摘要:
To identify the importance of IRAK2 kinase activity in TLR-mediated signaling pathways, we constructed a retroviral vector harboring either a mouse IRAK2 gene (IRAK2-WT) or with its mutant with loss of function of its ATP-binding site (IRAK2-KD). Further, we comparatively analyzed for the gain of function and modulations in TLR-mediated signaling pathways in IRAK2 knockout (IRAK2-KO) macrophages upon introduction of the IRAK2-WT retroviral constructs. The pBS/IRAK2-KD with the ATP-binding site mutation in IRAK2 was obtained by using site-specific mutagenesis. The recombinants were identified with appropriate double digestion and sequence analysis. The recombinant vector constructs were transfected by lipofection into phoenix packaging cells. The viral vectors (10<sup>7</sup> cfu/mL) with the construct were allowed to infect IRAK2-KO macrophages. The results showed that IRAK2-WT gene overexpressed in the IRAK2-KO macrophages exhibited a modified IRAK2 expression upon LPS induction. However, the modification was absent with IRAK2-KD construct on LPS stimulation;instead, the IRAK2 protein stability was reduced considerably. The results further show that the LPS-induced effect on the stability of IRAK2 is dependent of IRAK4 stimulation.<br/> ©2018, Springer Science+Business Media, LLC, part of Springer Nature.

摘要:
Neuronal growth, differentiation, extension, branching and neural network activity are strongly influenced by the mechanical property of extracellular matrix (ECM). However, the mechanism by which substrate stiffness regulates a neural network activity, and the importance of ECM composition in conferring substrate stiffness sensing have not been explored. To address this question, the hippocampal neurons were seeded on the polydimethylsiloxane (PDMS) substrate with different stiffness, which were coated with fibronectin and laminin respectively. Our results show that voltage-gated Ca2+ channel currents are greater in neurons on the stiff substrate than on the soft substrate. In addition, the neurons exhibit a greater increase of Ca2+ currents on laminin-coated stiff substrate than on those coated with fibronectin, indicating that the composition of ECM can modulate responses to substrate stiffness of neurons. Paired patch-clamp recordings have shown that upregulation of neural effective synaptic connectivity is greater on the laminin-coated stiff substrate than on the fibronectin-coated ones. Consistently, laminin-coated stiff substrate enhances Ca2+ oscillations of neurons is greater that compared with the fibronectin-coated ones. Our study demonstrates that a direct role for substrate stiffness in regulating neuronal network activity and indicate that this modulation is dependent on a specific type of ECM protein, which should be taken into account for the design of biomaterials for neuronal tissue engineering. © 2018 Elsevier B.V.

摘要:
Development of sensitive assay for detection of hotspot mutations of cancer driving gene is crucial for circulating tumor DNA analysis. This study tested the possibilities of applying restriction enzyme digestion and dephosphorylation coupled with blue/white screening technology for analyzing a hotspot point mutation in codon 13 of KRAS gene. The present study has documented that the combination of PCR with restriction digestion, dephosphorylation, blue/white screening and Sanger's sequencing can identify rare mutations with sensitivities at 0.003%. This novel assay with high sensitivity may have application in the diagnosis of early cancer targeting ctDNAs.

摘要:
As an important tumor marker, the expression level of premicroRNA-21 (pre-miR-21) can provide important information for early diagnosis, drug treatment and prognosis of tumor. Thus, developing pre-miR-21 detection is of great importance for diagnosis of disease. Herein, a direct, simple and rapid method for quantitative detection of pre-miR-21 was developed. This detecting system contained the tool of molecular beacon which could hybridize specifically with pre-miR-21 and form duplex to induce fluorescence signal change. When loop bases hybridized with the pre-miR-21 in the solution to form a double-stranded duplex, the fluorophore and the quencher was separated and caused fluorescence recovery. We developed the new method for pre-miR-21 assay with detection limit of 0.5 nM. Under the optimal conditions, the method was used to detect the level of pre-miR-21 in different tumor cells and tissues. The results showed that pre-miR-21 level was tightly related with tumor cells origins and malignancy degree. In 16 cases of gastric cancer tissues and adjacent tissues, the level of pre-miR-21 in 12 cases of cancer tissues was higher than that of adjacent tissues; 3 cases had lower expression level than that of adjacent tissues, and 1 case had no significant difference. Furthermore, Quantitative real-time PCR (qRT-PCR) method was used in to confirm the reliability of the detection results. The consistent results of the two different methods clearly showed that the molecular beacon assay with advantages of simplicity and rapidity for pre-miR-21 detection was hopeful for the wide usability in the function research and clinical diagnosis of pre-microRNA. © 2018 Changchun Institute of Applied Chemistry, Chinese Academy of Sciences

摘要:
In the past decades, significant progresses have been achieved in genetic engineering of nucleases. Among the genetically engineered nucleases, zinc finger nucleases, transcription activator-like (TAL) effector nucleases, and CRIPSPR/Cas9 system form a new field of gene editing. The gene editing efficiency or targeting effect and the off-target effect are the two major determinant factors in evaluating the usefulness of a new enzyme. Engineering strategies in improving these gene editing enzymes, particularly in minimizing their off-target effects, are the focus of this paper. Examples of using these genetically engineered enzymes in genome modification are discussed in order to better understand the requirement of engineering efforts in obtaining more powerful and useful gene editing enzymes. In addition, the identification of naturally existed anti-Cas proteins has been employed in minimizing off-target effects. Considering the future application in human gene therapy, optimization of these well recognized gene editing enzymes and exploration of more novel enzymes are both required. Before people find an ideal gene editing system having virtually no off-target effect, technologies used to screen and identify off-target effects are of importance in clinical trials employing gene therapy.

Nowadays, there is lack of effective serological detection method for Mycoplasma pneumoniae (M. pneumoniae) infection in clinic. In this study, the mimic epitopes of M. pneumoniae were screened to evaluate the role in the serodiagnosis of M. pneumoniae infection. The M. pneumoniae-positive serum was used as the target for biopanning to phage display random 7-peptide library. The positive phage clones were selected and the DNA were sequenced and analyzed by BLAST. The representative phages were identified using dot immunoblotting and ELISA. The exogenous heptapeptides were synthesized and their reactions with M. pneumonia-positive serum were tested by indirect ELISA. Two heptapeptides, namely heptapeptide 1: TVNFKLY and heptapeptide 2: LPQRLRT, were screened out from the randomly selected 40 phages after the four bio-panning rounds. They had high homologies to some M. pneumoniae antigens. Besides, the representative bacteriophage containing heptapeptide 1 or 2 could react with the M. pneumonia- positive serum. The sensitivities of heptapeptide 1 and heptapeptide 2 for the diagnosis of M. pneumoniae infection were 90.1 and 80.0%, respectively, and the specificities were 94.3 and 97.1%, respectively. Therefore the two heptapeptides were the mimic epitopes of M. pneumoniae and might have potential serological diagnosis value for M. pneumoniae infection.

Nanotechnology has been widely used for developing sensors with higher sensitivity and better selectivity. In this study, a molecularly imprinted electrochemical sensor based on nitrogen-doped carbon nanosheet frameworks decorated with Fe (Fe-NCNF) was proposed for simultaneous detection of mebendazole (Meb) and catechol (CC). Fe-NCNF was prepared by chemical blowing, and it was coated with molecularly imprinted polymer (MIP) to construct a novel hybrid nanomaterial, which is named Fe-NCNF/MIP. Meb-MIP was electropolymerized with methacrylic acid (MAA) as monomer via cyclic voltammetry (CV) method. This sensor architecture can detect Meb because of the high binding affinity of MIP. With the large specific surface area, Fe-NCNF can adsorb and electrochemically oxidize CC. The good selectivity of Fe-NCNF/MIP for CC and Meb was proven by anti-interference test, for which the oxidation potentials were set at 0.15 and 0.82 V respectively. The linear range for determination of CC was from 0.5 to 25 μM with a low limit of detection (LOD) of 0.06 μM (S/N = 3), and that for Meb was 0.01–1.5 μM with a low LOD of 0.004 μM (S/N = 3). Furthermore, this MIP electrochemical sensor was applied for the simultaneous detection of CC and Meb in tap water and river water. These results demonstrated that this method is reliable for selective, sensitive and feasible determination of Meb and CC. © 2018 Elsevier B.V.

APC gene mutations are associated with the development of many types of cancers and also related to cancers' sensitivities to selected drugs. Genetic testing of APC mutation using cancer tissues have been well developed, but assays targeting circulating tumor DNA of APC gene mutations are to be developed in order to meet the requirements by clinical application. This study tested the possibilities of applying restriction enzyme digestion combined with blue/white screening technology in analyzing a hotpot point mutation in APC gene. The present study has documented that the combination of restriction enzyme digestion, PCR, blue/white screening, and Sanger's sequencing can identify rare mutations in sensitivities at or higher than 0.01%. One advantage of this new assay is that it is false positive free with the confirmation by Sanger's sequencing. Additionally, this new strategy may be applied in the analysis of other hotspot mutations with the involvement of transformable stop codon in situations when the loss of a restriction enzyme site is destroyed by the mutation.

Chemotherapy is the most common therapeutic strategy for the treatment of unresectable hepatocellular carcinoma. However, the therapeutic efficacy is limited by the low delivery efficiency of chemotherapeutics and severe toxicity towards healthy tissues. To address these challenges, active-targeting mesoporous silica nanoparticles conjugating a platinum(IV) prodrug were developed as a therapy for liver cancer for the first time. Taking advantage of liver-targeting lactobionic acid (LA), the smart nano-carriers not only enhanced the circulation time, but also effectively concentrated at the liver tumor site. Moreover, the conjugated platinum(IV) could be reduced in the reductive tumor environment for the fast release of active platinum(II). The novel targeting and self-responsive drug-loading system offers new prospects for liver cancer chemotherapy.

In recent years, some two-dimensional nanomaterials, such as graphene and molybdenum disulfide, cause extensive interests from scientists due to their excellent performances. In this paper, MoS<inf>2</inf>was synthesized by a simple hydrothermal method without any assisted reagents, the MoS<inf>2</inf>nanosheets display relatively high photocatalytic activity of p-nitrophenol degradation under visible light illumination (λ>420 nm). Some experimental reaction conditions were optimized, the highest degradation efficiency of 55.0% is obtained within 120 min at initial 10.0 mg L<sup>−1</sup>p-nitrophenol when the MoS<inf>2</inf>is prepared at 200 °C for 20 h with the mole ratio of S/Mo = 1/1. A probable mechanism of photo-degradation was discussed. To our knowledge, there are no other reports on MoS<inf>2</inf>photocatalytic degradation of phenolic compounds including p-nitrophenol so far. Some two-dimensional nanomaterials, such as graphene and molybdenum disulfide, are presently being intensively investigated due to their excellent and unique performances. In the field of photocatalysis, MoS<inf>2</inf>is considered as a promising alternative to noble metal Pt for the hydrogen evolution reaction (HER). However, its poor electrical conductivity restricts its catalytic activity in the HER. In this work, MoS<inf>2</inf>was modified with graphene (G) by a simple hydrothermal method. The prepared G/MoS<inf>2</inf>composite was characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). The results show that graphene modification does not influence the crystal phase of MoS<inf>2</inf>, but makes the latter more dispersed. The HER performance of G/MoS<inf>2</inf>was evaluated using eosin Y (EY) as a photosensitizer, and triethanolamine (TEOA) as a sacrificial electron donor under visible-light irradiation (λ>420 nm, 250 W high pressure Hg lamp as light source). The EY sensitized G/MoS<inf>2</inf>composite displays enhanced hydrogen evolution in terms of not only activity but also stability. The average HER activity (9.1 μmol h<sup>-1</sup>) is three times that of EY sensitized pure MoS<inf>2</inf>over 10 h. It is believed that the incorporation of graphene enhances the charge transfer ability and retards the self-degradation path of EY<sup>-</sup>, ultimately improving the HER. ©2017 The Royal Society of Chemistry.

An innovative electrochemical sensor, based on a carbon paste electrode (CPE) modified with graphene (GR) and a boron-embedded duplex molecularly imprinted hybrid membrane (B-DMIHM), was fabricated for the highly sensitive and selective determination of lamotrigine (LMT). Density functional theory (DFT) was employed to study the interactions between the template and monomers to screen appropriate functional monomers for rational design of the B-DMIHM. The distinct synergic effect of GR and B-DMIHM was evidenced by the positive shift of the reduction peak potential of LMT at B-DMIHM/GR modified CPE (B-DMIHM/GR/CPE) by about 300 mV, and the 13-fold amplification of the peak current, compared to a bare carbon paste electrode (CPE). The electrochemical reduction mechanism of lamotrigine was investigated by different voltammetric techniques. It was illustrated that square wave voltammetry (SWV) was more sensitive than different pulse voltammetry (DPV) for the quantitative analysis of LMT. Thereafter, a highly sensitive electroanalytical method for LMT was established by SWV at B-DMIHM/GR/CPE with a good linear relationship from 5.0×10<sup>−8</sup>to 5.0×10<sup>−5</sup>and 5.0×10<sup>−5</sup>to 3.0×10<sup>−4</sup> mol L<sup>−1</sup>with a lower detection limit (1.52×10<sup>−9</sup> mol L<sup>−1</sup>) based on the lower linear range(S/N=3). The practical application of the sensor was demonstrated by determining the concentration of LMT in pharmaceutical and biological samples with good precision (RSD 1.04–4.41%) and acceptable recoveries (92.40–107.0%). ©2017 Elsevier B.V.

KRAS (Kirsten rat sarcoma viral oncogene homolog) mutations are associated with the development of many types of cancers and are also related to the cancer sensitivities to selected drugs. Genetic testing of KRAS mutation using cancer tissues has been well developed, but assays targeting circulating tumor DNA (ctDNA) of KRAS mutations are to be developed in order to meet the requirements for clinical applications. This study tested the possibilities of applying blue/white screening technology in analyzing point mutations at codon 13 of KRAS gene. The present study has documented that the combination of PCR, subcloning, blue/white screening and Sanger's sequencing can identify rare mutations with sensitivities up to 0.1%. One advantage of this new assay is that it is free of false positives. Higher sensitivity of the assay could be achieved using magnatic nanobeads to trap the wild typed fragments before, during, and after the amplification procedure. Furthermore, the quick blue/white assay can be used for the optimization of hybridization probes linked onto the nanobeads. Additionally, this new strategy may be applied to the analysis of hotspot mutations with the involvement of transformable stop codon of other genes.

Highly selective determination of enantiomers by electrochemical sensors with simple recognition elements is of great interest in pharmaceutics, biochemistry and medical fields. The present work developed an innovative electrochemical sensor, which based on boron-embedded molecularly imprinted hybrid film (B-MIHF) modified carbon paste electrode (CPE), for enantioselective determination of S-ornidazole (S-ONZ). A boron-embedded conductive copolymer was firstly electro polymerized on carbon paste electrode (CPE), and then molecularly imprinted polysiloxanes (MIP) were hydrolyzed and condensed over the conductive copolymer layer via a sol-gel process. The as-prepared B-MIHF, comprised the advantages of boron-embedded conductive copolymer and molecularly imprinted polysiloxanes, could not only implement direct signal transformation between imprinted cavities and sensing elements, but show amazing selectivity to template S-ornidazole. Above all, the introduction of coordinate interaction formed between trisubstituted boron atoms embedded in B-MIHF matrix and nitrogen atoms of template endow the sensor with intensive enantioselectivity. Under optimum experiments conditions, the reduction peak currents of S-ONZ by differential pulse voltammetry were linear to S-ONZ concentrations in the range from 6.0 x 10(8) to 1.0 x 10(6) mol L-1 and 1.0 x 10(-6) to 3.5 x 10(4) mol L-1 with a detection limit of 1.1 x 10(-8) mol L-1. The B-MIHF-based sensor has been successfully applied for the determination of S-ONZ in pharmaceutical and biological samples with good precision (RSD 2.25-4.11%) and acceptable recoveries (96.00-108.00%). Long non-coding RNAs (lncRNAs) are transcripts longer than 200 nucleotides without protein-coding potential. Although these molecules were initially considered as "junk products" of transcription without biological relevance, recent advances in research have shown that lncRNA plays an important role, not only in cellular processes such as proliferation, differentiation, and metabolism, but also in the pathological processes of cancers, diabetes, and neurodegenerative diseases. In this review, we focus on the potential regulatory roles of lncRNA in diabetes and the complications associated with diabetes. © 2017 Published by NRC Research Press.

摘要:
As an endogenous gaseous mediator, H2S exerts antioxidative, antiapoptotic, and cytoprotective effects in livers. This study was designed to investigate the protective role of H2S against uranium-induced hepatotoxicity in adult SD male rats after in vivo effect of uranium on endogenous H2S production was determined in livers. The levels of endogenous H2S and H2S-producing enzymes (CBS and CSE) were measured in liver homogenates from uranium -intoxicated rats. In rats injected intraperitoneally (i.p.) with uranyl acetate or NaHS (an H2S donor) alone or in combination, we examined biochemical parameters to assess liver function, revealed hepatic histopathological alteration, investigated oxidative stress markers, and explored apoptotic signaling in liver homogenates. The results suggest that uranium-intoxication in rats decreased CBS and CSE protein expression, H2S synthesis capacity, and endogenous H2S generation. NaHS administration in uranium-intoxicated rats produced amelioration in liver biochemical indices and histopathological effects, decreased MDA content, and increased GSH level and antioxidative enzymes activities like SOD, CAT, GPx, and GST. NaHS administration in uranium-intoxicated rats attenuated uranium-activated phosphorylation state of JNK. NaHS treatment in uranium-intoxicated rats increased antiapoptotic Bcl-2 but decreased pro-apoptotic Bax, resulting in the rise of Bcl-2/Bax ratio. NaHS treatment in uranium-intoxicated rats reduced the apoptosis mediator caspase-3 and cytochrome c release and elevated ATP contents. Taken together, these data implicate that H2S can afford protection to rat livers against uranium-induced adverse effects mediated by up-regulation of antioxidant and antiapoptotic signaling. BACKGROUND: Photocatalytic degradation of organic compounds, including nitrophenol, is one of the most popular approaches to pollution control and disposal. Graphene-based nanocomposites have attracted considerable attention in photocatalytic research owing to their excellent electrical, thermal and mechanical properties. In this work, TiO<inf>2</inf>material was modified with graphene by a hydrothermal method, the photocatalytic performance of the prepared catalyst was evaluated by degradation of 4-nitrophenol, which is a highly toxic, stable and carcinogenic organic pollutant. RESULTS: The characterization results showed that the low amount of graphene disperses well over TiO<inf>2</inf>and it does not influence the crystal phase, but makes the specific surface area of graphene/TiO<inf>2</inf>greater than that of TiO<inf>2</inf>. Compared with TiO<inf>2</inf>, graphene/TiO<inf>2</inf>exhibits enhanced photocatalytic activity for the degradation of 4-nitrophenol under simulated solar irradiation. CONCLUSION: The enhanced activity of photo-degradation could be attributed to the involved graphene being beneficial to charge transportation and separation of photogenerated charge carriers, and providing more surface active sites for degradation reaction of the target pollutant. The present result highlights the important application of graphene as a novel carbon material in treating highly toxic pollutant through photocatalysis technology. ©2017 Society of Chemical Industry. ©2017 Society of Chemical Industry

This research work presents two microbial synthesis approaches of silver nanoparticles (AgNPs) from the bacterial strain Pseudomonas aeruginosa CI109 through bio-reduction of silver ions. The antibacterial efficacy of the AgNPs on multi-drug resistant (MDR) clinical pathogens was also confirmed. The synthesized AgNPs were characterized by ultraviolet-visible (UV-vis) spectroscopy, scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX), transmission electron microscope (TEM) and laser particle size analyzer. Furthermore, the antibacterial characteristics of the synthesized AgNPs against MDR strains were investigated by an agar disk diffusion test. The UV analysis demonstrated that the diameters of the synthetic silver nanoparticles using the two biosynthetic methods increased gradually from 12 h to 72 h as its characteristic absorption peaks showed a bathochromic effect with elapsed time. EDX indicated the distinctions of the two types of the silver nanoparticles in the localized elemental information. By SEM and TEM, synthetic silver nanoparticles were found to be spherical or ellipsoidal in shape and their diameter using the extracellular solution of P. aeruginosa as a reducing agent (Av. 14 nm) was larger than when the P. aeruginosa cells (Av. 6 nm) was used. The two types of the silver nanoparticles showed good antibacterial effects against clinical MDR pathogens, and their relevant antimicrobial properties demonstrated a dose-dependent manner.

Chronic unpredictable mild stress (CUMS) induces hippocampal oxidative stress. H<inf>2</inf>S functions as a neuroprotectant against oxidative stress in brain. We have previously shown the upregulatory effect of H<inf>2</inf>S on BDNF protein expression in the hippocampus of rats. Therefore, we hypothesized that H<inf>2</inf>S prevents CUMS-generated oxidative stress by upregulation of BDNF-TrkB pathway. We showed that NaHS (0.03 or 0.1 mmol/kg/day) ameliorates the level of hippocampal oxidative stress, including reduced levels of malondialdehyde (MDA) and 4-hydroxy-2-trans-nonenal (4-HNE), as well as increased level of glutathione (GSH) and activity of superoxide dismutase (SOD) in the hippocampus of CUMS-treated rats. We also found that H<inf>2</inf>S upregulated the level of BDNF and p-TrkB protein in the hippocampus of CUMS rats. Furthermore, inhibition of BDNF signaling by K252a, an inhibitor of the BDNF receptor TrkB, blocked the antioxidant effects of H<inf>2</inf>S on CUMS-induced hippocampal oxidative stress. These results reveal the inhibitory role of H<inf>2</inf>S in CUMS-induced hippocampal oxidative stress, which is through upregulation of BDNF/TrkB pathway. ©2016 Min Hu et al.

摘要:
地震反应的有效估计是液化场地桥梁结构设计、加固改造的必要环节,但通过数值模拟手段获得的结果易受模型参数变化的影响.为探究影响液化场地桥梁结构地震反应的控制性参数,该文首先建立了某离心机试验的场地-结构整体有限元模型,通过数值分析与试验结果的对比验证了数值方法的可靠性;然后,建立了上覆粘土层-松砂层-密砂层的典型倾斜液化场地-桩柱式桥梁结构有限元模型,采用龙卷风图和一次二阶矩方法,以场地和结构的主要物理性质为分析参数,进行场地、结构地震反应的敏感性分析.结果表明:场地、结构的地震反应与各分析参数大体上呈线性或低非线性的变化关系;在所研究的参数中,液化松砂层的内摩擦角、土层倾角和地震峰值加速度为控制性参数,而密砂层各参数和钢筋、混凝土强度参数的敏感性均很低.研究结果可对液化场地桥梁结构地震反应的准确估算,振动台、离心机模型试验的工况优化提供科学参考.
