Bladder cancer (BC) is a familiar malignancy with high morbidity and mortality. The effect of treatment is unsatisfactory after the metastasis and invasion of BC. Hence, more studies should be carried out to explore the metastasis of BC. RT-qPCR or/and western blot was conducted to evaluate miR-494-3p, KLF9, and RGS2 expression. Cell proliferation and invasion were estimated by MTT assay and transwell assay, respectively. Cell migration was tested by wound healing assay and transwell assay. Dual-luciferase reporter gene assay was employed to validate the interplay between miR-494-3p and KLF9 mRNA. The interaction between KLF9 and RGS2 promoter was verified using dual-luciferase reporter gene assay and chromatin immunoprecipitation (ChIP) assay. miR-494-3p expression was upregulated, whereas KLF9 and RGS2 were downregulated in BC cells. miR-494-3p inhibition was competent to limit the growth of BC cells. KLF9 knockdown abolished the miR-494-3p depletion-mediated inhibitory growth of BC cells. Mechanistically, we found that KLF9 was a downstream gene of miR-494-3p and could bind to the promoter region of RGS2 to promote the expression of RGS2. Moreover, RGS2 knockdown abrogated the suppressive effects of miR-494-3p knockdown on the proliferation, migration, and invasion of BC cells. Notably, miR-494-3p inhibition obstructed the tumor growth in nude mice. miR-494-3p silencing inhibited the progression of BC by regulating the KLF9/RGS2 axis in vitro and in vivo, which laid the foundation for experiments of miR-494-3p in BC and provided therapeutic targets for BC.

Objective. To explore the emergency nursing countermeasures and nursing experience of patients with primary liver cancer nodule rupture and hemorrhage. Methods. 30 patients with primary liver cancer nodule rupture and hemorrhage treated in our hospital since January 2020 after the implementation of emergency nursing countermeasures were selected as the observation group, and another 30 patients with primary liver cancer nodule rupture and hemorrhage treated in our hospital before January 2020 were selected as the control group. The control group received basic nursing intervention, while the observation group received emergency nursing measures. The hemoglobin level, blood oxygen saturation monitoring value, and partial pressure of oxygen of patients with hemorrhagic shock due to nodular rupture of primary liver cancer were compared between the two groups at admission and after nursing care. All indexes of patients during the perioperative period were recorded. The incidence of complications, mortality, and nursing satisfaction rates of the patients' families were compared between the two groups. Results. After nursing care, the observation group's patients' hemoglobin level, blood oxygen saturation monitoring value, and partial pressure of oxygen were higher than those of the control group's patients (P < 0.05). The intraoperative bleeding volume, shock correction time, and discharge time of patients in the observation group were lower than those of patients in the control group (P < 0.05). The incidence of complications and mortality in the observation group was significantly lower than those in the control group (P < 0.05). The nursing satisfaction rate of patients in the observation group was higher than that of the control group (P < 0.05). Conclusion. The results of emergency nursing intervention in patients with primary liver cancer rupture and hemorrhage are reliable, which can significantly improve perioperative indicators of patients, reduce complications and mortality, improve nursing satisfaction, and effectively shorten the hospital stay of patients.

Vascular smooth muscle cells (VSMCs), the main cells constructing blood vessels, are important in the regulation of the pathophysiology of vascular systems; however, relatively few studies have investigated the influence of nanomaterials (NMs) on VSMCs. In this study, we found that the interaction between graphene oxide and human VSMCs led to the cytotoxicity and morphological changes of cells. Because transcriptomic data suggested that graphene oxide decreased anti-viral signaling pathways via decreasing Toll-like receptor 3 (TLR3), we further verified that graphene oxide decreased interferon induced protein with tetratricopeptide repeats 1 (IFIT1) and the radical S-adenosyl methionine domain containing 2 (RSAD2), and TLR3-downstream genes involved in anti-viral responses. Due to the involvement of RSAD2 in lipid dysfunction, we also verified that graphene oxide disrupted lipid homeostasis and increased adipose triglyceride lipase (ATGL). Adding TLR3 agonist polyinosinic:polycytidylic acid (Poly IC) partially increased TLR3-downstream protein interleukin-8 (IL-8) and some lipid classes, particularly lysophosphatidylcholine (LPC) and lysophosphatidylethanolamine (LPE), in graphene oxide-exposed VSMCs. In mice receiving repeated intravenous injection of graphene oxide, significantly decreased TLR3, IFIT1 and RSAD2 but increased ATGL proteins were observed in aortas. We conclude that graphene oxide altered anti-viral signaling pathways and lipid metabolism via decreasing TLR3 in VSMCs.

py g Small nucleolar RNA host gene 16 (SNHG16) participates in some cancers as microRNA (miR)-1303 is also Delivered by Ingenta reported to function on proliferation of various cancer cells. This study aimed to assess SNHG16 and miR-1303's role in renal cell carcinoma (RCC) and its underlying mechanism. RT-qPCR was used to determine SNHG16 and miR-1303 expression in RCC cells (A498, 786-O, ACHN and OS-RC-2) and normal kidney epithelial cells (HK-2). Functional experiment was established to detect the role of miR-1303. After synthesis of nanoparticles carrying miR-1303 and transfection, CCK-8 method and assays were used to evaluate cell growth and apoptosis. The target genes for miR-1303 were predicted using bioinformatics software, and binding of miR-1303 to SNHG16 was evaluated using the dual-luciferase reporter assay. miR-1303 was lowly expressed in RCC cells, with lowest level in A498 (p < 0.05). Overexpressed miR-1303 significantly reduced proliferation ability of RCC cells and induced apoptosis (p < 0.05). Besides, transfection of NP carrying miR-1303 mimic resulted in dramatically decreased migrated cells when reducing the expression of SNHG18 mRNA. The presence of NPs strengthened the inhibitory effect of miR-1303 on RCC. Furthermore, the miR-1303+ SNHG16-WT co-transfection group had lower relative luciferase activity compared with miR-1303+ SNHG16-MUT co-transfection group (p < 0.05). miR-1303 was down-regulated in RCC and NP delivery of miR-1303 inhibited RCC cell proliferation and differentiation through regulation of SNHG16. These findings suggest miR-1303 may become a potential molecular target for RCC.

Persistent infection of Chlamydia trachomatis is thought to be responsible for the debilitating sequelae of blinding trachoma and infertility. Inhibition of host cell apoptosis is a persistent C. trachomatis infection mechanism. ZEB1-AS1 is a long non-coding RNA (lncRNA), which was up-regulated in persistent C. trachomatis infection in our previous work. In this study, we investigated the role of ZEB1-AS1 in persistent infection and the potential mechanisms. The results showed that ZEB1-AS1 was involved in the regulation of apoptosis, and targeted silencing of ZEB1-AS1 could increase the apoptosis rate of persistently infected cells. Mechanically, interference ZEB1-AS1 caused an apparent down-regulation of the Bcl-2/Bax ratio and the repression of the mitochondrial membrane potential with the remarkable release of cytochrome c, resulting in the significant elevation level of caspase-3 activation. Meanwhile, the luciferase reporter assay confirmed that ZEB1-AS1 acted as a sponge for miR-1224-5p to target MAP4K4. The regulatory effect of miR-1224-5p/MAP4K4 on persistent infection-induced antiapoptosis was regulated by ZEB1-AS1. In addition, ZEB1-AS1 inhibited the apoptosis of Chlamydia-infected cells by activating the MAPK/ERK pathway. In conclusion, we found a new molecular mechanism that the ZEB1-AS1/miR-1224-5p/MAP4K4 axis contributes to apoptosis resistance in persistent C. trachomatis infection. This work may help understand the pathogenic mechanisms of persistent C. trachomatis infection and reveal a potential therapeutic strategy for its treatment.

An isolated ultra-short gamma-ray pulse is a unique tool for measuring ultrafast-physics processes, such as imaging infra-nuclear dynamics and inner-shell electron dynamics Here, we propose an all-optical efficient scheme for generating isolated ultra-short gamma-ray pulse from a laser-driven nanofoil. When a few-cycle circularly polarized laser pulse with an intensity of 10(22) W cm(-2) irradiates a nanofoil, the electrons in the nanofoil are pushed forwards collectively, forming a single relativistic electron sheet (RES) with a charge of nC. The electrons are substantially accelerated to high energies by the super-ponderomotive force of the laser. Then, a counter-propagating laser pulse with a peak intensity of 10(21) W cm(-2) collides with the RES, resulting in the generation of an isolated sub-femtosecond gamma-ray pulse via nonlinear Compton scattering. The effect of laser polarization on the polarization degree of gamma-rays is investigated by using a proof-of-principle calculation. It is shown that a highly polarized isolated gamma-ray pulse with a cut-off energy of 100 MeV can eventually be generated in a head-on collision configuration when the scattering laser is linearly polarized. Such an isolated ultra-short polarized gamma-ray source would provide critical applications in high-energy physics, laboratory astrophysics and nuclear physics.

Understanding the effects of green credit on green total factor productivity (GTFP) is conductive to promoting the sustainable economy development. This paper examines the total effects, influence mechanism, and heterogeneous impacts of green credit on GTFP based on GTFP data of 30 provinces in China from 2008 to 2017. The findings show that, firstly, on the whole, green credit significantly increases GTFP, which is tested by the panel regression model. Secondly, according to the result of the panel quantile model, the increasing effect of green credit on GTFP is strengthened by the improvement of GTFP. Thirdly, green credit has heterogeneous impact on GTFP, which is reflected in economic development with different level, especially for different degrees of environmental regulation. Fourthly, under the full samples, green credit impacts GTFP through green technology innovation, but it has no effect on energy consumption structure. Besides, the influence mechanism is heterogeneous in the variation of sample characteristics. Finally, some significant policy recommendations are provided for policymakers based on these conclusions.

Due to its unique physical and chemical properties, MXene has recently attracted much attention as a promising candidate for wastewater treatment. However, the low water permeation flux of MXene membrane remains a challenge that has not been fully solved. In this study, attapulgite was used to increase the flux of MXene membrane through a facile one-pot method, during which the MXene nanosheets were self-assembled while being intercalated by the attapulgite nanorods to finally form the composite membranes. Under optimal conditions, an increase of water permeation flux of 97.31% could be observed, which was attributed to the broadened nano-channel upon the adequate intercalation of attapulgite nanorods. Its permeation flux and rejection rate for methylene blue (MB) were further studied for diverse applications. In contrast to bare MXene, the permeation flux increased by 61.72% with a still high rejection rate of 90.67%, owing to the size rejection. Overcoming a key technique barrier, this work successfully improved the water permeability of MXene by inserting attapulgite nanorods, heralding the exciting prospects of MXene-based lamellar membrane in dye wastewater treatment.

Growing evidence indicates a connection between cancer-associated fibroblasts (CAFs) and tumor microenvironment (TME) remodeling and tumor progression. Nevertheless, how patterns of CAFs impact TME and immunotherapy responsiveness in triple-negative breast cancer (TNBC) remains unclear. Here, we systematically investigate the relationship between TNBC progression and patterns of CAFs. By using unsupervised clustering methods in the Molecular Taxonomy of Breast Cancer International Consortium (METABRIC) dataset, we identified two distinct CAF-associated clusters that were related to clinical features, characteristics of TME, and prognosis of patients. Then, we established a CAF-related prognosis index (CPI) by the least absolute shrinkage and selection operator (LASSO)-Cox regression method. CPI showed prognostic accuracy in both training and validation cohorts (METABRIC, GSE96058, and GSE21653). Consequently, we constructed a nomogram with great predictive performance. Moreover, the CPI was verified to be correlated with the responsiveness of immunotherapy in three independent cohorts (GSE91061, GSE165252, and GSE173839). Taken together, the CPI might help us improve our recognition of the TME of TNBC, predict the prognosis of TNBC patients, and offer more immunotherapy strategies in the future.

The enhancement of vacancies in catalysts involving Fenton-like reactions is a promising way to remove organic pollutants in wastewater, but sulfur vacancies are rarely involved. In this work, MoS2 containing defect sites were synthesized by a simple high-temperature treatment and then applied for activating peroxymonosulfate to eliminate organic pollutants in wastewater. The structure was characterized by several techniques such as XRD, BET, and XPS. Important influencing factors were systemically investigated. The results indicated that MoS2 with sulfur vacancies possessed a higher catalytic activity than that of the parent MoS2. The annealing temperature of the catalyst had a great effect on the removal of organic pollutants. Besides, the catalytic system had a wide pH range. Quenching and electron paramagnetic resonance (EPR) experiments indicated that the reaction system contained radical and non-radical species. The characterization results revealed that the defect sites in catalysts mainly strengthened the activity of catalysts. This study offers a new heterogeneous catalyst for the removal of organic pollutants via the peroxymonosulfate-based Fenton-like reactions.

Accurately identifying and delineating urban functional areas has seen increasing demand in smart urban planning, landscape design, and resource allocation. Recently, POI (point of interest) data have been increasingly applied to identify urban functional areas. However, heterogeneity in urban spaces or the corresponding POI data has not been fully considered in previous studies. In this study, we proposed a new scheme for urban-functional-area identification by combining POI data, OpenStreetMap (OSM) datasets, and high-resolution remote-sensing imagery. A function-intensity index that integrates the quantitative-density index and average-nearest-neighbor index (ANNI) of POIs was built for representing the urban function. The results show that the proposed function-intensity index can balance the impact of the spatial heterogeneity of each type of POI on determining the functional characteristics of the urban units. In Futian District, Shenzhen, China, the method was effective in distinguishing functional areas with fewer POI amounts but high ANNIs from those functional areas with dense POIs. The overall accuracy of the proposed method is about 11% higher than that of the method using the POI density only. This paper argues for considering both the quantitative density and spatial heterogeneity of POIs to improve urban-functional-area identification.
