Upregulation of SDF-1 is associated with atherosclerosis lesions induced by LDL concentration polarization

摘要:
Previous numerical simulations on low-density lipoprotein (LDL) concentration polarization in the arterial system indicated that LDL concentration polarization might play an important role in the genesis and development of atherosclerosis. To date, no in vivo experiments have examined this question directly, and the molecular mechanisms are unknown. In this study, ten rabbits were treated with gel- silica loop to develop a defined local stenosis in the straight segment of the left carotid artery. Both numerical simulation and experiment measurements showed that the concentration of LDL was about 35% higher at the blood/arterial wall interface than in the lumen on the distal side of the stenosis. Atherosclerotic lesions with abundant lipid deposits were observed and stromal derived factor-1 (SDF-1) was detected at the distal end of the stenosis, while the straight segment was plaque-free. In vitro studies demonstrated that LDLinduced SDF-1 expression in endothelial cells and increased monocyte adhesion to endothelial cells in a dose-dependent manner. The adhesion was suppressed when endothelial cells were pretreated with SDF-1 antibody. These results suggested LDL concentration polarization contributed to the localization of atherosclerosis and to the expression of SDF- 1. In turn, SDF-1 facilitated plaque formation. © 2011 Biomedical Engineering Society.

摘要:
目的观察氧化型低密度脂蛋白对人脐静脉内皮细胞自噬的影响及其调节机制。方法氧化型低密度脂蛋白孵育人脐静脉内皮细胞24h, RT-PCR检测TET2 mRNA表达,Western blot检测TET2以及自噬标记物Beclin 1、LC3的表达。TET2 siRNA转染人脐静脉内皮细胞,Western blot检测Beclin 1、LC3的表达。结果人脐静脉内皮细胞经不同浓度氧化型低密度脂蛋白孵育24h后,浓度依赖性地下调TET2 mRNA以及蛋白的表达(P<0.05),并且下调Beclin 1表达以及LC3Ⅱ/LC3Ⅰ的比值。TET2 siRNA转染人脐静脉内皮细胞后,Beclin 1表达以及LC3Ⅱ/LC3Ⅰ的比值明显降低。结论氧化型低密度脂蛋白促进人脐静脉内皮细胞自噬,可能与下调TET2表达相关。

摘要:
Proprotein convertase subtilisin/kexin 9 (PCSK9), a member of the protein-converting enzyme family, is highly expressed in adult hepatocytes and small intestinal enterocytes. To our knowledge, in this study, we demonstrate for the first time that PCSK9 is upregulated in a dose-dependent manner via oxidized low-density lipoprotein (oxLDL) stimulation in THP-1-derived macrophages. PCSK9 small interfering RNA (siRNA) suppresses the oxLDL-induced inflammatory cytokine expression in THP-1-derived macrophages. The exposure of macrophages to oxLDL markedly increased the expression of NF-kappa B protein in the nucleus. However, this effect was significantly attenuated by PCSK9 si RNA. These findings indicate that PCSK9 expression is induced by ox LDL, and that PCSK9 si RNA protects against inflammation via the inhibition of NF-kappa B activation in oxLDL-stimulated THP-1-derived macrophages. Our results suggest that PCSK9 may be used as a therapeutic target for the treatment of atherosclerosis since PCSK9 siRNA suppresses oxLDL-induced I kappa B-alpha degradation and NF-kappa B nuclear translocation into THP-1-derived macrophages.

目的观察组织蛋白酶L在动脉粥样硬化病变中的表达以及对巨噬细胞脂质蓄积的影响。方法颈总动脉套环并饲以高脂饮食建立兔颈总动脉粥样硬化模型,免疫组织化学方法检测斑块内组织蛋白酶L的表达。氧化型低密度脂蛋白孵育RAW264.7巨噬细胞24h,逆转录聚合酶链反应、Western blot以及荧光分光光度法检测组织蛋白酶L表达和活性。组织蛋白酶L抑制剂预处理巨噬细胞,油红O染色以及高效液相色谱法观察细胞内脂质蓄积。结果正常血管中不表达组织蛋白酶L,斑块内组织蛋白酶L高表达。巨噬细胞经不同浓度氧化型低密度脂蛋白孵育24h后,组织蛋白酶L mRNA的表达量在各组间无明显差异(P>0.05),但蛋白表达量以及活性明显增加(P<0.05)。组织蛋白酶L抑制剂剂量依赖性地抑制巨噬细胞内脂质积蓄。结论组织蛋白酶L在动脉粥样硬化斑块中高表达并参与巨噬细胞脂质代谢。

目的观察组织蛋白酶L(Cathepsin L)在异常剪切切应力诱导的动脉粥样硬化斑块中的表达,探讨Cathepsin L在动脉粥样硬化病变中的作用。方法采用套环法建立兔颈总动脉局部狭窄的动物模型,喂养4周;数值模拟法模拟局部狭窄远心端和近心端流场特性以及剪切应力分布;酶法测定血浆中总胆固醇、甘油三酯以及HDL水平;HE染色观察病理学改变;油红O染色观察病变处脂质的蓄积;免疫组织化学法检测Cathepsin L的表达。结果 4周后,兔血脂水平没有明显改变。套环形成局部狭窄后,颈总动脉血流流场发生显著的扰动,狭窄远心端有涡流以及二次流形成;狭窄近心端形成局部高剪切应力区域,而在远心端形成振荡的低剪切应力区域(0~0.3 Pa)。HE染色显示颈总动脉狭窄的近心端和远心端均有明显的内膜增生以及动脉粥样硬化斑块形成,近心端比远心端病变更严重。油红O染色显示有大量的脂质沉积;免疫组织化学染色结果表明病变中有大量的Cathepsin L表达,主要分布于斑块中的巨噬细胞和平滑肌细胞,并且近心端Cathepsin L的表达量明显高于远心端量,而对照侧血管仅有微量的Cathepsin L表达。结论切应力特性调节Cathepsin L的表达,Cathepsin L可能在异常剪切应力诱导的动脉粥样硬化斑块的发生发展中起着重要的调节作用。

Six Talent stent-grafts were harvested at reoperations (N=5) and autopsy (N=1). The explants were observed nondestructively, including gross morphology, X-rays, CT scans and closed pressure system analysis. The Nitinol frames in three devices harvested at reoperations and another harvested at autopsy were intact. One had a stent fracture of the proximal bare stent, and one had a wire fracture of a thin proximal external supporting stent as well as a hole in the fabric just above the bifurcation. For the three devices structurally intact, reoperations were performed for a type 1A endoleak (one patient) and aorto-enteric fistulas (two patients). The healing characteristics were poor or absent. The fabric in the main body of the grafts harvested after aorto-enteric fistula was devoid of biological deposits. Two of the grafts harvested at reoperation demonstrated fabric holes of up to 4 mm2. The device obtained at autopsy showed an almost continuous internal capsule with variable thickness. The luminal surface was smooth, but the capsule detached easily. The devices explanted at reoperations showed various levels of impaired biofunctionality associated with adverse outcomes. The stentgraft retrieved from autopsy was intact. ©2011 by Begell House, Inc.

目的组织蛋白酶L(Cat L)具有水解纤维连接蛋白、胶原和弹性蛋白的特性,在动脉粥样硬化性斑块中高表达。研究表明,Cat L基因敲除小鼠的动脉粥样硬化斑块中脂核明显减小,但其机制尚不清楚。方法 0、2、10及50 mg/L ox-LDL孵育RAW264.7巨噬细胞24 h,RT-PCR、Western blot以及荧光活性检测试剂盒分别检测Cat L的表达和活性。采用不同浓度的CatL抑制剂预处理巨噬细胞,然后再各加入ox-LDL至终浓度为50 mg/L,孵育24 h,油红O染色观察细胞内脂质蓄积。结果不同浓度ox-LDL孵育的细胞间Cat L mRNA的表达无明显差异(P>0.05);但随着ox-LDL浓度的增加,Cat L蛋白的表达量与活性都明显增加。油红O染色结果表明,Cat L抑制剂呈剂量依赖性地降低细胞内脂质蓄积。结论 ox-LDL上调巨噬细胞Cat L的表达和活性,Cat L抑制剂抑制巨噬细胞内脂质蓄积。

In this study, we investigated the effect of low density lipoprotein (LDL) on endothelial cell adhesion in shear stress. The endothelial cells were incubated with four different concentrations of low density lipoprotein for 24h. Thereafter, the content of cholesterol ester in endothelial cells was assayed with High Performance Liquid Chromatography (HPLC). The lipid within endothelial cells was detected with oil red O. F-actin was examined with fluorescence staining of rhodamine-phalloidin. A micropipette aspiration technique was employed to investigate the viscoelasticity of endothelial cells treated by LDL. After treated with LDL, endothelial cells were subjected to flow shear stress of 15Pa for 24h. Compared to control, LDL significantly increased the ester level in endothelial cells (p<0.05). Observation by fluorescence microscopy showed that the fibers of F-actin in normal endothelial cells concentrated along the cell membrane and no dense fibers were observed in the cytoplasm. The abnormal F-actin were appeared in LDL-treated endothelial cells, while most of the peripheral fibers disappeared and dense stress fibers were observed in the cytoplasm with high dose LDL. After exposed to 1.5 Pa shear stress the conformance and the adhesion of endothelial cells which incubated with 150 mg/L LDL decreased comparing with control. It is concluded that LDL causes lipid accumulation and the cytoplasm damaged, which may relate to endothelial cell conformance and adhesion. © 2010 International Federation for Medical and Biological Engineering.

In this study, we designed a animal model to detect the effect of LDL concentration polarization on atherosclerosis and explore the molecular mechanism. A method to induce a defined local stenosis in straight segment of rabbit left carotid artery was developed. The rabbits were fed with normal or high fat/cholester food Numerical simulation and experimental measurement were used to analyze the feature of the blood flow and the LDL distribution. The arterial lesions were evaluated by using HE staining, collagen staining, Oil red O staining and High performance Liquid Chromatography. The expression of stromal derived factor-1 (SDF-1) in the lesions and cultured endothelial cells were analyzed with immunohistochemistry, RT-PCR and Western blotting, respectively. Results showed that the blood flow was disturbed and the shear stress decreased at the distal of stenosis significantly. The LDL concentration polarization took place at the distal of stenosis and the concentration of LDL was 35% higher in the arterial wall than in the lumen. The atherosclerotic lesion was formed and SDF-1 was expressed in the arterial segment distal to the stenosis, whereas no atherosclerotic lesion and expression of SDF-1 were observed in the straight part of the segment. It was in vitro cell culture experiment showed that LDL induced monocyte adherence to the endothelial cells and such effect could be inhibited by SDF-1 antibody. © 2010 International Federation for Medical and Biological Engineering.

To test the hypothesis that concentration polarization of atherogenic lipids may occur in the arterial system and probe the effect of endothelial cells (ECs). A modifier with local stenosis was developed. The luminal surface LDL concentration at the distal of stenosis was analyzed with numerical stimulation and was measured by confocal microscopy. ECs were incubated with low density lipoprotein(LDL). Then, ECs were exposed to shear stress(15dyn/cm2) and F-actin was examined by fluorescence staining. Numerical stimulation showed that the luminal surface LDL concentration was higher than the bulk concentration. The relative luminal surface LDL concentration varied with the velocity and degree of stenosis. The experimental detection result was very good agreement with the numerical analysis. After treated with LDL, F-actin reorganization was observed. Under flow, the adhesion and retention of ECs were negatively relative to LDL concentration. Therefore provided concentration polarization of LDL occurs at the distal end of stenosis, and in turn effect the adhesion of ECs under flow.

The aim of this study was to determine the molecular mechanism underlying the inhibition of vascular restenosis by Radix Sophorae Flavescentis (RSF) extract after balloon dilatation injury. In a rat carotid model of balloon dilatation injury, the RSF extract showed a significant inhibitory effect on vascular restenosis. Immunohistochemistry showed that the expression of tumor necrosis factor-α(TNF-α) in the vascular focus was markedly reduced upon treatment with the RSF extract, whereas the expression of proliferating cell nuclear antigens (PCNA) was mostly unaffected. Colorimetric assays of methylene blue incorporation demonstrated that the proliferation of cultured vascular smooth muscle cells (VSMCs) was not inhibited with serum from rats treated with RSF extract. However, the expression of TNF-α in cultured VSMCs was significantly downregulated by serum from rats treated with RSF extract. These results suggested that RSF extract has an inhibitory effect on vascular restenosis after balloon dilatation injury which might be, in part, attributable to its inhibition of TNF-α expression. © Georg Thieme Verlag KG Stuttgart.

摘要:
为研究pcsk9基因沉默后对氧化型低密度脂蛋(oxLDL)诱导THP-1源性巨噬细胞凋亡的影响,用不同浓度oxLDL处理THP-1源性巨噬细胞48 h,Hoechst33258染色检测细胞凋亡,RT-PCR、Western blot分别检测pcsk9 mRNA、NARC-1蛋白的表达.应用Lipofectamine2000转染3对pcsk9 siRNAs进THP-1源性巨噬细胞中,筛选出最有效的siRNA再转染入THP-1源性巨噬细胞,24 h后加入oxLDL处理48 h,Hoechst33258染色观察细胞评价细胞凋亡,流式细胞术计数检测细胞凋亡率.结果发现,75 mg/L oxLDL处理THP-1源性巨噬细胞48 h后,Hoechst33258染色可见大量凋亡细胞.同时RT-PCR、Western blot检测发现,pcsk9 mRNA和NARC-1蛋白质表达量均随oxLDL浓度的增加而增加,75 mg/L oxLDL组增加最明显.不同浓度siRNA转染THP-1源性巨噬细胞后,RT-PCR筛选出3对siRNAs的终浓度为80 nmol/L均可出现明显的沉默效应.选取此浓度在蛋白质水平检测基因抑制情况,筛选出最有效的一对siRNA.将筛选出来的siRNA转染细胞24 h后,再用oxLDL处理48 h,Hoechst33258染色及流式细胞计数结果显示,转染siRNA组凋亡明显被抑制.结果表明,在本研究的浓度范围内,随着oxLDL浓度增加pcsk9的表达随之增加,同时,THP-1源性巨噬细胞凋亡也明显增加,75 mg/L oxLDL最明显,pcsk9 mRNA和蛋白质的表达也在该浓度最高.提示pcsk9 siRNA能有效抑制pcsk9基因的表达,从而有效抑制由oxLDL诱导的THP-1源性巨噬细胞的凋亡.

摘要:
目的 观察基质细胞衍生因子-1α(SDF-1α)对体外培养的大鼠骨髓源性内皮祖细胞(EPCs)动员和增殖的影响.方法 微孔法获取大鼠骨髓EPCs并采用荧光显微镜和流式细胞术鉴定EPCs特异性标记物.不同浓度SDF-1α处理EPCs后,采用细胞培养、MTT检测EPCs的克隆形成、细胞增殖.结果 通过MTT法检测,埘照组与(1、10、100 μg/L)SDF-1α处理组的OD值分别为0.311±0.054、0.587±0.072、0.813±0.056、1.029±0.078,通过统计学方法 分析,对照组与SDF-1α处理组比较差异均有统计学意义(n=5,P<0.05);100 μg/L SDF-1α处理组次级内皮祖细胞集落单位数目是对照组近3倍(4.67±1.577比14.33±3.055,n=5,P<0.01).结论 SDF-1α剂量依赖性地促进EPCs增殖,并显著增强EPCs克隆肜成能力.

摘要:
目的:进一步探讨雷帕霉素抗血管内再狭窄的分子机制.方法:球囊拉伤大鼠颈总动脉建立血管内膜增生的动物模型,口服雷帕霉素(25 mg/kg),4周后处死动物.HE染色观测血管病变,免疫组化检测增生内膜中基质细胞衍生因子-1(SDF-1)的表达,小室迁移系统观察SDF-1对平滑肌细胞迁移的影响.结果:球囊拉伤明显诱导血管内膜增生,增生内膜中有明显的SDF-1的表达,雷帕霉素能显著下调增生内膜中SDF-1的表达,而SDF-1浓度依赖性地诱导平滑肌细胞的迁移.结论:雷帕霉素可能通过下调SDF-1的表达从而抑制血管再狭窄.

为研究低密度脂蛋白(LDL)、氧化修饰的低密度脂蛋白(oxLDL)对THP-1源性巨噬细胞中PCSK9、 LDLR表达的影响及两者之间的关系.分别用0mg/L、10mg/L、20mg/L、30mg/L的LDL和0mg/L、10mg/L、20mg/L、30mg/L的oxLDL处理THP-1巨噬细胞,油红O染色检测细胞荷脂情况,免疫荧光检测THP-1巨噬细胞上PCSK9蛋白表达及分布情况,RT-PCR、 Western blot检测THP-1巨噬细胞上PCSK9、 LDLR mRNA、蛋白质的表达.结果发现,不同浓度LDL处理THP-1巨噬细胞后,随着LDL浓度的增大,细胞内脂滴数目略有增多.免疫荧光染色发现,PCSK9在THP-1巨噬细胞上的表达随LDL浓度的增加而增多,胞浆内定位于某一特定细胞器中;RT-PCR、 Western blot检测发现,LDL可以呈浓度依赖性下调THP-1巨噬细胞中LDLR的表达和上调PCSK9的表达.不同浓度oxLDL处理THP-1巨噬细胞后,随oxLDL浓度的增大,脂滴颗粒明显增加;oxLDL处理对THP-1巨噬细胞上PCSK9、 LDLR mRNA、蛋白质的表达影响均不明显.研究结果表明:THP-1巨噬细胞上,同时有PCSK9和LDLR的表达,且PCSK9定位于胞浆中某一特定细胞器;oxLDL对THP-1巨噬细胞LDLR和PCSK9表达没有影响;LDL能够降低THP-1巨噬细胞表面LDLR的表达,同时上调PCSK9表达,初步说明在THP-1巨噬细胞中,两者有一定的相关性.
