Melatonin receptors in diabetes: A potential new therapeutical target?

摘要:
Melatonin is synthesized and secreted mainly by the pineal gland in a circadian fashion, and it thus mediates endogenous circadian rhythms and influences other physiological functions. Both the G-protein coupled receptors MT1 (encoded by MTNR1A) and MT2 (encoded by MTNR1B) in mammals mediate the actions of melatonin. Evidence from in vivo and in vitro studies proved a key role of melatonin in the regulation of glucose metabolism and the pathogenesis of diabetes, as further confirmed by the recent studies of human genetic variants of MTNR1B. Remarkably, it was also suggested that genetic variations within MTNR1B disordered beta-cells function directly, i.e. insulin secretion. This indicated the functional link between MT2 and T2D risk at the protein level, and it may represent the prevailing pathomechanism for how impaired melatonin signaling causes metabolic disorders and increases the T2D risk lt is speculated that melatonin and its receptors may be a new therapeutic avenue in diabetes. 摘要:
The interaction of blood glucose with heme proteins plays a key role in inducing diabetes, a serious disease threatening human health. In this study, we investigated the non-covalent interaction between glucose and myoglobin (Mb), both theoretically and experimentally, using molecular dynamics (MD) simulation combined with spectroscopic studies. It revealed that glucoses can occupy the side pocket of Mb, and bind closely to one of the xenon cavities in Mb, by hydrogen bonding interactions with two propionate groups of heme as well as surrounding amino acids. These interactions alter the conformation of the heme active site slightly and lead to an enhanced peroxidase activity of Mb, as determined by kinetic studies. This study provides general information for glucose-heme proteins interactions, and also for blood glucose-protein interactions for patients with diabetes.

Uranium is harmful to human health due to its radiation damage and the ability of uranyl ion (UO22+) to interact with various proteins and disturb their biological functions. Cytochrome b<inf>5</inf> (cyt b<inf>5</inf>) is a highly negatively charged heme protein and plays a key role in mediating cytochrome c (cyt c) signaling in apoptosis by forming a dynamic cyt b <inf>5</inf>-cyt c complex. In previous molecular modeling study in combination with UV-Vis studies, we found that UO22+ is capable of binding to cyt b <inf>5</inf> at surface residues, Glu37 and Glu43. In this study, we further investigated the structural consequences of cyt b<inf>5</inf> and cyt c, as well as cyt b<inf>5</inf>-cyt c complex, upon uranyl binding, by fluorescence spectroscopic and circular dichroism techniques. Moreover, we proposed a uranyl binding site for cyt c at surface residues, Glu66 and Glu69, by performing a molecular modeling study. It was shown that uranyl binds to cyt b<inf>5</inf> (K<inf>D</inf> = 10 μM), cyt c (K<inf>D</inf> = 87 μM), and cyt b <inf>5</inf>-cyt c complex (K<inf>D</inf> = 30 μM) with a different affinity, which slightly alters the protein conformation and disturbs the interaction of cyt b<inf>5</inf>-cyt c complex. Additionally, we investigated the functional consequences of uranyl binding to the protein surface, which decreases the inherent peroxidase activity of cyt c. The information of uranyl-cyt b <inf>5</inf>/cyt c interactions gained in this study likely provides a clue for the mechanism of uranyl toxicity. ©2013 Elsevier B.V. All rights reserved.

摘要:
Melatonin is biosynthesized in the pineal gland and secreted into the bloodstream. Evidences indicate a role of melatonin in the regulation of glucose metabolism. The objective of this study was to investigate the effect of melatonin on insulin sensitivity in insulin resistant adipocytes. Following a preincubation with melatonin or vehicle for 30 min, insulin resistant cells of 3T3-L1 adipocytes were induced by palmitic acids (300 mu M, 6 h). Our results showed that palmitic acids inhibited both the basal and insulin-stimulated uptake of [H-3]-2-Deoxyglucose, down-regulated the levels of IRS-1 and GLUT-4. However, compared to the vehicle group, melatonin pre-treatment increased significantly the uptake of [H-3]-2-Deoxyglucose as well as the level of GLUT-4, and decreased phosphorylated IRS-1 (Ser307) although total IRS-1 did not change significantly. These data suggest that palmitic acids impair insulin signal via down-regulating the expressions of IRS-1 and GLUT-4; whereas melatonin can ameliorate insulin sensitivity by inhibiting Ser307 phosphorylation in IRS-1 and increasing GLUT-4 expressions in insulin resistant 3T3-L1 adipocytes. We conclude that melatonin regulates the insulin sensitivity and glucose homeostasis via inhibiting Ser-phosphorylation and improving function of IRS-1. (C) 2014 Elsevier Masson SAS. 摘要:
Forkhead box protein A1 (FoxA1) is a transcription factor that is involved in embryonic development and cell differentiation. In this study, we show that hydrogen peroxide (H2O2) treatment upregulated expression of FoxA1 and UCP2 in the A549 cell line. Overexpression of FoxA1 by full-length complementary DNA reduced UCP2 expression, while silencing of FoxA1 expression by small interfering RNA significantly increased UCP2 levels. FoxA1 binds to a site from -919 to -913 bp relative to the UCP2 transcription start site. The overexpression of FoxA1 promoted the DNA binding activity and attenuated the transcription of UCP2 promoter as shown by electromobility shift, chromatin immunoprecipitation assays, and luciferase reporter assay. These data indicate an important role of FoxA1 in regulating expression of UCP2. © 2013 Cell Stress Society International.

A novel derivative β-cyclodextrin (β-CD) bonded silica gel (β-CD/SiO2) was prepared via carbodiimide activation and used as a chiral stationary phase (CSP). The β-CD/SiO2 CSP was characterized by X-ray photoelectron spectroscopy and FTIR spectroscopy to prove the successful immobilization of β-CD onto the surface of the silica gel. The thermal behavior of the CSP was studied by differential scanning calorimetry and then used for the chiral separation of racemic naproxen using HPLC. Several factors affecting the separation efficiency of naproxen enantiomers were investigated systematically. The following parameters were selected: methanol–0.01 M phosphate buffer solution (85 + 15, v/v, pH 3.5) was the mobile phase, the flow rate was 1.0 mL/min, and the column temperature was 35°C. Under optimal conditions, the racemic naproxen was efficiently separated with a resolution factor (R S) of 1.70 and a separation factor (α) of 1.25. This method was successfully used for the enantiomeric separation and purity assessment of naproxen in commercial naproxen sustained release tablets.

摘要:
A direct fluorescence spectra method was applied for the determination of metallothioneins at nanomolar levels. In Britton-Robison (B-R) buffer (pH 7.0), the interaction of bis(1,10-phenanthroline)copper(II) complex cation [Cu(phen)2]2+ and metallothioneins enhanced the fluorescence intensity of system. The fluorescence enhancement at 365 nm was proportional to the concentration of metallothioneins. The mechanism was studied and discussed in terms of the fluorescence and UV-absorption spectra. Under the optimal experimental conditions, at 365 nm, there was a linear relationship between the fluorescence intensity and the concentration of the metallothioneins in the range of 8.30 × 10-9 - 7.70 × 10-7 mol L-1. The linear regression equation was ΔF = 8.96 + 38.01c (mol L-1), with a correlation coefficient of r = 0.998 and detection limit 2.50 × 10-9 mol L-1. The relative standard deviation was 0.47% (n = 11), and the average recovery 97.2%. The proposed method was successfully reliable, selective and sensitive in determining trace metallothioneins in fish visceral organ samples with the results in good agreement with those obtained by HPLC. © The Japan Society for Analytical Chemistry.

摘要:
Chronic sleep deprivation may speed the onset or increase the severity of age-related conditions such as Type 2 diabetes, high blood pressure and obesity. Piromelatine (Neu-P11) is a novel melatonin agonist, which has been developed for the treatment of insomnia. Animal studies have suggested possible efficacy of piromelatine in sleep maintenance, anxiety and depression. In addition, piromelatine has been shown to inhibit weight gain and improve insulin sensitivity in high-fat/high-sucrose-fed (HFSD) rats. The objective of this study was to investigate the effects of piromelatine on insulin sensitivity in sleep restricted rats. Sleep restriction was established by rotating cages intermittently for 20 h thereby sleeping time of rats was limited to 4 h per day. During 8 days of sleep restriction, rats were injected intraperitoneally with piromelatine (20 mg/kg), melatonin (5 mg/kg) or a vehicle. The results showed that sleep restriction increased plasma glucose, fasting insulin, total cholesterol (TC), triglycerides (TG) and oxidative stress markers while HDL-cholesterol (HDL-C) level and glucose tolerance were decreased. However, under piromelatine or melatonin treatment, the levels of plasma glucose, TG, TC decreased and HDL-C, glucose tolerance and antioxidative potency increased when compared with the vehicle-treated group. These data suggest that chronic sleep restriction in rats induce metabolic dysfunction, oxidative stress and insulin resistance, and these symptoms were improved by treatment with piromelatine or melatonin. We conclude that piromelatine could regulate metabolic profiles and insulin sensitivity, and attenuate insulin resistance induced by sleep restriction. © 2014 Elsevier B.V.

The chelating reaction of bis-salophen with uranyl to form binuclear complex uranyl-bis-salophen (UBS) was studied by fluorescence spectroscopy. The coordination reaction of UBS with fructose 1,6-bisphosphate (F-1,6-BP) to form supramolecular polymer was then studied by resonance light scattering (RLS) spectroscopy. The reaction of bis-salophen with uranyl results in a remarkable enhancement of fluorescence intensity. The maximum emission wavelength of the fluorescence is at 471 nm. The reaction of UBS with F-1,6-BP results in a remarkable enhancement of RLS intensity. The maximum scattering wavelength of the RLS is at 460 nm. The two reactions were used to establish fluorescence method for the determination of uranium (VI) and RLS method for the determination of F-1,6-BP, respectively. Under optimum conditions, the linear ranges for the detection of uranium (VI) and F-1,6-BP are 0.003-0.35 nmol/mL and 0.05-5.0 nmol/mL, respectively. The detection limits are 0.0017 nmol/mL and 0.020 nmol/mL, respectively. The proposed fluorescence method has been successfully applied for the determination of uranium (VI) in environmental water samples with the recoveries of 97.0-104.0%. The proposed RLS method has also been successfully applied for the determination of F-1,6-BP in medicine injection samples with the recoveries of 98.5-102.3%. ©2013 Elsevier B.V. All rights reserved.

摘要:
A double-receptor sandwich method for the fluorescence determination of adenosine triphosphate (ATP) is proposed in this paper. The solid phase receptor on the surface of glass slides is a molecularly imprinted membrane (MIM) containing an artificial nanocavity. It is constructed by a molecular imprinting technique using adenosine monophosphate (AMP) as a template molecule. The labeled receptor is a uranyl-salophen complex containing a fluorescent group or uranyl-salophen-fluorescein (USF). It is synthesized with salophen, 5-aminofluorescein, and uranyl. In a procedure of determining ATP, ATP in sample solution is first adsorbed on the surface of the glass slide through the combination of the AMP group in ATP with the nanocavity in MIM. Then, the adsorbed ATP binds USF through the coordination reaction of the phosphate group in ATP with uranyl in USF to form a sandwich-type structure of MIM-ATP-USF. The amount of ATP is detected through the fluorescence determination of USF bound on the slide. Under optimal conditions, the linear range for the determination of ATP is 0.3 to 4.8 nmol/mL with a detection limit of 0.041 nmol/mL. The proposed method has been successfully employed for the determination of ATP in real samples with the recoveries of 98.5 to 102.5 %. © 2013 Springer-Verlag Berlin Heidelberg.

摘要:
Human soluble guanylate cyclase (hsGC), a NO sensor/NO receptor of a heterodimeric hemoprotein, plays a critical role in the NO-sGC-cGMP signaling pathway, and also reveals a novel nitrite reductase activity. This indicates that hsGC could activate itself by catalytic reduction of nitrite to NO instead of receiving NO from nitric oxide synthase (NOS), which provides valuable insight into the physiological function of the homodimeric hsGC.

Determination of fructose 1,6-bisphosphate using a double-receptor sandwich type fluorescence sensing method based on uranyl–salophen complexes

In this paper, we report a double-receptor sandwich type fluorescence sensing method for the determination of fructose bisphosphates (FBPs) using fructose 1,6-bisphosphate (F-1,6-BP) as a model analyte based on uranyl-salophen complexes. The solid phase receptor is an immobilized uranyl-salophen (IUS) complex which is bound on the surface of glass slides by covalent bonds. The labeled receptor is another uranyl-salophen complex containing a fluorescence group, or uranyl-salophen-fluorescein (USF). In the procedure of determining F-1,6-BP in sample solution, F-1,6-BP is first adsorbed on the surface of the glass slide through the coordination reaction of F-1,6-BP with IUS. It then binds USF through another coordination reaction to form a sandwich-type structure of IUS-F-1,6-BP-USF. The amount of F-1,6-BP is detected by the determination of the fluorescence intensity of IUS-F-1,6-BP-USF bound on the glass slide. Under optimal conditions, the linear range for the detection of F-1,6-BP is 0.05-5.0nmolmL-1 with a detection limit of 0.027nmolmL-1. The proposed method has been successfully applied for the determination of F-1,6-BP in real samples with satisfactory results. © 2013 Elsevier B.V.

Aims: This study aims to examine the possible associations between high density lipoprotein (HDL) subclass distribution and APOA5-1131T>C polymorphism in hypertriglyceridemia. Methods: The distribution of HDL subclasses was quantified by 2-dimensional electrophoresis in conjunction with immunodetection method. The APOA5-1131T>C polymorphism was identified in 95 hypertriglyceridemic (HTG) patients and 102 healthy subjects by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP). Results: The APOA5-1131C (C) allele frequency was higher in the HTG group than in the control group. Plasma triglycerides (TG) were significantly higher and apoA5 was significantly lower in patients with the C allele when compared to patients with the APOA5-1131T (T) allele, even more dramatically so in the APOA5-1131CC homozygote. In both the HTG group and the control group, the frequency of the C allele was positively correlated with levels of TG, total cholesterol (TC), low density lipoprotein cholesterol (LDL-C) and apolipoprotein B100 (apoB100), and negatively correlated with levels of high density lipoprotein cholesterol (HDL-C), apolipoprotein A1 (apoA1) and apolipoprotein A5 (apoA5) (P < 0.001). In all subjects, the frequency of the C allele was positively correlated with the level of small-sized HDL (pre beta(1)-HDL and HDL3a), and negatively correlated with levels of HDL2a and HDL2b. Conclusion: Changes in HDL subclass distributions in HTG may be related to the APOA5-1131T>C polymorphism. This polymorphism leads to a general shift towards smaller-sized HDL. A high performance liquid chromatography method with visible detection (HPLC-VIS) for the determination of malachite green (MG), crystal violet (CV), leucomalachite green (LMG), and leucocrystal violet (LCV) in fish has been developed after clean-up through an immunoaffinity column (IAC). Residues were simultaneously extracted from fish muscle with acetonitrile and ammonium acetate buffer. The leuco-forms, LMG and LCV, were oxidized quantitatively to the chromic CV and MG by reaction with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone. Extracts were then purified on an IAC which prepared by immobilizing the anti-MG-CV antibodies by the sol-gel method. Finally, the eluents were analyzed by HPLC-VIS. The limits of detection were 0.15, 0.1, 0.18 and 0.14 ng/g for MG, CV, LMG and LCV, respectively. The average recoveries in samples fortified with MG, CV, LMG and LCV over the range 0.5-10 ng/g were from 71.6% to 96.8% with RSDs of 5.1-12.3% (n = 6). This novel method was confirmed by liquid chromatography-tandem mass spectrometry with electrospray interface in positive mode using multiple reaction monitoring. (c) 2012 Elsevier B.V. 摘要:
Piromelatine, a novel investigational multimodal sleep medicine, is developed for the treatment of patients with primary and co-morbid insomnia. Piromelatine has been shown to inhibit weight gain and improve insulin sensitivity in high-fat/high-sucrose-fed (HFHS) rats. Considering that piromelatine has also been implicated in lowering of triglyceride levels in HFHS rats, this work elucidated whether this effect involves in the regulation of adipose triglyceride lipase (ATGL) and hormone-sensitive lipase (HSL) in triglyceride (TG) metabolism. In this study, we investigated the effects of piromelatine and MT2 receptors inhibition on TG content, insulin-stimulated glucose uptake, and the expressions of ATGL and HSL in 3T3-L1 adipocytes preincubated in high glucose and high insulin (HGI) conditions. Our results showed that culturing 3T3-L1 adipocytes under HGI conditions increased triglyceride accumulation with concomitant decrease of ATGL and HSL expression, inducing insulin resistance in 3T3-L1 adipocytes. We also found that triglyceride accumulation was significantly inhibited and the levels of ATGL/HSL increased after melatonin or piromelatine treatment. The effects of melatonin/piromelatine (10 nM) were counteracted by pretreatment with the relatively selective MT2 receptor antagonist luzindole (100 nM). In this study, our data demonstrate that piromelatine reverses high glucose and high insulin-induced triglyceride accumulation in 3T3-L1 adipocytes, possibly through up-regulating of ATGL and HSL expression via a melatonin-dependent manner. © 2013 Elsevier Masson SAS. Thorium(IV) biosorption is investigated by citric acid treated mangrove endophytic fungus Fussarium sp. #ZZF51 (CA-ZZF51) from South China Sea. The biosorption process was optimized at pH 4.5, equilibrium time 90min, initial thorium(IV) concentration 50mgL<sup>-1</sup> and adsorbent dose 0.6gL<sup>-1</sup> with 90.87% of removal efficiency and 75.47mgg<sup>-1</sup> of adsorption capacity, which is obviously greater than that (11.35mgg<sup>-1</sup>) of the untreated fungus Fussarium sp. #ZZF51 for thorium(IV) biosorption under the condition of optimization. The experimental data are analyzed by using isotherm and kinetic models. Kinetic data follow the pseudo-second-order model and equilibrium data agree very well with the Langmuir model. In addition, FTIR analysis indicates that hydroxyl, amino, and carbonyl groups act as the important roles in the adsorption process. ©2013 Elsevier Ltd.

摘要:
In this study, to enhance the capability of metal ions disturbing the orientation of liquid crystals (LCs), we designed a new label-free LC biosensor for the highly selective and sensitive detection of heavy metal ions. This strategy makes use of the target-induced DNA conformational change to enhance the disruption of target molecules for the orientation of LC leading to an amplified optical signal. The Hg2+ ion, which possesses a unique property to bind specifically to two DNA thymine (T) bases, is used as a model heavy metal ion. In the presence of Hg2+, the specific oligonucleotide probes form a conformational reorganization of the oligonucleotide probes from hairpin structure to duplex-like complexes. The duplex-like complexes are then bound on the triethoxysilylbutyraldehyde/N,N- dimethyl-N-octadecyl (3-aminopropyl) trimethoxysilyl chloride (TEA/DMOAP)-coated substrate modified with capture probes, which can greatly distort the orientational profile of LC, making the optical image of LC cell birefringent as a result. The optical signal of LC sensor has a visible change at the Hg 2+ concentration of low to 0.1 nM, showing good detection sensitivity. The cost-effective LC sensing method can translate the concentration signal of heavy metal ions in solution into the presence of DNA duplexes and is expected to be a sensitive detection platform for heavy metal ions and other small molecule monitors. © 2012 American Chemical Society.

Despite the established efficacy of statin therapy, the risk of cardiovascular events remains high in many patients. We examined high-density lipoprotein (HDL) subclass distribution profiles among statin-treated coronary heart disease (CHD) patients undergoing percutaneous coronary intervention (PCI). Plasma HDL subclasses were measured in 85 patients with established CHD and quantified by two-dimensional gel electrophoresis and immunoblotting. In CHD patients with statin treatment, the mean value of total cholesterol (TC) reached the desirable level and the triacylglycerol level (TAG) was borderline high. Moreover, low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), apolipoproteinA-I, and apolipoproteinB-100 levels in these patients resembled those in normolipidemic healthy subjects. The HDL subclass did not show a normal distribution and was characterized by the lower large-sized HDL(2b) contents and higher contents of small-sized prebeta(1)-HDL in CHD patients, compared to those in normolipidemic control subjects. Multiple stepwise regression analysis revealed that the severity of coronary stenosis, determined by the Gensini Score, was significantly and independently predicted by HDL(2b) and HDL(3b). Statin therapy was effective in modifying plasma lipids levels, but not adequate as a monotherapy to normalize the HDL subclass distribution phenotype of patients with CHD undergoing PCI. The HDL subclass distribution may aid in risk stratification, especially in patients with CHD and therapeutic LDL-C and HDL-C levels.

Heme proteins are among the most abundant and important metalloproteins, exerting diverse biological functions including oxygen transport, small molecule sensing, selective C-H bond activation, nitrite reduction, and electron transfer. Rational heme protein designs focus on the modification of the heme-binding active site and the heme group, protein hybridization and domain swapping, and de novo design. These strategies not only provide us with unique advantages for illustrating the structure-property-reactivity-function (SPRF) relationship of heme proteins in nature but also endow us with the ability to create novel biocatalysts and biosensors. All roads lead to Rome: During the last few decades, various approaches for rational heme protein design have emerged. These techniques not only enhance our understanding of the structure and function of heme proteins in nature but also facilitate the creation of novel biocatalysts and biosensors beyond nature.
