai Kiadó, Budapest, Hungary.

摘要:
Myoglobin (Mb) is an ideal scaffold protein for rational protein design mimicking native enzymes. We recently designed a nitrite reductase (NiR) based on sperm whale Mb by introducing an additional distal histidine (Leu29 to His29 mutation) and generating a distal tyrosine (Phe43 to Tyr43 mutation) in the heme pocket, namely L29H/F43Y Mb, to mimic the active site of cytochrome cd
1 NiR from Ps. aeruginosa that contains two distal histidines and one distal tyrosine. The molecular modeling and dynamics simulation study herein revealed that L29H/F43Y Mb has the necessary structural features of native cytochrome cd
1 NiR and can provide comparable interactions with nitrite as in native NiRs, which provides rationality for the protein design and guides the protein engineering. Additionally, the present study provides an insight into the relatively low NiR activity of Mb in biological systems.

摘要:
Uranium is toxic to human body with mechanisms not fully understood. The structural and functional consequences of uranyl ions (UO22+) interacting with an axial mutant of cytochrome b5, His39Ser (cyt b5 H39S), were investigated by both spectroscopic and molecular modeling methods. Although slightly disturbs protein folding, UO22+ binding to cyt b5 H39S leads to a decrease of peroxidase activity. A uranyl binding site was further proposed in the heme-binding domain at Glu37 and Glu43. The impacts of UO22+ binding to cyt b5 H39S studied herein provide valuable insights into the toxicity mechanism of UO22+ towards membrane heme proteins.

摘要:
A wireless sensing method for the measurement of gamma radiation dose has been developed based on the fact that gamma rays can initiate the polymerization of acrylamide, which causes an increase in solution viscosity that can be detected with a wireless magnetoelastic sensor. The magnetoelastic sensor is able to wirelessly detect the resonance frequency shifts of a magnetoelastic foil in response to changes in solution viscosity. There is a linear relationship between the resonance frequency shift and gamma radiation dose in the range of 0–50 Gy (under optimal conditions) with a detection limit of 0.25 Gy. This method has the advantage of providing real-time, continuous measurement in situ. The method has been used successfully to determine the gamma radiation dose in real exposure scenarios, with satisfactory results.

以一种铀酰配合物为模板,制备了一种用于从水相选择性分离铀酰的新型印迹聚合物。用5-氯甲基水杨醛和3,4-二氨基苯甲酸合成四齿配体salophen,再合成铀酰与salophen的配合物铀酰-salophen。然后以甲基丙烯酸甲酯为功能单体,二甲基丙烯酸乙二醇酯为交联剂,铀酰-salophen为模板,合成铀酰的印迹聚合物。聚合物中的铀酰通过酸浸除去。用平衡吸附法研究了聚合物对铀酰的吸附性能和选择识别能力,且探讨了实验条件对印迹聚合物吸附性能的影响。结果表明,在竞争金属离子存在下,印迹聚合物对铀酰具有较高的吸附能力和选择识别能力,选择性系数大于200,且可以循环使用。

摘要:
By retaining the native distal His64 in sperm whale myoglobin (Mb), a second distal histidine was engineered in Mb by mutating Leu29 to His29. The resultant mutant of L29H Mb exhibits an unusual enhanced peroxidase activity with a positive cooperativity in comparison to that of wild type Mb. The new enzyme with two cooperative distal histidines has not been found in native peroxidase, which emphasizes a creation of the rational protein design. © 2012 Ying Wu Lin.

In this study, we report a double-receptor sandwich supramolecule sensing method for the determination of adenosine triphosphate (ATP). One receptor is a uranyl-salophen complex which can bind the triphosphate group in ATP selectively, and another is an anti-adenosine aptamer which is a single-stranded oligonucleotide and can recognize the adenosine group in ATP specifically. The uranyl-salophen complex was immobilized on the surface of amino-silica gel particles and used as the solid phase receptor of ATP. The anti-adenosine aptamer was labeled with a fluorescent group and used as the labeled receptor of ATP. In the procedure of ATP detection, ATP was first combined with the solid phase receptor and then conjugated with the labeled receptor to form a sandwich-type supramolecule. The conditions of fabricating solid phase receptor and the influence of manifold variables on the determination were studied. The experimental results demonstrate that the method has a number of advantages such as high selectivity, high sensitivity, good stability and low cost. Under optimal conditions, the linear range for detection of ATP is 0.2-5.0. nmol/mL with a detection limit of 0.037. nmol/mL. The proposed method was successfully applied for the determination of ATP in real samples with the recoveries of 96.8-103.3%. © 2012 Elsevier B.V.

A double-receptor sandwich supramolecule method for the separation and determination of trace uranium was proposed in this paper. One receptor is a salophen which can react with uranyl to form a uranyl-salophen complex, and another receptor is an oligonucleotide which can bind uranyl to form oligonucleotide-uranyl-salophen supramolecule. The salophen was immobilized on the surface of silica gel particles and used as the solid phase receptor for separating uranium from solution. The oligonucleotide was labeled with a fluorescent group and used as the labeled receptor for quantitatively analyzing uranium. In the procedure of separation and determination, uranyl ion was first combined with the solid phase receptor and then conjugated with the labeled receptor to form the sandwich-type supramolecule. The labeled receptor in the sandwich supramolecule was then eluted and determined by fluorescence analysis. The experimental results demonstrate that this method has a number of advantages such as high selectivity, excellent pre-concentration capability, high sensitivity, good stability and low cost. Under optimal conditions, the linear range for the detection of uranium is 0.5-30.0ngmL -1 with a detection limit of 0.2ngmL -1. The proposed method was successfully applied for the separation and determination of uranium in real samples with the recoveries of 95.0-105.5%. © 2012 Elsevier B.V.

Sperm whale myoglobin (swMb) is a well-studied heme protein, both experimentally and theoretically. Comparatively, little attention has been paid to another member of Mb family, Aplysia limacina myoglobin (apMb). swMb and apMb have the same overall structure and perform the same biological function, i.e., O 2 carrier, while using a distinct heme active site. To provide insights into the structure-function relationship for these two Mbs, we herein made a comparison in terms of their dynamics properties using molecular dynamics simulation. We analyzed the overall structure and protein motions, as well as the intramolecular contacts, namely salt-bridges and hydrogen bonds, especially the interactions between the heme propionate groups and the polypeptide chain. The internal cavities in apMb were also compared to the well-known xenon and other cavities in swMb. Based on current simulations, we propose a unique "arginine gate" for apMb, which has a similar function to the histidine gate observed for swMb in previous studies. This study provides valuable information for understanding the homology of heme proteins, and also aids in rational design of structural and functional heme proteins by alternating the heme active site. © Springer-Verlag 2011.

5), with heme bound noncovalently, has been converted into a cyt c-like protein (cyt b
5 N57C) by constructing a thioether linkage between the heme and the engineered cysteine residue. With no X-ray or NMR structure available, we herein performed a molecular modeling study of cyt b
5 N57C. On the other hand, using amino acid sequence information for a newly discovered member of the cyt b
5 family, domestic silkworm cyt b
5 (DS cyt b
5), we predicted the protein structure by homology modeling in combination with MD simulation. The modeling structure shows that both Cys57 in cyt b
5 N57C, and Cys56, a naturally occurring cysteine in DS cyt b
5, have suitable orientations to form a thioether bond with the heme 4-vinyl group, as the heme is in orientation A. In addition to providing structural information that was not previously obtained experimentally, these modeling studies provide insight into the formation of cyt c-like thioether linkages in cytochromes, and suggest that c-type cyt b
5 maturation involves a b-type intermediate.

A novel method for determination of trace amounts of microorganism populations in solution was developed by using a multiwalled carbon nanotube (MWNT) modified glassy carbon electrode and square wave voltammetry (SWV). The simultaneous combination of MWNT and SWV allowed the electrochemical signal of electro-active materials in Escherichia coli O157:H7 (E. coli) to be dramatically amplified. Compared with a bare glassy carbon electrode, the MWNT-modified glassy carbon electrode showed catalytic properties in the oxidation of electro-active materials on cell surfaces. Moreover, SWV was proved to be more sensitive than cyclic voltammetry (CV) for investigation of the electrochemical behavior of cells. In this paper, a linear relationship was obtained between the SWV peak current and the cell concentration in the range 2 × 102–2 × 108 cell mL−1 with a detection limit of 2 × 102 cell mL−1. The effect of antibiotic drug Gentamycin Sulfate injection (GSI) on the growth of E. coli was also investigated.

摘要:
Heme transfer is commonly observed from one heme protein to the other such as from cytochrome b<inf>5</inf> (cyt b<inf>5</inf>) to apo-myoglobin. In this study, instead of to another heme protein, we observed the heme transfer from wild-type (WT) cyt b<inf>5</inf>, H39C cyt b<inf>5</inf> with heme axial ligand His39 mutated to Cys39, and DME cyt b<inf>5</inf> with heme replaced by protoporphyrin IX dimethyl ester, to a heme DNA aptamer, PS2.M, respectively, with a different rate constant. The heme transfer was further confirmed by the enhancement of peroxidase activity of the cyt b<inf>5</inf>s-PS2.M system due to the formation of catalytic PS2.M-heme complex. This study provides valuable insights into both cyt b<inf>5</inf>-heme and PS2.M-heme interactions and shows that heme transfer from heme protein to heme-aptamer can be used to evaluate the relative stability of heme proteins. In addition, this study sheds light on the maturation of heme proteins in vivo by interacting with DNA/RNA enzymes. ©2012 Elsevier B.V. All rights reserved.

5 was only slightly affected, UO

binding to cyt b
5 H39S with a K
D of 2.5 μM resulted in obvious alteration of the heme active site, and led to a decrease in peroxidase activity. Theoretically, molecular simulation proposed a uranyl ion binding site for cyt b
5 at surface residues of Glu37 and Glu43, revealing both coordination and hydrogen bonding interactions. The information gained in this study provides insights into the mechanism of uranyl toxicity toward membrane protein at an atomic level.

摘要:
应用密度泛函理论(DFT),在B3LYP/6-31+G(d)水平上对含有5~24个碳原子的部分液相链烷烃进行结构优化和量化计算其分子中各个原子间空间拓扑距离,并构建拓扑距离矩阵.结合分子中各原子的支化度,应用原子的平衡电负性对分子图进行着色,得到新型结构拓扑指数PX<,m>(m=1,2,3).采用多元线性回归技术建立液相链烷烃热导率γ与PX[m](m=1,2,3)的定量关系拓扑模型,并用该模型对热导率γ进行预测与估算,结果表明实验值与预测值、估算值均很吻合.同时,采用留一法(1eave-one-out)和外检验方法测试模型的内部稳定性和外部预测能力.与文献结果比较,本文所用参数少,计算简便,为液相链烷烃导热的研究提供了一种新方法.

A wireless immunosensor for the detection of Mycoplasma genitalium was fabricated by immobilizing polyclonal antibody onto the surface of a magnetostrictive strip. In response to a time-varying magnetic field, the immunosensor longitudinally vibrates at a resonance frequency, emitting magnetic flux that can be remotely detected by a pickup coil. No physical connections between the immunosensor and the detection system are required, facilitating wireless aseptic operation. The binding of M. genitalium to the immunosensor surface resulted in a decrease in the resonance frequency of the immunosensor. When solutions with varying concentrations of the bacteria were tested, the shift of the resonance frequency was proportional to the concentration of M. genitalium. Under the optimized conditions, the linear range for the determination of M. genitalium was 2.0×103 to 2.9×104 color change units (ccu)/ml with a detection limit of 3.4×102 ccu/ml. The immunosensor was successfully applied to real samples containing M. genitalium with results similar to those previously obtained by the color change unit method. © 2010 Elsevier Inc.

Uranium is toxic to the human body with mechanisms not fully understood. The interaction between uranyl ion (UO22+) and ubiquitin (Ub) was predicted by molecular modeling, molecular dynamics (MD) simulation, and steered molecular dynamics (SMD) simulation. The structural and dynamics consequences, as well as protein unfolding behavior, upon uranyl binding to two surface residues of Ub, Glu18 and Asp21, were revealed at an atomic level. This in silico study exhibits a possibility of UO22+ binding to Ub, providing a possible mechanism of uranyl toxicity in vivo. © 2011 The Chemical Society of Japan.
