Objective To establish a rapid simple,efficient method for extracting human genomic DNA from whole blood. Method Whole blood is lysed by adding a chaotropic agent NaI,then genomic DNA is directly extracteded with chloroform:isoamyl alcohol(24∶1). Result The genomic DNA extracted with this method is of large molecular weight and A 260 nm/280 nm =1.83±0.13,33.5±3.55 μg DNA can be gained from per milliliter whole blood.The results of PCR and restriction endonucleases digestion for the DNA are good.
