摘要:
A highly sensitive, low-cost colorimetric aptasensor was developed for the determination of 8-hydroxy-2′-deoxyguanosine (8-OHdG) in human urine. The method is based on a conformational switching of the 8-OHdG aptamer to form a G-quadruplex structure in the presence of 8-OHdG. The resulting G-quadruplex assembles into a peroxidase-like DNAzyme with hemin, which effectively catalyzes the oxidation of 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS2-) by H2O2 to ABTS +, resulting in an increase in the absorption signal at 416 nm along with a color change of the solution. The response signals linearly correlated with the concentration of 8-OHdG, ranging from 466 pM to 247 nM with a detection limit of 141 pM. The relative standard deviation and the recovery were 1.97-3.47% (n = 11) and 98.8-100.2%, respectively. The proposed method avoids the label and derivatization steps in common methods and allows direct analysis of the samples by the naked eye without costly instruments, which is reliable, inexpensive, and sensitive. © 2014 Elsevier Inc. Magnetic carboxymethylcellulose (CMC/Fe3O4) was used as a framework adsorption material to remove uranium ions from aqueous solutions. Carboxyl functional groups were grafted onto the CMC/Fe3O4. The maximum adsorption capacity of the magnetic composite toward U(VI) was 122.48 mg/g. Kinetic and thermodynamic parameters of the adsorption process were estimated. The pseudo-second-order model was more suitable and it proved to be an endothermic and spontaneous process. The Langmuir and Freundlich models were applied to evaluate the adsorption isotherm. The data matched well with Langmuir model after equilibrium was reached and with Freundlich model before equilibrium was reached.

A novel method for the determination of trace formaldehyde in food samples has been established using a catalytic kinetic fluorimetry technique. It is based on the fact that the oxidation of pyronine Y by sodium periodate is catalyzed by formaldehyde in phosphoric acid. When the excitation wavelength was set at 546 nm, it was observed that decreased intensity (ΔF) of fluorescence at 563 nm was proportional to the concentration of formaldehyde in the range 0 to 0.30 μg mL-1. The limit of detection and the average recovery for formaldehyde were 2.0 × 10-2 ng mL -1 and 101.3% (n = 6), respectively. The present method has been applied to the determination of trace formaldehyde in aqueous products and the results observed were in good agreement with those obtained by distillation-spectrophotometry. A review of recently published catalytic or inhibiting kinetic methods for the determination of HCHO is presented for comparison. 摘要:
A novel method for the determination of metallothioneins (MTs) in urine was developed by fluorescence quenching strategy. The response signals linearly correlated with the concentration of MTs in the ranges of 3.12 ×10 <sup>-8</sup>-1.23 ×10<sup>-6</sup> mol L<sup>-1</sup>, and the limit of detection (LOD) was 9.36 ×10<sup>-9</sup> mol L<sup>-1</sup>. The proposed method avoids the label and derivatization steps in common methods, and is reliable, inexpensive and sensitive. Furthermore, the interaction of MTs and 8-hydroxyquinoline-5-sulphonic acid (HQS)-Cd(II) chelate was investigated, and a static quenching mode was proposed to be primarily responsible for the fluorescence quenching event. It could provide a promising potential for the detection of the biomacromolecules which have no native fluorescence, and be benefit to extend the application of fluorescence strategy. ©2013 Elsevier B.V. Urinary trichloroacetic acid (TCAA) and baseline blood trihalomethanes (THMs) have been measured as biomarkers of exposure to drinking water disinfection by-products (DBPs) that have been associated with increased risk of cancers and adverse reproductive outcomes. This study aimed to identify predictors of urinary TCAA and baseline blood THMs among men in China. Urine samples, blood samples, and information on socio-demographic factors and water-use activities were collected from 2216 men who participated in a cross-sectional study of exposure to drinking water DBPs and reproductive health during 2011 to 2012. Urinary TCAA and baseline blood THMs including chloroform (TCM), bromodichloromethane (BDCM), dibromochloromethane (DBCM), and bromoform (TBM) were analyzed. Multivariable linear regression was used to evaluate predictors of urinary TCAA and baseline blood THM concentrations. Tap water consumption was significantly associated with creatinine-adjusted urinary TCAA concentration (beta = 0.23 mu g/g creatinine per log10 unit; 95% CI: 0.12, 035). Men with surface water source had 0.13 (95% CI: 0.00, 027) higher mean creatinine-adjusted urinary TCAA concentrations than those with ground water source. Smoking was associated with lower concentration of creatinine-adjusted urinary TCAA. Age was significantly associated with baseline blood Br-THM (sum of BDCM, DBCM, and TBM) concentration (beta = 0.01 ng/L per unit; 95% CI: 0.00, 0.02). Increased household income was associated with decreased concentrations of baseline blood BDCM and Br-THMs. Our results suggest that tap water consumption, water source, smoking, age, and household income as the primary determinants of exposure to drinking water DBPs should be considered in exposure assessment. 摘要:
In order to develop an effective and economical method for removing U(VI) from the low concentration radioactive wastewater with the U(VI) concentration of less than 1 mg L<sup>-1</sup>, the biomass of Aspergillus niger was prepared and modified with ethylenediamine, and the biosorption of uranium from the low concentration radioactive wastewater by the unmodified and the modified biomasses was investigated in a batch system. The modified biomass exhibited the adsorption efficiency of 99.25 % for uranium under the optimum conditions that pH was 5.0, the contact time was 150 min, and the biosorbent dose was 0.2 g L<sup>-1</sup>. The adsorption fitted well to Langmuir isotherm, and the maximum sorption capacity of the modified biomass for U(VI) was determined to be 6.789 mg g<sup>-1</sup> which increased by 36.45 % compared with the unmodified biomass. The adsorption kinetics was better depicted by pseudo-second-order kinetic model. The Gibbs free energy change (ΔG <sup>0</sup>), enthalpy change (ΔH <sup>0</sup>), and entropy change (ΔS <sup>0</sup>) showed that the process of U(VI) adsorption was spontaneous, endothermic, and feasible. The changes in the groups, morphology, and the presence of U(VI) on the surface of the adsorbents which were characterized by FT-IR, SEM, and EDS, demonstrated that the U(VI) was successfully adsorbed onto the modified biomass. Moreover, the UO<inf>2</inf> <sup>2+</sup> absorbed on the modified biomass can be released by 0.1 mol L<sup>-1</sup> HNO<inf>3</inf> with high desorption efficiency of 99.21 %. The results show that the modified biomass can remove U(VI) from low concentration radioactive wastewater more effectively than the unmodified biomass.

摘要:
In this study, the size distribution of atmospheric aerosol in Beijing was monitored by the scanning mobility particle sizer spectrometer and the optical particle sizer. The size of particles in atmospheric aerosol was primarily distributed in the range of less than 1 μm. It showed different changes with the mass concentrations of particulate matters with an aerodynamic diameter of ⩽2.5 μm (PM2.5) for different sizes of fine particles. The amount of ultrafine particles (less than about 60 nm) decreased while the larger ones (>60 nm) increased along with the mass concentration of PM2.5 in atmospheric aerosol. This was because of the formation of the secondary atmospheric aerosol. The polylactic acid (PLA) nanofibers were prepared for filtering the aerosol by electrospinning. PLA nanofiber mats were used as the middle layer to design the composite filter membranes. Atmospheric aerosol was used as dust source in the filtration test. The results showed that the filtration efficiency of the composite filter media increased along with the thickness of nanofiber mats, which was controlled by the collection time during electrospinning. Filtration efficiency can be improved obviously by compositing with a thin layer of nanofibers.

摘要:
A direct fluorescence spectra method was applied for the determination of metallothioneins at nanomolar levels. In Britton-Robison (B-R) buffer (pH 7.0), the interaction of bis(1,10-phenanthroline)copper(II) complex cation [Cu(phen)2]2+ and metallothioneins enhanced the fluorescence intensity of system. The fluorescence enhancement at 365 nm was proportional to the concentration of metallothioneins. The mechanism was studied and discussed in terms of the fluorescence and UV-absorption spectra. Under the optimal experimental conditions, at 365 nm, there was a linear relationship between the fluorescence intensity and the concentration of the metallothioneins in the range of 8.30 × 10-9 - 7.70 × 10-7 mol L-1. The linear regression equation was ΔF = 8.96 + 38.01c (mol L-1), with a correlation coefficient of r = 0.998 and detection limit 2.50 × 10-9 mol L-1. The relative standard deviation was 0.47% (n = 11), and the average recovery 97.2%. The proposed method was successfully reliable, selective and sensitive in determining trace metallothioneins in fish visceral organ samples with the results in good agreement with those obtained by HPLC. © The Japan Society for Analytical Chemistry.

Based on ciprofloxacin (CIP)-Cu(II) system, the novel methods for the detection of metallothioneins (MTs) have been developed by fluorescence (FL) and resonance light scattering (RLS) strategies. The FL strategy avoids the label and derivatization steps in common methods, while the RLS strategy can be applied for determining bio-macromolecules and small molecules without native fluorescence. The response signals linearly correlated with the concentration of MTs over the ranges of 1.03×10<sup>-8</sup>-1.23×10<sup>-6</sup> mol L<sup>-1</sup> for FL, and of 2.56×10<sup>-7</sup>-1.54×10 <sup>-6</sup> mol L<sup>-1</sup> for RLS. The limits of detection (LOD) are 3.1×10<sup>-9</sup> mol L<sup>-1</sup> for FL and 7.68×10 <sup>-8</sup> mol L<sup>-1</sup> for RLS. This study represents the comparison of these two methods using the same CIP-Cu<sup>2+</sup>-MTs system. They not only allow practical application for MTs detection but also serve as a potential choice for the operators according to their concrete needs. In addition, the mechanisms for FL and RLS enhancement of the system were also discussed. ©2013 Elsevier B.V.

In this study, to enhance the capability of metal ions disturbing the orientation of liquid crystals (LCs), we designed a new label-free LC biosensor for the highly selective and sensitive detection of heavy metal ions. This strategy makes use of the target-induced DNA conformational change to enhance the disruption of target molecules for the orientation of LC leading to an amplified optical signal. The Hg2+ ion, which possesses a unique property to bind specifically to two DNA thymine (T) bases, is used as a model heavy metal ion. In the presence of Hg2+, the specific oligonucleotide probes form a conformational reorganization of the oligonucleotide probes from hairpin structure to duplex-like complexes. The duplex-like complexes are then bound on the triethoxysilylbutyraldehyde/N,N- dimethyl-N-octadecyl (3-aminopropyl) trimethoxysilyl chloride (TEA/DMOAP)-coated substrate modified with capture probes, which can greatly distort the orientational profile of LC, making the optical image of LC cell birefringent as a result. The optical signal of LC sensor has a visible change at the Hg 2+ concentration of low to 0.1 nM, showing good detection sensitivity. The cost-effective LC sensing method can translate the concentration signal of heavy metal ions in solution into the presence of DNA duplexes and is expected to be a sensitive detection platform for heavy metal ions and other small molecule monitors. © 2012 American Chemical Society.

摘要:
This research investigated the ionizing radiation effect on thiamine hydrochloride (TH) and its usability as an irradiation dosimeter. The fundamental principle is to determine the concentration variations of TH solutions with high-performance liquid chromatography (HPLC) after exposing to γ-rays. The decreasing peak area of TH in the HPLC chromatogram forms a linear relationship with the rising radiation dose. We investigated the characteristics and suitable application range of the TH as a new radiation dosimeter. The influence factors and mechanism of the reaction induced by radiation were also discussed. According to the correlation between the concentration and the radiation dose, 0.3 g/L of a TH solution is suitable for the 0.1 - 10 kGy dose range, and 2 g/L is appropriate for 0.1 - 20 kGy. The easy availability and the simple, but stable, chemical structure of thiamine makes it a potential candidate for radiation dose research. 摘要:
为了解决目前依靠转向灯识别换道意图时因转向灯开启率低而识别遗漏的问题,探索运用驾驶人在不同任务条件下眼动行为存在的差异来识别换道意图.在真实道路环境条件下进行实车试验,采集了16名驾驶人车辆保持阶段和换道意图阶段的眼动行为数据,运用数理统计方法对比分析驾驶人在不同阶段的基本眼动参数、注意力分配和视觉搜索模式3个方面眼动行为参数.研究结果表明:平均注视时间、平均扫视幅度、车道注意力分配、后视镜注意力分配、仪表盘注意力分配、区域间注视转移概率、水平视觉搜索广度等参数在2个阶段存在显著差异;与车道保持阶段相比,驾驶人在换道意图阶段对目标车道和后视镜的关注明显增加,对当前车道和车内仪表的关注减少,在各注视区域间注视点转移更加灵活,水平方向视觉搜索区域增大.

摘要:
A novel method of resonance light scattering (RLS) was developed for the analysis of trace metallothioneins (MTs) in human urine. In a CH <inf>3</inf>COOH-CH<inf>3</inf>COONa buffer solution of pH 4.5, the formation of a complex between levofloxacin (LEV)-Pd and MTs led to enhance the RLS intensity of the system, and the enhanced RLS intensity at 468 nm was proportional to the concentration of MTs in the range of 0.059-22.4 μg mL<sup>-1</sup>. The linear regression equation was ΔI = 127.5 ρ(μg mL<sup>-1</sup>)-88.02 with a correlation coefficient of 0.9992, and the detection limit of 17.8 ng mL<sup>-1</sup>. The relative standard deviation and the average recovery were 3.8-5.4% (n = 11) and 92.15%, respectively. The proposed method is convenient, reliable and sensitive, and has been used successfully for the determination of trace MTs in human urine samples. ©2012 Elsevier B.V. All rights reserved.

摘要:
MicroRNAs (miRNAs) are highly conserved, small, noncoding RNAs that regulate gene expression at the posttranscriptional level. Their actions affect numerous important biological processes, including adipocyte differentiation and function, sugar and lipid metabolism, and insulin production and secretion. Recent reports suggest miRNAs may also be involved in the pathogenic processes of obesity, diabetes, and insulin resistance. In this review, we summarize research progresses on adipocyte miRNAs and their physiological and pathological implications. © 2013 Published by NRC Research Press.

摘要:
A wireless magnetoelastic sensor (WMS) for uranyl detection has been developed using DNAzyme-graphene oxide (GO) and gold nanoparticles (AuNPs)-based amplification. The WMS is fabricated by immobilizing a layer of GO film on a magnetoelastic ribbon-like sensor. When the substrate strand of DNAzyme was cleaved in the presence of UO<inf>2</inf><sup>2+</sup>, the single-stranded DNA (ssDNA) was adsorbed onto the GO film of the WMS, resulting in a decrease of resonance frequency of the WMS. HAuCl<inf>4</inf> can be reduced by ascorbic acid (AA) to atom Au coated on AuNPs in cetyltrimethyl ammonium bromide (CTMAB) solution, leading to the signal amplification of the system. When the molar ratio of AA and HAuCl<inf>4</inf> was 3:2, the response signals linearly correlated with the concentration of UO<inf>2</inf><sup>2+</sup> over the range of 9.57 ×10<sup>-9</sup> to 1.20 ×10<sup>-7</sup> M in the MES buffer solution of pH 5.0 with a detection limit of 2.87 ×10<sup>-9</sup> M. The proposed WMS has additional advantages of being less time-consuming, low cost and remote query, and avoids the interference of some metals such as Cu<sup>2+</sup>, Fe<sup>2+</sup>, Fe<sup>3+</sup>, Hg<sup>2+</sup>, and Tb <sup>3+</sup>. It has been successfully applied for detecting UO <inf>2</inf><sup>2+</sup> in real samples with a satisfactory result. A polyacrylamide gel electrophoresis (PAGE) assay, scanning electron microscopy (SEM) and UV-vis spectra were used to confirm the mechanism for structure switching and signal amplification. ©2013 Elsevier B.V.

A resonance light scattering (RLS) method has been developed using a uranyl (UO22+) specific DNAzyme and gold nanoparticles (AuNPs). In this strategy, the cleavage of the substrate strand (SDNA) of DNAzyme results in releasing a shorter duplex in the presence of UO22+, leading to the aggregation of AuNPs and the increase of RLS intensity. The response signals linearly correlated with the concentration of UO22+ over the range of 1.36 ×10<sup>-8</sup>-1.50 ×10<sup>-7</sup> mol L<sup>-1</sup>. The limit of detection (LOD) is 4.09 ×10<sup>-9</sup> mol L<sup>-1</sup>. The method has excellent selectivity and higher sensitivity. It could provide a promising potential for the detection of metal ions, and be benefit to extend the application of RLS method. ©2013 Elsevier B.V. All rights reserved.

A simple and sensitive method for label-free, colorimetric detection of metallothioneins (MTs) has been developed by using a thymine (T)-rich oligonucleotide (TRO)-Hg-AuNP system. In this colorimetric strategy, the thiol groups of MTs could interact with mercury from the T-Hg2+-T complex to release TRO, resulting in a color change of the system. The response signals linearly correlated with the concentration of MTs over the range of 2.56 × 10-8 to 3.08 × 10-7 mol L-1, and the limit of detection was 7.67 × 10-9 mol L-1. The relative standard deviation and the recovery were 2.3-4.8% (n = 11) and 94.2-103.9%, respectively. The proposed method avoids the label and derivatization steps in common methods, allows direct analysis of the samples by the naked eye without costly instruments, and is reliable, inexpensive, and sensitive. © 2013 Elsevier Inc. In this paper, a core-shell structured nanocomposite material was prepared for the detection, adsorption and removal of Hg(II) ions in aqueous solution. The core was made from Fe<inf>3</inf>O<inf>4</inf> nanoparticles with superparamagnetic behavior and the outer shell was made from amorphous silica modified with pyrene-based sensing-probes. The material could detect and adsorb Hg(II) ions in aqueous solution due to its surface being modified with pyrene-based sensing-probes, and could easily be removed from the solution by magnetic force because of its core being made from magnetic Fe<inf>3</inf>O <inf>4</inf> nanoparticles. This multifunctional core-shell structure was confirmed and characterized by TEM, IR spectra, TGA, XRD and N<inf>2</inf> adsorption/desorption isotherms. Experiments were conducted on its functions of detection, adsorption and removal of Hg(II) ions in aqueous solution. The experimental results showed that this composite material had high sensitivity and unique selectivity to Hg(II), and that it could easily be removed from the solution. 摘要:
Melamine, a basic organic chemical intermediate, can cause renal failure because of the formation of insoluble melamine cyanurate crystals in the kidneys. This study examined a novel fluorescence spectrometry for the determination of trace melamine in dairy. The method was based on the inhibitory effect of melamine on the decolorizing reaction of the uncatalytic oxidation of acridine red by potassium permanganate in a sulfuric acid medium. The calibration graph was linear for melamine concentrations of 2.1 × 10−4–1.6 mg/L, and a detection limit of 61.5 ng/L was obtained. The melamine was determined in dairy samples by the proposed method after liquid–liquid and solid phase extraction. The results correlated with the high-performance liquid chromatography. The recoveries were within the range of 99.6–103.3% for liquid milk and 98.5–111.1% for milk powder. The possible mechanism of the reaction was also investigated by ultraviolet spectra.

A novel method for determination of trace amounts of microorganism populations in solution was developed by using a multiwalled carbon nanotube (MWNT) modified glassy carbon electrode and square wave voltammetry (SWV). The simultaneous combination of MWNT and SWV allowed the electrochemical signal of electro-active materials in Escherichia coli O157:H7 (E. coli) to be dramatically amplified. Compared with a bare glassy carbon electrode, the MWNT-modified glassy carbon electrode showed catalytic properties in the oxidation of electro-active materials on cell surfaces. Moreover, SWV was proved to be more sensitive than cyclic voltammetry (CV) for investigation of the electrochemical behavior of cells. In this paper, a linear relationship was obtained between the SWV peak current and the cell concentration in the range 2 × 102–2 × 108 cell mL−1 with a detection limit of 2 × 102 cell mL−1. The effect of antibiotic drug Gentamycin Sulfate injection (GSI) on the growth of E. coli was also investigated.
