摘要:
采用水蒸气蒸馏法提取野菊花挥发油,利用气相色谱-质谱法( GC-MS)测定挥发油化学成分,同时结合直观推导式演进特征投影法解析重叠色谱峰,并采用总体积积分法进行定量。鉴定出66种化合物,占总含量的75.32%,主要成分为石竹烯氧化物、匙叶桉油烯醇、6-isopropenyl-4,8a-dimethyl-1,2,3,5,6,7,8,8a-octahydro-naph-thalen-2-ol和反式长松香芹醇。安徽产野菊花挥发油的化学成分主要为单萜烯类、倍半萜烯类及其含氧衍生物等。采用GC-MS 联用技术结合直观推导式演进特征投影法分析安徽产野菊花挥发油,提高了定性定量结果的准确性。

摘要:
采用两步法合成磁性胺肟基功能化CMC(简称PAO/CMC/Fe_3O_4),用批示法研究了PAO/CMC/Fe_3O_4对U(VI)的吸附行为,考察了pH值、吸附时间、吸附剂用量等对吸附效果的影响。结果表明,PAO/CMC/Fe_3O_4对U(VI)的吸附容量大,速度快,最大吸附容量119.21 mg/g,30 min左右达到吸附平衡,平衡数据符合Langmuir等温方程,符合伪二阶动力学吸附模型,说明PAO/CMC/Fe_3O_4对U(VI)的化学吸附过程是速率控制步骤。

摘要:
以Cu/ZSM_5为催化剂,在新型微波催化反应器上进行了微波协同催化分解NO反应.初步探讨了微波协同催化分解NO反应的宏观动力学,并与常规加热条件下催化分解NO反应的宏观动力进行了对比.计算结果表明:在微波辐射条件下,该反应的活化能为14.6 kJ/mol.而常规加热条件下,该反应的活化能为75.6 kJ/mol.微波辐射条件下该反应活化能仅仅是常规加热条件下该反应活化能的1/5.177.这说明微波作用不仅具有致热效应,而且大大降低了反应活化能,具有微波催化效应.

摘要:
Uranium is harmful to human health due to its radiation damage and the ability of uranyl ion (UO22+) to interact with various proteins and disturb their biological functions. Cytochrome b<inf>5</inf> (cyt b<inf>5</inf>) is a highly negatively charged heme protein and plays a key role in mediating cytochrome c (cyt c) signaling in apoptosis by forming a dynamic cyt b <inf>5</inf>-cyt c complex. In previous molecular modeling study in combination with UV-Vis studies, we found that UO22+ is capable of binding to cyt b <inf>5</inf> at surface residues, Glu37 and Glu43. In this study, we further investigated the structural consequences of cyt b<inf>5</inf> and cyt c, as well as cyt b<inf>5</inf>-cyt c complex, upon uranyl binding, by fluorescence spectroscopic and circular dichroism techniques. Moreover, we proposed a uranyl binding site for cyt c at surface residues, Glu66 and Glu69, by performing a molecular modeling study. It was shown that uranyl binds to cyt b<inf>5</inf> (K<inf>D</inf> = 10 μM), cyt c (K<inf>D</inf> = 87 μM), and cyt b <inf>5</inf>-cyt c complex (K<inf>D</inf> = 30 μM) with a different affinity, which slightly alters the protein conformation and disturbs the interaction of cyt b<inf>5</inf>-cyt c complex. Additionally, we investigated the functional consequences of uranyl binding to the protein surface, which decreases the inherent peroxidase activity of cyt c. The information of uranyl-cyt b <inf>5</inf>/cyt c interactions gained in this study likely provides a clue for the mechanism of uranyl toxicity. ©2013 Elsevier B.V. All rights reserved.

摘要:
Forkhead box protein A1 (FoxA1) is a transcription factor that is involved in embryonic development and cell differentiation. In this study, we show that hydrogen peroxide (H2O2) treatment upregulated expression of FoxA1 and UCP2 in the A549 cell line. Overexpression of FoxA1 by full-length complementary DNA reduced UCP2 expression, while silencing of FoxA1 expression by small interfering RNA significantly increased UCP2 levels. FoxA1 binds to a site from -919 to -913 bp relative to the UCP2 transcription start site. The overexpression of FoxA1 promoted the DNA binding activity and attenuated the transcription of UCP2 promoter as shown by electromobility shift, chromatin immunoprecipitation assays, and luciferase reporter assay. These data indicate an important role of FoxA1 in regulating expression of UCP2. © 2013 Cell Stress Society International.

摘要:
A novel highly sensitive and specific electrochemical biosensor for detecting uranium based on specific Deoxyribozymes and gold nanoparticles (AuNPs) is reported. In this work, AuNPs provide excellent electrochemical signal transduction and a large surface area for immobilising numerous Deoxyribozymes, so a low detection limit of 3.24ngL(-1) uranium and a good linear relationship over the range 5.94-35.1ngL(-1) (r=0.994) were obtained. The proposed biosensor presents high specificity and selectivity for uranium and is not affected by other metal ions. Thus, the biosensor protocol offers good selectivity, rapid speed and operational convenience for detection uranium in liquid waste.

摘要:
A computer simulation approach was developed to screen functional monomers for the rational design of molecularly imprinted polymers (MIPs). The proposed approach is based on a comparison of the binding energy of complexes, which are formed by the interaction between a template molecule and various functional monomers. According to the results of theoretical calculations, MIPs using thymopentin (TP5) as a template were prepared by a solution polymerization method, in which 2-methylacrylamide, N,N′-methylenebisacrylamide and poly(methylacrylic acid)/SiO2 were used as the functional monomer, cross-linker and support, respectively. Solid-phase extraction experiments were performed, and static and dynamic binding properties of the synthesized MIP (i.e. TP5-MIP) for TP5 were studied. Results show that TP5-MIP exhibits excellent binding affinity and specific selectivity for the template molecule (i.e. TP5). © 2014, Multi-Science Publishing Co. Ltd. All rights reserved.

摘要:
目的:探讨兰索拉唑对幽门螺杆菌(helicobacter pylori,Hp)感染的胃上皮细胞的体外抗炎效应。方法体外培养胃上皮细胞系AGS,加入兰索拉唑和培养的Hp感染不同时间。ELISA检测IL-1β和IL-8的产生,Western blot检测NF-κB p 65亚基的核转位以及红素氧合酶1(Heme oxygenase-1, HO-1)蛋白表达水平。结果兰索拉唑孵育8 h后,能显著抑制Hp诱导的IL-1β和IL-8。同时,兰索拉唑能抑制NF-κB p 65亚基的核转位,并能上调HO-1蛋白的表达。结论兰索拉唑可能通过抑制NF-κB的激活,上调HO-1的表达而发挥对胃上皮细胞的抗炎作用。

摘要:
代谢组学是功能基因组学和系统生物学研究不可或缺的重要组成部分,是通过考察生物体系受刺激或扰动前后代谢产物的动态变化,研究生物体系的代谢网络的一种技术。应用代谢组学高通量、整体性的研究思路来理解中药的作用过程,与中医药的整体、辩证观点是一致的。代谢组学已成为中药研发的一个重要途径和手段,为中药现代化在技术上提供巨大支持,有助于为中药现代化研究寻找更多有效的突破口。本文在前人综述的基础上,着重概括了中药代谢组学研究方法近3年来在中药有效物质基础和作用机制、药物作用模型的鉴别和确证、毒性研究和中药安全性评价等方面的应用情况,同时展望了代谢组学方法所面临的机遇和挑战。

摘要:
结合高等教育‘以人为主体、促进人的全面发展’要求,提出了“以教为纲,以学为主,以选为标”的教学管理理念。并进一步探索了以‘限选-修课’与‘自选-修课’代替现有‘必修课’与‘选修课’课程名称称谓模式的可行性。讨论了以教学名师构建课程地图、学生构建符合自身条件的专业知识树,从以教为主向以学为主转变的课程教学模式;分析了课程体系与学生自身职业生涯规划之间的联系,阐述了学生依据课程地图结合自身职业生涯规划进行自主选课的意义。“以学生为主体”的课程体系构建,既能使其知晓课程体系的完整性与重要性,又能促使学生形成专业知识迁移的能力,最终实现终身学习、自我完善的全面发展。

摘要:
红外热成像仪同步跟踪橡胶弹性体材料的循环形变过程,几种特殊的热效应特征能被观察到:(1)拉伸时明显的热弹转变效应,即橡胶试样在拉伸时要经历一个明显的温度降低,然后温度才逐渐升高;(2)试样初始拉伸-回复时,温度变量具有部分可逆性,而随后的拉伸-回复温度变化几乎完全可逆;(3)拉伸时温度升高具有速率依赖性,即拉伸速率越快,温升越高。热效应特征能反映形变分子聚集结构的变化,显然,橡胶的受力形变过程除构象变化以外,还具有其它特殊的分子结构变化。

摘要:
目的建立水中痕量镉(Ⅱ)的荧光素-曙红Y能量转移荧光猝灭测定法。方法取0.50ml的水样置于5.0 ml的比色管中,分别加入1×10~(-4) mol/L荧光素溶液0.30 ml,1×10~(-5) mol/L曙红Y溶液0.10 ml,BR缓冲液(pH 6.5)0.50 ml,定容后在502 nm处测定溶液荧光强度(F)值,以内标法定量。结果在3.43×10~(-7) ~1.246×10~(-5) mol/L的线性范围为,该方法所得镉(Ⅱ)的线性回归方程为ΔF=14.221+546.81×10~(-5)c,r=0.999 5。该方法的检出限为1.03×10~(-8) mol/L,平均回收率为94.53%~98.15%,RSD为2.44%~4.67%。结论该方法操作简便、灵敏、选择性好,适用于水中镉(Ⅱ)的测定。

摘要:
Two series of 7-O-modified chrysin derivatives were prepared from 7-O-carboxymethyl chrysin(2a), 7-O-carboxypropylchrysin(2b) and short-chain alcohols by using 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride(EDCI), N-hydroxybenzotriazole(HOBt) and 4-dimethylamiopryidine(DMAP) as coupling reagents. Taking cisplatin as a reference substance, their anti-proliferative activities in vitro against human gastric carcinoma MGC-803 cells were evaluated by the standard 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide(MTT) method. The results showed that among the compounds tested, compound hepty 4-(5-hydroxy-4-oxo-2-phenyl-4H-chromen-7-yloxy) acetate(3f) displayed the most potent growth-inhibitory effect on MGC-803 cells with half maximal inhibitory concentration(IC50) value of 3.23 μmol/L. The preliminary mechanism of inhibitory effect of compound 3f was also detected by flow cytometry(FCM), and the compound exerted anticancer activity via inducing the apoptosis of MGC-803 cells in a dose dependent manner, which suggested that compound 3f would be a potential anti-cancer agent. © 2014, Jilin University, The Editorial Department of Chemical Research in Chinese Universities and Springer-Verlag GmbH.

Room-temperature ionic liquids (RTILs) have recently received increasing attention as solvent alternatives for possible application in spent fuel reprocessing, particularly in the extraction of metal ions from high-level radioactive aqueous waste, due to their unique physical and chemical properties. Herein, the solvent extraction of the uranyl ions (UO2 2+) was performed using N,N- diethyl-N,N N-di(para)tolyl-dipicolinamide (Et(p)TDPA) as the extractant in a commonly used ionic liquid 1-butyl-3- methylimidazolium hexafluorophosphate ([C4mim][PF6]). The effects of vortexing time, phase ratio and the concentration dependence of Et(p)TDPA, nitric acid and sodium nitrate on the extraction were studied in detail. The extraction mechanismwas deduced based on the slope analysis and UV-vis measurement. The distribution ratio of U from 3mol/Lnitricacidby0.3mol/LEtTDPA/C4mimPF6 is found to be almost 100. Conventional log-log plot analysis of the extraction equilibrium data suggests that the ions are extracted as a complex in 2 : 3 ratio of UO2 2+ to extractant, and the extractionmost likely occurs by a cationexchange mode since the concentration of C4mim+ in the aqueous phase increases linearly with the percent extraction of UO2 2+ evidenced by UV-vis measurement. This work promises to provide new efficient media based on RTILs for separation of uraniumfromthe radioactive aqueous waste. © 2014 Oldenbourg Wissenschaftsverlag GmbH.
