摘要:
Bacterial infection is one of the most common problems after orthopedic implant surgery. If not prevented, bacterial infection can result in serious and life threatening conditions such as osteomyelitis. Thus, in order to reduce chances of such serious complication, patients are often subjected to antibiotic drug therapy for 6-8 weeks after initial surgery. The antibiotics are systemically delivered either intravenously, intramuscularly or topically. Systemic antibiotic delivery entails certain drawbacks such as systemic toxicity and limited bioavailability. Further, in order for the drug to be effective at the site of implantation, high doses are required, which can result in undesired side effects in patients. Thus, local antibiotic therapy is the preferred way of administering drugs. To that end, we have developed Ti-Al intermetallic compounds porous material for local delivery of antibiotics off-implant at the site of implantation. The Ti-Al intermetallic compounds porous materials were fabricated with reactively synthesizing. The fabrication strategies allow us to precisely control the dimension of aperture, thus enabling us to load different amounts of drugs and control the release rates. In this work we have fabricated Ti-Al intermetallic compounds porous material with 40 similar to 200 nm and 2 similar to 5 mu m apertures. We have loaded these tubes with 180, 360 and 540 mu g of penicillin G. The penicillin G release kinetics from the apertures and its effect on Staphylococcus aureus ssp. anaerobius (S. aureus) adhesion were investigated. Further, an osteosarcoma cell line called MG-63 was cultured on penicillin G-loaded aperture to evaluate the effect of Ti-Al intermetallic compounds porous material on cell functionality. Our results indicate that we can effectively fill the apertures with the drug and the drug eluting apertures significantly reduced bacterial adhesion on the surface. Also, there is enhanced MG-63 differentiation on apertures filled with penicillin G.

摘要:
该文采用便利取样法,选取2013级教学一班81名同学作为试验组,教学二班85名同学作为对照组。试验组采用逆向案例教学法施教,对照组采用传统教学法施教。教学方法的优劣通过比较测试成绩、调查学生对逆向案例教学方法的感受来评价。结果表明,应用逆向案例教学法学生的期中、期末成绩高于传统教学法的学生,并且学生认为逆向案例教学法可以提高学习兴趣和主动参与教学过程的积极性。在此基础上,该文结合逆向案例教学方法在分析化学课程教学中的应用及效果,提出了几点思考,认为逆向案例教学法教学效果较为理想,可以在更多的课程教学中进行推广。

摘要:
In this paper, we report a resonance light scattering (RLS) method for the determination of fructose bisphosphates (FBPs) in water solution using fructose 1,6-bisphosphate (F-1,6-BP) as a model analyte without the procedure of extracting target analyte. The method used a type of modified gold nanoparticles (GNPs) as optical probe. The modified GNPs are uranyl-salophen-cysteamine-GNPs (U-Sal-Cy-GNPs) which are obtained through the acylation reaction of carboxylated salophen with cysteamine-capped GNPs (Cy-GNPs) to form Sal-Cy-GNPs and then the chelation reaction of uranyl with tetradentate ligand salophen in the Sal-Cy-GNPs. A FBP molecule is used easily to connect two U-Sal-Cy-GNPs to cause the aggregation of the GNPs by utilizing the specific affinity of uranyl-salophen complex to phosphate group, resulting in the production of strong RLS signal from the system. The amount of FBPs can be determined through detecting the RLS intensity change of the system. A linear range was found to be 2.5 to 75 nmol/L with a detection limit of 0.91 nmol/L under optimal conditions. The method has been successfully used to determine FBPs in real samples with the recoveries of 96.5-103.5 %. © 2015 Springer-Verlag Berlin Heidelberg.

摘要:
Understanding uranium-protein interaction is important for revealing the mechanism of uranyl ion (UO_2~(2+)) toxicity. In this study, we investigated the interaction between UO_2~(2+) and a quadruple mutant of cytochrome b5 (E44/48/56A/D60A cyt b_5, namely 4A cyt b_5) by spectroscopic approaches. The four mutated negatively-charged surface residues of cyt b_5 have been considered to be the interactive sites with cytochrome c (cyt c). Also, we studied the interaction between UO_1~(2+) and the protein-protein complex of 4A cyt b5-cyt c. The results were compared to the interaction between UO_2~(2+) and cyt b5, and the interaction between cyt c and cyt b_5-cyt c complex, from previous studies. It was found that the interaction of UO_2~(2+)-cyt b_5, i.e., uranyl ion binding to cyt b_5 surface at Glu37 and Glu43 as previously proposed by molecular modeling, is regulated by both surface mutations of cyt b_5 and its interacting protein partner cyt c. These provide valuable information on metal-protein-protein interactions and clues for understanding the mechanism of uranyl toxicity.

A heme-protein cross-link is a key post-translational modification (PTM) of heme proteins. Meanwhile, the structural and functional consequences of heme-protein cross-links are not fully understood, due to limited studies on a direct comparison of the same protein with and without the cross-link. A Tyr-heme cross-link with a C-O bond is a newly discovered PTM of heme proteins, and is spontaneously formed in F43Y myoglobin (Mb) between the Tyr hydroxyl group and the heme 4-vinyl group in vivo. In this study, we found that with an additional distal His29 introduced in the heme pocket, the double mutant L29H/F43Y Mb can form two distinct forms under different protein purification conditions, with and without a novel Tyr-heme cross-link. By solving the X-ray structures of both forms of L29H/F43Y Mb and performing spectroscopic studies, we made a direct structural and functional comparison in the same protein scaffold. It revealed that the Tyr-heme cross-link regulates the heme distal hydrogen-bonding network, and fine-tunes not only the spectroscopic and ligand binding properties, but also the protein reactivity. Moreover, the formation of the Tyr-heme cross-link in the double mutant L29H/F43Y Mb was investigated in vitro. This study addressed the key issue of how Tyr-heme cross-link fine-tunes the structure and functions of the heme protein, and provided a plausible mechanism for the formation of the newly discovered Tyr-heme cross-link. © The Royal Society of Chemistry 2015.

Two metal-free organic sensitizers containing two N,N-diethylaniline (DEA) moieties and a twisted 1,1,2,2-tetraphenylethene (TPE) structure, dye SD with one anchoring group and dye DD with two anchoring groups, were synthesized and applied in dye-sensitized solar cells (DSSCs). The introduction of a nonplanar TPE structure was used to form a series of propeller-like structures and reduce the tendency of dyes to randomly aggregate on TiO2 surface, but without importing an aggregation-induced emission (AIE) property. The thermal stabilities, UV-vis absorption spectra, electrochemical properties, and photovoltaic parameters of DSSCs with these two dyes were systematically studied and compared with each other. The overall conversion efficiencies (eta) of 4.56% for dye SD and 6.08% for dye DD were obtained under AM 1.5 G irradiation.

A resonance light scattering (RLS) method for the detection of fluoride ions in aqueous solution is reported in this paper. Uranyl-bis-salophen (UBS), a binuclear uranyl complex prepared by the reaction of uranyl with a ditopic tetradentate Schiff base ligand bis-salophen, was used for the experiment. It is found that after UBS self-assembled with pyrophosphate to form a uranyl coordination supramolecular polymer (SP), the system produced a strong RLS signal. When fluoride ions coexisted with UBS, the formation of SP was inhibited due to which the fluoride ions occupied the coordination sites in UBS. This resulted in RLS quenching. Based on these findings, we established a RLS method for the detection of fluoride ions in aqueous solution. Under optimal conditions, the linear range was found to be 0.005 to 1.5 nmol mL-1 with a detection limit of 0.0028 nmol mL-1. The method was applied to determine fluoride ions in environmental water samples with relative standard deviations of 1.3 to 3.1% and recoveries of 98.7-101.3%. © the Partner Organisations 2014.

摘要:
Protein nitration is an important post-translational modification regulating protein structure and function, especially for heme proteins. Myoglobin (Mb) is an ideal protein model for investigating the structure and function relationship of heme proteins. With limited structural information available for nitrated heme proteins from experiments, we herein performed a molecular dynamics study of human Mb with successive nitration of Tyr103, Tyr146, Trp7 and Trp14. We made a detailed comparison of protein motions, intramolecular contacts and internal cavities of nitrated Mbs with that of native Mb. It showed that although nitration of both Tyr103 and Tyr146 slightly alters the local conformation of heme active site, further nitration of both Trp7 and Trp14 shifts helix A apart from the rest of protein, which results in altered internal cavities and forms a water channel, representing an initial stage of Mb unfolding. The computational study provides an insight into the nitration of heme proteins at an atomic level, which is valuable for understanding the structure and function relationship of heme proteins in non-native states by nitration. © 2014 Elsevier Ltd. 摘要:
The novel quantum topological indices PY<inf>1,2</inf>were derived from molecular structure combined with the effect of atom space, the character of bonding atoms (such as equilibrium electro-negativity) and the branching effect between the atoms. The quantitative structure-property relationships (QSPRs) were proposed between PY<inf>1,2</inf>and the thermodynamic properties (<inf>f</inf>H<sup>θ</sup>, <inf>f</inf>G<sup>θ</sup>and Δ<inf>f</inf>G<inf>R</inf><sup>θ</sup>) of phenoxazine (Phx) and 135 kinds of polychlorinated phenoxazines (PCPXs), by Multiple linear regression (MLR) analysis method. The high-quality prediction models were evidenced by the correlation coefficient R, the standard error of estimate S, the Fisher statistic value and the cross-validated correlation coefficient RCV. With the new QSPR model, we are able to predict a wide range of thermodynamic properties of an extensive number of molecules. And the model is statistically significant and shows good stability for data variation as tested by the leave-one-out cross-validation (LOO-CV). ©2014 Published by Elsevier B.V.

Electrochemical behaviors of U(VI) in 1-butyl-3-methylimidazolium chloride (C4MimCl) with various water contents investigated by chronopotentiometry and cyclic voltammetry. The electrochemical reduction of U(VI) was identified to follow two processes: a lower valence intermediate U(V) was initially formed at the potential of ca. -0.2 V(vs. Ag wire). Then, further deposition of UO2 was followed at around -0.8 V. Little amount of water (1–4 wt%) in C4MimCl, however, has an effect on the U(VI) reduction by changing the current density of the redox reaction and the diffusion coefficient of U(VI) in C4MimCl. The deposited product by potentiostatic electrolysis on the surface of stainless steel electrode was characterized by the scanning electron microscopy (SEM), energy dispersive X-ray (EDX) and X-ray diffraction (XRD) methods. Although the electrodeposited black film was amorphous, the electrochemical reduced product of U(VI) can be still confirmed to be UO2 by XRD after the crystallization of the amorphous deposits at 1,073 K in nitrogen atmosphere. © 2014,Akadémiai Kiadó, Budapest, Hungary

摘要:
采用密度泛函理论(DFT)中的B3LYP方法,在6-31G、6-31G~(**)、6-311G~(**)三种基组水平上,对三种铀酰-Salophen配合物的分子结构进行优化,计算了它们的振动频率和化学位移,并对得到的几何构型、电子结构、红外光谱、热力学性质和化学位移进行了讨论分析。

亚洲玉米螟地理种群不同食物及温度下实验种群的蛹重研究

摘要:
为明确亚洲玉米螟Ostrinia furnacalis (Guenée)不同地理自然种群及不同食物和不同温度条件下实验种群蛹重的差异,对云南不同玉米种植区亚洲玉米螟自然种群、不同食料及不同温度下饲养的实验种群的雌、雄蛹重进行了研究.结果表明:云南昭通、会泽、宣威、寻甸、建水、玉溪、元江和陇川县不同地理种群雌蛹和雄蛹重量之间均存在显著性差异,从同一地理来源的雌蛹、雄蛹重量来看,雌蛹重量明显大于雄蛹重量.室内21℃、25℃和29 ℃下饲养的实验种群雌蛹重量均显著高于雄蛹重量,21 ℃与25℃下雌蛹、雄蛹重量大于29 ℃的雌蛹、雄蛹重量,而21℃和25℃下实验种群雌、雄蛹重无显著差异.室内常温条件下人工饲料饲养种群与玉米秆饲养种群雌蛹、雄蛹重量无显著差异.综合以上结果,亚洲玉米螟雌蛹重大于雄蛹,且随着不同地理来源而存在一定差异,而室内利用人工饲料和玉米秆饲养种群的雌、雄蛹重间无显著差异,在21-25℃条件下种群雌雄蛹重量大于29℃条件下雌雄蛹重量,该结果将为亚洲玉米螟对环境的适应性及其机理研究提供理论依据.

摘要:
A resonance light scattering (RLS) method for the direct detection of uranium (VI) or uranyl in aqueous solution without separation procedure has been reported in this paper. Sulfo-salophen, a water-soluble tetradentate Schiff base ligand of uranyl, reacted with uranyl to form a complex. The complex reacted further with oxalate to form supramolecular dimer with large molecular volume, resulting in a production of strong RLS signal. The amount of uranium (VI) was detected through measuring the RLS intensity. A linear range was found to be 0.2-30.0 ng/mL under optimal conditions with a detection limit of 0.15 ng/mL. The method has been applied to determine uranium (VI) in environmental water samples with the relative standard deviations of less than 5 % and the recoveries of 98.8-105.8 %. The present technique is suitable for the assay of uranium (VI) in environmental water samples collected from different sources. © 2014 Akadémiai Kiadó.

以无机非金属材料专业为例,指出了当前工程教育人才培养模式的一些不足。针对节能环保及产业转型对无机非金属材料专业发展提出的新要求,变革当前培养模式:通过多元化培养方案,完善教材体系,强化专业选修课的教学,实现学生的层次化个性发展;通过科技孵化平台/实验教学平台/创新平台/重点实验室的建设,满足实习、实践教学的需要,为产业转型背景下的多元化高级技术人才培养提供良好的实验、实习、实践教学条件和环境。

“分析化学”是化学工程与工艺等专业的必修学科基础课程,在工科人才培养中起着重要作用。目前中外教育交流日益频繁,教育国际化趋势明显,如何合理借鉴海外先进教学经验,是一个值得探索的课题。以所在地方高校为例,坚持以学生为中心,进行“分析化学”小组讨论式双语教学模式改革,将课内与课外、科学与人文、教学与研究结合起来,形成独具特色的卓越工程师人才培养模式,满足时代发展的需求,为实现国际化人才培养目标打下良好基础。

摘要:
Uranium is harmful to human health due to its radiation damage and the ability of uranyl ion (UO22+) to interact with various proteins and disturb their biological functions. Cytochrome b<inf>5</inf> (cyt b<inf>5</inf>) is a highly negatively charged heme protein and plays a key role in mediating cytochrome c (cyt c) signaling in apoptosis by forming a dynamic cyt b <inf>5</inf>-cyt c complex. In previous molecular modeling study in combination with UV-Vis studies, we found that UO22+ is capable of binding to cyt b <inf>5</inf> at surface residues, Glu37 and Glu43. In this study, we further investigated the structural consequences of cyt b<inf>5</inf> and cyt c, as well as cyt b<inf>5</inf>-cyt c complex, upon uranyl binding, by fluorescence spectroscopic and circular dichroism techniques. Moreover, we proposed a uranyl binding site for cyt c at surface residues, Glu66 and Glu69, by performing a molecular modeling study. It was shown that uranyl binds to cyt b<inf>5</inf> (K<inf>D</inf> = 10 μM), cyt c (K<inf>D</inf> = 87 μM), and cyt b <inf>5</inf>-cyt c complex (K<inf>D</inf> = 30 μM) with a different affinity, which slightly alters the protein conformation and disturbs the interaction of cyt b<inf>5</inf>-cyt c complex. Additionally, we investigated the functional consequences of uranyl binding to the protein surface, which decreases the inherent peroxidase activity of cyt c. The information of uranyl-cyt b <inf>5</inf>/cyt c interactions gained in this study likely provides a clue for the mechanism of uranyl toxicity. ©2013 Elsevier B.V. All rights reserved.
