摘要:
采用生物吸附法去除废水中Cu~(2+),研究了南海红树林内源真菌Fusarium sp.#ZZF51吸附Cu~(2+)的行为特性、吸附模型及吸附机理。结果表明:受试菌吸附Cu~(2+)的吸附平衡时间为90 min,常温常压下吸附最佳pH值为6.5,Cu~(2+)初始浓度50 mg/L,吸附时间90 min,此时受试菌对Cu~(2+)的吸附率和吸附容量分别为82.14%和20.53 mg/g。Langmuir方程比Freundlich方程能更好地描述该受试菌对Cu~(2+)的平衡吸附行为。通过比较受试菌吸附Cu~(2+)前后红外光谱图得知,在吸附过程中,羟基和羰基都起着十分重要的作用。

摘要:
A novel and simple sensor is developed in this paper for melamine detection, which is based on an electropolymerized molecularly imprinted polymer (MIP) of para-aminobenzoic acid (pABA). The poly(para-aminobenzoic acid) (P-pABA) film was deposited in a pABA solution by potentiodynamic cycling of potential with and without the template (melamine) on a glassy carbon electrode. The surface feature of the modified electrode was characterized by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). The molecular imprinted sensor was tested by differential pulse voltammetry (DPV) to verify the changes in redox peak currents of hexacyanoferrate. Several important parameters controlling the performance of the P-pABA were investigated and optimized. In the optimal conditions, the relative redox peak currents of hexacyanoferrate were linear. The concentration of melamine ranged from 4.0 μM to 0.45 mM, with a linear correlation coefficient of 0.9992. The detection limit was 0.36 μM (S/N = 3). The MIP sensor was successfully applied to the determination of melamine in milk products and showed high selectivity, sensitivity, and reproducibility. The results of this research demonstrate that it is feasible to use the molecular imprinting methodology when preparing sensing devices for analytes that are electrochemically inactive. © 2011 Elsevier Ltd. 摘要:
A copper-catalyzed N-N bond-forming reaction was performed by a dehydrogenative homocoupling of N-alkylanilines, affording N,N-dialkyl-N,N- diphenylhydrazines in 72-88% yields. This new strategy has the advantages of direct synthesis from N-alkylanilines, using air as the oxidant, convenient manipulations, mild reaction conditions and moderate to good yields. A possible mechanism by coordination and reductive elimination has been proposed. © Georg Thieme Verlag Stuttgart New York.

摘要:
This work aimed to investigate the cellular response of human airway epithelial cells (A549) to oxidative stress induced by benzo(a)pyrene [B(a)P]. Levels of intracellular reactive oxygen species (ROS) and lipid peroxidation were investigated in A549 cells treated with varying concentrations of B(a)P. A comparative proteomic analysis of total proteins was performed in cells treated with 1 μM B(a)P [B(a)P-1] and untreated cells. The expression of Mn superoxide dismutase (Mn SOD), one of the identified down-regulated proteins in B(a)P-1 cells, was then analyzed by Western blotting. The total antioxidant activity, total superoxide dismutase activity, catalase (CAT) activity, and glutathione reductase (GR) activity were all analyzed after B(a)P treatment. Our results demonstrated that 1 μM B(a)P could induce ROS generation and lead to lipid peroxidation in A549 cells, and 23 differentially expressed proteins were identified. The expression levels of Mn SOD and the total SOD were induced at 0.1 μM and suppressed at 1 μM and 10 μM. Up-regulation of CAT and GR activity resulted in an increase in total antioxidant activity in A549 after exposure to B(a)P. These findings provide a basis for understanding the mechanisms of mitochondrial dysfunction and perturbation of antioxidant status induced by B(a)P on airway epithelial cells. © 2011 Informa Healthcare USA, Inc.

摘要:
采用离子交换法、固相分散法和微波固相法等不同方法制备了Cu/HZSM-5催化剂,以BET、XRD和XPS等手段对催化剂样品进行了表征。结果表明,不同方法制备的Cu/HZSM-5催化剂上Cu物种的落位分布状态不同,离子交换法制备的催化剂Cu物种更多地落位于分子筛孔道内,微波固相法和固相分散法制备的催化剂Cu物种较多地落位分布在分子筛外表面。固相分散法制备的样品未能使铜物种完全分散于分子筛表面,在13.1°、16.8°、35.5°和38.0°等处仍存在CuO的晶相衍射峰。催化分解NO反应的活性考察结果表明,用微波固相法制备的催化剂催化分解NO的活性及稳定性明显超过另两种方法所制备的催化剂,在无氧条件下NO最初转化率高达89.2%,经反应25h后,转化率仍维持在70%以上;在富氧气氛下催化分解NO活性降低速率低于由离子交换法制备的催化剂。结合表征结果可以得出,落位于分子筛外表面以离子交换态形式存在的Cu物种对催化分解NO反应更为有利,而且催化稳定性更好。

摘要:
应用密度泛函理论(DFT),在B3LYP/6-31+G(d)水平上对含有5~24个碳原子的部分液相链烷烃进行结构优化和量化计算其分子中各个原子间空间拓扑距离,并构建拓扑距离矩阵.结合分子中各原子的支化度,应用原子的平衡电负性对分子图进行着色,得到新型结构拓扑指数PX<,m>(m=1,2,3).采用多元线性回归技术建立液相链烷烃热导率γ与PX[m](m=1,2,3)的定量关系拓扑模型,并用该模型对热导率γ进行预测与估算,结果表明实验值与预测值、估算值均很吻合.同时,采用留一法(1eave-one-out)和外检验方法测试模型的内部稳定性和外部预测能力.与文献结果比较,本文所用参数少,计算简便,为液相链烷烃导热的研究提供了一种新方法.

摘要:
以甲醛为交联剂,将胱氨酸修饰到啤酒酵母菌(SC)上,并采用海藻酸钠和明胶固定化,得到一种新型的生物吸附剂——修饰啤酒酵母菌( MSC).通过红外光谱(IR)分别表征了两种吸附剂的结构,考察了其吸附铀的主要影响因素即溶液pH值、吸附时间等.结果表明:MSC细胞表面具有大量吸附铀的基团,MSC和SC吸附铀的最佳条件是:pH值为6.0,吸附时间分别为1.8、1.5h.对吸附动力学模型和吸附等温模型进行了分析,MSC和SC对铀的吸附动力学模型较好地符合了准二级动力学模型,MSC和SC的相关系数均大于0.99;吸附等温线均能符合Langmuir和Freundlich等温线模型,说明该吸附体系是一个单层覆盖与多层吸附相结合的吸附模式,且MSC的最大吸附量是SC的6.5倍.

摘要:
应用密度泛函理论(DFT),在B3LYP/6-31+G(d)基组上优化和振动分析计算了多氯联苯醚(PCDEs)所有209种可能的分子空间几何结构,得到其各原子之间空间拓扑距离,并建立拓扑空间距离矩阵.结合分子中各原子的支化度,应用原子的平衡电负性对分子图进行着色,得到量子拓扑指数PX<,1>、PX<,2>.采用多元线性回归技术建证联苯醚和209种可能结构的多氯联苯醚PCDEs 3种少见报道的热力学性质--标准生成热、标准生成自由能和相对自由能与PX<,1>、PX<,2>的定量关系拓扑模型,并用该模型分别对不同热力学性质进行预测与估算.结果表明,本方法的预测值、估算值与文献[3]的计算值均相吻合.同时,采用留一法(leave-one-out)和外检验方法测试模型的内部稳定性和外部预测能力,测试结果显示模型具有良好的稳定性和较强的预测能力.

摘要:
利用分子印迹技术制备了重金属铅(Ⅱ)的印迹聚合物,并对其识别性能进行定量描述。以铅(Ⅱ)离子为模板,水杨醛肟为功能单体,二甲基丙烯酸乙二醇酯为交联剂,偶氮二异丁腈为引发剂,采用本体聚合法制备印迹聚合物。结果表明,与未印迹的聚合物相比较,印迹聚合物对铅(Ⅱ)离子具有较好的识别选择性,达到吸附平衡的时间为40min,Pb2+初始浓度为10mg/L时,最大吸附量为949μg/g,并推测了该印迹聚合物和铅离子的识别机理。

采用高效液相色谱法检测牛奶和奶粉中的6种玉米赤霉醇及其类似物,该方法准确、可靠、灵敏度高。样品用乙腈提取,经免疫亲和柱净化后.用高效液相色谱-紫外检测器检测。结果表明,牛奶和奶粉中添加6种玉米赤霉醇及其类似物回收率均为80%-110%,变异系数均小于12%。

摘要:
为研究二苯乙烯苷对过氧化氢(H_2O_2)诱导人脐静脉内皮细胞凋亡的影响并初步探讨二苯乙烯苷抗凋亡的可能机制,采用不同浓度H_2O_2和二苯乙烯苷处理内皮细胞24 h,以MTT法检测细胞生长活力、Hoechst33258染色观察细胞形态、流式细胞仪检测凋亡率等方法筛选造模内皮细胞凋亡的H_2O_2浓度和二苯乙烯苷最佳的抗内皮细胞凋亡作用浓度.RT-PCR、Western-blot分别检测Caspase-3、PARP mRNA及蛋白质的表达.结果发现,与空白对照组相比,300μmol/L H_2O_2作用后,内皮细胞增殖明显受到抑制,Hoechst33258染色可见大量凋亡细胞,细胞凋亡率显著增加,流式细胞仪检测出明显的凋亡峰,Caspase-3和多聚腺苷酸二磷酸核糖基聚合酶(PARP)的表达量显著增加.经二苯乙烯苷处理后,随着二苯乙烯苷浓度增加,细胞的增殖率随之增加,凋亡细胞数减少,凋亡率逐渐降低,与H_2O_2组比较,10μmol/L的二苯乙烯苷能够显著提高细胞增殖率,降低细胞凋亡率,并显著减少Caspase-3和PARP表达.以上结果表明,二苯乙烯苷能够抑制由H_2O_2诱导的人脐静脉内皮细胞凋亡,增加细胞生长活性,降低细胞凋亡率,其作用机制可能与下调Caspase-3和多聚腺苷酸二磷酸核糖基聚合酶(PARP)的表达有关。

Rhodium(III)-catalyzed oxidative olefination of N-(1-naphthyl)sulfonamides has been achieved at the peri position. Three categories of olefins have been successfully applied. Activated olefins reacted to afford five-membered azacycles as a result of oxidative olefination-hydroamination. Unactivated olefins reacted to give the olefination product. 2-fold oxidative C-C and C-N coupling was achieved for allylbenzenes. © 2011 American Chemical Society.

Notwithstanding the continuous progress in instrument's playability and impressive potential of modem LC-MS based bio-analytical methodologies, sample preparation still remains as the key technical bottleneck step, mainly due to the time-consuming and error-prone process that is highly dependent on the analyte nature and the matrix type.Nowadays, there is a clear trend in the field of residue analysis toward the use of a straightforward generic method to cover a broad array of analyte/matrix combinations by the simultaneous extraction of a vast number of target compounds (multi-analyte) from various types of sample matrixes (multi-matrix) in a single procedure.

摘要:
以无机铝盐A1(NO_3)_3为原料,以乙二胺和聚乙二醇为协同模板导向剂,与氨水反应,经过“沉淀-超声-煅烧”等过程,制得介孔纳米氧化铝。用TEM, XRD,N_2吸附-脱附等测试技术对样品进行了测试和表征。研究结果表明,所得样品具有相当高的比表面积(510 m~2?G~(-1))、较大孔容(1.13 cm~3?g~(-1))和较为集中的孔径(5.8 nm)分布。根据乙二胺和氧化铝表面的溶液化学反应及在不同pH条件下的溶液组分分布,探讨了它们在介孔氧化铝制备过程中的协同模板作用机理。

摘要:
Rational design of functional enzymes is a powerful strategy to gain deep insights into more complex native enzymes, such as nitric oxide reductase (NOR). Recently, we engineered a functional model of NOR by creating a two His and one Glu (2-His-1-Glu) non-heme iron center in sperm whale myoglobin (swMb L29E, F43H, H64, called FeBMb(-His)). It was found that FeBMb(-His) adopts a low-spin state with bis-His coordination in the absence of metal ions binding to the designed metal center. However, no structural information was available for the variant in this special spin state. We herein performed molecular modeling of FeBMb(-His) and compared with the X-ray structure of its copper bound derivative, Cu(II)-CN−-FeBMb(-His), resolved recently at a high resolution (1.65 Å) (PDB entry 3MN0). The simulated structure shows that mutation of Leu to Glu at position 29 in the hydrophobic heme pocket alters the folding behavior of Mb. The hydrogen bond between Glu29 and His64 further plays a role in stabilizing the bis-His (His64/His93) coordination structure. This study offers an excellent example of using molecular modeling to gain insights in rational design of both structural and functional proteins. Proteins 2011. © 2010 Wiley-Liss, Inc.

摘要:
We report on a carbon paste electrode that was modified with a binuclear manganese(II) complex by the drop-coating method. A study on the mechanism of the electro-oxidation of tryptophan (Trp) at this electrode indicated that it enables Trp to be determined with good sensitivity and selectivity. Second-order derivative linear sweep voltammetry at pH 4. 1 revealed that a sensitive anodic peak appears at 812 mV (vs. SCE) whose current is proportional to the concentration of Trp in the concentration range from 0. 1 to 1. 0 μmol L -1 and 1. 0 to 80 μmol L -1 , with a detection limit (S/N = 3) of 0. 08 μmol L -1 (60 s of accumulation). The method was applied to the determination of Trp in amino acid injection solutions with satisfactory results. © 2011 Springer-Verlag.

摘要:
采用火焰原子吸收光谱法测定了谷粒马尾藻、鼠尾藻、海青菜、匍枝马尾藻、裙带菜、大叶藻、矶石鸡冠藻(红、白)这8种藻类植物中钙、镁、铁、锰、铜、锌、钾7种元素的含量。结果表明,这些海藻中含有较丰富的钙、镁、铁、钾元素,而铜、锰、锌元素含量相对较少。钙元素的含量最为丰富,铜的含量最少。该方法的加标回收率在95.7%—101.1%之间,RSD值在0.054%—1.764%之间,具有较好的准确度和精密度。本实验结果为这些藻类植物的食用,药用以及饲养价值提供了依据。
