摘要:
目的分析北京、吉林、新疆、陕西、湖南、河南6省份结核分枝杆菌耐药性数据,了解耐药性结核病分布特征及影响因素。方法在6省份采用问卷调查收集背景资料,并使用比例法药物敏感性试验对4种一线抗结核药物(异烟肼、利福平、链霉素、乙胺丁醇)进行检测,再利用 SPSS 20.0软件分析其危险因素。结果总体耐药率和耐多药率分别为23.42%和13.51%。北京、吉林、湖南、河南、陕西、新疆分离株耐药率分别为21.50%、12.24%、36.27%、42.86%、27.78%、 24.39%,耐多药率分别为4.67%、8.16%、24.51%、26.53%、15.28%、14.15%。χ~2检验结果显示,各省份间单种药物耐药率、总体耐药率和耐多药率差异均有统计学意义(P=0.000)。单因素分析结果显示,结核病的复发、治疗史与耐药结核病的产生具有明显的相关性,差异有统计学意义(P<0.05)。结论我国耐药结核病的形势非常严峻,调查地区结核病耐药性存在明显差异,应针对各地区主要的危险因素加强预防干预。

摘要:
Syphilis is a multistage disease caused by the invasive spirochete Treponema pallidum subsp. pallidum, and accurate diagnosis is important for the prevention and treatment of syphilis. Here, to identify appropriate diagnostic antigens for serodiagnosis of syphilis, 6 recombinant proteins were expressed in Escherichia coli and purified, including flagellins (FlaB1 [Tp08681, FlaB2 [Tp0792], and FlaB3 [Tp0870]), Tp0463, Tp0751, and Tp1038. The sensitivities were determined by screening sera from individuals with primary (n = 82), secondary (n = 115), latent (n = 105), and congenital (n = 65) syphilis. The specificities were determined by screening sera from uninfected controls (n = 30) and potentially cross-reactive infections including Lyme disease (n = 30), leptospirosis (n = 5), and hepatitis B (n = 30). Our data showed that FlaB1, FlaB2, FlaB3, Tp0463, and Tp1038 exhibited higher overall sensitivities and specificities for detecting IgG antibody, with 95.4% and 98.9%, 92.6% and 95.8%, 95.1% and 95.8%, 92.6% and 97.9%, and 95.9% and 98.9%, respectively. In contrast, Tp0751 demonstrated only an overall sensitivity of 39.2%. For comparison, the sensitivity and specificity of Architect Syphilis TP were determined to be 98.1% and 93.7%, respectively. In addition, FlaB1, FlaB2, FlaB3, and Tp0463 demonstrated excellent performance for detecting IgM antibody in primary and congenital syphilis, with sensitivities of 76.8% and 83.1%, 72.0% and 87.7%, 74.4% and 89.2%, and 64.6% and 75.3%, respectively. These results indicate that FlaB1, FlaB2, FlaB3, and Tp0463 could be as novel diagnostic candidates for serodiagnosis of syphilis. (C) 2016 Elsevier Inc. All rights reserved.

摘要:
目的:探讨传代选择对鼠衣原体质粒缺失株CMUT3菌株培养特性的影响及其相应机制。方法采用“无辅助感染”和“辅助感染”交替方式体外传代培养CMUT3菌株,而后观察传代菌株和亲代感染细胞时的吸附效率以及对离心因素的依赖性,并利用下一代测序技术对传代菌株CMUT3G40和亲代CMUT3G0进行全基因组测序分析。结果传代菌株CMUT3G40在感染HeLa细胞时对离心因素依赖性降低,吸附试验表明CMUT3G40菌株对细胞的吸附能力增强;比较基因组学分析结果表明 CMUT3G40菌株与亲代 CMUT3G0在 tc0237基因存在差异, TC0237Q117E错义突变出现于CMUT3G40菌株基因组( CMUT3G40,99.7%;CMUT3G0,0%)。结论通过体外传代培养CMUT3菌株可获得吸附能力增强的菌株,该表型与传代选择所致的TC0237Q117E密切相关。

<正>微生物耐药性问题日益严重,很多病原微生物,例如结核分枝杆菌和恶性疟原虫等对人类生命健康造成了极大的威胁,开发新的抗菌药物迫在眉睫。微生物作为抗生素的重要来源,在发掘抗耐药菌新型抗生素的研究中承担了重要角色,许多微生物来源的天然化合物展现了显著的抗耐药菌活性。而大量病原微生物基因组的测序完成,又为寻找针对病原微生物的分子药物靶标提供了有利条件。

摘要:
目的分析我国结核分枝杆菌复合群(MTBC)临床分离株中GlnA1、Mpt70、LppX、 GroES和LpqH5种抗原编码基因多态性及其人T细胞表位的多态性。方法选取13个省份临床分离的173株MTBC,采用PCR扩增5种抗原基因,并运用BioEdit软件进行序列比对,分析其人T细胞表位与非表位的变异情况。利用Mega 6.0软件分别计算同义突变率(dS)和非同义突变率(dN)及其比值。结果173株菌的基因glnA1非表位区出现2个非同义突变位点;基因mpl70表位区出现1个非同义突变位点;基因lpqH表位区表现为1个非同义突变位点和1个同义突变位点; groES在整个基因中未发现任何突变;基因lppX表位区表现为5个非同义突变和1个同义突变位点,其中152位的氨基酸相同位点有9株菌发生了同义突变,表现较高的多态性。同时基因lppX的15个T细胞抗原表位中有7个表位发生了改变,其dN/dS值为0.19。结论结核分枝杆菌抗原Mpt70、LppX和LpqH的人T细胞表位区具有多态性,反映了此抗原可能参与逃避宿主免疫的分化选择。GlnAl的非表位区的多态性,对该菌株的免疫反应影响较小。GroES序列相对保守,不具有明显的多态性,可能对结核分枝杆菌的鉴定、诊断及新型疫苗的研制具有重要作用。

摘要:
Chlamydia psittaci (C. psittaci), an obligate intracellular agent of psittacosis, causes an atypical pneumonia in humans. The transmembrane head proteins (TMH) of C. psittaci, putatively belong to the Inc family and presumably play similar roles. CPSIT_0844 and CPSIT_0846 were the putative TMH proteins of C psittaci. To identify these two proteins, antisera were raised with fusion proteins which were prokaryotic expressed in Escherichia coli and purified. By immunofluorescence assay, CPSIT_0844 and CPSIT_0846 were localized in the inclusion membrane of C psittaci-infected cells. By RT-PCR and western blot analysis to detect the temporal expression, CPSIT_0844 and CPSIT_0846 were detected as early as 12 h post-infection (p.i.) and 6 h p.i., separately; meanwhile, in secretions monitored with immunofluorescence assay, these proteins were observed in the inclusion membrane at 18 h p.i. and remained in the inclusion membrane throughout the growth cycle. CPSIT_0844 and CPSIT_0846 could specifically be recognized by the antiserum of C psittaci but failed to react with the antiserums of Chlamydia trachomatis and Chlamydia pneumoniae, which is consistent with the fact that they had no significant orthologs in C trachomatis and C pneumoniae. These results revealed that CPSIT_0844 and CPSIT_0846, the putative TMH family proteins, might be unique to C psittaci and could be used to diagnose the infection caused by C psittaci. Moreover, CPSIT_0844 and CPSIT_0846 could induce the expression of the inflammatory cytokines IL-1 beta, IL-6 and TNF-alpha in THP-1 cells, which might contribute to chlamydia-induced inflammatory pathologies. Pulmonary diseases caused by nontuberculous mycobacteria (NTM) are increasing in incidence and prevalence worldwide. In this study, we identified NTM species of the clinical isolates from 8 provinces in China, in order to preliminarily provide some basic scientific data in the different species and distribution of NTM related to pulmonary disease in China. A total of 523 clinical isolates from patients with tuberculosis (TB) diagnosed clinically from 2005 to 2012 were identified to the species using conventional and molecular methods, including multilocus PCR, rpoB and hsp65 PCR-PRA, hsp65, rpoB, and 16S-23S internal transcribed spacer region sequencing. The isolates were identified into 3 bacterium genera, including NTM, Gordonia bronchialis, and Nocardia farcinica, and, for the 488 NTM isolates, 27 species were identified. For all the 27 species of NTM which were found to cause pulmonary infections in humans, the most prevalent species was M. intracellulare, followed by M. avium and M. abscessus. And seven other species were for the first time identified in patients with TB in China. NTM species identification is very important for distinguishing between tuberculosis and NTM pulmonary diseases, and the species diversity drives the creation of diverse and integrated identification methods with higher accuracy and efficacy. © 2016 Haican Liu et al.

目的 研究我国结核分枝杆菌复合群(MTBC)抗原mpt83、mpt70、lpqH、pstS3、groES及其人B细胞表位的多态性.方法 从中国疾病预防控制中心传染病所结核室菌株库随机选取我国的179株MTBC临床分离株,培养后提取DNA,基因扩增并测序,根据H37Rv标准序列和免疫表位数据库(IEDB)的抗原表位信息,比较、分析各基因B细胞表位序列的变异情况,最后用MEGA6软件计算各基因的非同义突变率(dN)、同义突变率(dS)及其比值.结果 在179株MTBC菌株中,mpt83基因有7个单核苷酸多态性(SNP)位点,该基因及其表位区、非表位区的dN/dS分别为0.88、0.34和0.14;mpt70基因有1个SNP位点,为非同义突变;lpqH基因及其B细胞表位区dN/dS均为0.38,非表位区dN、dS均为0;pstS3基因及B细胞非表位区的dN/dS分别为1.74和1.40;groES基因未发现突变位点.结论 结核分枝杆菌mpt83、lpqH、pstS3的基因序列及B细胞表位具有较高的多态性,而mpt70和groE基因B细胞表位较保守.

Epstein–Barr virus (EBV) is a human oncogenic herpesvirus associated with lymphoma and nasopharyngeal carcinoma. Because the susceptible hosts of EB virus are limited to human and cotton-top tamarins (Saguinus oedipus), there have been no appropriate animal models until the lymphoma model induced by EBV in human peripheral blood lymphocyte (hu-PBL)/SCID chimeric mice was reported. However, it is still controversial whether the EBV-associated lymphoma induced in hu-PBL/SCID mice is a monoclonal tumor. In this study, we transplanted normal human peripheral blood lymphocytes (hu-PBL) from six donors infected with EBV into SCID mice to construct hu-PBL/SCID chimeric mice. The induced tumors were found in the mediastinum or abdominal cavity of SCID mice. Microscopic observation exhibited tumor cells that were large and had a plasmablastic, centroblastic or immunoblastic-like appearance. Immunophenotyping assays showed the induced tumors were LCA-positive, CD20/CD79a-positive (markers of B cells), and CD3/CD45RO-negative (markers of T cells). A human-specific Alu sequence could be amplified by Alu-PCR. This confirmed that induced tumors were B-cell lymphomas originating from the transplanted human lymphocytes rather than mouse cells. EBER in situ hybridization detected positive signals in the nuclei of the tumor cells. Expression of EBV-encoded LMP1, EBNA-1, and EBNA-2 in the tumors was significantly positive. PCR-based capillary electrophoresis analysis of IgH gene rearrangement revealed a monoclonal peak and single amplification product in all six cases of induced tumors. This indicated that EBV can induce monoclonal proliferation of human B lymphocytes and promotes the development of lymphoma. J. Med. Mycoplasma are the smallest prokaryotic microbes present in nature. These wall less, malleable organisms can pass through cell filters, and grow and propagate under cell free conditions in vitro. Of the pathogenic Mycoplasma Mycoplasma pneumoniae has been examined the most. In addition to primary atypical pneumonia and community acquired pneumonia with predominantly respiratory symptoms, M. pneumoniae can also induce autoimmune hemolytic anemia and other diseases in the blood, cardiovascular system, gastrointestinal tract and skin, and can induce pericarditis, myocarditis, nephritis and meningitis. The pathogenesis of M. pneumoniae infection is complex and remains to be fully elucidated. The present review aimed to summarize several direct damage mechanisms, including adhesion damage, destruction of membrane fusion, nutrition depletion, invasive damage, toxic damage, inflammatory damage and immune damage. Further investigations are required for determining the detailed pathogenesis of M. pneumoniae.

Background: The lack of Treponema pallidum-specific antigens with highly accurate diagnosis makes the diagnosis of syphilis challenging. Methods: A soluble recombinant version of a new diagnostic protein Tp0663 has been produced. The serodiagnostic potential of this protein was assessed by screening 3326 serum samples simultaneously evaluated by rapid plasma reagin and T. pallidum particle agglutination tests. Kappa (κ) coefficients were used to compare the concordance between clinical diagnosis and the Tp0663-based ELISA or the ARCHITECT Syphilis TP chemiluminescent immunoassay (Abbott GmbH and Co. KG). Results: Using the results of clinical diagnosis as the gold standard, the sensitivity and specificity of Tp0663 were found to be 98.83% (95% confidence interval (CI) 96.61-99.60%) and 100% (95% CI 99.88-100%), respectively. In comparison, the ARCHITECT Syphilis TP assay was found to have a lower sensitivity (97.27%, 95% CI 94.46-98.67%) and specificity (99.61%, 95% CI 99.32-99.78%). In particular, the ARCHITECT Syphilis TP exhibited a false-positive rate of 0.39%. Moreover, the ELISA was in perfect agreement with the gold standard, with a κ value of 0.99, comparable to that of ARCHITECT Syphilis TP (0.96). Conclusion: These results identified Tp0663 as a novel serodiagnostic candidate with great potential for developing novel tests for the diagnosis of syphilis. © 2015 The Authors.

Chlamydia psittaci is a zoonotic pathogen with a broad host range that can lead to severe respiratory and systemic disease in humans. Currently, an effective commercial vaccine against C. psittaci infection is not available. The chlamydial plasmid is an important virulence factor and encodes plasmid proteins that play important roles in chlamydial infection and the corresponding immune response. In this study, we assessed the efficacy of vaccination with plasmid proteins at preventing C. psittaci lung infection in a murine model. BALB/c mice were immunized intraperitoneally, three times at 2-week intervals, with purified recombinant CPSIT_p8 protein and then infected with C. psittaci. Immunization significantly decreased chlamydial load in the lungs of infected mice, resulted in a lower level of IFN-γ, and reduced the extent of inflammation. In vivo or in vitro neutralization of C. psittaci with sera collected from immunized mice did not reduce the amount of viable C. psittaci in the lungs of mice, indicating that CPSIT_p8-specific antibodies do not have neutralizing capacity. Furthermore, confocal fluorescence microscopy using a mouse anti-CPSIT_p8 antibody revealed that CPSIT_p8 was localized inside the inclusion of C. psittaci 6BC-infected cells. Our results demonstrate that CPSIT_p8 protein induces significant protective immunity against challenge with C. psittaci in mice and represents a promising new vaccine candidate for the prevention of C. psittaci infection. © 2016, Springer-Verlag Berlin Heidelberg.

Background Over the past decade, the incidence of syphilis and widespread macrolide resistance in its etiological agent, Treponema pallidum subsp. pallidum, have become a major health concern across countries, including China. Regional trends in subtypes and antibiotic resistance can be monitored effectively by molecular surveillance programs. In this study, whole blood samples were used to assess circulating T. pallidum strains collected from various regions of Hunan, China, between 2013 and 2015. Methods Traditional polymerase chain reaction, targeting polA, tpp47, bmp, and tp0319 genes, was used as preliminary screening assay. About 455 polymerase chain reaction-positive specimens were obtained from 2253 whole blood samples of patients with secondary or latent syphilis. Molecular subtyping was performed using a Centers for Disease Control and Prevention-based typing method combined with an analysis of the variable region of tp0548 gene. Resistance to macrolides was analyzed by examining point mutations in 23S rRNA, and the presence of the G1058C point mutation within 16S rRNA associated with decreased susceptibility to doxycycline was assessed. Results Circulating T. pallidum strains were resolved into 32 subtypes, among which subtype 14d/f was predominant. A2059G mutation in 23S rRNA, and the G1058C mutation in 16S rRNA was absent, but the prevalence of A2058G mutation in 23S rRNA was 97.5%. Conclusions We found that it is possible to use whole blood to evaluate molecular subtypes and monitor antibiotic resistance in circulating T. pallidum strains, especially when chancres are absent. High frequency of macrolide-resistant T. pallidum indicates that macrolide antibiotics, such as azithromycin, should be avoided as a treatment option for syphilis in Hunan, China. Death domain associated protein (Daxx), a multi-functional protein, plays an important role in transcriptional regulation, cell apoptosis, carcinogenesis, anti-virus infection and so on. However, its regulatory mechanisms for both cell survival and apoptosis remain largely obscure. Our review of recent studies shows that Daxx has many interesting functional dualities and can provide a reference for further research on Daxx.

Chlamydophila psittaci (C. psittaci) is a human zoonotic pathogen, which could result in severe respiratory disease. In the present study, we investigated the role and mechanism of the type III secretion system (T3SS) of C. psittaci in regulating the inflammatory response in host cells. C. psittaci-infected THP-1 cells were incubated with the specific T3SS inhibitor INP0007, inhibitors of ERK, p38, or JNK, and the levels of inflammatory cytokines were analyzed using Q-PCR and ELISA. The levels of ERK, p38, and JNK phosphorylation were analyzed by Western blot. Our results verified that INP0007 inhibited chlamydial growth in vitro, but the coaddition of exogenous iron completely reversed the growth deficit. INP0007 inhibited the growth of C. psittaci and decreased the levels of IL-8, IL-6, TNF-, and IL-1β. Exogenous iron restored the chlamydial growth but not the production of inflammatory cytokines. These results demonstrated that the expression of inflammatory cytokines during infection was associated with the T3SS which reduced by incubation with ERK and JNK inhibitors, but not with p38 inhibitor. We concluded that the T3SS elicited inflammatory responses by activating the JNK or ERK signaling pathways in the infection of C. psittaci. © 2015 Qing-zhi He et al.

Chlamydophila psittaci is an important intracellular pathogen. Persistent infection is an important state of the host-parasite interaction in this chlamydial infection, which plays a significant role in spreading the organism within animal populations and in causing chronic chlamydiosis and serious sequelae. In this study, a C. psittaci persistent infection cell model was induced by penicillin G, and real-time quantitative PCR was used to study the transcriptional levels of 10 C. psittaci genes (dnaA, dnaK, ftsW, ftsY, grpE, rpsD, incC, omcB, CPSIT_0846, and CPSIT_0042) in acute and penicillin-G-induced persistent infection cultures. Compared with the acute cultures, the penicillin-G-treated cultures showed a reduced chlamydial inclusion size and a significantly decreased number of elementary body particles. Additionally, some enlarged aberrant reticulate body particles were present in the penicillin- G-treated cultures but not the acute ones. The expression levels of genes encoding products for cell division (FtsW, FtsY) and outer membrane protein E encoding gene (CPSIT_0042) were downregulated (p < 0.05) from 6 h post-infection onward in the persistent infection cultures. Also from 6 h post-infection, the expression levels of DnaA, DnaK, IncC, RpsD, GrpE, and CPSIT_0846 were upregulated (p < 0.05); however, the expression level of OmcB in the persistent infection was almost the same as that in the acute infection (p > 0.05). These results provide new insight regarding molecular activities that accompany persistence of C. psittaci, which may play important roles in the pathogenesis of C. psittaci infection. © 2015, by The Korean Society for Microbiology and Biotechnology.

【目的】探讨鼠衣原体(Chlamydia muridarum,Cm)标准株与减毒株全基因组序列中存在的差异,筛选毒力基因并建立不同基因型的单克隆菌株,为后续致病机制的研究奠定基础。【方法】将Cm标准株G0和减毒株G28以高通量测序法进行全基因组测序,通过全基因组序列比对分析并筛选潜在的毒力相关基因;经空斑形成实验(Plaque assay)在混合菌G0和G28中大量挑取空斑,以毒力靶基因的PCR测序鉴定从G0和G28中筛选含有不同毒力基因型的单克隆菌株。【结果】全基因测序结果显示Cm G0与G28的TC0412、TC0237和TC0668基因明显不同。通过挑取空斑初步筛选了111个空斑样品,通过3个毒力基因的PCR测序鉴定最终获得了G0和G28来源的56个单克隆菌株,并根据TC0412蛋白型分成B3、D1和E1三组,每组中含有TC0237和TC0668基因差异性菌株。【结论】Cm的致病能力可能与TC0412、TC0237和TC0668基因有关,筛选获得的单克隆菌株通过分组匹配后可用于后续的毒力基因功能研究。

目的预测和鉴定梅毒螺旋体(Treponema pallidum, Tp)Tp92蛋白的Th细胞表位,为深入探讨这些表位在梅毒表位疫苗中的作用奠定基础。方法采用RANKPEP和SYFPEITHI软件分析工具联合预测Tp92蛋白的Th细胞表位,人工合成5条表位多肽,以5条表位多肽(同时设ConA阳性对照和RPMI-1640阴性对照)分别刺激梅毒患者和正常人外周血单个核细胞(PBMC),用CCK-8法鉴定预测的Th细胞表位,进一步以ELISA检测INF-γ和IL-4的分泌水平,区分Th细胞表位的类型。结果软件预测显示,Tp92蛋白的P1(103-118AA)、P2(694-712AA)、P3 (668-680AA)、P4 (300-313AA)、P5 (396-410AA)氨基酸序列最可能为其Th细胞表位。CCK-8法检测显示,P2、P3、P5能诱导梅毒患者淋巴细胞明显增殖,而正常人淋巴细胞不反应;ELISA检测细胞因子产生情况发现:P2、P3、P5可刺激梅毒患者淋巴细胞产生较高水平IFN-γ,而不产生IL-4。结论P2、P3、P5为Tp92蛋白潜在的HLA-DRBl限制性特异性Th1型细胞表位。
