摘要:
Objective: To test the effects of capsaicin on learning and memory ability in Alzheimer's disease (AD) mice. Results: Capsaicin (CAP) has no effect on spontaneous activity in APP/PS1 transgenic mice. CAP reduces the average escape latency and average swimming distance of Morris water maze in APP/PS1 transgenic mice. CAP adds academic record of Y maze in APP/PS1 transgenic mice. Conclusion: Capsaicin improves the function of learning and memory in AD mice.

Object: To observe the influence of silencing the expression of YB-1 gene by RNA interference cell on proliferation and migration of prostate cancer. Methods: The YB-1 siRNA was transfected into PC-3 cells at logarithmic growth by liposome, expression levels of YB-1 mRNA and protein were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Wesretn-blot, proliferation and invasion of PC-3 cell was detected by MTT assay and Transwell experiments. Results: After transfection for 48 h, compared with PC-3 cells non-transfected with plasmid, YB-1 mRNA levels and YB-1 protein levels in PC-3 cells transfected with recombinant plasmid pGenesil-1-YB-1-1 and recombinant plasmid pGenesil-1-YB-1-2 were significantly decreased (P< 0.05). YB-1 mRNA inhibition rates in PC-3 cells transfected with recombinant plasmid pGenesil-1-YB-1-1 and recombinant plasmid pGenesil-1-YB-1-2 were 36.23% and 39.42%, while YB-1 protein inhibition rates were 41.56% and55.33%. MTT results showed that the proliferation of PC-3 cells transfected with recombinant plasmid pGenesil-1YB- 1-1 and recombinant plasmid pGenesil-1-YB-1-2 was significantly decreased when compared with PC-3 cells non-transfected with plasmid (P< 0.05). Transwell migration experiments showed that the number of cells go through the membrane in PC-3 cells non-transfected, PC-3 cells transfected with pGenesil-1-YB-NC, PC-3 cells transfected with recombinant plasmid pGenesil-1-YB-1-1 and PC-3 cells transfected with recombinant plasmid pGenesil-1-YB-1-2, were(123.71+/-13.25),(135.73+/-14.18),(67.73+/-8.82) and (51.73+/-6.75) respectively (P<0.05). The migration of PC-3 cells was reduced by downregulating the expression of YB-1, also the invasiveness of PC-3 cells vitro was inhibited. Conclusions: cell proliferation and migration of prostate cancer was inhibited by RNA interference silencing the expression of YB-1 gene.

Retroviral transduction of cultured schistosomes was explored in our study. The vesicular stomatitis virus glycoprotein (VSVG)-pseudotyped pantropic retroviral vector pBABE-puro was modified to incorporate the human telomerase reverse transcriptase gene (hTERT) as a reporter gene under the control of retroviral long terminal repeat (LTR). Pseudotyped virions were employed to transduce Schistosoma japonicum (Sj) to investigate the utility of retrovirus mediated Sj transgenesis and to investigate the activity as human telomerase as a reporter transgene in schistosomes. The presence of transgene hTERT was detected by PCR. Furthermore, analysis of RNAs from transduced parasites, immunohistochemistry of thin sections and immunoblot analysis revealed expression of hTERT transgene in the transduced worms. These findings indicated that Sj could be effectively transduced by VSVG pseudotyped retrovirus carrying the hTERT gene.

摘要:
To test the hypothesis that concentration polarization of atherogenic lipids may occur in the arterial system and probe the effect of endothelial cells (ECs). A modifier with local stenosis was developed. The luminal surface LDL concentration at the distal of stenosis was analyzed with numerical stimulation and was measured by confocal microscopy. ECs were incubated with low density lipoprotein(LDL). Then, ECs were exposed to shear stress(15dyn/cm2) and F-actin was examined by fluorescence staining. Numerical stimulation showed that the luminal surface LDL concentration was higher than the bulk concentration. The relative luminal surface LDL concentration varied with the velocity and degree of stenosis. The experimental detection result was very good agreement with the numerical analysis. After treated with LDL, F-actin reorganization was observed. Under flow, the adhesion and retention of ECs were negatively relative to LDL concentration. Therefore provided concentration polarization of LDL occurs at the distal end of stenosis, and in turn effect the adhesion of ECs under flow.
