MicroRNAs (miRNAs) play essential roles in many biological processes. It is known that aberrant miRNA expression contributes to some pathological conditions. However, it is not known whether miRNAs play any role in the development of insulin resistance in adipocytes, a key pathophysiological link between obesity and diabetes. To investigate the function of miRNAs in the development of insulin resistance, using miRNA microarray analysis we compared miRNA expression profiles between normal insulin-sensitive 3T3-L1 adipocytes and 3T3-L1 adipocytes rendered insulin resistant following treatment with high glucose (25 mmol/L) and high insulin (1 μmol/L). Furthermore, adipocytes were transfected with specific antisense oligonucleotides against miRNA-320 (anti-miR-320 oligo) and the effects on the development of insulin resistance were evaluated. We identified 50 upregulated and 29 downregulated miRNAs in insulin-resistant (IR) adipocytes, including a 50-fold increase in miRNA-320 (miR-320) expression. Using bioinformatic techniques, the p85 subunit of phosphatidylinositol 3-kinase (PI3-K) was found to be a potential target of miR-320. In experiments with anti-miR-320 oligo, insulin sensitivity was increased in IR adipocytes, as evidenced by increases in p85 expression, phosphorylation of Akt and the protein expression of the glucose transporter GLUT-4, as well as insulin-stimulated glucose uptake. These beneficial effects of anti-miR-320 oligo were observed only in IR adipocytes and not in normal adipocytes. In conclusion, the miRNA profile changes in IR adipocytes compared with normal 3T3-L1 adipocytes. Anti-miR-320 oligo was found to regulate insulin resistance in adipocytes by improving insulin-PI3-K signalling pathways. The findings provide information regarding a potentially new therapeutic strategy to control insulin resistance. © 2009 Blackwell Publishing Asia Pty Ltd.

An iodine and boron co-doped TiO<inf>2</inf> photocatalyst was prepared by the hydrolyzation-precipitation method. X-ray diffraction (XRD), ultraviolet-visible diffuse reflectance spectroscopy (UV-Vis DRS), and X-ray photoelectron spectroscopy (XPS) were applied to characterize the crystalline structure, light absorbing ability, and the chemical state of iodine and boron in the photocatalysts. The results of photocatalytic degradation of methyl orange demonstrated that the I-B-TiO<inf>2</inf> catalyst prepared at 400 °C for 3 h exhibited the highest photocatalytic activity with a methyl orange degradation ratio of 61% under visible-light (λ&le 420 nm) irradiation for 120 min. The characterization results revealed that I-B-TiO<inf>2</inf> is in conformity with the anatase TiO<inf>2</inf> and that the doping of iodine and boron ions could efficiently inhibit the grain growth. Doped iodine was present in the multivalent forms of I<sup>7+</sup>, I<sup>-</sup> and I<sup>5+</sup>. Doped boron was present as B<sup>3+</sup> in an as-prepared sample, forming a possible chemical environment such as B-O-Ti. Overall, the doping of I and B enhanced the ability of TiO<inf>2</inf> to absorb visible-light, and it was observed that the photocatalytic activity of I-B-TiO<inf>2</inf> was enhanced by the synergistic effect of I and B. Neuropathy target esterase (NTE) was proposed as the initial target during the process of organophosphate-induced delayed neuropathy (OPIDN) and adult hens are the animal model of OPIDN. However, little has been known about the sequence and characteristics of chicken NTE. Here, we firstly identified the full length cDNA of chicken NTE (cNTE), which contained an open reading frame of 3966 nucleotides encoding 1321 amino acids. Chicken NTE had two distinct regions, one was the regulatory domain (cNTER) and the other was the catalytic domain (cNEST). Over-expression of cNTER in mammalian cells did not show any NTE activity, whereas cNEST had NTE activity. Cells expressing cNTER tagged with green fluorescent protein (GFP) showed accumulation of cNTER-GFP in an endoplasmic reticulum-like localization pattern. In addition, macroautophagy and the proteasome pathways were found to be involved in the degradation of cNTER, but not cNEST. These results first showed that cNTE was an ER-anchored protein and degraded by macroautophagy as well as the proteasome. © 2009 Elsevier B.V. All rights reserved.

Aims: Neuropathy target esterase (NTE) was proposed as the initial target during the process of organophosphate-induced delayed neuropathy (OPIDN) in humans and some sensitive animals. NTE was recently identified as a novel phospholipase B that is anchored to the cytoplasmic side of the endoplasmic reticulum. However, little is known about the degradation of NTE. In this study, we have investigated the role of the macroautophagic-lysosomal pathway in NTE degradation in neuronal and non-neuronal cells. Main methods: Macroautophagy inhibitors and activators were used to interrupt the lysosomal pathway, and NTE protein level was followed using western blotting analysis. A fluorescent microscopy assay was used to determine the co-localization of NTE and lysosomes. Key findings: Western blotting analysis showed that the macroautophagy inhibitors 3-methyladenine and ammonium chloride increased the levels of a heterologously expressed NTE-GFP fusion protein as well as endogenous NTE. Starvation had the opposite effect. The role of macroautophagy in NTE degradation was further Supported by the co-localization of exogenous NTE with lysosomes in starved COS7 cells. Furthermore, the contribution of NTE activity and protein domains to the degradation of NTE by macroautophagy was investigated, showing that both the transmembrane and regulatory domains played a role in the degradation of NTE and that the catalytic domain, and thus NTE activity, was not involved. Significance: Our findings clearly demonstrate, for the first time, that the macroautophagy/lysosome pathway plays a role in controlling NTE quantity, providing a further understanding of the function of NTE. 摘要:
A novel method for the simultaneous determination of 1-hydroxypyrene (1-OHP), β-naphthol (β-NAP) and 9-hydroxyphenanthrene (9-OHPe) in human urine has been established by using synchronous fluorescence spectrometry. It was based on the fact that synchronous fluorescence spectrometry can resolve the broad-band overlapping of conventional fluorescence spectra, which arise from their similar molecular structures. Only one single scan is needed for quantitative determination of three compounds simultaneously when Δλ = 15 nm is chosen. The signals detected at these three wavelengths, 369.6, 330.0 and 358.0 nm, vary linearly when the concentration of 1-OHP, β-NAP and 9-OHPe is in the range of 2.16 × 10-8-1.50 × 10-5 mol L-1, 1.20 × 10-7-1.10 × 10-5 mol L-1 and 1.07 × 10-7-3.50 × 10-5 mol L-1, respectively. The correlation coefficients for the standard calibration graphs were 0.994, 0.999 and 0.997 (n = 7) for 1-OHP, β-NAP and 9-OHPe, respectively. The limits of detection (LOD) for 1-OHP, β-NAP and 9-OHPe were 6.47 × 10-9 mol L-1, 3.60 × 10-8 mol L-1 and 3.02 × 10-8 mol L-1with relative standard deviations (R.S.D.) of 4.70-6.40%, 2.80-4.20%, 3.10-4.90% (n = 6), respectively. The method described here had been applied to determine traces of 1-OHP, β-NAP and 9-OHPe in human urine, and the obtained results were in good agreement with those obtained by the HPLC method. In addition, the interaction modes between β-cyclodextrin (β-CD) and 1-OHP, β-NAP or 9-OHPe, as well as the mechanism of the fluorescence enhancement were also discussed. © 2009 Elsevier B.V. The resonance Rayleigh scattering (RRS), second-order scattering (SOS) and frequency-double scattering (FDS) spectra of sodium dodecylbenzene sulfonate (SDBS) (anionic surfactant (AS)) with acridine orange (AO) system were studied. Experimental results showed that when λ<inf>em</inf> = λ<inf>ex</inf> = 537 nm, the RRS peak of AO was greatly enhanced with the increase of SDBS concentration at a pH range of 1.8-4.0. The linear range of the calibration curve for SDBS was 0.028-8.71 mg L<sup>-1</sup> with a detection limit of 8.36 μg L<sup>-1</sup> when the AO concentration was 2.5 ×10<sup>-5</sup>mol L<sup>-1</sup>. The method has been applied to the determination of trace amount of AS in environmental water samples with satisfactory results. In addition, when λ<inf>em</inf> = 321 nm and λ<inf>ex</inf> = 642 nm, the intensity of FDS was proportional to the SDBS concentration ranging from 0.014 to 8.71 mg L<sup>-1</sup> and the correlation coefficient was 0.993 with a detection limit of 4.31 μg L<sup>-1</sup>;when λ<inf>em</inf> = 642 nm and λ<inf>ex</inf> = 321 nm, the intensity of SOS was proportional to the SDBS concentration ranging from 0.050 to 8.71 mg L<sup>-1</sup>, and the correlation coefficient was 0.993 with a detection limit of 14.9 μg L<sup>-1</sup>. ©2008 Elsevier B.V. 摘要:
To investigate the genotoxic potencies of extractable organic matter (EOM) in aerosols, fine air particulate matter (PM<inf>2.5</inf>) was collected simultaneously at a roadside (1.2 m above ground) and at a rooftop location (50 m above ground) in urban Guangzhou (China) during a nonhaze period in September 2006 and a haze period in October 2006. Particle-bound organics were extracted and separated into aliphatic, aromatic, and polar fractions. The genotoxicity of total and fractionated extracts were tested by single-cell electrophoresis (comet assay) with human blood lymphocytes. The PM<inf>2.5</inf> concentrations usually exceeded the U.S. National Ambient Air-Quality Standard level (65 μg/m<sup>3</sup>) at both the roadside and the rooftop. During nonhaze days, the roadside samples showed substantially higher PM<inf>2.5</inf> levels (108-130 μg/m<sup>3</sup>) and significantly higher genotoxic effects of total and fractionated EOM (p <0.05 for >10 m<sup>3</sup> air equivalent/ml) than the rooftop samples. During haze days, however, PM <inf>2.5</inf> levels and genotoxic potencies of rooftop samples were drastically elevated and comparable to those of roadside samples, implying that during haze episodes, most people in the urban area are exposed to PM <inf>2.5</inf> pollution as serious as in the heavily polluted roadside microenvironment. All total EOM samples showed significant (p <0.05) dose-response effects, and their effects as olive tail moment were less than the sums of the three fractions. Aromatic fractions of EOM exhibited the greatest genotoxic potencies, but polar fractions also contributed substantially to DNA-damaging effects. Polycyclic aromatic hydrocarbons and nitrated derivatives likely are the most important species responsible for the genotoxicity of EOM in PM<inf>2.5</inf>. ©2008 SETAC.

It was discovered that a second order spectra data matrix of Sudan I produced from the solvent components gradual change-visible absorption spectra can be expressed as the combination of two bilinear data matrices. Based on this discovery, a new method for the determination of Sudan I in gray systems using second order calibration algorithms has been developed. The second order calibration algorithms were based on the popular parallel factor analysis (PARAFAC) and rank annihilation factor analysis (RAFA), respectively. In the method described here, the components of the solvent were changed gradually by adding ethanol into cyclohexane, the absorption spectra of Sudan I and chilli samples in a series of cyclohexane-ethanol mixed solvents with various ethanol volume fractions were recorded, and then the second order data were obtained from the solvent components gradual change-visible absorption spectra. Thus, the concentration of Sudan I in a gray system could be determined from the spectra matrices using second order calibration algorithms. This method is simple, convenient and dependable. The method has been used to determine Sudan I in chilli powder with satisfactory results. © 2007 Elsevier B.V. All rights reserved.

A novel method for the determination of trace naphthols was proposed based on naphthols, potassium bromate and pyronine Y in dilute sulfuric acid medium can react to form ion-association complexes, which not only resulted in the changes of the three-dimension fluorescence spectra, absorption spectra and the quenching of fluorescence, but also resulted in the great enhancement of resonance light scattering (RLS). The characteristics of RLS spectra, the effective factors and optimum conditions of reaction were studied. It was found that the enhanced intensity of RLS at 396 nm was proportional to the concentration of 1-naphthol and 2-naphthol in the range of 41.4 similar to 3024 ng mL(-1) and 38.3 similar to 3068 ng mL-1, respectively. The urine samples analysis of the relative standard deviation was 3.1-7.1% and the average recovery was 96.0% (n = 6). The present method had been applied to the determination of trace naphthols in human urine, the obtained results were in good agreement with those obtained by the HPLC method; moreover, we had carried on an initial study of the reaction mechanism in terms of spectral characteristics. 摘要:
It has been known that tri-ortho-cresyl phosphate (TOCP) can induce delayed neurotoxicity in humans and sensitive animal species: however, it also has influence on the developing central nervous system or differentiating neuronal cells. In this study, the effects of TOCP on cell proliferation and cell cycle regulation and the mechanisms that contribute to this effect were investigated by using human neuroblastoma SH-SY5Y cell line. Treatment of the cells with TOCP suppressed cell proliferation and reduced cell viability in a dose- and time-dependent manner. Analysis of cell cycle profile indicated that TOCP blocked cell cycle progression by arresting the cell cycle at G(1) phase. The data of determination of cell cycle regulated molecules at mRNA and protein levels showed that TOCP decreased cyclin D I and increased p21 expression, while did not affect the p53 and p27 levels. Thus, these results indicated that TOCP might induce potential neurodevelopmental toxicity, and a possible mechanism of this toxicity might be the disturbance of cell proliferation by disrupting cell cycle regulatory proteins cyclin D1 and p21 expression. A novel method for the rapid and sensitive analysis of 1-hydroxypyrene (1-OHP) in human urine has been developed that uses a resonance light scattering (RLS) technique. The assay was based on the interaction of ethyl violet (EV) with 1-hydroxypyrene to form an ion-associate complex, which resulted in the enhancement of RLS intensity and the appearance of new RLS spectra. In the presence of anionic surfactant, the maximum RLS peak of the system was located at 396 nm at pH 8.0. Under the optimum conditions, it was found that the enhanced RLS intensity was directly proportional to the concentration of 1-hydroxypyrene in the range of 4.0 - 982 μg l-1. The detection limit was 1.2 μg l-1 and the recoveries of 1-hydroxypyrene were 92.8 - 102.3% (n = 6). The proposed method was successfully applied to the analysis of human urine samples. The results of 1-hydroxypyrene were in agreement with those obtained by the method of high-performance liquid chromatography. 2007 © The Japan Society for Analytical Chemistry.

Previous studies have shown that rapid eye movement (REM) sleep deprivation (RSD) exerts a detrimental effect on some memory tasks. However, whether post-learning RSD impairs memory for fear extinction, an important model of inhibitory learning, remains to be elucidated. The present study examined the effects of post-extinction RSD from 0 to 6 h and 6 to 12 h on recall of fear extinction tested 24 h after extinction training. We found that RSD from 0 to 6 h significantly increased freezing when recall of extinction of cued fear was tested in the context in which rats received extinction training whereas RSD from 6 to 12 h had no effect (experiments 1 and 2, two hippocampus-independent memory tasks). RSD at either time point had no effect on freezing when recall of extinction of cued fear was tested in the context different from that in which extinction training occurred (experiment 3, a hippocampus-dependent memory task). Additionally, we observed no effect of RSD at either time point on freezing during recall test for extinction of contextual fear (experiment 4, a hippocampus-dependent memory task). These results suggest that the effects of post-extinction RSD on memory for fear extinction are complex. RSD impairs recall of fear extinction in hippocampus-independent tasks, but does not affect recall of fear extinction in hippocampus-dependent tasks. Our findings extend previous research on the effects of RSD on learning and memory and support the notion that REM sleep is involved in memory process of certain tasks. (c) 2006 IBRO. Purpose: To explore the relationship between social support and posttraumatic stress disorder (PTSD) among flood victims. Methods: A cross-sectional survey was carried out in 2000 among individuals who had suffered floods in 1998 in Hunan, China. Multistage sampling was used to select the subjects from the flood-affected areas. PTSD was diagnosed according to DSM-IV criteria, and social support was measured according to a social support rating scale. Data were collected through face-to-face interviews using a structured questionnaire. Multiple logistic regression analysis and confirmatory factor analysis was used to examine the relationship between social support and PTSD. Results: Out of a total of 25,478 subjects interviewed, 2336 (9.7%) were diagnosed as having PTSD. PTSD was significantly associated with total social support (odds ratio [OR] 0.80, 95% confidence interval [CI], 0.78-0.82), subjective support (OR 0.48, 95%CI, 0.44-0.52), and support utilization (OR 0.53, 95%CI, 0.49-0.57). Conclusion: PTSD in flood victims is significantly associated with social support; subjective support and support utilization may play more important roles in mitigating the impact of flood than objective support. © 2007 Elsevier Inc. 摘要:
Heavy metals in municipal solid waste incineration bottom ash (MSWIBA) may leach into soil and groundwater and pose long-term risks to the environment. In this study, toxicity characteristic leaching procedure (TCLP) was carried out on the MSWIBA from Macao. Heavy metals in leachates were determined by inductively coupled plasma mass spectrometry (ICP-MS) and inductively coupled plasma atomic emission spectrometry (ICP-AES), and genotoxicity of leachates was also evaluated by micronucleus (MN) assay with Vicia faba root tip cells. The results showed that the concentrations of aluminium (Al), manganese (Mn), cobalt (Co), cadmium (Cd) and mercury (Hg) in the leachates were less than 0.01 mg l<sup>-1</sup>, and those of iron (Fe), copper (Cu) and molybdenum (Mo) were less than 0.1 mg l<sup>-1</sup>. The concentrations of chromium (Cr), zinc (Zn), selemium (Se), strontium (Sr), barium (Ba) and caesium (Cs) were between 0.11 mg l<sup>-1</sup> and 2.19 mg l<sup>-1</sup>. Lead (Pb) concentrations, in particular, reached as high as 19.6 mg l<sup>-1</sup>, significantly exceeding the maximum concentration limit (5 mg l<sup>-1</sup> for lead by TCLP). Compared with the negative group, a significant increase of MN frequencies was observed in the leachate-exposed groups (P <0.05). With the increase of heavy metals in the leachates, the toxic effects on the Vicia faba root tip cells increased, implying that heavy metals were the main factors causing the genotoxic effects. Our results suggested that apart from chemical analysis, bioassays like the MN assay of Vicia faba root tip cells should also be included in a battery of tests to assess the eco-environmental risks of bottom ashes before decisions can be made on the utilization, treatment or disposal. ©2006 Elsevier Ltd. 摘要:
This paper investigated the influence of season and altitude on the occurrence of nematode-trapping fungi in cattle faeces. Six hundred and sixty samples of cattle faeces deposited on three plateau pastures with different altitudes in the west of Yunnan Province, China, were examined in 2004. A total of 17 species of nematode-trapping hyphomycetes were isolated from these samples. The predominant species from all three plateau pastures were Arthrobotrys oligospora, A. musiformis, Monacrosporium ellipsosporum, and M. thaumasium. Species with adhesive networks were the most frequently isolated. Overall, species diversity index was negatively correlated with altitude and was different among seasons within the same site. Levels of diversity were highest in the summer, followed by autumn, spring, and winter. The conidia of the hyphomycetes isolated here germinated normally on medium containing cattle faeces, with species developing adhesive networks having the highest rate of germination. However, the rate of conidial trap (CT) formation was lower in species with adhesive networks than those in other species.

摘要:
The sensitivity of cancer cells as well as normal cells in response to ionizing radiation (IR) is believed to be associated with the early inducible expression of specific genes. Using cDNA microarray technology, here we explored and compared the global transcriptional changes in human lymphoblastoid AHH-1 cells irradiated with 0.05-, 0.2-, 0.5-, 2.0- and 10-Gy doses of gamma-rays 4 h after exposure. A dose as low as 0.05 Gy was efficient in inducing a transcriptional response including the up-regulation of 25 genes, some of which are involved in signal transduction pathways, e.g. BMPR2, GPR124, MAPK8IP2 and AGGF1, and the down-regulation of 18 genes. Expression of some genes was altered only at a specific dose. Most importantly, we discovered a number of radiation-response genes, e.g. DNA repair gene XPC, tumor protein p53 inducible protein 3 gene (TP53I3), immediate early response 5 gene, whose transcriptional levels were increased or depressed by IR in a dose-dependent trend within the dose range 0.05-10 Gy. The dose-dependent induced expression of TP53I3 and XPC was confirmed by Northern blot analyses. Using quantitative real-time PCR, we further confirmed that XPC gene induction was dose dependent as well as time dependent, reaching a peak 4 h post-2 Gy and 10 h post-0.05 Gy. The maximum induced expression level of the XPC gene was higher after 2 Gy (3.2-fold) than 0.05 Gy (1.93-fold). The identification of these radiation-inducible genes, especially those exhibiting a dose-dependent response, not only expands our knowledge of the mechanisms underlying the diverse biological effects induced by IR, but provides candidates for developing novel biomarkers of radiation injury.

研究了吩噻嗪类染料甲苯胺蓝(TB)与DNA作用的共振光散射光谱,在pH 10~ll的范围内,加入 DNA导致甲苯胺蓝共振光散射增强,在350 nm处,存在一共振光散射增强峰,其强度与DNA浓度呈线性关系,据此建立了一种测定DNA的共振光散射法。对于ctDNA,方法的线性范围为0~900 ng·mL~(-1),检出限为6.75 ng·mL~(-1),RSD为3.7%;对于fsDNA,方法的线性范围为0~900 ng·mL~(-1),检出限为2.99 ng· mL~(-1),RSD为5.6%.已用于合成样品中DNA的测定。

制备镧型阳离子交换树脂,用于饮水除氟.用硝酸镧溶液将001×7强酸性阳离子交换树脂在优化的条件下转变为镧型树脂;利用氟与树脂上的镧作用除去水中氟.用硝酸镧溶液再生,树脂可反复用于水中除氟.镧型树脂的转型、除氟及再生优化条件:d树脂=0.315~0.600mm,V树脂∶V再生液=1∶6,t转型=48h,T=298.16K(25℃).动态、静态除氟容量分别为5.60、4.08mg/g,通过再生,镧型树脂可使用8个以上除氟周期.镧型树脂是饮水除氟的一种新型材料,设备简单,操作方便,除氟容量大、效率高、周期长.

摘要:
为揭示双酚A(BPA)的发育毒性及其可能的作用机制,采用植入后全胚胎培养模型,将8.5日龄BalB/C小鼠胚胎和9.5日龄大鼠胚胎分别与含BPA的大鼠即刻离心血清共培养48h(BPA终浓度为0、40、60、80、100 mg/L),观察双酚A对小鼠和大鼠胚胎生长发育和组织器官形态分化的影响.结果显示,随着BPA剂量的增加胚胎在体外的生长发育受影响越严重,呈现出明显的剂量-效应及剂量-反应关系.≥60 mg/L的BPA可诱发小鼠及大鼠胚胎的卵黄囊生长和血管分化不良、生长迟缓及形态分化异常,严重者出现体位异常、神经系统、鳃弓发育异常及小眼、小肢芽等畸形.提示在较高剂量下,BPA对体外培养的大鼠和小鼠胚胎具有一定的毒性作用,其中对卵黄囊胎盘的结构和功能的影响可能是其致畸作用的机制之一.
