Guinea pigs were injected by Evans blue through sublingual vein. After 30 minutes, 1,1/2 and 1/4 original culture medium of stably transfected cells were injected by hypodermic injections [33]. The injected dot diameter was about 5 mm. After 15 minutes, the permeability was detected by the subcutaneous blue dot emergence or not and their diameter were quantified. This protocol was supported by the Committee on Experimental Animal Care of the University of Nanhua (Province of Hunan, PR China).

The purpose of this investigation was to establish monoclonal cell lines of HUVEC with the stable expression of the VEGF121 gene. Such cells are likely to better adhere to the luminal surface of stents or grafts and to promote a complete endothelialization. The eukaryotic expression vector PCD2-VEGF121 was transfected into cell lines of HUVEC mediated by lipofect AMINE. The positive clones were obtained by the screening of G418.
