摘要:
肺炎嗜衣原体(Chlamydophila pneumoniae,C.pneumoniae,Cpn)及鹦鹉热嗜衣原体(Chlamydophila psittaci,C.psittaci,Cps)是严格细胞内寄生,具有独特发育周期的人兽共患的原核微生物,主要引起呼吸道尤其是肺部感染,并在世界各地广泛流行.为了优化Cpn、Cps临床株的分离培养的关键技术及敏感细胞;建立Cpn及Cps呼吸道感染的动物模型.本研究从患者呼吸道及禽鸟肝组织采集标本,抽提总DNA,PCR法体外扩增种Cpn、Cps ompA基因,经进一步酶切、测序初步鉴定Cpn、Cps阳性标本;同时将阳性标本接种到Hep-2和Vero细胞中培养,通过多次亚培养与传代,感染的单层细胞用甲醇固定,Giemsa和免疫荧光染色法鉴定衣原体包涵体;将临床株扩大培养后,用2×104、2× 105、2×106IFU三个剂量鼻内接种小鼠,分别于感染后5d和10d处死小鼠,显微镜观察受染小鼠各脏器病理变化.结果显示:采用PCR技术从患者呼吸道标本中检测到Cpn阳性标本15例,禽鸟标本中检测到cps阳性标本6例;并成功地在Hep-2及、vero细胞中培养出2株CPn及3株Cps床分离株;发现Hep-2及、Vero细胞内的Cpn. Cps包涵体数目较多,结构致密,连续传代中更稳定,对衣原体感染引起的宿主细胞溶解耐受性较强,更适合用于Cpn, Cps.的分离培养及体外研究。

摘要:
目的表达、鉴定梅毒螺旋体(Tp)重组蛋白Tp0993(rTp0993),为进一步评价其在梅毒血清学诊断中的意义奠定基础。方法 PCR扩增去除信号肽核苷酸序列的Tp0993基因,构建原核重组体pET-28a/Tp0933并诱导表达蛋白;Ni-NTA法纯化重组蛋白,Western blot检测表达产物的抗原性。结果成功构建了原核重组体pET-28a/Tp0993,经诱导表达了一分子量大约为40 Ku的重组蛋白;Western blot结果显示纯化蛋白能被梅毒患者血清特异性识别。结论 rTp0993有较好的抗原性,为进一步研究其在梅毒血清学诊断中的作用奠定了基础。

摘要:
目的 探讨肺炎嗜衣原体(CPN)蛋白酶样活性因子(CPAF)重组蛋白在CPN感染实验室诊断中的应用价值.方法 构建CPAF免疫优势区基因重组质粒,诱导表达并纯化重组蛋白,分析其抗原特异性;间接 ELISA法检测CPN参考血清、临床血清标本中的特异性IgM和IgG抗体.以及沙眼衣原体(Ct)阳性血清.结果 高效表达和有效纯化出一相对分子质量(Mr)约为5.13×10~3的特异性重组蛋白;间接ELISA法测定用其免疫兔血清中的特异性IgG和IgM抗体效价,分别高达1:16 000和1:8 000;检测Cpn IgM和IgG参考血清,阴性和阳性结果符合率均为100.0%;与"金标准"方法MIF比较,检测300份呼吸道感染患者抗血清,IgM、IgG符合率分别为98.3%、99.0%;检测Ct阳性参考血清和阳性临床血清标本,无阳性结果.结论 表达的CPN CPAF免疫优势区重组蛋白具有良好的抗原性,在CPN感染的实验室诊断中具有较高的应用价值.

摘要:
The tissue destruction characteristic of syphilis infection may be caused by inflammation due to Treponema pallidum and the ensuing immune responses to the pathogen. T. pallidum membrane proteins are thought to be potent inducers of inflammation during the early stages of infection. However, the actual membrane proteins that induce inflammatory cytokine production are not known, nor are the molecular mechanisms responsible for triggering and sustaining the inflammatory cascades. In the present study, Tp0751 recombinant protein from T. pallidum was found to induce the production of proinflammatory cytokines, including TNF-α, IL-1βand IL-6, in a THP-1 human monocyte cell line. The signal transduction pathways involved in the production of these cytokines were then further investigated. No inhibition of TNF-a, IL-1β, or IL-6 production was observed following treatment with the SAPK/JNK specific inhibitor SP600125 or with an ERK inhibitor PD98059. By contrast, anti-TLR2 mAb, anti-CD14 mAb, and the p38 inhibitor SB203580 significantly inhibited the production of all three cytokines. In addition, pyrrolidine dithiocarbamate (PDTC), a specific inhibitor of NF-κB, profoundly inhibited the production of these cytokines. Tp0751 treatment strongly activated NF-κB, as revealed by Western blotting. However, NF-κB translocation was significantly inhibited by treatment with PDTC. These results indicated that TLR2, CD14, MAPKs/p38, and NF-κB might be implicated in the inflammatory reaction caused by T. pallidum infection. © Science China Press and Springer-Verlag Berlin Heidelberg 2010.

摘要:
The Chlamydia pneumoniae genome-ncoded open reading frames Cpn0146, Cpn0147, and Cpn0308 were expressed as recombinant proteins for detecting C. pneumoniae-specific antibodies in samples from three groups of individuals including 183 with C. pneumoniae-associated respiratory infection (group 1), 60 healthy blood donors (group 11), and 32 with no known respiratory infection (group 111). The recombinant Cpn0146 was recognized by 71 (38.8% positive recognition rate), 15 (25%) and 1 (3.1%), Cpn0147 by 75 (40.9%), 14 (23.3%), and 2 (6.3%), and Cpn0308 by 82 (44.8%), 16 (26.7%), and 0 (0%) samples from groups I, II, and IIIs, respectively. The positive recognition rates with any of the three antigens were significantly higher in group I than those in groups II and III, suggesting that more individuals from group I were likely infected with C. pneumoniae. This conclusion was confirmed with a commercially available whole organism-based ELISA kit (Savyon Diagnostics Ltd., Ashdod, Israel), which detected C. pneumoniae antibodies in 98 (64.1%), 26 (43.3%), and 4 (12.5%) samples from group I, II, and III, respectively. Comparing to the commercial kit, the recombinant antigen-based detection assays displayed >97% of detection specificity and >87% of sensitivity, suggesting that these recombinant antigens can be considered alternative tools for aiding in serodiagnosis of C. pneumoniae infection. 摘要:
MyD88, a key adaptor molecule required for many innate immunity receptor-activated signaling pathways, was evaluated in a Chlamydia muridarum urogenital tract infection model. Compared with wild-type mice, MyD88 knockout (KO) mice failed to produce significant levels of inflammatory cytokines in the genital tract during the first week of chlamydial infection. MyD88 KO mice developed a Th2-dominant whereas wild-type mice developed a Th1/Th17-dominant immune response after chlamydial infection. Despite the insufficient production of early inflammatory cytokines and lack of Th1/Th17-dominant adaptive immunity, MyD88 KO mice appeared to be as resistant to chlamydial intravaginal infection as wild-type mice based on the number of live organisms recovered from vaginal samples. However, significantly high numbers of chlamydial organisms were detected in the upper genital tract tissues of MyD88 KO mice. Consequently, MyD88 KO mice developed more severe pathology in the upper genital tract. These results together have demonstrated that MyD88-dependent signaling pathway is not only required for inflammatory cytokine production in the early phase of host response to chlamydial infection but also plays a critical role in the development of Th1/Th17 adaptive immunity, both of which may be essential for limiting ascending infection and reducing pathology of the upper genital tract by chlamydial organisms. 目的探讨生殖支原体(Mg)脂质相关膜蛋白(LAMPs)能否经Toll样受体2(TLR2)激活核转录因子KB(NF-KB).方法提取Mg LAMPs刺激THP-I细胞,ELISA检测NF-KBp65活化水平,RT-PCR检测THP-1细胞TLR2 mRNA的表达,随后ELISA检测TLR2中和抗体对LAMPs激活 NF-KBp65的影响,最后用LAMPs刺激瞬时共转染pFLAG -TLR2、pNF-KB-luc、pRL-TK的293T细胞,双荧光素酶报告基因分析TLR2在LAMPs激活NF-KB中的作用.结果 LAMPs能诱导THP-I细胞激活NF-KBp65,并且与LAMPs浓度呈明显的剂量依赖性,当LAMPs为4.0μg/ml时NF-KBp65活化程度最高;LAMPs能上调THP-1细胞TLR2 mRNA的表达;TLR2中和抗体能使LAMPs激活NF-KBp65的活化程度降低60%;共转染pFLAG-TLR2浓度的增加,NF-KB的活化程度明显增加,并且与TLR2的转染量呈剂量依赖性.结论Mg LAMPs能经TLR2激活NF-KB,TLR2介导的激活在LAMPs发挥致病性过程起着重要作用

摘要:
目的表达梅毒螺旋体(Tp)重组蛋白Tp0971(rTp0971),评价其作为候选抗原在梅毒血清学诊断中的意义。方法 PCR扩增Tp0971基因,构建原核重组体pET-28a/Tp0971并诱导表达蛋白,Western blot检测表达产物的抗原性;建立rTp0971-ELISA,检测238份TPPA阳性和120份TPPA阴性血清的Tp特异性IgG,并与TPPA法比较。结果成功高效表达了rTp971;Western blot显示纯化的rTp0971能被梅毒患者血清识别。用rTp0971-ELISA检测临床血清标本的特异性为100%,敏感性为89.1%,总符合率为92.7%。结论 rTp0971有较好的抗原性,可望作为梅毒血清学检测的候选诊断抗原。

摘要:
目的 探讨联合检测外周血端粒酶活性和组织多肽特异性抗原对乳腺癌患者的临床价值.方法 采用聚合酶链反应(PCR)-端粒酶重复序列扩增反应(TRAP)-酶联免疫吸附测定(ELISA)法,分别检测67例乳腺癌患者、31例乳腺良性病变患者及36例体检健康者的外周血单个核细胞的端粒酶活性;同时对其血清应用ELISA法检测组织多肽特异性抗原的表达情况.结果 外周血端粒酶活性和组织多肽特异性抗原的表达均与乳腺癌淋巴结转移密切相关(P<0.05).其中,外周血端粒酶活性的表达还与癌组织的浸润相关(P<0.001).结论 外周血端粒酶活性和组织多肽特异性抗原的表达预示乳腺癌淋巴结转移,并且外周血端粒酶活性的表达还与乳腺癌的局部浸润生长有关.联合检测这2项指标,对患者预后的评估及治疗方案的制定具有重要的意义.

摘要:
Infection with Chlamydia muridarum in the mouse urogenital tract can induce both protective immunity and inflammatory pathologies, which has been used as a model for understanding the immune and pathogenic mechanisms of C. trachomatis infection. We compared the roles of CD28- and CD40 ligand (CD40L)-mediated costimulation in C. muridarum infection. Mice with CD28 or CD80/CD86 gene knockout (KO) displayed an infection course similar to that of wild-type mice during both primary and secondary infection, suggesting that CD28-mediated costimulation is not required for protection against C. muridarum infection. However, mice deficient in CD40L or CD40 displayed a prolonged infection course after primary or secondary infection, suggesting that CD40-CD40L costimulation plays an essential role in the development of anti-C. muridarum immunity. Interestingly, the CD28- or CD80/CD86-deficient mice displayed significantly lower levels of in-flammatory pathologies in the upper genital tracts after primary infection, although the attenuation in inflammation was no longer significant during secondary infection. However, the CD40L or CD40 KO mice developed inflammatory pathologies as severe as those in wild-type mice following either primary or secondary infection despite the obvious deficits in adaptive immunity in these KO mice. The resistance of CD28 or CD80/CD86 KO mice to chlamydial infection correlated with production of gamma interferon, while the development of inflammatory pathologies in CD40L or CD40 KO mice correlated with the production of other proinflammatory cytokines in mouse urogenital tracts during the early stages of the infection. These observations together suggest that C. muridarum-induced protective immunity and inflammatory pathologies can be mediated by distinct costimulatory signals. 摘要:
Mycoplasmas, the smallest free-living, self-replicating bacteria with diameters of 200 to 800 nm, have been reported to be associated with human diseases. It is well known that the mycoplasma lipoprotein/peptide is able to modulate the host immune system, whose N-terminal structure is an important factor in inducing immunity and distinguishing Toll-like receptors (TLRs). However, there is still no clear elucidation about the pathogenic mechanism of mycoplasma lipoprotein/peptide and the signaling pathway. Some researchers have focused on understanding the structures of these proteins and the relationships between their structure and biological function. This review provides an update on the research in this field.
