摘要:
Acute lung injury (ALI) is a severe pulmonary disease that causes a high number of fatalities worldwide. Studies have shown that FoxA1 expression is upregulated during ALI and may play an important role in ALI by promoting the apoptosis of alveolar type II epithelial cells. However, the mechanism of FoxA1 overexpression in ALI is unclear. In this study, an in vivo murine model of ALI and alveolar type II epithelial cells injury was induced using lipopolysaccharide (LPS). LPS upregulated FoxA1 in the lung tissue of the in vivo ALI model and in LPS-challenged type II epithelial cells. In contrast, miR-17 was significantly downregulated in these models. After miR-17 antagomir injection, the expression of FoxA1 was significantly increased in ALI mice. MiR-17 mimics could significantly inhibit FoxA1 mRNA and protein expression, whereas the miR-17 inhibitor could significantly increase FoxA1 mRNA and protein expression in LPS-induced type II epithelial cells. Thus, our results suggest that the downregulation of miR-17 expression could lead to FoxA1 overexpression in ALI. © 2014 Elsevier Inc. All rights reserved.

Aims: This study aims to examine the possible associations between high density lipoprotein (HDL) subclass distribution and APOA5-1131T>C polymorphism in hypertriglyceridemia. Methods: The distribution of HDL subclasses was quantified by 2-dimensional electrophoresis in conjunction with immunodetection method. The APOA5-1131T>C polymorphism was identified in 95 hypertriglyceridemic (HTG) patients and 102 healthy subjects by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP). Results: The APOA5-1131C (C) allele frequency was higher in the HTG group than in the control group. Plasma triglycerides (TG) were significantly higher and apoA5 was significantly lower in patients with the C allele when compared to patients with the APOA5-1131T (T) allele, even more dramatically so in the APOA5-1131CC homozygote. In both the HTG group and the control group, the frequency of the C allele was positively correlated with levels of TG, total cholesterol (TC), low density lipoprotein cholesterol (LDL-C) and apolipoprotein B100 (apoB100), and negatively correlated with levels of high density lipoprotein cholesterol (HDL-C), apolipoprotein A1 (apoA1) and apolipoprotein A5 (apoA5) (P < 0.001). In all subjects, the frequency of the C allele was positively correlated with the level of small-sized HDL (pre beta(1)-HDL and HDL3a), and negatively correlated with levels of HDL2a and HDL2b. Conclusion: Changes in HDL subclass distributions in HTG may be related to the APOA5-1131T>C polymorphism. This polymorphism leads to a general shift towards smaller-sized HDL. 摘要:
目的 研究二苯乙烯苷(TSG)对过氧化氢(H2O2)诱导的人脐静脉内皮细胞凋亡的作用以及TSG是否通过调节X连锁凋亡抑制蛋白(XIAP)的表达抗内皮细胞凋亡.方法 预先用不同剂量TSG(1~ 100 μmol/L)处理人脐静脉内皮细胞24h,再用300 μmol/L H2O2作用细胞24h.采用MTT法检测细胞活力,Hoeschest染色及流式细胞仪检测细胞凋亡.RT-PCR及Western blot检测TSG、XIAP抑制剂Embelin作用细胞后XIAP的表达.结果 300 μmol/L H2O2作用HUVECs细胞24 h可有效降低细胞活力,诱导细胞凋亡;1 ~ 100 μmol/L TSG预处理24 h均可提高细胞存活率,抑制细胞凋亡;30 μmol/L Embelin预处理细胞6h后再用TSG预处理24h,细胞凋亡率较TSG预处理组明显上升,细胞活力显著降低.RT-PCR、Western blot结果显示H2O2处理后,细胞XIAP表达水平显著降低;与H2O2组相比,1~ 100 μmol/L TSG预处理24h后,XIAP表达升高;与TSG预处理组相比,Embelin预处理6h后再用TSG预处理24h,XIAP的表达降低.结论 TSG能抑制H2O2诱导的人脐静脉内皮细胞凋亡,其作用机制与上调XIAP的表达有关.

摘要:
An efficient method for the copper-catalyzed trifluoromethylation of terminal alkenes with an electrophilic trifluoromethylating reagent has been developed. The reactions proceeded smoothly to give trifluoromethylated alkenes in good to excellent yields. The results provided a versatile approach for the construction of Cvinyl-CF3 bonds without using prefunctionalized substrates. © 2013 Wang et al.

摘要:
目的 探讨代谢综合征(metabolic syndrome,MS)患者血浆高密度脂蛋白(high density lipoprotein,HDL)亚类含量变化及与甘油三酯水平的关系.方法 采用双向电泳-免疫印迹检测法测定352例MS患者和187例正常人血浆HDL亚类的相对含量,全自动生化分析仪测定血脂含量,ELISA法检测载脂蛋白浓度.结果 与对照组相比,MS患者血浆甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDLC)、载脂蛋白B100 (ApoB100)、preβ1-HDL、HDL3b含量及ApoB100/A1和LDLC/HDLC比值均显著性增高,HDLC、ApoA1、HDL2b、 HDL2a显著降低(P <0.001).根据ATPⅢ文件,将MS患者按TG浓度分4组,即正常组(< 1.7 mmol/L)、临界高组(1.7~2.3 mmol/L)、高组(2.3~5.69 mmol/L)、极高组(≥5.7 mmol/L),随血浆TG水平的升高,小颗粒的preβ1-HDL、HDL3a和HDL3b含量逐渐升高,而大颗粒的HDL2b和HDL2a含量逐渐降低.结论 代谢综合征患者血浆HDL亚类颗粒有变小的趋势,并且随着TG水平的升高,HDL颗粒减小更显著.

摘要:
The ligand-free trifluoromethylation of arylboronic acids with a [Ph 2SCF 3] +[OTf] -/Cu(0) system has been carefully investigated. Aryl-, alkenyl- and heteroarylboronic acids with a variety of functional groups were suitable substrates for this reaction. It is suggested that a CuCF 3 species is formed under the reaction conditions. © 2011 The Royal Society of Chemistry.

摘要:
为研究二苯乙烯苷对过氧化氢(H_2O_2)诱导人脐静脉内皮细胞凋亡的影响并初步探讨二苯乙烯苷抗凋亡的可能机制,采用不同浓度H_2O_2和二苯乙烯苷处理内皮细胞24 h,以MTT法检测细胞生长活力、Hoechst33258染色观察细胞形态、流式细胞仪检测凋亡率等方法筛选造模内皮细胞凋亡的H_2O_2浓度和二苯乙烯苷最佳的抗内皮细胞凋亡作用浓度.RT-PCR、Western-blot分别检测Caspase-3、PARP mRNA及蛋白质的表达.结果发现,与空白对照组相比,300μmol/L H_2O_2作用后,内皮细胞增殖明显受到抑制,Hoechst33258染色可见大量凋亡细胞,细胞凋亡率显著增加,流式细胞仪检测出明显的凋亡峰,Caspase-3和多聚腺苷酸二磷酸核糖基聚合酶(PARP)的表达量显著增加.经二苯乙烯苷处理后,随着二苯乙烯苷浓度增加,细胞的增殖率随之增加,凋亡细胞数减少,凋亡率逐渐降低,与H_2O_2组比较,10μmol/L的二苯乙烯苷能够显著提高细胞增殖率,降低细胞凋亡率,并显著减少Caspase-3和PARP表达.以上结果表明,二苯乙烯苷能够抑制由H_2O_2诱导的人脐静脉内皮细胞凋亡,增加细胞生长活性,降低细胞凋亡率,其作用机制可能与下调Caspase-3和多聚腺苷酸二磷酸核糖基聚合酶(PARP)的表达有关。

摘要:
目的:研究二苯乙烯苷(TSG)对过氧化氢(H2O2)诱导人脐静脉内皮细胞(HUVECs)凋亡的保护作用.方法:运用四甲基偶氮唑盐还原法(MTT法)和流式细胞术筛选建立细胞凋亡模型的H202合适浓度以及检测不同浓度TSG时H2O2诱导HUVECs的增殖率和凋亡率;Hoeehst33258染色观察细胞凋亡形态.结果:MTT及流式法筛选300μmol/L为H2O2作用于细胞的最适凋亡浓度.MTT和流式结果显示,与H2O2(300μmol/L)损伤组比较,10μmol/L与100μmol/L TSG预处理组细胞的增殖率增加(P<0.05),凋亡率显著降低(P<0.01);Hoechst33258染色观察TSG能降低H2O2诱导的细胞凋亡,使细胞凋亡数减少.结论:TSG能抑制H2O2诱导的HUVECs凋亡,从而起到保护血管内皮细胞的作用.

摘要:
目的:构建编码Smad4 mRNA的shRNA真核表达载体,并筛选出基因沉默效果最明显的shRNA质粒表达载体。方法:根据GenBank提供的人Smad4基因的mRNA序列构建2个shRNA质粒表达载体和1个阴性对照质粒表达载体,并通过基因测序进行鉴定。经鉴定后转染体外培养人成纤维细胞,western-blot检测Smad4蛋白水平抑制表达效果。结果:构建质粒表达载体测序结果显示,插入片段测序结果与合成的shRNA序列一致;靶向Smad4基因shRNA质粒表达载体对所转染的人成纤维细胞中Smad4蛋白水平表达均有抑制作用,其中shRNA1最为明显。结论:成功构建了靶向Smad4基因shRNA质粒表达载体,其中抑制Smad4基因表达效果最为明显的是shRNA1,为进一步研究Smad4基因的RNA干扰奠定了基础。

摘要:
目的:以Smad4基因为靶标构建小干扰RNA(siRNA)真核表达载体.方法:根据GenBank公布的人Smad4核酸序列及SiRNA设计原则,用AmbionRNAi在线软件,筛选得到2个19 bp片段为靶序列,合成两端带有Bam H Ⅰ、HindⅢ酶切位点的发夹结构寡核苷酸序列,经过退火,将此序列克隆到真核表达质粒pSilencerTM3.1-H1 neo vector中,构建成重组质粒,酶切及测序验证.脂质体介导转染人原代增生性瘢痕成纤维细胞,经G418筛选细胞克隆,并用RT-PCR检测Smad4基因的表达.结果:成功构建了pSilencerTM3.1.H1 neo-Smad4 shRNA表达载体克隆,插入片段测序结果与合成的siRNA序列一致.RT-PCR结果显示转染shRNA1、shRNA2重组质粒的成纤维细胞内Smad4 mRNA水平均降低,其中以pSilencerTM3.1-H1-Smad4 shRNA1更为明显(P<0.01).结论:构建P-Smad4 shRNA袁达载体成功,为进一步研究Smad4基因的RNA干扰奠定了基础.

摘要:
Forkhead box protein A1 (Foxa1) is an evolutionarily conserved winged helix transcription factor. In this study, the effect of Foxa1 on the expression of HSP72 was examined by RT-PCR and Western blot in Foxa1 overexpression or deficient cells. The results showed overexpression of Foxa1 promoted the expression of HSP72, while Foxa1 depletion, induced by antisense oligonucleotides, decreased the expression of HSP72 in MCF-7 cells under normal and heat stress condition. Electrophoretic mobility shift assay and chromatin immunoprecipitation revealed that Foxa1 bound to HSP72 promoter, and heat stress promoted its DNA binding activity. Luciferase reporter showed that Foxa1 also increased the transcription activity of HSP72 promoter. These results indicate an important role for Foxa1 as a novel regulator of expression of HSP72. © 2009 Elsevier Inc. All rights reserved.

目的:观察HDL、LPS和(HDL+LPS)对小鼠血液SOD及MDA的影响,研究HDL抗LPS的作用.方法:(1)用不同浓度的PEG-6000离心人血浆脂蛋白,提取HDL并脱脂;(2)给小鼠注射HDL,LPS或HDL+LPS,对照组小鼠注射生理盐水;观察小鼠存活时间,测定其血液中SOD活性及MDA含量.结果:(1)相比对照组及HDL组,LPS组和HDL+LPS组小鼠的存活时间明显缩短,且后两者之间存在显著性差异;(2)LPS组小鼠血浆中SOD活性降低,MDA含量升高,均和其他三组有显著性差异.结论:LPS能使内毒素损伤小鼠血浆中SOD活性降低,MDA含量升高,HDL有抗内毒素损伤的作用.

Adrenaline inhibited cell proliferation and regulated expression of TGF-beta1 and bFGF in cultured human hypertrophic scar fibroblasts via alpha-receptor

摘要:
Adrenaline has been shown to modulate proliferation of mouse fibroblasts, adventitial fibroblasts and synovial B (fibroblasts-like) cells. However, little is known about the response of cultured human hypertrophic scar fibroblasts to adrenaline. In this study, we investigated cell proliferation and involved mechanisms in hypertrophic scar fibroblasts in response to adrenaline. Population doubling time (PDT) assay and MTT assay were performed to determine the cell proliferation and cell viability, respectively. The expression of bFGF and TGF-beta 1 was measured by reverse transcriptase-polymerase chain reaction (RT-PCR) and enzyme linked immunosorbent assay (ELISA). The results showed that adrenaline inhibited proliferation of normal and hypertrophic scar fibroblasts in a dose-dependent manner. Moreover, adrenaline up-regulated the expression of bFGF and down-regulated the expression of TGF-beta 1 in normal and hypertrophic scar fibroblasts. Interestingly incubation with the a receptor antagonist regitine indicated that adrenaline mediated inhibition of cell proliferation and regulation of TGF-beta 1 and bFGF in cultured normal and hypertrophic scar fibroblasts were mediated by the a receptor. These studies suggest that adrenaline inhibits proliferation and alters the expression of TGF-beta 1 and bFGF in human hypertrophic scar fibroblast involving an a receptor mediated pathway.

参芪扶正注射液对肺癌小鼠顺铂化疗后免疫功能的影响

摘要:
目的 探讨参芪扶正注射液对肺癌小鼠顺铂化疗后免疫功能的影响.方法 采用顺铂(DDP)3 mg/(kg·d)连续7 d腹腔注射建立肺癌小鼠化疗后动物模型.然后随机分为对照组和治疗组,治疗组给予参芪扶正注射液40,80ml/(kg·d)分别腹腔注射给药,连续7 d;对照组予生理盐水40 ml/(kg·d).末次给药后24 h采用免疫器官重量法检测胸腺指数、脾脏指数;同时采用放射免疫法测定小鼠血清中IL-2和TNF-α的含量.结果 治疗组小鼠胸腺指数、脾脏指数均高于对照组(P<0.05);治疗组小鼠血清IL-2水平显著高于对照组,而治疗组TNF-α低于对照组(P<0.05).结论 参芪扶正注射液可提高肺癌小鼠顺铂化疗后的免疫功能,临床上可试用参芪扶正注射液对肺癌化疗后免疫功能低下患者进行治疗.
