摘要:
The intake of contaminated foods is an important exposure pathway for endocrine disrupting chemicals (EDCs). However, data on the occurrence of EDCs in foodstuffs are sporadic and the resultant risk of co-exposure is rarely concerned. In this study, 450 food samples representing 7 food categories (mainly raw and fresh food), collected from three geographic cities in China, were analyzed for eight EDCs using high performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS). Besides estrone (E-1), other EDCs including diethylstilbestrol (DES), nonylphenol (NP), bisphenol A (BPA), octylphenol (OP), 17 beta-estradiol (E-2), 17 alpha-ethinylestradiol (EE2), and estriol (E-3) were ubiquitous in food. Dose-dependent relationships were found between NP and EE2 (r = 0.196, p < 0.05), BPA (r = 0.391, p < 0.05). Moreover, there existed a correspondence between EDCs congener and food category. Based on the obtained database of EDCs concentration combined with local food consumption, dietary EDCs exposure was estimated using the Monte Carlo Risk Assessment (MCRA) system. The 50th and 95th percentile exposure of any EDCs isomer were far below the tolerable daily intake (TDI) value identically. However, the sum of 17 beta-estradiol equivalents (Sigma EEQs) exposure in population was considerably larger than the value of exposure to E-2, which implied the underlying resultant risk of multiple EDCs in food should be concern. In conclusion, co-exposure via food consumption should be considered rather than individual EDCs during health risk evaluation. (C) 2015 Elsevier Ltd. All rights reserved.

摘要:
The paper first reports the use of a kind of positive electrode material (graphite/graphene-oxide composite) in zinc-cerium redox flow battery (RFB). It exhibits excellent electrochemical performance toward Ce3+/Ce4+ redox couple in zinc-cerium RFB. The material was characterized with X-ray diffraction (XRD), scanning electron microscopy (SEM), Raman spectroscopy, and X-ray photoelectron spectroscopy (XPS). The electrochemical activity of this kind of material to Ce3+/Ce4+ redox reaction was studied with cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The results showed that the electrochemical performance of the electrode was improved significantly when 2.5 wt% graphene oxide (GO) was added into graphite electrode. The redox peak current of Ce3+/Ce4+ couple on the composite electrode was significantly larger than that on the graphite electrode, and the charge transfer resistance on the composite electrode is also obviously reduced. The enhanced electrochemical activity could be attributed to the presence of plentiful oxygen-containing functional groups on the basal planes and sheet edges of the GO and large specific areas introduced by the GO. The energy efficiency of zinc-cerium RFB using graphite/GO composite as positive electrode is around 76.3 %, which is larger than that using graphite (65.9 %). The excellent results demonstrate that the graphite/GO composite holds great promise in the application of zinc-cerium RFB.

Indoor air quality has great impact on the human health. An increasing number of studies have shown that printers could release particulate matters and pose adverse effects on indoor air quality. In this study, a thorough investigation was designed to assess the aerosol printer particle total number concentration (TNC) and size distribution in normal office environment, one copy center, and a clean chamber. Particle analyzers, SMPS, OPS, and CPC3007 were used to monitor the total printing process. In normal office environment, 37 laser printers out of all surveyed 55 printers were classified as high particle emitters. Comparing to laser printers, 5 inkjet printers showed no particle emission. Particle emission level in a copy center increased slightly with TNC elevating to about 2 times of the aerosol background. Simulating test in a clean chamber indicated that printer-emitted particles were dominated by particles in nanoscale (diameter of particle, D-p < 100 nm). These particles in a sealed clean chamber attenuated so slowly that it still held at high level with the concentration of 1.5 x 10(4) particles/cm(3) after printing for 2.5 hours. Our present results demonstrate that printers indeed release particulates which keeping at a high concentration level in the indoor environment. Special care should be taken to this kind of widely applied machines and effective controls of particle emission at printing processes are necessary.

We report on an enzyme-free and label-free strategy for the ultrasensitive determination of adenosine. A novel multipurpose adenosine aptamer (MAAP) is designed, which serves as an effective target recognition probe and a capture probe for malachite green. In the presence of adenosine, the conformation of the MAAP is converted from a hairpin structure to a G-quadruplex. Upon addition of malachite green into this solution, a noticeable enhancement of resonance light scattering was observed. The signal response is directly proportional to the concentration of adenosine ranging from 75 pM to 2.2 nM with a detection limit of 23 pM, which was 100-10,000 folds lower than those obtained by previous reported methods. Moreover, this strategy has been applied successfully for detecting adenosine in human urine and blood samples, further proving its reliability. The mechanism of adenosine inducing MAAP to form a G-quadruplex was demonstrated by a series of control experiments. Such a MAAP probe can also be used to other strategies such as fluorescence or spectrophotometric ones. We suppose that this strategy can be expanded to develop a universal analytical platform for various target molecules in the biomedical field and clinical diagnosis. © 2015 Elsevier B.V.

A facile strategy has been developed to fabricate Cu(OH)2 supercages (SCs) as an artificial enzyme system with intrinsic peroxidase-mimic activities (PMA). SCs with high catalytic activity and excellent recyclability were generated via direct conversion of amorphous Cu(OH)2 nanoparticles (NPs) at room temperature. More specifically, the process that takes a single nanoparticle to a 3D supercage involves two basic steps. First, with addition of a copper-ammonia complex, the Cu2+ ions that are located on the surface of amorphous Cu(OH)2 NPs would evolve into a fine lamellar structure by coordination and migration and eventually convert to 1D nanoribbons around the NPs. Second, accompanied by the migration of Cu2+, a hollow cavity is generated in the inner NPs, such that a single nanoparticle eventually becomes a nanoribbon-assembled 3D hollow cage. These Cu(OH)2 SCs were then engineered as an artificial enzymatic system with higher efficiency for intrinsic PMA than the peroxidase activity of a natural enzyme, horseradish peroxidase. © 2015 American Chemical Society.

Silver nanoparticles (AgNPs) attract considerable public attention both for their antimicrobial properties and their potential adverse effects. In the present study, endoplasmic reticulum (ER) stress was used as a sensitive and early biomarker to evaluate the toxic potential of AgNPs in three different human cell lines in vitro and in vivo in mice. In 16HBE cells, the activation of ER stress signaling pathway was observed by upregulated expression including xbp-1s, chop/. DDIT3, TRIB3, ADM2, BIP, Caspase-12, ASNS and HERP at either the mRNA and/or protein levels. However, these changes were not observed in HUVECs or HepG2 cells. Furthermore, mice experiments showed that different tissues had various sensitivities to AgNPs following intratracheal instillation exposure. The lung, liver and kidney showed significant ER stress responses, however, only the lung and kidney exhibited apoptosis by TUNEL assay. The artery and tracheal tissues had lower ER stress and apoptosis after exposure. The lowest observable effect concentrations (LOEC) were proposed based on evaluation of AgNP induced ER stress response in cell and mouse models. In summary, preliminary evaluation of AgNP toxicity by monitoring the ER stress signaling pathway provides new insights toward the understanding the biological impacts of AgNPs. The adverse effects of exposure to AgNPs may be avoided by rational use within the safe dose. © 2015 Elsevier Ltd.

Neuropathy target esterase (NTE) is an endoplasmic reticulum membrane-associated phospholipase B, which is essential for embryonic and nervous system development. However, the regulation of NTE at the protein level had not been thoroughly investigated. Our previous study showed that NTE was degraded not only by the macroautophagy-lysosome pathway but also by the ubiquitin-proteasome pathway. Here we further reveal that androgen receptor-associated protein 54 (ARA54) regulated the ubiquitin-proteasome degradation of NTE. We find that deletion of the regulatory domain of NTE, which possesses a putative destruction box and thus is essential for its degradation by the proteasome, prevented its degradation by the proteasome. In addition, we demonstrate that ARA54, which has a RING finger domain and E3 ligase activity, interacts directly with NTE. Overexpression of ARA54 downregulates the protein level of NTE, and knockdown of ARA54 inhibits the degradation of NTE. The mutation in the RING domain of ARA54 blocks the degradation of NTE by ARA54, which indicates that the RING domain is essential for ARA54's E3 activity. These findings suggest that ARA54 acts as the ubiquitin ligase to regulate the ubiquitin-proteasome degradation of NTE. © 2015 American Chemical Society.

We proposed a novel strategy which combines graphene oxide-based background reduction with RCDzyme-based enzyme strand recycling amplification for ultrahigh sensitive detection of uranyl. The RCDzyme is designed to contain a guanine (G)-rich sequence that replaces the partial sequence in an uranyl-specific DNAzyme. This multifunctional probe can act as the target recognition element, DNAzyme and the primer of signal amplification. The presence of UO22+ can induce the cleavage of the substrate strands in RCDzyme. Then, each released enzyme strand can hybridize with another substrate strands to trigger many cycles of the cleavage by binding uranyl, leading to the formation of more G-quadruplexes by split guanine-rich oligonucleotide fragments. The resulting G-quadruplexes could bind to N-methyl-mesoporphyrin IX (NMM), causing an amplified detection signal for the target uranyl. Next, graphene oxide-based background reduction strategy was further employed for adsorbing free ssDNA and NMM, thereby providing a proximalis zero-background signal. The combination of RCDzyme signal amplification and proximalis zero-background signal remarkably improves the sensitivity of this method, achieving a dynamic range of two orders of magnitude and giving a detection limit down to 86. pM, which is much lower than those of related literature reports. These achievements might be helpful in the design of highly sensitive analytical platform for wide applications in environmental and biomedical fields. © 2015 Elsevier B.V.

进行4个不同厚度、不同配筋方式的34桩厚承台的模型试验,分析承台的破坏模式、开裂荷载、破坏荷载以及桩顶反力分布,基于模型试验结果,以7个厚承台模型进行数值模拟分析,研究大型集群桩基厚承台的受力特性,验证空间桁架模型的合理性,并讨论不同承台厚度和配筋形式对承台的承载性能及桩顶反力分布的影响。结果表明大型集群桩基厚承台符合空间桁架模型的受力特点,承台厚度适当增大和桩顶区域集中配筋均能增强承台的承载力,桩顶反力分布受承台厚度和配筋方式的影响较小,数值模拟的结果与试验结果基本吻合,数值模拟的结果具有一定的参考性。

A novel spectrofluorimetric method has been developed for the simultaneous determination of leucomalachite green (LMG), leucocrystal violet (LCV), malachite green (MG) and crystal violet (CV) by combining the sensitivity of molecular fluorescence and the selectivity of the second-order calibration. Residues of LMG, LCV, MG and CV were simultaneously extracted from fish and shrimp muscle with acetonitrile. The non-fluorescent CV and MG were then reduced to the corresponding fluorescent LMG and LCV by reacting with sodium borohydride. After preconcentration with dispersive liquid-liquid microextraction technique, the extracts were analyzed by using excitation-emission matrix fluorescence coupled with second-order calibration methods based on parallel factor analysis (PARAFAC) and alternating trilinear decomposition (ATLD) algorithms. The limits of detection obtained were 2.21-2.65 ng g-1 by PARAFAC and 2.30-2.86 ng g-1 by ATLD, respectively. The developed method was successfully applied to simultaneous determination of the four analytes in grass carp and shrimp samples with recoveries of 90.53-103.03% for PARAFAC and 90.40-102.75% for ATLD. The accuracy of this novel method was also verified by high performance liquid chromatography. © 2015 Elsevier Ltd.

The interactions of metallothioneins with Eosin Y were studied by the absorption spectra and resonance light scattering (RLS) spectra methods. A direct RLS spectra method was applied for the determination of trace metallothioneins. The interaction of Eosin Y and metallothioneins enhanced the RLS intensity of system in Britton-Robinson buffer (pH 3.91), with the help of anionic surfactant (sodium dodecyl benzene sulphate). The mechanism was studied and discussed in terms of the RLS and UV-absorption spectra. Under optimal experimental conditions, at 366 nm, there was a linear relationship between the RLS intensity and the concentration of the metallothioneins in the range of 0.04–14.0 μg mL<sup>−1</sup>, with a correlation coefficient of r = 0.9985 and detection limit of 11.9 ng mL<sup>−1</sup>. The relative standard deviation was 3.1% (n = 11), and the average recovery was 95.2%. The method proposed was reliable, selective and sensitive in determining trace metallothioneins in human urine samples with the results in good agreement with those obtained by high-performance liquid chromatography. ©2015 Taylor & Francis.

摘要:
目的:研究卷烟烟气所致人支气管上皮细胞DNA损伤以及茶多酚二甲基亚砜合剂的保护作用,为卷烟烟气对机体损伤的防护提供理论及实验依据。方法将细胞分为5组:空白组、卷烟烟气组、茶多酚保护组、二甲基亚砜保护组、茶多酚二甲基亚砜合剂保护组;通过检测微核和多核细胞观察染色体损伤,彗星实验及H2AX蛋白磷酸化检测DNA的损伤,多核细胞法检测细胞HPRT突变。结果BEAS-2B细胞经卷烟烟气染毒后,细胞的微核率、多核率、拖尾率升高,H2AX蛋白磷酸化表达增高, HPRT突变率增高,差异均有统计学意义(P<<0.05),茶多酚保护组、二甲基亚砜保护组能降低卷烟烟气诱发的DNA损伤,合剂保护组的防护作用最好。结论卷烟烟气对BEAS-2B细胞DNA具有损伤作用,茶多酚二甲基亚砜合剂有较强防护作用。

Exposure to metals, including essential and nonessential elements, is widespread and may be associated with altered semen quality. This study aimed to examine the association between urinary metal concentrations and semen quality in a Chinese population. We measured semen quality parameters (sperm concentration, count, motility, normal morphology, and abnormal head) and 13 metals [arsenic (As), cadmium (Cd), cobalt (Co), chromium (Cr), copper (Cu), iron (Fe), lead (Pb), manganese (Mn), molybdenum (Mo), mercury (Hg), nickel (Ni), selenium (Se), and zinc (Zn)] in the urine of 394 men from an infertility clinic. Multivariable logistic and linear regressions were used to assess the relationship between the creatinine-adjusted urinary metal concentrations and semen quality parameters. We found a significant trend for decreased odds ratios (ORs) for below-reference sperm count with increasing Se quartiles (p for trend = 0.04) and a significant trend for increased sperm percent abnormal head with increasing Ni quartiles (p for trend = 0.03). These associations persisted, even when considering multiple metals. Our results suggest that Ni exposure may be associated with deteriorated sperm morphology and that Se exposure may be associated with better semen quality. However, our findings warrant further studies in a larger and general population. © 2015 American Chemical Society.

Amidoxime modified Aspergillus niger (AMAN) was prepared by the oximation reaction. The effects of the initial pH, contact time, initial U(vi) concentration and biosorbent dose on the adsorption of U(vi) ions from radioactive wastewater in U(vi) concentrations of less than 1 mg L<sup>-1</sup>by AMAN and the raw Aspergillus niger (RAN) were investigated. The maximum adsorption efficiency by AMAN for the 0.5 mg L<sup>-1</sup>U(vi) solution amounted to 98.85% under the optimum adsorption conditions, while the maximum adsorption efficiency by RAN was only 77.83%. The adsorption equilibrium data were found to be best fitted to Langmuir isotherm model, and the maximum biosorption capacity of AMAN for U(vi) was estimated to be 621 mg g<sup>-1</sup>at 298 K. The biosorption kinetics followed the pseudo-second order model and intraparticle diffusion equation. The Gibbs free energy change (ΔG°), enthalpy change (ΔH°) and entropy change (ΔS°) showed that the adsorption process of U(vi) was spontaneous, feasible and endothermic. The SEM-EDS study indicated that much more U(vi) ions were adsorbed by AMAN than by RAN. FT-IR study showed that the -NH<inf>2</inf>and =N-OH groups of amidoxime were the dominant ones for binding UO<inf>2</inf><sup>2+</sup>ions. Moreover, AMAN was found to have excellent selective adsorption capability of U(vi) due to amidoxime groups. The UO<inf>2</inf><sup>2+</sup>ions adsorbed by AMAN could be desorbed using 0.1 M HCl, and the desorption efficiency reaching 87.28% at the 8th cycle of adsorption and desorption. ©The Royal Society of Chemistry.

The environmental pollution associated with electrical discharge machining is not yet clearly understood. Airborne exposure to nanoscale and respirable particles were investigated with regard to the aerosol characteristics of a wire electrical discharge machining (WEDM) workshop. The total number concentration of the aerosol was multimodal, with the highest peak maxima during the working hours of 10:00 am and 3:00 pm. The majority of the released particles were smaller than d = 100 nm, with the maximum amount sized 40 nm. A large quantity of metallic elements, including Fe, Al and Cu, were found in the aerosol particulates coming from WEDM processing. Furthermore, the aerosol particles exhibited higher cellular toxicity and ROS producing ability in human alveolar epithelial cells (16HBE) when compared to the atmospheric background. Our results indicate substantial hazards arising from exposure to polluted atmosphere of a WEDM workshop. Effective exposure controls and protections are thus strongly recommended. © Taiwan Association for Aerosol Research.

根据目标物诱导核酸分子构象发生变化,导致体系共振光散射强度增大的原理建立了检测Hg~(2+)的新方法。当溶液中有Hg~(2+)存在时,基于"T-Hg~(2+)-T"特异性结合作用,促使Hg~(2+)特异核酸探针的构象发生变化,由单链状态变为刚性双链结构,使体系共振光散射强度增大。在波长566 nm处,当汞离子浓度在7.2 ×10~(-9)~9×10~(-8)mol? L~(-1)范围内时,体系的共振光散射增强程度△I与汞离子的浓度(c)具有良好的线性关系。其线性方程为△I=5.12c+3.55(r=0.999 5)。将该方法用于环境水样中汞离子的测定,相对标准偏差(RSD)小于1.9%;样品加标回收率为99.4%~104.3%。该方法以Hg~(2+)的高度特异性核酸探针作为识别元件,通过控制Hg~(2+)浓度的变化调节共振光散射强度的变化,将Hg~(2+)的检测转化为对DNA分子构象变化的检测,该转化有利于增强体系共振光散射强度,提高方法灵敏度,该共振光散射检测方法能检测到浓度为2.16×10~(-9)mol?L~(-1)的Hg~(2+)。同时,由于"T-T"错配碱基对对Hg~(2+)的特异结合能力,显著提高了该分析方法对Hg~(2+)的选择性。另外,该方法操作简便、不需标记。

This study investigated the chromatographic characteristics of irradiated vitamins and their feasibility for dose measurement by using high performance liquid chromatography (HPLC). Water-soluble vitamins in the B family, including thiamine hydrochloride (TH), riboflavin (RF) and nicotinic acid (NA), were used for comparative study. The results showed that riboflavin was sensitive to irradiation, and the content changes of RF were linearly correlated with irradiation doses. Combining the analyses of ultraviolet and fluorescence spectra, the suitable application range for dose determination was confirmed from 100 to 2000 Gy; moreover, the influencing factors were further discussed. The content of RF could maintain good stability before and after irradiation under low-light conditions. These characteristics make it possible for RF to be used as a material for dose measurement. In addition, this work also provides references for irradiation nutrition research due to the favourable separation and analysis characteristics of HPLC. © 2014 The Royal Society of Chemistry.

The fabrication of tiron-copper complex as a novel fluorescence probe for the sensitive directly detection of metallothioneins at nanomolar levels was demonstrated. In Britton-Robinson (B-R) buffer (pH 7.50), the interaction of bis(firon)copper(II) complex cation [Cu(tiron)(2)](2+) and metallothioneins enhanced the fluorescence intensity of the system. The fluorescence enhancement at 347 nm was proportional to the concentration of metallothioneins. The mechanism was studied and discussed in terms of the fluorescence spectra. Under the optimal experimental conditions, at 347 nm, there was a linear relationship between the fluorescence intensity and the concentration of the metallothioneins in the range of 8.80 x 10(-9)-7.70 x 10(-7) mol L-1, with a correlation coefficient of r = 0.995 and detection limit 2.60 x 10(-9) mol L-1. The relative standard deviation was 0.77% (n = 11), and the average recovery 94.4%. The method proposed was successfully reliable, selective and sensitive in determining of trace metallothioneins in fish visceral organ samples with the results in good agreement with those obtained by HPLC. We developed a highly sensitive label-free assay of 8-hydroxy-2'-deoxyguanosine (8-OHdG) using 8-OHdG-aptamer (Apt) as the recognition element. The Apt was adsorbed onto the surface of gold nanoparticles (AuNPs), which prevents them from aggregating under high-salt conditions. Upon addition of 8-OHdG, the conformation of the Apt changes to form a G-quadruplex structure, which leads to the aggregation of the AuNPs along with the increase of the resonance light scattering intensity. The mechanism of 8-OHdG that induces Apt to form G-quadruplexes structure was studied by circular dichroism. The response signals linearly correlated with the concentration of 8-OHdG ranging from 90.8. pM to 14.1. nM with a detection limit of 27.3. pM, which is much lower than that obtained by other methods. This method does not need any label steps and sophisticated equipment. The application for detection of 8-OHdG in real samples further demonstrated its reliability. This strategy would be helpful for developing a universal analytical method by simply replacing corresponding aptamers for various target molecules. © 2014 Elsevier B.V.

The novel quantum topological indices PY<inf>1,2</inf>were derived from molecular structure combined with the effect of atom space, the character of bonding atoms (such as equilibrium electro-negativity) and the branching effect between the atoms. The quantitative structure-property relationships (QSPRs) were proposed between PY<inf>1,2</inf>and the thermodynamic properties (<inf>f</inf>H<sup>θ</sup>, <inf>f</inf>G<sup>θ</sup>and Δ<inf>f</inf>G<inf>R</inf><sup>θ</sup>) of phenoxazine (Phx) and 135 kinds of polychlorinated phenoxazines (PCPXs), by Multiple linear regression (MLR) analysis method. The high-quality prediction models were evidenced by the correlation coefficient R, the standard error of estimate S, the Fisher statistic value and the cross-validated correlation coefficient RCV. With the new QSPR model, we are able to predict a wide range of thermodynamic properties of an extensive number of molecules. And the model is statistically significant and shows good stability for data variation as tested by the leave-one-out cross-validation (LOO-CV). ©2014 Published by Elsevier B.V.
