摘要:
Mycoplasma genitalium (Mg) is the smallest and simplest self-replicating bacteria lacking of cell wall and is a human pathogen causing various diseases. This paper describes the real-time, long-term and in situ monitoring of the growth of Mg and evaluation of the effect of the antibiotics tetracycline and levofloxacin on the growth using a wireless magnetoelastic sensor. The sensor is fabricated by coating a magnetoelastic strip with a polyurethane protecting film. In response to a time-varying magnetic field, the sensor longitudinally vibrates at a resonance frequency, emitting magnetic flux that can be remotely detected by a pick-up coil. No physical connections between the sensor and the detection system are required. The wireless property facilitates aseptic operation. The adhesion of Mg on the sensor surface results in a decrease in the resonance frequency, which is proportional to the concentration of Mg. The shift of the resonance frequency-time curves shows that under routine culture condition the growth curve of Mg is composed of three phases those are lag, logarithmic and stationary phase, respectively. In the presence of the antibiotics, the lag phase in the growth inhibition curves is prolonged obviously and the stationary phase is substituted by a decline phase. The growth inhibition of Mg is related to the concentration of the antibiotics. The MIC50 (minimal inhibitory concentration) of Mg incubated in the presence of the antibiotics for 120 h is calculated to be 1.5 and 0.5 μg/mL for tetracycline and levofloxacin, respectively. © 2008 Elsevier B.V. 摘要:
Mycoplasma genitalium is a leading pathogen of nongonoccocal chlamydia-negative urethritis, which has been implicated directly in numerous other genitourinary and extragenitourinary tract pathologies. The pathogenesis of infection is attributed in part to excessive immune responses. M. genitalium-derived lipid-associated membrane proteins (LAMPs) are a mixture of bacterial lipoproteins, exposed at the surface of mycoplasma, that are potent inducers of the host innate immune system. However, the interaction of M. genitalium-derived LAMPs as pathogenic agents with Toll-like receptors (TLRs) and the signaling pathways responsible for active inflammation and NF-κB activation have not been fully elucidated. In this study, LAMPs induced the production of tumor necrosis factor alpha (TNF-α) and interleukin-6 (IL-6) in a dose-dependent manner. Blocking assays showed that TLR2- and CD14-neutralizing antibodies reduced the expression of TNF-α and IL-6 in THP-1 cells. Furthermore, LAMP-induced NF-κB activation was increased in 293T cells transfected with TLR2 plasmid. The activity of NF-κB was synergically augmented by cotransfected TLR1, TLR6, and CD14. Additionally, LAMPs were shown to inhibit NF-κB expression by cotransfection with dominant-negative MyD88 and TLR2 plasmids. These results suggest that M. genitalium-derived LAMPs activate NF-κB via TLR1, TLR2, TLR6, and CD14 in a MyD88-dependent pathway. Copyright © 2009, American Society for Microbiology. 摘要:
循环核酸是指存在于血液(血浆或血清)中的细胞外游离DNA和RNA.肿瘤患者血循环中核酸含量明显高于正常人,研究证实其来源于肿瘤细胞,并且与原发肿瘤有着结构或功能上的某些一致性,因此循环核酸对肿瘤的诊断、治疗和预后评估都可能有重要价值.本文对当前血浆(血清)DNA如Ras和p53基因突变、微卫星改变、肿瘤抑癌基因启动子的高甲基化和肿瘤相关病毒DNA等以及肿瘤相关RNA如酪氨酸酶RNA、端粒酶成分RNA、不同肿瘤相关基因编码的mRNA和病毒RNA等检测在肿瘤诊断和预后中的应用及其意义进行了综述,并展望循环核酸测定的应用前景.

摘要:
目的 表达梅毒螺旋体黏附蛋白Tp0751,纯化表达产物并进行免疫活性分析,为探索Tp0751重组蛋白在梅毒致病过程中的作用奠定基础.方法 通过生物信息学分析,去除Tp0751信号肽序列,构建原核表达体进行诱导表达;Ni亲和层析柱纯化重组蛋白,Western blot检测其免疫反应性,用重组蛋白免疫新西兰家兔,评价其免疫原性.结果 成功构建了pET-28a(+)-0751原核表达载体,经表达、纯化后获得了相对分子质量约为26×10~3的融合蛋白;Western blot检测其能与梅毒患者阳性血清发生特异性反应;利用纯化的Tp0751重组蛋白免疫新西兰家兔,能诱导家兔产生特异性免疫应答,ELISA法测定免疫血清中特异性抗体滴度在1∶10 240以上.结论 重组表达的Tp0751黏附蛋白具有良好的免疫活性,为进一步研究其在梅毒致病过程中的作用和生物学功能奠定了基础.

摘要:
确定沙眼衣原体CT358蛋白在衣原体感染细胞中的位置并初步鉴定其生物学功能.采用PCR方法从D型沙眼衣原体的基因组中扩增CT358基因,并克隆入pGEX和pDSRedC1表达载体中,将重组质粒pGEX-CT358转化到XL1-blue宿主菌,并诱导表达融合蛋白GST-CT358.纯化后的CT358融合蛋白免疫小鼠制备抗体,应用间接免疫荧光技术对CT358蛋白在衣原体感染细胞内的定位及表达模式进行分析.同时,pDSRedC1-CT358重组质粒瞬时转染HeLa细胞,观察CT358蛋白对衣原体感染的影响.实验结果证明CT358蛋白为沙眼衣原体包涌体膜蛋白.该蛋白质在衣原体感染12 h后就表达定位于包涵体膜上,直至持续到整个感染周期,转基因在胞浆表达的CT358融合蛋白不影响其后的衣原体感染.该研究为深入研究衣原体与宿主细胞间相互作用提供了新的线索,并可为衣原体性的治疗、预防提供新方向.

摘要:
目的 探讨半巢式聚合酶链反应(PCR)扩增梅毒螺旋体(Tp)的DNA多聚酶Ⅰ基因(polA)以探讨检测全血标本中微量Tp的可行性. 方法 应用半巢式PCR和常规PCR分别扩增165例可疑梅毒及非梅毒患者全血中Tp的polA,与血清学方法比较,探讨半巢式PCR方法在扩增全血中Tp DNA的意义. 结果 待测的165例标本中,与血清学方法比较,半巢式PCR检测Tp的敏感性和特异性分别为62.7%和95.9%,两者符合率为82.4%;两种PCR方法检测结果差异有显著性(P<0.01),半巢式PCR敏感性远高于常规PCR. 结论 半巢式polA PCR检测全血中Tp较常规PCR灵敏,是血清学方法诊断梅毒的有效补充,但其检测的灵敏度仍有待进一步提高.

摘要:
Background. C. trachomatis organisms carry a cryptic plasmid that encodes 8 open reading frames designated as pORF1 to 8. It is not clear whether all 8 pORFs are expressed during C. trachomatis infection in humans and information on the functionality of the plasmid proteins is also very limited. Results. When antibodies from women urogenitally infected with C. trachomatis were reacted with the plasmid proteins, all 8 pORFs were positively recognized by one or more human antibody samples with the recognition of pORF5 protein (known as pgp3) by most antibodies and with the highest titers. The antibody recognition of the pORFs was blocked by C. trachomatis-infected HeLa but not normal HeLa cell lysates. The pgp3 fusion protein-purified human IgG detected the endogenous pgp3 in the cytosol of C. trachomatis-infected cells with an intracellular distribution pattern similar to that of CPAF, a chlamydial genome-encoded protease factor. However, the human antibodies no longer recognized pgp3 but maintained recognition of CPAF when both antigens were linearized or heat-denatured. The pgp3 conformation is likely maintained by the C-terminal 75% amino acid sequence since further deletion blocked the binding by the human antibodies and two conformation-dependent mouse monoclonal antibodies. Conclusion. The plasmid-encoded 8 proteins are both expressed and immunogenic with pgp3 as the most immunodominant antigen during chlamydial infection in humans. More importantly, the human anti-pgp3 antibodies are highly conformation-dependent. These observations have provided important information for further understanding the function of the plasmid-encoded proteins and exploring the utility of pgp3 in chlamydial diagnosis and vaccination. © 2008 Li et al; licensee BioMed Central Ltd.

摘要:
目的研究粪肠球菌临床株表面蛋白基因(esp)的流行情况及esp基因存在状况与粪肠球菌耐药的相关性。方法用PCR法扩增粪肠球菌esp基因并克隆、测序;用琼脂稀释法检测5种常用抗菌药物对粪肠球菌临床株的最低抑菌浓度(MIC)。结果61株粪肠球菌中,esp基因阳性率为42.6%;氨苄西林、环丙沙星、高浓度庆大霉素MIC〉500 mg/L,红霉素耐药粪肠球菌中,esp阳性率分别为33.3%、54.8%、70.6%、51.0%;敏感株中esp阳性率分别为43.6%、30.0%、7.4%、8.3%。结论esp基因的存在状况与粪肠球菌对氨苄西林耐药无明显相关性,而与环丙沙星、红霉素、高浓度庆大霉素耐药有显著相关性;esp基因有可能成为粪肠球菌耐药株的分子表面标志物。

肺炎克雷伯菌中超广谱β内酰胺酶和AmpC酶的检测

摘要:
[目的]观察对头孢菌素类耐药的肺炎克雷伯菌对抗菌素的敏感性及其产ESBLs和AmpC酶的情况.[方法]利用ATB G-5试条法测定细菌对抗菌素的敏感性,用双纸片协同试验及纸片法确证试验检测ESBLs,用三维试验检测AmpC酶.[结果]从头孢菌素类耐药的肺炎克雷伯菌菌中共检出产ESBLs和AmpC酶分别为90.5%和5.5%, ESBLs和AmpC酶共同存在者为4.6%.产酶菌株对3代头孢菌素高度耐药,对复方磺胺甲噁唑、环丙沙星等有较高的耐药性,对亚胺培南抗生素敏感,加酶抑抑制剂的抗菌素对产AmpC酶菌株无亲和性.[结论]肺炎克雷伯菌存在比较严重的耐药情况,ESBLs和AmpC酶是肺炎克雷伯菌对头孢菌素类耐药耐药的重要机制,实验室应加强对其检测和对抗菌素的耐药性监测,对指导临床合理用药至关重要.

肺炎嗜衣原体(Cpn)是二十世纪八十年代新发现的一种重要的呼吸系统感染的病原体,它不仅可以引起非典型肺炎,而且还有可能通过直接或间接的机制参与动脉粥样硬化的发生与发展,因而受到了人们越来越多的关注。近年来,Cpn感染的血清学诊断也成为人们关注的焦点之一,目前常用于诊断Cpn感染的抗原有全菌抗原、脂多糖和主要外膜蛋白等属特异性抗原。因此本文就Cpn感染诊断候选抗原的研究进展作一综述。

Mycoplasma genitalium lipid-associated membrane proteins (LAMPg) can induce human monocytic cell line THP-1 to produce proinflammatory cytokines, including tumor necrosis factor-α (TNF-α), IL-1β and IL-6, as demonstrated by an enzyme-linked immunosorbent assay and reverse transcription-PCR (RT-PCR). This study also investigated the signaling transduction pathways involved in the production of these cytokines. THP-1 cells were stimulated with LAMPg and then examined for the activation of MAPKs, such as SAPK/JNK, p38, extracellular signal-regulated kinase (ERK)1/2 and NF-κB and AP-1. Western blot clearly showed that stress-activated protein kinase (SAPK)/c-Jun-N-terminal kinase (JNK), p38 and ERK1/2 were activated in response to LAMPg, peaking at 30 min. SAPK/JNK-specific inhibitor SP600125 slightly suppressed IL-6 production although no evident effects were obtained for TNF-α and IL-1β; ERK1/2 inhibitor PD98059 blocked both TNF-α and IL-1β, but not IL-6 production. However, p38 inhibitor SB203580 abrogated TNF-α, IL-1β and IL-6 production. The DNA-binding activity of NF-κB and AP-1 was also assessed by an electrophoretic mobility gel shift assay, and an NF-κB specific inhibitor, pyrrolidine dithiocarbamate, profoundly inhibited the synthesis and production of the proinflammatory cytokines. Based on these results, this study concludes that MAPKs, NF-κB and AP-1 may play important roles in the genital tract inflammatory reaction after mycoplasma infection. © 2008 Federation of European Microbiological Societies.

This study was designed to investigate the molecular mechanisms responsible for the induction of proinflammatory cytokines gene expression and apoptosis in human monocytic cell line THP-1 stimulated by lipoproteins ( LPs) prepared from Mycoplasma genitalium. Cultured cells were stimulated with M. genitalium LP to analyze the production of proinflammatory cytokines and expression of their mRNA by ELISA and RT-PCR, respectively. Cell apoptosis was also detected by Annexin V-FITC-propidium iodide ( PI) staining and acridine orange ( AO)-ethidium bromide ( EB) staining. The DNA-binding activity of nuclear factor-kappa B ( NF-kappa B) was assessed by electrophoretic mobility shift assay ( EMSA). Results showed that LP stimulated THP-1 cells to produce tumor necrosis factor-alpha ( TNF-alpha), interleukin-1 beta ( IL-1 beta), and IL-6 in a dose-dependent manner. The mRNA levels were also upregulated in response to LP stimulation. LPs were also found to increase the DNA-binding activity of NF-kappa B, a possible mechanism for the induction of cytokine mRNA expression and the cell apoptosis. These effects were abrogated by PDTC, an inhibitor of NF-kappa B. Our results indicate that M. genitalium-derived LP may be an important etiological factor of certain diseases due to the ability of LP to produce proinflammatory cytokines and induction of apoptosis, which is probably mediated through the activation of NF-kappa B. Although the Chlamydia trachomatis genome is predicted to encode 50 inclusion membrane proteins, only 18 have been experimentally localized in the inclusion membrane of C. trachomatis-infected cells. Using fusion proteins and anti-fusion protein antibodies, we have systematically evaluated all 50 putative inclusion membrane proteins for their localization in the infected cells, distribution patterns, and effects on subsequent chlamydial infection when expressed ectopically, as well as their immunogenicity during chlamydial infection in humans. Twenty-two of the 50 proteins were localized in the inclusion membrane, and 7 were detected inside the inclusions, while the location of the remaining 21 was not defined. Four (CT225, CT228, CT358, and CT440) of the 22 inclusion membrane-localized proteins were visualized in the inclusion membrane of Chlamydia-infected cells for the first time in the current study. The seven intra-inclusion-localized proteins were confirmed to be chlamydial organism proteins in a Western blot assay. Further characterization of the 50 proteins revealed that neither colocalization with host cell endoplasmic reticulum nor inhibition of subsequent chlamydial infection by ectopically expressed proteins correlated with the inclusion membrane localization. Interestingly, antibodies from women with C. trachomatis urogenital infection preferentially recognized proteins localized in the inclusion membrane, and the immunodominant regions were further mapped to the region predicted to be on the cytoplasmic side of the inclusion membrane. These observations suggest that most of the inclusion membrane-localized proteins are both expressed and immunogenic during C. trachomatis infection in humans and that the cytoplasmic exposure may enhance the immunogenicity. 目的 克隆、表达肺炎嗜衣原体(Cpn)蛋白酶样活性因子(CPAF)的免疫优势区基因(CPAFm),初步评价其在血清学诊断中的应用价值.方法 构建pGEX6p-2/CPAFm重组质粒,诱导表达重组蛋白,十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)鉴定结果;将纯化蛋白免疫新西兰兔,间接酶联免疫吸附试验(ELISA)检测其免疫原性;与人抗Cpn抗血清反应分析其免疫反应性.间接ELISA法检测Cpn IgG和沙眼衣原体(Ct)阳性血清.结果 高效表达和纯化出一相对分子质量约为51.3×103的重组蛋白,蛋白质印迹法(Western blot)证明其能与人抗Cpn IgG抗体发生反应;在被免疫的新西兰兔体内,特异性IgG抗体的滴度为1∶16 000以上,间接ELISA法检测40例Cpn IgG参考血清,阴性和阳性结果的符合率均为100.0%(20/20);与微量免疫荧光法(MIF)对照,检测300例临床血清标本中的IgG抗体,符合率为98.3%;检测Ct阳性血清未见交叉反应.结论 表达的Cpn CPAF免疫优势区重组蛋白具有良好的免疫活性,在Cpn的血清学诊断中具有较高的应用价值.
