It must be admitted that the incidence of colorectal cancer (CRC) was on the rise all over the world, but the related treatment had not caught up. Further research on the underlying pathogenesis of CRC was conducive to improving the survival status of current CRC patients. Differentially expressed genes (DEGs) screening were conducted based on “limma” and “RobustRankAggreg” package of R software. Weighted gene co-expression network analysis (WGCNA) was performed in the integrated DEGs that from The Cancer Genome Atlas (TCGA), and all samples of validation were from Gene Expression Omnlbus (GEO) dataset. The terms obtained in the functional annotation for primary DEGs indicated that they were associated with CRC. The MEyellow stand out whereby showed the significant correlation with clinical feature (disease), and 4 hub genes, including ABCC13, AMPD1, SCNN1B and TMIGD1, were identified in yellow module. Nine datasets from Gene Expression Omnibus database confirmed these four genes were significantly down-regulated and the survival estimates for the low-expression group of these genes were lower than for the high-expression group in Kaplan-Meier survival analysis section. MEXPRESS suggested that down-regulation of some top hub genes may be caused by hypermethylation. Receiver operating characteristic curves indicated that these genes had certain diagnostic efficacy. Moreover, tumor-infiltrating immune cells and gene set enrichment analysis for hub genes suggested that there were some associations between these genes and the pathogenesis of CRC. This study identified modules that were significantly associated with CRC, four novel hub genes, and further analysis of these genes. This may provide a little new insights and directions into the potential pathogenesis of CRC.

A simulation calculation model in the case of a severe accident such as a loss of coolant accident (LOCA) was established and calculated by MELCOR code in a marine pressurized water reactor. The effects of severe accidents on the hydrogen source term, release of the radionuclide source term, and the accident process were investigated, and the radiation consequences were analyzed. LOCA sensitivity analysis was conducted with different break sizes; the release and diffusion of source terms were analyzed. The results indicated that the hydrogen yield in the reactor core was dependent on the temperature, and on the residual water in the reactor core, which has no direct relationship with the break size. The break size directly affected the accident progression. However, when the break exceeded a certain size, it did not affect the LOCA process. The break size may not change to change the release amount and migration rule of radionuclides such as Xe and I, and the fission product CsI, as final containment is in dynamic equilibrium in LOCA conditions. The contents of fission products released into the containment vessel, major loop, cavity, and the environment are sensitive to the break size. However, no special correlation is observed between the amount released and the break size. Primary liver cancer is the second-most commonly occurring cancer and has resulted in numerous deaths worldwide. Hepatic resection is of two main types, i.e., anatomic resection (AR) and non-anatomic resection (NAR). The oncological outcomes of hepatocellular carcinoma (HCC) patients after AR and NAR are still considered controversial. Therefore, we aimed to compare the impact of AR and NAR on the oncological outcomes of HCC patients with tumor diameters ≤ 5 cm using the propensity score matching method and research-based evidence. A systematic literature search was conducted. The main outcomes were disease-free survival (DFS), overall survival (OS), intrahepatic recurrence rate, and extrahepatic metastasis rate. Relative risk (RR) was calculated from forest plots and outcomes using random-effects model (REM). AR significantly improved DFS at 1, 3. and 5 years after surgery, compared to NAR (RR = 1.09, 95% CI = 1.04–1.15, P = 0.0003; RR = 1.16, 95% CI = 1.07–1.27, P = 0.0005; RR = 1.29, 95% CI = 1.07–1.55, P = 0.008). However, both of the difference in DFS at 7 years and OS at 1 and 3 years after AR versus that after NAR were not statistically significant. Nevertheless, the long-term OS associated with AR (5, 7, and 10 years) was superior to that associated with NAR (RR = 1.12, 95% CI = 1.03–1.21, P = 0.01; RR = 1.19, 95% CI = 1.04–1.36, P = 0.01; RR = 1.18, 95% CI = 1.05–1.34, P = 0.008). The difference in the intrahepatic recurrence rate after AR versus that after NAR was not statistically significant, but the extrahepatic metastasis rate after AR was significantly lower than that observed after NAR (RR = 0.61, 95% CI = 0.40–0.94, P = 0.03). Therefore, AR should be the preferred surgical approach for HCC patients with tumor diameters ≤ 5 cm. PROSPERO registration number CRD42022330596.

Background: C-X-C chemokine receptor type 4 (CXCR4) plays a crucial role in mediating podocyte dysfunction, proteinuria and glomerulosclerosis. However, the underlying mechanism remains poorly understood. Here we studied the role of beta-catenin in mediating CXCR4-triggered podocyte injury. Methods: Mouse models of proteinuric kidney diseases were used to assess CXCR4 and beta-catenin expression. We utilized cultured podocytes and glomeruli to delineate the signal pathways involved. Conditional knockout mice with podocyte-specific deletion of CXCR4 were generated and used to corroborate a role of CXCR4/beta-catenin in podocyte injury and proteinuria. Results: Both CXCR4 and beta-catenin were induced and colocalized in the glomerular podocytes in several models of proteinuric kidney diseases. Activation of CXCR4 by its ligand SDF-1 alpha stimulated beta-catenin activation but did not affect the expression of Wnt ligands in vitro. Blockade of beta-catenin signaling by ICG-001 preserved podocyte signature proteins and inhibited Snail1 and MMP-7 expression in vitro and ex vivo. Mechanistically, activation of CXCR4 by SDF-1 alpha caused the formation of CXCR4/beta-arrestin-1/Src signalosome in podocytes, which led to sequential phosphorylation of Src, EGFR, ERK1/2 and GSK-3 beta and ultimately beta-catenin stabilization and activation. Silencing beta-arrestin-1 abolished this cascade of events and inhibited beta-catenin in response to CXCR4 stimulation. Podocyte-specific knockout of CXCR4 in mice abolished beta-catenin activation, preserved podocyte integrity, reduced proteinuria and ameliorated glomerulosclerosis after Adriamycin injury. Conclusion: These results suggest that CXCR4 promotes podocyte dysfunction and proteinuria by assembling CXCR4/beta-arrestin-1/Src signalosome, which triggers a cascade of signal events leading to beta-catenin activation.

Objective. To explore the risk factors of lower extremity deep venous thrombosis (LEDVT) after surgery and discuss the treatment and nursing countermeasures. Methods. A retrospective analysis was conducted on 268 surgical patients admitted between July to December 2021. The factors associated with LEDVT were analyzed using the Logistic regression model. Further, LEDVT patients were assigned to a research group treated with targeted nursing to prevent LEDVT and a control group that used routine care. Coagulation function and inflammatory cytokines before and after nursing intervention were compared between groups. The assessment of patients' mobility employed the lower limb motor function part of the Fugel-Meyer Assessment (FMA), Harris Hip Score (HHS), and Barthel index (BI), and their psychological status was evaluated using the Kolcaba's General Comfort Questionnaire (GCQ) and Self-rating Anxiety/Depression Scale (SAS/SDS). Finally, patient satisfaction with the treatment service was investigated. Results. Logistic regression analysis showed that hypertension, limb paralysis, central venous catheterization of lower limbs, and bedridden time affect postoperative LEDVT in an independent way (P < 0.05). After the intervention, the coagulation function and inflammatory reaction were improved in both groups, with more significant improvement in the research group (P < 0.05). The research group also showed higher FMA, Harris, GCQ, and BI scores while lower SAS and SDS scores than the control group postnursing intervention (P < 0.05). Finally, a higher satisfaction rate was identified in the research group as compared to the control group (P < 0.05). Conclusion. Hypertension, limb paralysis, CVC of lower limbs, and bedridden time are all independent risk factors for LEDVT after surgery. The implementation of targeted nursing strategies for the above factors can effectively alleviate the hypercoagulable state of patients after operation, reduce inflammatory responses, and improve patient comfort, which is of great significance for preventing the occurrence of LEDVT.

This study aims to compare the differences between obstructed and unobstructed total anomalous pulmonary venous connection (TAPVC) using echocardiography, and to evaluate the clinical and echocardiographic parameters associated with pulmonary venous obstruction (PVO). We conducted a retrospective study of 70 patients with TAPVC between 2014 and 2019. The morphologic and hemodynamic echocardiographic parameters of patients were observed and measured, and the parameters between obstructed and unobstructed TAPVC were compared. The clinical and echocardiographic parameter differences between the two groups were used for ROC curve analysis. Obstructed TAPVC was found in 30 (42.9%) of 70 patients. Between obstructed and unobstructed TAPVC, there were significant differences in atrial septal defect size, pulmonary artery maximum velocity (PA V-max), peak E velocity of mitral valve, left ventricular fractional shortening, left ventricular ejection fraction, stroke volume and the incidence of patent ductus arteriosus, but there was no significant difference in birth weight. The first diagnosis age of obstructed TAPVC was earlier than unobstructed type. The ROC curve analysis for the first diagnosis age showed the sensitivity and specificity were 76.7%, 80% respectively. The ROC curve analysis for the PA V-max showed the sensitivity and specificity were 88.5%, 67.6% respectively. Patients with TAPVC had a high incidence of PVO. The presence of PVO can affect the size of atrial septal defect and the closure of the ductus arteriosus, cause significant changes in PA V-max, peak E velocity of mitral valve, left ventricular fractional shortening, left ventricular ejection fraction, stroke volume, lead to earlier symptoms and earlier first diagnosis age. The first diagnosis age and PA V-max were excellent values since they associated with PVO.

Objective: The aim of our study was to investigate the role of three-dimensional arterial spin labeling (3D-ASL) perfusion in the diagnosis and follow-up of children with viral encephalitis.Methods: Twenty-five consecutive children with viral encephalitis and 25 healthy children of similar age were recruited for the study between 2017 and 2020. Conventional magnetic resonance imaging and 3D-ASL were performed for all subjects, and a color map of cerebral blood flow (CBF) was generated. The images were classified into three groups depending on the time points at which the magnetic resonance examinations were conducted, including the initial admission scan, inpatient review, and follow-up review. Clinical, neuroradiologic, and follow-up features were studied.The CBF values of the lesion area in the diseased brain group and the bilateral temporal cortex in the control group were measured and the differences between the two groups were compared.Results: Perfusion was significantly increased in the acute cerebral disease group, and CBF and normalized cerebral blood flow(nCBF) were significantly higher than in the control group (124.5 vs 70.3 mL/100 g/min, 2.85 vs 1.36). Follow-up revealed that brain tissue perfusion in the lesion area of nine children decreased gradually after treatment, as their condition improved.Conclusion: Brain tissue perfusion in children with viral encephalitis increases during the acute stage and decreases when the condition improves. 3D-ASL provides a reference for diagnosis and follow-up of viral encephalitis in children.

With the increase in the amount of 3D point cloud data and the wide application of point cloud registration in various fields, the question of whether it is possible to quickly extract the key points of registration and perform accurate coarse registration has become a question to be urgently answered. In this paper, we proposed a novel semantic segmentation algorithm that enables the extracted feature point cloud to have a clustering effect for fast registration. First of all, an adaptive technique was proposed to determine the domain radius of a local point. Secondly, the feature intensity of the point is scored through the regional fluctuation coefficient and stationary coefficient calculated by the normal vector, and the high feature region to be registered is preliminarily determined. In the end, FPFH is used to describe the geometric features of the extracted semantic feature point cloud, so as to realize the coarse registration from the local point cloud to the overall point cloud. The results show that the point cloud can be roughly segmented based on the uniqueness of semantic features. The use of a semantic feature point cloud can make the point cloud have a very fast response speed based on the accuracy of coarse registration, almost equal to that of using the original point cloud, which is conducive to the rapid determination of the initial attitude.
