摘要:
A magnetic functionalised calix[4]arene composite consisting of Fe3O4 and calix[4]arene phosphonate derivative (CPD) was prepared through a facile self-assembly method. The composite was characterised by scanning electron microscopy (SEM), powder X-ray diffraction (PXRD) and Fourier transform infrared spectroscopy (FTIR). The as-synthesised Fe3O4/CPD composite was used to remove UVI from aqueous solutions under different conditions. Meanwhile, the adsorption isotherm, kinetics and thermodynamics were fitted and analysed. The results show that the Fe3O4/CPD composite may be a promising adsorption material for the separation and enrichment of UVI from aqueous solutions in the cleanup of environmental pollution. © 2019 CSIRO.

摘要:
目的 对我国原发性高血压患者血压控制的微信干预效果进行Meta分析.方法 检索中国学术文献总库、 维普中文科技期刊数据库、 万方数据知识服务平台等中文数据库以及PubMed、The Cochrane Library等外文数据库,收集有关微信干预对我国原发性高血压患者血压控制效果评价的随机对照试验的文献,发表时间为2011—2018年.根据研究目的筛选文献,并进行质量评价和信息提取,采用Stata12.0软件进行Meta分析.结果 检索到880篇文献,最终纳入11篇中文文献,包括1174例原发性高血压患者,试验组和对照组各587例.Meta分析结果显示,与常规健康教育相比,微信干预能有效降低原发性高血压患者的收缩压(WMD=14.77,95%CI:11.76~17.77)和舒张压(WMD=8.17,95%CI:5.67~10.67);亚组分析结果显示,微信干预时间越长,对原发性高血压患者收缩压和舒张压的降低效果越明显(P<0.05).结论 微信干预较常规健康教育对原发性高血压患者的血压控制效果更好.

Despite intensive research activities, there are still many major knowledge gaps over the potential adverse effects of titanium dioxide nanoparticles (TiO2-NPs), one of the most widely produced and used nanoparticles, on human cardiovascular health and the underlying mechanisms. In the present study, alkaline comet assay and cytokinesis-block micronucleus test were employed to determine the genotoxic potentials of four sizes (100, 50, 30, and 10 nm) of anatase TiO2-NPs to human umbilical vein endothelial cells (HUVECs) in culture. Also, the intracellular redox statuses were explored through the measurement of the levels of reactive oxygen species (ROS) and reduced glutathione (GSH) with kits, respectively. Meanwhile, the protein levels of nuclear factor erythroid 2-related factor 2 (Nrf2) were also detected by western blot. The results showed that at the exposed levels (1, 5, and 25 mu g/mL), all the four sizes of TiO2-NPs could elicit an increase of both DNA damage and MN frequency in HUVECs in culture, with a positive dose-dependent and negative size-dependent effect relationship (T100 < T50 < T30 < T10). Also, increased levels of intracellular ROS, but decreased levels of GSH, were found in all the TiO2-NP-treated groups. Intriguingly, a very similar manner of dose-dependent and size-dependent effect relationship was observed between the ROS test and both comet assay and MN test, but contrary to that of GSH assay. Correspondingly, the levels of Nrf2 protein were also elevated in the TiO2-NP-exposed HUVECs, with an inversely size-dependent effect relationship. These findings indicated that induction of oxidative stress and subsequent genotoxicity might be an important biological mechanism by which TiO2-NP exposure would cause detrimental effects to human cardiovascular health.

Concerns over the health risk of the widely distributed, commonly used silica nanoparticles (SiNPs) are increasing worldwide. Yet, up to now, there are still many major knowledge gaps over the potential adverse effects of SiNP exposure on human cardiovascular health and the underlying mechanisms. In this study, comet assay and micronucleus test were employed to determine the genotoxic potentials of four sizes (10, 25, 50, 100 nm) of SiNPs to human umbilical vein endothelial cells (HUVECs) in culture. The intracellular redox statuses were explored through the determination of the levels of reactive oxygen species (ROS) and reduced glutathione (GSH) with kits, respectively. The protein levels of nuclear factor erythroid 2-related factor 2 (Nrf2) were also detected by Western blot. 摘要:
目的系统评价个体化营养支持对鼻咽癌同期放化疗患者营养相关指标的影响。方法计算机检索中国知网、万方、维普、Cochrane Library、web of science、pubmed数据库,收集从建库起至2018年3月国内外发表的关于个体化营养支持对鼻咽癌同期放化疗患者营养相关指标影响且符合纳排标准的相关原始研究。应用stata12.0软件进行数据分析,采用I2检验评估各研究结果间的异质性,漏斗图定性评估发表偏倚。结果共纳入符合纳排标准的9篇随机对照研究,总样本量727例,实验组(个体化营养干预)361例,对照组(常规饮食)366例。结果显示,个体化营养治疗的总白蛋白[SMD=1.12, 95%CI(0.85, 1.39),P<0.001]、血红蛋白[SMD=0.81, 95%CI(0.59, 1.03), P<0.001]、白蛋白[SMD=0.61,95%CI(0.37, 0.85), P<0.001]、体重[SMD=0.67, 95%CI(0.47,0.86), P<0.001]、体重指数[OR=0.20, 95%CI(0.11, 0.35), P<0.001]均明显高于对照组。结论与常规饮食相比,个体化营养支持能有效改善鼻咽癌同步放化疗患者营养状况,进而减少患者营养不良发生率,从而提高患者的生活质量、治疗耐受力及疗效。

Concerns over the health risk of the widely distributed, commonly used titanium dioxide nanoparticles (nano-TiO2) are increasing worldwide. Yet, up-to-now, our understanding in their potential effects on the cardiovascular system is very limited and the toxicological mechanisms are still unclear. In the present study, the CCK-8 assay was performed to determine the cytotoxicity of four sizes (10, 30, 50, and 100 nm) of anatase nano-TiO2 on human umbilical vein endothelial cells (HUVECs) in culture, and the flow cytometry was employed to investigate the potential of these nano-TiO2 to induce the apoptosis of HUVECs. The apoptotic pathway was also probed through the determination of the protein expression and activation of p53, Bax, Bcl-2, caspases-9, -7, -3, and PARP by western blot. The results showed that at the administrative levels (1, 5, 25 μg/mL), all the four sizes of nano-TiO2 could significantly inhibit the viability of HUVECs and elicit significant apoptosis in them, compared with the negative control (P <.05, P <.01). Moreover, the apoptotic rates of HUVECs were increased respectively with the elevating levels and decreasing sizes of the administrative nano-TiO2, showing a clear dose- and size-dependent effect relationships. Interestingly, the increasing phosphorylation of p53, decreasing ratio of Bcl-2/Bax, and enhancing activation of the downstream proteins caspase-9, -7, -3, and PARP, were also observed with the decreasing sizes of the administrative nano-TiO2 in the western blot, indicating that the intracellular approach of apoptosis, the p53-caspase pathway, is the major way of the nano-TiO2-mediated apoptosis in HUVECs in culture and that the size is an important parameter that may determine the potential of nano-TiO2 to induce cellular response. In conclusion, these results suggested that high levels of nano-TiO2 exposure may pose potential risks to human cardiovascular health by inducing cardiovascular EC apoptosis. © 2018 Wiley Periodicals, Inc.

With the growing production and applications of silica nanoparticles (SiNPs), human exposure to these nanoparticles continues to increase. However, the possible hazards that SiNP exposure may pose to human cardiovascular system and the underlying mechanisms remain unclear. In the present study, the flow cytometry was employed to investigate the potential of four sizes (10, 25, 50, 100 nm) of SiNPs to induce the apoptosis of human umbilical vein endothelial cells (HUVECs) in culture. The apoptotic pathway was also explored through the determination of the protein expression and/or activation of p53, Bcl-2, Bax, caspases-9, -7, -3, and PARP by western blot. The results showed that all the four sizes of SiNPs could significantly elicit apoptosis in HUVECs at the tested concentrations (1, 5, 25 μg/mL), compared with the negative control (p < 0.05, p < 0.01). Moreover, the apoptotic rates were increased with the elevating levels and decreasing sizes of administrative SiNPs, showing both dose- and size-dependent effect relationships. Interestingly, the enhancing phosphorylation of p53 protein (Ser15), decreasing ratio of Bcl-2/Bax protein, and elevating activation of the downstream proteins, caspase-9, -7, -3 and PARP, were also observed with the decreasing sizes of tested SiNPs, indicating that the p53-caspase pathway is the main way of the SiNP-mediated apoptosis in HUVECs and that the size is an important parameter that determines the SiNPs' potential to induce cellular response. The SiNPs may cause significant apoptosis to HUVECs in culture by activating the p53-caspase pathway, and the size is an important parameter that determines their potential to elicit cellular response. © 2017 Elsevier Ltd

摘要:
P-carboxyphenyl azo calix[4]arene phosphate derivative (P-CACPD) was successfully synthesized using calix[4]arene as raw material. The P-CACPD was characterized by FT-IR and 1H-NMR. The effects of pH, extraction time and temperature on the extraction of U(VI) by P-CACPD were investigated. The results show that pH value, extraction time and temperature have an effect on the extraction of U(VI) by P-CACPD. When the initial pH is 5, temperature is 25 °C, extraction time is 2 h, the extraction rate of U(VI) by P-CACPD reaches the maximum. Both kinetics parameters and thermodynamic parameters of the extraction process were calculated. The calculated data indicate that the extraction process of uranium by P-CACPD is an exothermic spontaneous process, which well fits with the pseudo-second-order kinetic model. © 2018, Akadémiai Kiadó, Budapest, Hungary.

目的 系统评估和定量分析人类乳头瘤病毒(HPV)感染与肺癌的潜在关系.方法 相关文献源于中国知网、PubMed等数据库1979年1月至2015年4月的文献,提取文献中与HPV有关的数据,使用调整的OR值及采用固定或随机效应模型进行分析.结果 按照纳入、排除标准,选出有效研究22篇,合计病例1 645例,对照733例.合并结果显示HPV与肺癌有关(OR =5.53,95% CI:4.22~7.23,P<0.001);鳞癌、腺癌与之相似(鳞癌:OR =8.29,95% CI:5.23~13.15,P<0.001;腺癌:OR =4.70,95% CI:2.73~8.09,P<0.001).此外,HPV16和18型亦分别与肺癌相关(HPV16:OR =5.63,95%CI:3.37~9.38,P<0.001;HPV18:OR =4.05,95%CI:2.40~6.83,P<0.001).结论 HPV感染与肺癌有关,尤其是鳞癌和腺癌;HPV亚型中与肺癌有关的亚型主要为16和18型,但仍需进一步深入研究以证实.

摘要:
The p-tert-butyl calix[4]arene symmetrical sulfide derivatives (3a and 3b) were successfully synthesized using p-tert-butyl calix[4]arene as the raw material and were characterized by FT-IR. The effects of pH value, temperature, and contact time on the U(VI) extraction with compounds 3a and 3b from aqueous solution were systematically investigated. The results showed that the optimum pH value for uranium extraction was 4.0 and the extraction equilibration time was 90 min. It is also found that the pseudo-second-order kinetic model (R
2 > 0.999) was better fitted to the extraction process. The thermodynamic parameters of the enthalpy (ΔH
θ) were all negative, indicating that the extraction reaction were exothermic reaction.

Concerns have arisen about the health and environmental impacts of the increasing commercial use of silver nanoparticles (AgNPs). However, the toxic mechanisms and target tissues of AgNPs have not been fully defined. In this paper, we investigated the tissue toxicity of mice after intravenous administration of AgNPs at a single-dose of 0.2, 2 or 5 mg per kg (body weight), respectively. Biodistribution, endoplasmic reticulum stress, and oxidative stress were examined in mouse organs at eight hours after exposure. Stress markers, e.g. HSP70, BIP, p-IRE1, p-PERK, chop and xbp-1s proteins/genes, were significantly upregulated in a dose-dependent manner. In the liver, spleen, lung and kidney, high stress accompanied by apoptosis occurred. Low stress levels were observed in the heart and brain. Thus, it is proposed that the liver, spleen, lung and kidney are dominant target tissues of AgNP exposure. The lower stress and toxicity in the heart and brain were in agreement with lower AgNP accumulation. The present results demonstrated that AgNP exposure eventually resulted in permanent toxic damage by gradually imposing stress impacts on target organs. These findings highlight the potent applications of stress markers in future risk evaluation of silver nanoparticle toxicity.

A facile strategy has been developed to fabricate Cu(OH)2 supercages (SCs) as an artificial enzyme system with intrinsic peroxidase-mimic activities (PMA). SCs with high catalytic activity and excellent recyclability were generated via direct conversion of amorphous Cu(OH)2 nanoparticles (NPs) at room temperature. More specifically, the process that takes a single nanoparticle to a 3D supercage involves two basic steps. First, with addition of a copper-ammonia complex, the Cu2+ ions that are located on the surface of amorphous Cu(OH)2 NPs would evolve into a fine lamellar structure by coordination and migration and eventually convert to 1D nanoribbons around the NPs. Second, accompanied by the migration of Cu2+, a hollow cavity is generated in the inner NPs, such that a single nanoparticle eventually becomes a nanoribbon-assembled 3D hollow cage. These Cu(OH)2 SCs were then engineered as an artificial enzymatic system with higher efficiency for intrinsic PMA than the peroxidase activity of a natural enzyme, horseradish peroxidase. © 2015 American Chemical Society.

The aim of the study was to explore the interactions of human papilloma virus 16 (HPV16) E2 protein and Daxx. The localization or co-localization of PML and E2 with Daxx in Caski cells was observed by indirect immunofluorescence. The interaction of E2 and Daxx was analyzed by co-immunoprecipitation, Western-blot and yeast-two hybrid assays. In Caski cells the fluorescence of Daxx and PML was mainly distributed in the cytoplasm or nucleus, respectively, and in the align image their signals did not overlap. However, when the red signal of HPV16 E2 and the green signal of Daxx in the cytoplasm of Caski cells were merged, the yellow signal appeared. The yeast co-transformed with pGBKT7/Daxx and pGADT7/E2 or pGADT7/E2 TAD can grow on SD/-Trp-Leu-His and SD/-Trp-Leu-His-Ade plates. So Daxx is not colocated with PML but with HPV16 E2 mainly in the cytoplasm of Caski cells. On the base of the results one can propose that HPV16 E2, in particularly its transcription-activity domain (TAD), interacts with Daxx.

摘要:
A novel signal enhanced liquid crystal biosensor based on using AuNPs for highly sensitive DNA detection has been developed. This biosensor not only significantly decreases the detection limit, but also offers a simple detection process and shows a good selectivity to distinguish perfectly matched target DNA from two-base mismatched DNA.

目的:建立一种高灵敏度的共振荧光光谱法测定奶粉中三聚氰胺的新方法.方法:对B-R缓冲溶液-CPC表面活性剂介质中的甲酚紫高氯酸盐进行同步扫描,应用三维荧光和吸收光谱分析同步扫描光谱峰的共振荧光特性;基于三聚氰胺对甲酚紫高氯酸盐的共振荧光强度的抑制作用,建立阻抑共振荧光测定三聚氰胺的分析方法.结果:在9.6×10-8~ 2.8×10 -6mol/L浓度范围内,体系的△F值与三聚氰胺浓度(c,10 -6mol/L)之间具有良好的线性关系,线性回归方程为△F=106.59c - 5.44,相关系数r=0.9993.检出限为2.87×10-8 mol/L.奶粉样品测定的相对标准偏差为1.9%~2.7%,加标回收率为95.0%~103.1%.结论:方法灵敏,操作简便,分析成本低,用于奶粉中三聚氰胺的测定,结果满意.

目的:建立一种共振瑞利散射方法测定食品中三聚氰胺的新方法.方法:在pH =5.5的B-R缓冲溶液中,三聚氰胺与巴比妥在四氢呋喃介质中以非共价键结合聚集形成大分子,在λ=296 nm下,激发产生强烈的共振瑞利散射.体系散射强度增强值(△I)与三聚氰胺浓度呈现良好的线性关系,据此建立了共振瑞利散射法测定乳制品中三聚氰胺的新方法.结果:在优化的实验条件下,浓度为3.03×10-8 mol/L ~1.60×10-6mol/L时,体系的共振光散射强度(△I)值与三聚氰胺的浓度成线性关系,线性回归方程为△I=1594.4c+9.70(c为三聚氰胺的浓度,10 -6mol/L),相关系数r=0.9991,检出限为9.1×10-9 mol/L.实验对4.0×10-8、8.0×10-8、1.2×10-7 mol/L的三聚氰胺标准溶液分别进行11次平行测定,相对标准偏差分别为4.54%、3.31%、4.61%,样品加标回收率为95.0% ~97.5%.结论:方法精密度.准确度好,灵敏,操作简便,用于食品中三聚氰胺的测定,结果满意.

摘要:
A new and highly sensitive inhibitory kinetic fluorescence method for the determination of arsenic (III) has been established based on its inhibitory effect on the oxidation reaction of Acridine red (ADR) by KBrO<inf>3</inf> in sulphuric acid medium. The reaction has been followed by measuring the enhancement of fluorescence at 550 nm. It relies on the linear relationship where the change in the fluorescence (ΔF) versus added As(III) amounts in the range of 0-0.450 μgmL<sup>-1</sup> is plotted, under the optimum conditions. The sensitivity of the proposed method, i.e. the limit of detection, is 2.1 ×10<sup>-2</sup> ngmL<sup>-1</sup>. The method is featured with good accuracy and reproducibility for arsenic (III) determination. This method was successfully applied for the quantitative determination of arsenic (III) in food products samples, and the relative standard deviations and the recoveries were in ranges of 2.31-2.83% and 90.0-107.2%, respectively. A review of recently published catalytic or inhibiting kinetic methods for the determination of arsenic (III) has also been presented for comparison. Peptides, bound to the tegument of live Schistosoma japonicum schistosomula, were differentially screened by phage display in vitro using three rounds of reverse absorption and bio-panning. Three M13 phage peptides were isolated and identified by determination of their recovery rate, immunohistochemical localization, immunoblot analysis, and their anti-schistosomal effects in vivo and in vitro. Of the three, M13 phage peptide ZL4 (MppZL4, YSGLQDSSLRLR, 1.4 kDa, pI 8.8) bound to the tegument of mechanically transformed schistosomula and to other developmental stages of S. japonicum from the mammalian host. By contrast, MppZL4 did not bind to the surface of cercariae. To further examine its binding properties, MppZL4 was conjugated to Rhodamine B (RhB-YSGLQDSSLRLR, RhB-ZL4) and a peptide control (RhB-AIPYFSGILQWR, RhB-12P) was similarly synthesized. The binding capacities of RhB-ZL4 to the surface membrane of S. japonicum schistosomula in vitro and of S. japonicum adult worms in vivo were examined and revealed specificity for binding. When examined for anti-parasite activity, both MppZL4 and RhB-ZL4 exhibited a potent schistosomicidal effect in vitro. Further MppZL4 also affected the growth and development of schistosomula in vivo. These findings extend previous studies showing that phage display techniques can recover polypeptides that bind specifically to living schistosomes and, moreover, that these bound peptides have the potential to inhibit key physiological processes in these parasites. Our findings suggest further that ectogenic polypeptides, which can bind to the tegument of S. japonicum, might be adapted as vectors to deliver experimental probes and/or pharmacologically relevant compounds to the schistosome tegument, including drugs and immunological mediators. © 2011 2011 Elsevier B.V. In this work, a new method has been proposed to simultaneously determine the trace amount of malachite green and crystal violet from aqueous solution by spectrophotometry after cloud point extraction (CPE) using partial least squares regression. The optimal extraction and operating conditions, such as pH, reagents concentration and effect of time and temperature, and so on, have been investigated using the non-ionic surfactant Triton X-114. The maximum absorption wavelength for malachite green and crystal violet is 624 and 579 nm, respectively;linearity is obeyed in the range of 9.9-800 and 16-1000 ng mL<sup>-1</sup> with detection limit of 2.9 and 4.8 ng mL<sup>-1</sup>, and the root mean square error of prediction (RMSEP) are 0.0197 and 0.0343, respectively. The proposed method has been applied successfully to simultaneously determine the trace amount of malachite green and crystal violet in real matrix samples with the recoveries of 92.45-102.5%. ©2009 Elsevier B.V.
