Atherosclerosis is the leading cause of acute cardiovascular events, and vascular calcification is an important pathological phenomenon in atherosclerosis. Recently, many studies have shown that immune cells are closely associated with the development of atherosclerosis and calcification, but there are many conflicting viewpoints because of immune system complications, such as the pro-atherosclerotic and atheroprotective effects of regulatory B cells (Bregs), T helper type 2 (Th2) cells and T helper type 17 (Th17) cells. In this review, we summarize the studies on the roles of immune cells, especially lymphocytes and macrophages, in atherosclerotic calcification. Furthermore, we prepared graphs showing the relationship between T cells, B cells and macrophages and atherosclerotic calcification. Finally, we highlight some potential issues that are closely associated with the function of immune cells in atherosclerotic calcification. Based on current research results, this review summarizes the relationship between immune cells and atherosclerotic calcification, and it will be beneficial to understand the relationship of immune cells and atherosclerotic calcification.

Background: Programmed cell death-ligand 1 (PD-L1) expression has been associated with shorter progression-free survival (PFS) of crizotinib-treated patients with anaplastic lymphoma kinase (ALK) rearranged non-small cell lung cancer (NSCLC). However, the association between PD-L1 expression and overall survival (OS) in ALK-rearranged NSCLC remains unclear. In this study, we investigated the survival implication of baseline PD-L1 expression status in crizotinib-treated patients with ALK-rearranged advanced NSCLC. Methods: Between October 1, 2015, and October 31, 2021, we retrospectively analyzed the baseline PD-L1 expression levels using immunohistochemistry 22C3 assay of tissue samples from 128 patients with ALK-rear-ranged advanced lung adenocarcinoma who were treated with first-line crizotinib. Results: Of the 128 baseline tumor specimens analyzed, a majority (76.6%, n = 98) had low PD-L1 expression (tumor proportion score (TPS) < 50%), wherein 58.6% (n = 75) had < 1% and 18.0% (n = 23) had 1%-49%, and the remaining 23.4% (n = 30) had high PD-L1 expression level (TPS >= 50%). High baseline PD-L1 expression was not associated with any clinical characteristic examined. Patients with high baseline PD-L1 (n = 30) expression level had significantly shorter median PFS (6 vs 11 months, p = 0.011) and OS (17 vs 53 months, p = 0.023) on crizotinib treatment than those with low PD-L1 level (n = 98). Conclusions: A subset of patients with ALK-rearranged NSCLC having high baseline PD-L1 expression level (TPS of >= 50%) had poorer survival outcomes despite crizotinib therapy. Our study raises the need to investigate alternative treatment strategies to improve survival outcomes of this patient subset.

The generation of defects, such as large-sized brittle phases and cracks, remains challenging due to the high carbon diffusion during repairing cast iron. To resolve this problem, a new austenitic Fe-14Ni-19Cr material was designed to repair QT500-7 by laser cladding under N(2 )protective atmosphere. The results show that the defect formation was effectively suppressed, and a low-content carbon (similar to 0.18 %) was diffused from QT500-7 substrate into the laser-repaired layer. The microhardness increased steadily from the substrate of similar to 200 HV0.2 to the laser-repaired layer of similar to 257 HV0.2. The laser-repaired specimen exhibits mechanical properties with ultimate tensile strength of 490 MPa, yield strength of 401 MPa and elongation of 7.7 %, which is equivalent to the QT500-7 substrate. This study will be potential in repairing high-carbon cast iron offering high performance at low cost.

This review thoroughly presents the recent progress of Deinococcus radiodurans, recombinant D. radiodurans and immobilized D. radiodurans as superior adsorbents to efficiently remove toxic heavy metals ions and radionuclides. Finally, a summary and prospect on the opportunities and challenges of D. radiodurans are provided.

Despite activated autophagy ameliorating hepatocyte steatosis and metabolic associated fatty liver disease (MAFLD), mechanisms underlying the beneficial roles of autophagy in hepatic deregulation of lipid metabolism remain undefined. We explored whether autophagy can ameliorate oleic acid (OA)-induced hepatic steatosis by suppressing pyroptosis. Pyroptosis is involved in hepatocyte steatosis induced by OA. In addition, autophagy flux was blocked in OA-treated hepatocytes. Treatment with OA induced lipid accumulation in liver cell line L-02, which was attenuated by rapamycin (Rap), an autophagy agonist, while aggravated by autophagy inhibitor bafilomycin A1 (Baf A1). Inversely, treatment with pyroptotic agonist Nigericin aggravated OA-induced hepatic steatosis, while pyroptosis antagonist disulfiram ameliorated this effect. Mechanistically, treatment with Rap downregulated the expression of pyroptosis-related proteins, including NLRP3, Caspase-1, IL-18, GSDMD expression evoked by OA, thus improving pyroptosis in hepatic steatosis. Significantly, overexpression of ATG5 obviously downregulated cleaved caspase-1 expressions without altering the total caspase1 expressions in hepatic cell steatosis. Taken together, our studies strongly demonstrated that the activation of ATG5 inhibits pyroptosis to improve hepatic steatosis and suggest autophagy activation as a potential therapeutic strategy for pyroptosis-mediated MAFLD.

Fumarates (fumaric acid esters), primarily dimethyl fumarate (DMF) and monoethyl fumarate (MEF) and its salts, are orally administered systemic agents used for the treatment of psoriasis and multiple sclerosis. It is widely believed that the pharmaceutical activities of fumarates are exerted through the Keap1-Nrf2 pathway. Although it has been revealed that DMF and MEF differentially modify specific Keap1 cysteine residues and result in the differential activation of Nrf2, how the modification of DMF and MEF impacts the biochemical properties of Keap1 has not been well characterized. Here, we found that both DMF and MEF can only modify the BTB domain of Keap1 and that only C151 is accessible for covalent binding in vitro. Dynamic fluorescence scanning (DSF) assays showed that the modification of DMF to Keap1 BTB increased its thermal stability, while the modification of MEF dramatically decreased its thermal stability. Further crystal structures revealed no significant conformational variation between the DMF-modified and MEF-modified BTBs. Overall, our biochemical and structural study provides a better understanding of the covalent modification of fumarates to Keap1 and may suggest fundamentally different mechanisms adopted by fumarates in regulating the Keap1-Nrf2 pathway. Chlamydia trachomatis persistent infection is the leading cause of male prostatitis and female genital tract diseases. Inhibition of host cell apoptosis is the key to maintaining Chlamydia survival in vivo, and long noncoding RNAs (lncRNAs) play important roles in its developmental cycle and pathogenesis. However, it is not clear how lncRNAs regulate persistent Chlamydia infection. Here, using a microarray method, we identified 1718 lncRNAs and 1741 mRNAs differentially expressed in IFN-gamma-induced persistent C. trachomatis infection. Subsequently, 10 upregulated and 5 downregulated differentially expressed lncRNAs were verified by qRT-PCR to confirm the reliability of the chip data. The GO and KEGG analyses revealed that differentially regulated transcripts were predominantly involved in various signalling pathways related to host immunity and apoptosis response. Targeted silencing of three lncRNAs (MIAT, ZEB1-AS1 and IRF1) resulted in increased apoptosis rates. Furthermore, interference with lncRNA MIAT caused not only an obvious downregulation of the Bcl-2/Bax ratio but also a marked release of cytochrome c, resulting in a significantly elevated level of caspase-3 activation. Meanwhile, MIAT was involved in the regulation of chlamydial development during the persistent infection. Collectively, these observations shed light on the enormous complex lncRNA regulatory networks involved in mitochondria-mediated host cell apoptosis and the growth and development of C. trachomatis.

While deep learning models have contributed to the advancement of sensor-based human activity recognition (HAR), it usually requires large amounts of annotated sensor data to extract robust features. To alleviate the limitations of data annotation, contrastive learning has been applied to sensor-based HAR. One of the essential factors of contrastive learning is data augmentation, significantly impacting the performance of pretraining. However, current popular augmentation methods do not achieve competitive performance in contrastive learning for sensor-based HAR. Motivated by this issue, we propose a new sensor data augmentation method by resampling, which introduces variable domain information and simulates realistic activity data by varying the sampling frequency to maximize the coverage of the sampling space. The resampling augmentation method was evaluated in supervised learning and contrastive learning [SimCLR for HAR (SimCLRHAR) and MoCo for HAR (MoCoHAR)]. In the experiment, we use four datasets, UCI-HAR, MotionSense, USC-HAD, and MobiAct, using the mean F1-score as the evaluation metric for downstream tasks. The experimental results show that the resampling data augmentation outperforms all state-of-the-art augmentation methods in supervised learning and contrastive learning with a small amount of labeled data. The results also demonstrate that not all data augmentation methods have positive effects in contrastive learning frameworks.

Using a sample of (10.09 +/- 0.04) x 10(9) J/psi events collected with the BESIII detector operating at the BEPCII storage ring, a partial wave analysis of the decay J/psi -> gamma eta eta ' is performed. The first observation of an isoscalar state with exotic quantum numbers J(PC) = 1(-+), denoted as eta(1) (1855), is reported in the process J/psi -> gamma eta(1) (1855) with eta(1) (1855) -> eta eta '. Its mass and width are measured to be (1855 +/- 9(-1)(+6) MeV/c(2) and (188 +/- 18(-8)(+3) MeV, respectively, where the first uncertainties are statistical and the second are systematic, and its statistical significance is estimated to be larger than 19 sigma.
