Aggregation-induced photoluminescence enhancement of protamine-templated gold nanoclusters for 1-hydroxypyrene detection using 9-hydroxyphenanthrene as a sensitizer

摘要:
In this study, we confirmed that protamine-templated gold nanoclusters (PRT-AuNCs) exhibit aggregation-induced emission properties (AIE-PRT-AuNCs). 1-Hydroxypyrene (1−OHPy) further induced the aggregation of AIE-PRT-AuNCs via hydrogen bonding and electrostatic and hydrophobic interactions, resulting in the aggregation-induced photoluminescence enhancement of AIE-PRT-AuNCs. 9-Hydroxyphenanthrene was able to decrease the background signal, thus increasing the sensitivity of the method. Based on these findings, a cost-effective, highly sensitive and selective strategy was proposed for the quantitative detection of 1−OHPy. This method displayed a wide linear range of 0.924 – 74.1 nmol/L with a low detection limit of 0.277 nmol/L, showing great potential for the monitoring of 1−OHPy in human urine. This strategy may provide a theoretical basis for future studies of the AIE properties of metal nanoclusters and their applications in the field of chemical and biological sensing. © 2020 Elsevier B.V.

We report a label-free ultrasensitive and selective strategy for Pb(ii) assay by a multifunctional DNA probe (MDP)-mediated rolling-circle amplified synthesis of the G-quadruplexes. The MDP acted as a target recognition probe, a catalytic DNAzyme and a primer of rolling-circle amplification (RCA). The presence of Pb(ii) can induce the conformational switching of the MDP, resulting in the cleavage of the S-DNA in the MDP to release an E-DNA. The released E-DNA then initiated a RCA reaction with a reasonably devised padlock DNA template to produce an accumulated amount of repeated sequences. The RCA product in the present work was designed as a G-rich sequence, which could fold into thousands of G-quadruplex units. The G-quadruplex formed by RCA can specifically bind to NMM to result in an amplified fluorescence signal. Through these cascade amplifications, Pb(ii) ions can be detected at as low as 94.29 pM, which is much lower than those reported in related literature. We expect that this amplification strategy might be helpful in the design of a highly sensitive analytical platform for wide application in environmental and biomedical fields. © The Royal Society of Chemistry.

摘要:
We certify that protamine-gold nanoclusters (PRT-AuNCs) synthesized by one-pot method exhibit peroxidase-like activity. The catalytic activity of PRT-AuNCs followed typical Michaelis-Menten kinetics and exhibited higher affinity to 3,3',5,5'-tetramethylbenzidine (TMB) as the substrate compared to that of natural horseradish peroxidase. Meanwhile, we found that Hg(II) could dramatically and selectively enhance the peroxidase-like activity of PRT-AuNCs, and the enhanced mechanism by Hg(II) was demonstrated to be generation of the cationic Au species and the partly oxidized Au species (Au(delta+)) by Hg(2+)-Au(0)/Au(+) interaction. Based on this finding, quantitative determinations of Hg(II) via visual observation and absorption spectra were achieved. The proposed strategy displays high selectivity that arises from the strong aurophilic interaction of mercury towards gold. Moreover, the developed method is highly sensitive with a wide linear range and low detection limit of 1.16 nM. This strategy is not only helpful to develop effective nanomaterials-based artificial enzyme mimics but also irradiative to discover new applications of artificial mimic enzymes in bio-detection, medical diagnostics, and biotechnology. Hg(II) can stimulate the peroxidase-like activity of PRT-AuNCs selectively, enhancing their ability to catalyze the chromogenic reaction of TMB by H2O2.

摘要:
A novel wireless magnetoelastic sensor (WMS) strategy for Hg2+ assay was proposed using a specific aptamer probe and graphene oxide (GO) as an adsorbent. The introduction of Hg2+ results in the release of CDNA-biotin. This one can be adsorbed to the surface of GO that was immobilized on the WMS along with a decrease of the resonance frequency of the sensor. A biotin-streptavidin (SA)-gold nanoparticles( AuNPs) system designed in this work was used for amplifying response signal of the sensor. The signal intensity linearly correlated with Hg2+ concentration ranging from 2.80 to 88.9 nM with a detection limit of 0.885 nM. This WMS has additional advantages of being less time-consuming, low cost and remote query, and can be employed for the real-time and in situ detection of Hg2+ in real samples. The mechanism for structure switching and signal amplification was also studied by transmission electron microscopy and UV-vis spectra. (C) 2016 Elsevier B.V. All rights reserved.

Based on ciprofloxacin (CIP)-Cu(II) system, the novel methods for the detection of metallothioneins (MTs) have been developed by fluorescence (FL) and resonance light scattering (RLS) strategies. The FL strategy avoids the label and derivatization steps in common methods, while the RLS strategy can be applied for determining bio-macromolecules and small molecules without native fluorescence. The response signals linearly correlated with the concentration of MTs over the ranges of 1.03×10<sup>-8</sup>-1.23×10<sup>-6</sup> mol L<sup>-1</sup> for FL, and of 2.56×10<sup>-7</sup>-1.54×10 <sup>-6</sup> mol L<sup>-1</sup> for RLS. The limits of detection (LOD) are 3.1×10<sup>-9</sup> mol L<sup>-1</sup> for FL and 7.68×10 <sup>-8</sup> mol L<sup>-1</sup> for RLS. This study represents the comparison of these two methods using the same CIP-Cu<sup>2+</sup>-MTs system. They not only allow practical application for MTs detection but also serve as a potential choice for the operators according to their concrete needs. In addition, the mechanisms for FL and RLS enhancement of the system were also discussed. ©2013 Elsevier B.V.

摘要:
A wireless magnetoelastic sensor (WMS) for uranyl detection has been developed using DNAzyme-graphene oxide (GO) and gold nanoparticles (AuNPs)-based amplification. The WMS is fabricated by immobilizing a layer of GO film on a magnetoelastic ribbon-like sensor. When the substrate strand of DNAzyme was cleaved in the presence of UO<inf>2</inf><sup>2+</sup>, the single-stranded DNA (ssDNA) was adsorbed onto the GO film of the WMS, resulting in a decrease of resonance frequency of the WMS. HAuCl<inf>4</inf> can be reduced by ascorbic acid (AA) to atom Au coated on AuNPs in cetyltrimethyl ammonium bromide (CTMAB) solution, leading to the signal amplification of the system. When the molar ratio of AA and HAuCl<inf>4</inf> was 3:2, the response signals linearly correlated with the concentration of UO<inf>2</inf><sup>2+</sup> over the range of 9.57 ×10<sup>-9</sup> to 1.20 ×10<sup>-7</sup> M in the MES buffer solution of pH 5.0 with a detection limit of 2.87 ×10<sup>-9</sup> M. The proposed WMS has additional advantages of being less time-consuming, low cost and remote query, and avoids the interference of some metals such as Cu<sup>2+</sup>, Fe<sup>2+</sup>, Fe<sup>3+</sup>, Hg<sup>2+</sup>, and Tb <sup>3+</sup>. It has been successfully applied for detecting UO <inf>2</inf><sup>2+</sup> in real samples with a satisfactory result. A polyacrylamide gel electrophoresis (PAGE) assay, scanning electron microscopy (SEM) and UV-vis spectra were used to confirm the mechanism for structure switching and signal amplification. ©2013 Elsevier B.V.

采用水蒸气蒸馏法提取药对陈皮-青皮与其单味药的挥发油成分,利用气相色谱-质谱法(GC-MS)和化学计量学多元分辨方法——直观推导式演进特征投影法(HELP),同时结合程序升温保留指数(PTRI)进行挥发油成分定性定量分析。分别在陈皮、青皮和药对陈皮-青皮的挥发油中鉴定出27、27和23个成分,定性组分含量分别占三者挥发油总含量的93.64%/96.45%和85.28%,共有化合物19种;主要化学成分为D-柠檬烯、y-萜品烯、芳樟醇和а-法呢烯。

摘要:
The interaction of pyronine Y (PY) with nucleic acids was studied by resonance Rayleigh scattering (RRS) for nucleic acid detection. The enhanced RRS intensity of nucleic acids reacted with PY was proportional to the concentration of nucleic acids in the ranges of 27.0-625ngml -1 for fish sperm DNA, 39.0-500ngml - for calf thymus DNA, and 59.0-375ngml -1 for yeast RNA. The limits of determination were 0.2ngml -1 for fish sperm DNA, 0.6ngml -1 for calf thymus DNA, and 0.7ngml 1 for yeast RNA. The method had been successfully applied to the quick determination of nucleic acids in synthetic and natural samples. © 2012 Copyright Taylor and Francis Group, LLC.

摘要:
A novel method for determination of trace amounts of microorganism populations in solution was developed by using a multiwalled carbon nanotube (MWNT) modified glassy carbon electrode and square wave voltammetry (SWV). The simultaneous combination of MWNT and SWV allowed the electrochemical signal of electro-active materials in Escherichia coli O157:H7 (E. coli) to be dramatically amplified. Compared with a bare glassy carbon electrode, the MWNT-modified glassy carbon electrode showed catalytic properties in the oxidation of electro-active materials on cell surfaces. Moreover, SWV was proved to be more sensitive than cyclic voltammetry (CV) for investigation of the electrochemical behavior of cells. In this paper, a linear relationship was obtained between the SWV peak current and the cell concentration in the range 2 × 102–2 × 108 cell mL−1 with a detection limit of 2 × 102 cell mL−1. The effect of antibiotic drug Gentamycin Sulfate injection (GSI) on the growth of E. coli was also investigated.

利用水蒸气蒸馏法提取挥发油,采用GC-MS联用技术结合直观推导式演进特征投影法(HELP)对湖北和江西产香薷的挥发油化学成分进行比较分析。共鉴定出37种化合物,湖北香薷35种,江西香薷19种,分别占总含量的95.16%和96.40%。挥发油主要成分为百里香酚、柠檬烯、β-聚伞花素、百里香酚乙酸酯、香荆芥酚和α-丁香烯。

A novel small molecule probe, aptamer beacon (AB), was introduced for adenosine (Ade) recognition and quantitative analysis. The Ade aptamer was engineered into an aptamer beacon by adding a gold nanoparticle-modified nucleotide sequence which is complementary to aptamer sequence (FDNA) at the 3'-end of FDNA. The fluorescence signal "turning on" was observed when AB was bound to Ade, which is attributed to a significant conformational change in AB from a FDNA/QDNA duplex to a FDNA-Ade complex. The Ade measurement was carried out in 20 mmol L-1 Tris-HCl buffer solution of pH 7.4, Delta F signal linearly correlated with the concentration of Ade over the range of 2.0 x 10(-8) to 1.8 x 10(-6) mol L-1. The limit of detection (LOD) for Ade is 6.0 x 10(-9) mol L-1 with relative standard deviations (R.S.D) of 3.64-5.36%, and the recoveries were 98.6%, 100%, 102% (n = 6), respectively. The present method has been successfully applied to determine Ade in human urine samples, and the obtained results were in good agreement with those obtained by the HPLC method. Our investigation shows that the unique properties of the AB could provide a promising potential for small molecules detection, and be benefit to extend the application of aptamer beacon technique. Determination of trace formaldehyde in blood plasma by resonance fluorescence technology

摘要:
A novel method for the determination of trace formaldehyde in blood plasma has been established by using resonance fluorimetry technique. It was based on the fact that oxidation of pyronine Y by potassium bromate was catalyzed by formaldehyde in sulfuric acid. When the wavelength interval was at Δλ=0nm, it was found that the decreased intensity (ΔF) of resonance fluorescence at 574.6nm was proportional to the concentration of formaldehyde in the range of 1.27×10-2 to 2.28μgmL-1. The limit of detection and the average recovery for formaldehyde were 3.80ngmL-1 and 101.6% (n=6), respectively. The present method had been applied to the determination of trace formaldehyde in blood plasma, and the obtained results were in good agreement with those obtained by the resonance light scattering method. © 2011 Elsevier B.V.

A wireless magnetoelastic sensor has been developed for the determination of 2-naphthol (2-NAP) in human urine. This method is based on the precipitation of 2-NAP with diazonium salts produced by the diazo-reaction of sulfamethoxazole (SMZ) with nitrite under a weak alkaline condition, resulting in a descending of the resonance frequency of a wireless magnetoelastic sensor. The frequence shift values (ΔF) of the sensor were directly proportional to the concentration in the range of 1.13 - 139 μmol L -1 for 2-NAP with a correlation coefficient of 0.997 and a detection limit of 0.340 μmol L -1 . The relative standard deviations were 2.38, 2.40 and 2.44%, and the average recovery was 107% (n = 6). The proposed method has additional advantages of being less time-consuming, low cost and remote query, and can be applied for realtime and in situ monitoring of 2-NAP in human urine. It would be a benefit to extend the scope of applications of magnetoelastic sensing techniques. 2011 © The Japan Society for Analytical Chemistry.

A novel method for the simultaneous determination of -naphthol (-NAP), -naphthol (-NAP) and 1-hydroxypyrene (1-OHP) in human urine has been established by using synchronous fluorescence spectrometry. The measurement was carried out in sodium acetate-boroborax buffer solution (pH 1/45.0) with -cyclodextrin (-CD) enhancing fluorescence. At D 1/423 nm, 1-OHP and -NAP exhibit maximum signal with minimum interferences from -NAP. At D 1/4 175nm, the signal of -NAP is not influenced by the presence of 1-OHP and -NAP. The signals detected at these three wavelengths, 360.2 nm, 330.6 nm and 296.4 nm, vary linearly when the concentration of 1-OHP, -NAP and -NAP is in the range of 0.65-218.3 ngmL 1, 2.8-1441.0 ng mL 1 and 3.6-1586.0 ng mL 1, respectively. The limits of detection (LOD) for -NAP,-NAP and 1-OHP were 1.53 ng mL 1, 0.78ng mL 1 and 0.020ng mL 1 with relative standard deviations (RSD) of 2.3%, 2.4% and 1.8%, respectively. The proposed method was successfully applied for the simultaneous determination of -NAP, -NAP and 1-OHP in human urine samples, and the obtained results were in good agreement with those obtained by the method of high-performance liquid chromatography (HPLC). © 2011 Taylor & Francis.

A novel sensitive method was presented for the simultaneous determination of 1-naphthol (1-NAP) and 2-naphthol (2-NAP) in human urine samples. This method is based on the combination of the high sensitivity of quartz crystal microbalance (QCM) and excellent predictive ability of chemometrics. Nanocrystalline TiO<inf>2</inf> films were used as substrates for immobilizing β-cyclodextrin (β-CD) onto the QCM surface. The sensing layer consisting of β-CD and nanocrystalline TiO<inf>2</inf> particles was characterized by X-ray photoelectron spectroscopy (XPS), thermogravimetric analysis (TG) and IR spectroscopy. In the Britton-Robinson (B-R) buffer solution of pH 6.37, frequency shift (ΔF) of the QCM was directly proportional to the concentration of 1-NAP and 2-NAP as 1.0 ×10<sup>-6</sup> to 1.3 ×10<sup>-4</sup> mol L<sup>-1</sup>, and 1.0 ×10<sup>-6</sup> to 1.2 ×10<sup>-4</sup> mol L<sup>-1</sup>, respectively. The correlation coefficients were 0.993 for 1-NAP, and 0.990 for 2-NAP. The limits of detection (LOD) were found to be 8.362 ×10<sup>-7</sup> mol L<sup>-1</sup> for 1-NAP and 2.146 ×10<sup>-7</sup> mol L<sup>-1</sup> for 2-NAP. The proposed method has been successfully applied for detecting 1-NAP and 2-NAP in human urine with a satisfactory result. The QCM with β-CD/TiO<inf>2</inf> composite films coupled with chemometrics can lead to significant improvements in the sensor performance, and would be benefit to extend the scope of application of QCM and chemometrics. ©2009 Elsevier B.V. A novel method of first derivative synchronous fluorescence was developed for the rapid simultaneous analysis of trace 1-hydroxypyrene (1-OHP), 1-naphthol (1-NAP), 2-naphthol (2-NAP), 9-hydroxyphenanthrene (9-OHPe) and 2-hydroxyfluorene (2-OHFlu) in human urine. Only one single scan was needed for quantitative determination of five compounds simultaneously when Δλ= 10 nm was chosen. In the optimal experimental conditions, there was a linear relationship between the fluorescence intensity and the concentration of 1-OHP, 1-NAP, 2-NAP, 9-OHPe and 2-OHFlu in the range of 1.75 ×10<sup>-9</sup>to 4.50 ×10<sup>-6</sup>mol L<sup>-1</sup>, 3.64 ×10<sup>-8</sup>to 2.20 ×10<sup>-4</sup>mol L<sup>-1</sup>, 8.18 ×10<sup>-9</sup>to 1.20 ×10<sup>-4</sup>mol L<sup>-1</sup>, 3.26 ×10<sup>-9</sup>to 8.50 ×10<sup>-5</sup>mol L<sup>-1</sup>and 4.88 ×10<sup>-9</sup>to 5.50 ×10<sup>-6</sup>mol L<sup>-1</sup>, respectively. The limits of detection (LOD) were found to be 5.25 ×10<sup>-10</sup>mol L<sup>-1</sup>for 1-OHP, 1.10 ×10<sup>-8</sup>mol L<sup>-1</sup>for 1-NAP, 2.46 ×10<sup>-9</sup>mol L<sup>-1</sup>for 2-NAP, 9.77 ×10<sup>-10</sup>mol L<sup>-1</sup>for 9-OHPe and 1.46 ×10<sup>-9</sup>mol L<sup>-1</sup>for 2-OHFlu. The proposed method is reliable, selective and sensitive, and has been used successfully in the determination of traces of 1-OHP, 1-NAP, 2-NAP, 9-OHPe and 2-OHFlu in human urine samples, whose results were in good agreement with those gained by the HPLC method. ©2009 Elsevier B.V. Pyronin Y (PY) has a strong resonance fluorescence in sulfuric acid medium. The characteristics of the resonance fluorescence spectra and the factors affecting the spectra were studied. A catalytic resonance fluorometry method for the determination of hydroquinone was proposed based on the catalytic effect of hydroquinone on the oxidation of PY by potassium bromate. The oxidation of PY resulted in the decrease of the resonance fluorescence intensity. The influences of several variables on the sensitivity were studied. At the optimized conditions, the decrease of the resonance fluorescence intensity was in proportion to the concentration of hydroquinone in the range of 4.42 - 1.60 × 103 μg l-1, and the detection limit was 1.46 μg l-1. The proposed method was applied successfully for the determination of trace hydroquinone in environment samples. The relative standard deviation was less than 3.90% and the recoveries were in the range of 95.2 - 104.0%. 2009 © The Japan Society for Analytical Chemistry.

摘要:
二甲酚橙是微量分析中一种很重要的光度显色剂,在pH 6.0的磷酸-醋酸-硼酸缓冲溶液中,二甲酚橙与铅形成稳定的1∶1红色配合物,导致吸光度的增强.研究了铅与三苯甲烷类染料二甲酚橙的显色反应体系,发现在波长580 nm处吸光度的增强值与铅的加入量有很好的线性关系,据此建立了测定微量铅的新方法.该方法的摩尔吸光数为3.312×10~4 L/(mol · cm),铅离子的浓度在0.075~1.2 mg/L范围内符合比耳定律.方法简便、灵敏、选择性好,可广泛应用.

A molecular resonance fluorescence quenching method for the determination of arsenic was proposed. The method is based on the quenching effect of As (V) on the molecular resonance fluorescence of Rhodamine B (RhB) in sulfuric acid medium and in the presence of molybdate and PVA. The influences of acidity, chemical and manifold variables on the sensitivity were studied. At the optimized conditions, the linear range for the determination of arsenic was 2.0 ×10<sup>-3</sup>-0.12 μg ml<sup>-1</sup> with the detection limit of 1.20 ×10<sup>-9</sup> g ml<sup>-1</sup>. The relative standard deviations for the eleven replicate determinations of 0.006 μg ml<sup>-1</sup> and 0.060 μg ml<sup>-1</sup> of arsenic were 1.12% and 1.61%, respectively. The proposed method was applied successfully for the determination of trace arsenic in hair samples with the recoveries of 94.8-104.6%. ©2008 Elsevier B.V. All rights reserved.

建立检测MTs的新方法.在pH 2.5的缓冲溶液中,MTs-Hg(Ⅱ)-KI-PVA-XO体系在505 nm处共振光散射增强最大,且增强的强度与MTs的加入量呈线性关系,据此检测MTs.结果表明,检出限21.58 ng/mL,线性范围0.072~6.3 μg/mL,相关系数0.994 9,相对标准偏差3.67%~5.61%, 平均回收率95.6% (n=6).该方法用于测定尿样MTs结果满意.
